Sporothrix schenckii was isolated from a specimen from a 75-year-old woman with lymphatic sporotrichosis on the left forearm. A mass of fungi was cultured in liquid Sabouraud dextrose broth for three months. Crude lipids were extracted from the cultured fungal mass. Total fatty acids and free fatty acids were isolated, purified, and then analyzed using thin-layer chromatography, gas liquid chromatography, and mass spectrometry. Total fatty acid composition consisted of seven molecular species of C15:0 (2.5%), C16:1 (2.5%), C16:0 (64.5%), 2-Me-C16:0 (5.1%), C17:0 (2.0%), C18:1 (18.0%) and C18:0

The whole cells and purified cell walls of Sporothrix schenckii were extracted with various solvents. Using hydrolysis, the fractions obtained afforded mannose, glucose, rhamnose as main sugars, and galactose. The repartition of polysaccharides was different according to the solvent used. These polysaccharides could be used as antigens for diagnosis and to follow the evolution of the sporotrichosis.

The efficacy of voriconazole (VRC) was evaluated against two strains of each of the two most common species causing sporotrichosis, Sporothrix schenckii sensu stricto and Sporothrix brasiliensis, using a murine model of disseminated infection. Voriconazole was administered at doses of 20 or 40 mg kg(-1) per day by gavage. The drug showed some efficacy, especially at 40 mg kg(-1) per day, in prolonging the survival and reducing fungal load in spleen and liver in mice infected with S. schenckii, whereas in animals infected with S. brasiliensis the drug did not work.

Sporothrix schenckii is a dimorphic fungus existing as mould in the environment and as yeast in the host. The morphological shift between mycelial/yeast phases is crucial for its virulence, but the transcriptional networks implicated in dimorphic transition are still not fully understood. Here, we report the global transcriptomic differences occurring between mould and yeast phases of S. schenckii, including changes in gene expression profiles associated with these distinct cellular phenotypes. Moreover, we also propose a new genome annotation, which reveals a more complex transcriptional arch

Sporotrichosis is a serious fungal disease of humans and animals worldwide. The ongoing epidemic of cat-transmitted sporotrichosis caused by Sporothrix brasiliensis in South America raises major public health concerns, yet data on environmental persistence and disinfectant efficacy are scarce. Using the quantitative disk carrier method (ASTM 2197), we evaluated persistence and disinfectant susceptibility of Sporothrix schenckii and Sporothrix brasiliensis yeasts on hard, non-porous stainless-steel surfaces. Both species remained viable for at least 6 weeks, supporting the potential for fomite-

Inhibition of Δ(24)-sterol methyltransferase (24-SMT) in Sporothrix schenckii sensu stricto and Sporothrix brasiliensis was investigated in vitro. The effects on fungal growth and sterol composition of the 24-SMT inhibitor 22-hydrazone-imidazolin-2-yl-chol-5-ene-3β-ol (H3) were compared to those of itraconazole. MIC and MFC analysis showed that H3 was more effective than itraconazole against both species in both their filamentous and yeast forms. H3 showed fungistatic activity in a time-kill assay, with inhibitory activity stronger than that of itraconazole. GC analysis of cell sterol composit

Sporothrix, one of the anamorph genera of Ophiostoma, includes the important human pathogen S. schenckii and various fungi associated with insects and sap stain of wood. A survey of fungi from wood utility poles in South Africa yielded two distinct groups of Sporothrix isolates from different geographical areas. DNA sequence and morphological data derived in this study showed that isolates in these groups represent two novel species in the S. schenckii-O. stenoceras species complex. A new species isolated from pine poles and rosebush wood and phylogenetically closely related to S. pallida is d

Oxidative stress is key in immune defense against fungal infections, such as those caused by Sporothrix schenckii, the dimorphic fungus responsible for sporotrichosis. Phagocytic cells utilize oxidative stress as a crucial mechanism to control pathogen spread. During S. schenckii infection, phagocytic cells recognize pathogen-associated molecular patterns (PAMPs) on their surface through conserved transmembrane or soluble receptors, known as pattern recognition receptors (PRRs). This recognition triggers a cascade of immune responses, including the generation reactive oxygen species (ROS) esse

