PURPOSE: S. epidermidis is an ocular pathogen and a leading cause of keratitis. It produces hemolysins and at least 3 proteases. The purpose of the present study is to compare the secretion of hemolysins and proteases between 28 ocular isolates and one non-ocular strain and to determine their relationship to ocular virulence in selected strains using a rabbit model of infection.
MATERIALS AND METHODS: Culture supernatants were compared for protease production and hemolysis. Selected strains were injected into rabbit corneas and their virulence and pathology recorded. The major protease activi

Bacterial sepsis is a major killer in hospitalized patients. Coagulase-negative staphylococci (CNS) with the leading species Staphylococcus epidermidis are the most frequent causes of nosocomial sepsis, with most infectious isolates being methicillin-resistant. However, which bacterial factors underlie the pathogenesis of CNS sepsis is unknown. While it has been commonly believed that invariant structures on the surface of CNS trigger sepsis by causing an over-reaction of the immune system, we show here that sepsis caused by methicillin-resistant S. epidermidis is to a large extent mediated by

Using two different strains of Staphylococcus epidermidis in a rat model of experimental endocarditis, we examined the prophylactic efficacy of cefamandole (200 mg/kg/dose), cefazolin (200 mg/kg/dose), nafcillin (200 mg/kg/dose), and vancomycin (20 mg/kg/dose). In vitro susceptibility testing demonstrated that both test strains were resistant to methicillin and cefazolin and susceptible to cefamandole and vancomycin. A 10(6) cfu inoculum was used for both strains, an inoculum which produced endocardial infections in greater than 90% of rats. Initial doses of each antibiotic were given 45 min t

The in vitro activity of coumermycin was tested against oxacillin-susceptible and resistant strains of Staphylococcus aureus and Staphylococcus epidermidis, and compared with that of penicillin G, oxacillin, minocyclin, erythromycin, vancomycin, teicoplanin, rifampicin and LM 427. The MICs were measured using the agar dilution method with an inoculum of 10(5) cfu/spot. Coumermycin was the most active antibiotic with MIC90 of 0.025-0.2 mg/l. The MICs of coumermycin remained unchanged by further incubation for 48 h. The combination of coumermycin with ciprofloxacin, an inhibitor of subunit A of

BACKGROUND: Staphylococcus epidermidis is a prevalent common microorganism found on the skin of most mammals, including humans. However, S. epidermidis is also known as an opportunistic pathogen that can cause diseases if the skin microbiome becomes skewed to favor its overgrowth. This situation can be worsened if the causative strains are antibiotic-resistant. To explore potential alternatives to replace conventional antibiotics, we aim to use probiotics, specifically Lacticaseibacillus rhamnosus and Lactiplantibacillus plantarum, in controlling the growth, colonization, and biofilm formation

Staphylococcus epidermidis is a typically indolent pathogen that is often considered a blood culture contaminant. It is a rare and unexpected cause of osteomyelitis, especially in the absence of recent surgical intervention or orthopedic implants. We highlight a case in which a 90-year-old Caucasian male with no recent spine surgery was found to have osteomyelitis of the lumbar spine and repeat positive blood cultures for methicillin resistant Staphylococcus epidermidis (MRSE). Further investigation revealed a history of mitral valve replacement and a new diagnosis of endocarditis leading to p

BACKGROUND: Cutibacterium acnes has been identified as one of the main triggers of acne. However, increasing knowledge of the human skin microbiome raises questions about the role of other skin commensals, such as Staphylococcus epidermidis, in the physiopathology of this skin disease.
SUMMARY: This review provides an overview of current knowledge of the potential role of S. epidermidis in the physiopathology of acne. Recent research indicates that acne might be the result of an unbalanced equilibrium between C. acnes and S. epidermidis, according to dedicated interactions. Current treatments

Phenol-soluble modulins (PSMs) are multifunctional peptide toxins produced by many staphylococcal strains. PSMs have received much recent attention, owing to multiple reports underscoring their importance for staphylococcal pathogenesis. Members of the PSM family may be strongly cytolytic to neutrophils and other cell types; promote inflammatory, receptor-mediated responses in several human cell types; and contribute to biofilm structuring and detachment. Here we describe biochemical methods to isolate, purify, and quantitatively analyze Staphylococcus epidermidis PSMs.

