Combined extraction and spray-freeze-drying of Cannabis sativa Flos provides stable lyophilizate particles of cannabinoids.

The objective of this study was to explore non-aqueous spray-freeze-drying (SFD) as a formulation strategy to stabilize and improve the dissolution of Cannabis sativa full extracts. The medicinal use of Cannabis sativa has strongly intensified over the last decade. Here, Cannabis flos full extracts prepared by tert-butanolic solutions with 25% solid content were directly processed via spray-freeze-drying (SFD) with different excipients to increase the storage stability of the 4 main cannabinoids: tetrahydrocannabinol (THC), cannabidiol (CBD) and their corresponding acid precursors (THCA and CB

Kombucha Fermentation With Dried Starter Cultures: A Strategy for Microbial Stabilization via Spray and Freeze Drying.

Kombucha production relies on complex microbial consortia, which often vary across batches, posing challenges for fermentation reproducibility, product standardization, and industrial scalability. This study explored the effects of spray drying (SD) and freeze drying (FD) on the viability and metabolic activity of kombucha starter cultures, using maltodextrin (MD) and gum Arabic (GA) as drying carriers. The dried starter cultures exhibited favorable physicochemical properties, including low moisture content (2.22 - 3.62%), reduced water activity (0.135 - 0.230), and high solubility (> 90%). Fe

Improved drug loading via spray drying of a chalcone implant for local treatment of cutaneous leishmaniasis.

Current chemotherapy of cutaneous leishmaniasis (CL), even the mildest forms, encompasses multiple and painful injections with toxic drugs that cause systemic adverse effects. Recently, we showed the promising use of poly(lactic-co-glycolic acid) (PLGA) microparticles loaded with an antileishmanial nitrosylated chalcone (CH8) for effective, safe, local, and single-dose treatment of CL. Here, we proposed to optimize the delivery system by increasing the CH8 loading in PLGA-microparticles using spray drying instead of emulsification-solvent evaporation. The effect of solvent composition and poly

Aqueous-System-Enabled Spray-Drying Technique for the Synthesis of Hollow Polycrystalline ZIF-8 MOF Particles.

Zeolitic imidazolate framework-8 shares the same topology with sodalite zeolite but consists of Zn nodes bridged by imidazolate linkers to form a neutral open-framework structure. ZIF-8 has been recognized as a unique molecular sieving material with a flexible framework enabling interesting "gate-opening" functionality. Controlling the crystal size and shape is crucial for regulating the structural flexibilities and mass transport properties. The present study demonstrates that an aqueous-system-enabled spray-drying process enables the shape engineering of ZIF-8 with a hollow polycrystalline s

Atomizing into a chilled extraction solvent eliminates liquid gas from a spray-freeze drying microencapsulation process.

A spray-freeze drying encapsulation process using direct atomization into a chilled extraction solvent (ACES), in the absence of liquefied gas, was developed. Heat transfer models, developed to estimate droplet freezing time (t(f)), identified ACES conditions where solvent extraction, nonsolvent influx, and droplet deformation were minimized. Calculated t(f)'s for dichloromethane and dichloroethane droplets were 98 and 46 ms, respectively, using atomization into liquid nitrogen (ALN2). For droplets <100 microm, this was shorter than the calculated headspace residence time, indicating freezing

A new oral vaccine candidate based on the microencapsulation by spray-drying of inactivated Vibrio cholerae.

The aim of this work was to evaluate the microencapsulation by spray-drying of inactivated Vibrio cholerae, using methacrylic copolymers Eudragit® L30D-55 and FS30D. The microparticles obtained presented a particle size around 3.0 μm. The preparation temperature affected the morphology and the antigenicity of microparticles, but it did not affect the V. cholerae content. In vitro release studies showed that in acid medium less than 5% of bacteria was released, and in neutral medium, Eudragit® L30D-55 microparticles released 86% after 24 h, whereas FS30D released less than 30%. Rats inoculated

The spray drying of acetazolamide as method to modify crystal properties and to improve compression behaviour.

Acetazolamide shows a very poor compression ability and tablets must usually be produced through a wet granulation process. However, the possibility to obtain pure acetazolamide for direct compression could be interesting for industrial application. With the scope to obtain a material for direct compression, three different crystallisation methods were chosen, with respect to acetazolamide solvent solubility. (a) Acetazolamide was dissolved in an ammonia solution and then spray dried. It was possible to characterise the spherical particles as a mixture of two polymorphic forms, I and II by Pow

Process Optimization of Spray-Dried Aquafaba and Comparison with Freeze-Drying: Techno-Functional Performance and Structural Attributes.

This study aimed to optimize the spray-drying (SD) process for aquafaba and compare the physical and techno-functional properties of the resulting powder with those of its freeze-dried (FD) counterpart. A Box-Behnken design was employed to evaluate the effects of inlet air temperature (150-190 °C), air speed (3.5-4.3 m/s), and feed flow rate (0.3-0.5 L/h) on 14 quality responses. The optimized SD conditions were determined to be an inlet air temperature of 189 °C, an air speed of 4.2 m/s, and a feed flow rate of 0.3 L/h. Validation experiments demonstrated that the developed models had high pr

Urea's distinct role in agricultural spray deposition: modulating drying dynamics to enhance central deposition without wetting benefits.

BACKGROUND: Urea is frequently tank-mixed with pesticides, yet whether it functions as a conventional spray adjuvant or acts at another stage of droplet evolution remains unclear. This study examined how 0-2.0 wt% urea affects spray liquid properties, atomization, impact, wetting, evaporation, and final deposition. RESULTS: Urea caused only minor changes in equilibrium surface tension and viscosity. Across nozzle and pressure conditions, changes in volume median diameter were generally within ±3%, while impact, wetting, and the time required for 90% volume loss (55-60 min) were not appreciabl

Optimization of Spray Drying Conditions for Yield, Particle Size and Biological Activity of Thermally Stable Viral Vectors.

PURPOSE: This work examines the relevance of viral activity in the optimization of spray drying process parameters for the development of thermally stable vaccine powders. In some instances, the actual active pharmaceutical ingredient (API) is not included in the process optimization as it is deemed too costly to use until the final selection of operating conditions, however, that approach is inappropriate for highly labile biopharmaceutics. We investigate the effects of spray drying parameters on i) yield, ii) particle size and iii) viral vector activity of a mannitol/dextran encapsulated rec

Scaling up the spray drying process from pilot to production scale using an atomized droplet size criterion.

PURPOSE: The purpose of this study was to investigate the possibility of producing identical powders in pilot and production scale spray drying equipment by matching the droplet size distributions produced by two differently sized atomizers. METHODS: Particles were prepared by spray drying solutions of acetaminophen and polyvinylpyrrolidone K-30. The success of the up-scaling was evaluated by comparing the powders in terms of particle size distribution (laser diffraction), crystallinity (XPRD) and morphology (SEM). Furthermore, the influence of process parameters on other product characterist

Particle design of three-component system for sustained release using a 4-fluid nozzle spray-drying technique.

We prepared composite particles of acetaminophen (Act) with chitosan (Cht) and hydroxypropylmethylcellulose phthalate (HPMCP) as a carrier using a newly developed 4-fluid nozzle spray-dryer. Cht dissolves in acid solutions and forms a gel, but it is insoluble in alkaline solutions. On the other hand, HPMCP is insoluble in acid solutions, but it dissolves in alkaline solutions. Therefore, we tested a preparation of controlled release composite particles using the characteristics of these carriers. Act and Cht mixtures in prescribed ratios were dissolved in an acid solution. Composite particles