Sporotrichosis is a neglected fungal disease, affecting mammals. Here, a feline patient presented a small ulcerated lesion. Short treatment by itraconazole resulted in clinical cure. The isolate was identified as Sporothrix brunneoviolacea by sequencing. Despite proper growth, antifungal susceptibility testing by microbroth dilution could not be performed due to a lack of growth. This is the first report of sporotrichosis by S. brunneoviolacea, which was originally classified as an environmental and non-pathogenic species.

Sporotrichosis is a subacute, or chronic mycosis caused by traumatic inoculation of material contaminated with the fungus Sporothrix schenckii which is part of the Sporothrix spp. complex. The infection is limited to the skin, although its progression to more severe systemic or disseminated forms remains possible. Skin is the tissue that comes into contact with Sporothrix first, and the role of various cell lines has been described with regard to infection control. However, there is little information on the response of keratinocytes. In this study, we used the human keratinocyte cell line (Ha

Recently, sporotrichosis caused by Sporothrix brasiliensis transmitted during tattooing process was described in the epicenter of the Brazilian epidemic sporotrichosis. We report a similar case of this mycosis in a patient recently tattooed, but probably infected by a sick cat instead of via a contaminated procedure. Clinical cure was attained after two months of oral itraconazole. In the hyperendemic S. brasiliensis regions of sporotrichosis, health professionals must be aware of atypical transmissions of this fungus.

The development of mature biofilms is an aid in numerous aspects of the life cycle of fungi. It is well known that Sporothrix schenckii complex causes a benign subcutaneous mycosis, but recent studies have suggestedthat biofilm formation may be one of the important factors involved in its virulence. Here we report the study of the biomass organization and a model of the stages of S. schenckii biofilm development: adsorption, active adhesion, microcolony formation, maturation, and dispersal of biofilm fragments. During the development, the biofilm is surrounded by extracellular matrix, which co

Sporothrix brasiliensis usually causes infection limited to the skin, subcutaneous tissue and regional lymph nodes. Contamination occurs through inhalation or accidental inoculation from animal scratches and bites. Meningitis is rare and mostly occurs in immunosuppressed patients. Here, we describe an immunocompetent person who developed chronic meningitis and discuss the diagnosis, differential diagnosis and treatment of this rare condition.

Sporothrix schenckii sensu lato is a complex of thermally dimorphic species whose natural habitats are soil and plant materials. However, the traumatic implantation of the species into human skin is traditionally thought to be the route leading to the fungal disease sporotrichosis. The complex contains Sporotrhix mexicana, S. globosa, S. brasiliensis, S. luriei, in addition to S. schenckii sensu stricto. In this study we evaluated the differences among these species relative to their frequency in the environment and in human hosts, as well as discuss their remarkable diverse pathogenicity. Tod

An 87-year-old patient presented with a 6-week history of an isolated progressive destructive nodular eyelid mass, secondary nodular and ulcerative lesions, and regional painful lymphadenopathy. After 4 weeks, fungal cultures demonstrated Sporothrix schenckii. S. schenckii is a rare dimorphic fungus that can occasionally involve the periocular skin. The authors' case demonstrates typical clinical features, emphasizes the delay in diagnosis, and shows effective treatment with oral itraconazole.

Restriction fragment length polymorphism (RFLP) in mitochondrial DNA (mtDNA) from 25 environmental isolates of Sporothrix schenckii from northeastern China was investigated. Based on the mtDNA-RELP patterns with Hae III, 6 isolates were confirmed to be S. schenckii, while the other 19 isolates were confirmed to be species distinct from S. schenckii. The mtDNA RFLP patterns of the 19 non-S. schenckii were identical to each other. The non-S. schenckii isolates could not be discriminated from S. schenckii by their macro- or micro-morphological features, and were not pathogenic in guinea pigs. Ser