The inclusion of 5 mM salicylic acid (SAL) in medium inhibited both growth and biofilm production by Staphylococcus epidermidis by up to 55%. The inhibition was not due primarily to chelation of cations. Excess divalent cations restored growth and biofilm production in chelated medium and in medium containing EDTA but not in medium containing SAL. ELISA analyses demonstrated that SAL inhibited production of teichoic acid, slime-associated proteins, and type 1 antigen by as much as 95%. However, it inhibited polysaccharide/adhesin production by only 50%--a figure paralleling the reduction in gr

From two human populations (one pediatric and one adult), clinically diagnosed with Staphylococcus epidermidis (S. epidermidis) sepsis of similar severity, bacteria were isolated from pre-antibiotic blood samples and evaluated for virulence. The LD50 of the bacteria in a mouse model was performed, with evaluation of animals dying acutely following intravenous S. epidermidis administration. More simple assays of virulence were also performed, including bacterial adherence to a fibrin clot and carbohydrate specific lectin binding. The eight pediatric-host S. epidermidis isolates required a signi

Infections in immunocompromised patients and in patients with indwelling prosthetic devices are often caused by hospital strains of Staphylococcus epidermidis resistant to methicillin. Tests for the detection of methicillin resistance, indicating resistance to all beta-lactam antibiotics, were evaluated in order to define a suitable screening test. A broth tube breakpoint test with a large inoculum, 10(7) colony forming units (cfu), gave the highest recovery of resistant strains. False resistance due to hyperproduction of beta-lactamase was excluded. The results correlated completely with the

The ability to identify Staphylococcus epidermidis quickly and accurately has become increasingly important in clinical microbiology. Susceptibility to desferrioxamine, an iron-chelating agent, was investigated as a new test for the identification of S. epidermidis. All strains of S. epidermidis and S. hominis tested were susceptible to a 1000-micrograms disk of desferrioxamine when grown on brain heart infusion agar. All other strains of coagulase-negative staphylococci, S. aureus and micrococci were resistant. As a single test, susceptibility to desferrioxamine was 96.4% efficient in identif

124 newborns admitted to the Department within the period of 3 years with suspected sepsis were analysed. In 55.6% of children, boys prevailing (62.3%), generalized infection was confirmed with positive bacterial cultures. Late-onset disease predominance was found (72.8%). The mortality rate in this group was 5.8% (11.3% boys and 1.9% girls). Analysis of the causative organisms revealed more frequently the Gram positive bacteria (81% of all positive cultures) with a predominant role of Staphylococcus epidermidis (43.5%). In the literature many cases of Staphylococcus epidermidis (SE) complications are reported, but we have not found any reference about reactive arthritis secondary to SE. We report an unusual case of a patient with SE bacteriaemia, who developed elbow arthritis, asymmetrical sacroiliitis, keratoderma and restrictive cardiomyopathy. The clinical pictures, the instrumental and biochemical findings, in particular the positivity of HLA B27, allow us to set this case in the complex and heterogeneous chapter of reactive arthritis.

Methods to detect resistance to methicillin in Staphylococcus epidermidis were studied in order to find a rapid screening test suitable for routine use. One hundred and forty-nine clinical isolates, 16 isolates from skin of healthy people and two reference strains were studied. Hypersecretion of beta-lactamase as a cause of methicillin resistance was eliminated in the strains studied. Tube and microtitre breakpoint, agar breakpoint and disc diffusion methods were compared. The breakpoint for methicillin resistance used was 16 mg/L in broth and 10 mg/L in agar. The discs used contained 1 and 5

The effect of hydration on Staphylococcus epidermidis, the predominant resident bacterial flora, was studied on skin affected by leprosy and known to have impaired sweating. Normal areas served as control. Significantly higher bacterial counts were observed in affected areas compared with normal-looking skin in 16/19 of the patients. Artificial application of Staph. epidermidis on leprosy-affected and unaffected areas, however, showed equivocal results, as in only 50% of the patients were higher counts obtained in affected compared with unaffected sites. The possible responsible factors for th

Staphylococcus epidermidis, a major component of the skin flora, is usually considered a contaminant when recovered from diagnostic cultures. Since 1974 infections caused by gram-negative bacilli and S. aureus occurring among patients with granulocytopenic cancer have remained constant; infections due to S. epidermidis occurring before 1977 at a rate of 2.0 per 1000 days of hospitalization of patients with acute leukemia, increased to 14.6 per 1000 days in 1979. All S. epidermidis infections before 1977 originated from skin sites; since 1977 the respiratory tract and alimentary canal have beco

Prosthetic joint infections (PJI) are severe complications in Orthopedics, with Staphylococcus aureus and Staphylococcus epidermidis being the most commonly isolated pathogens. The variable antimicrobial susceptibility found in these microorganisms, along with the increasing number of methicillin-resistant strains, increases the difficulty of antibiotic selection and makes it necessary to perform individual susceptibility studies to select the optimal antibiotic treatment. The aim of this study was to evaluate the in vitro susceptibility pattern of 35 clinical strains isolated from PJI (17 S.
