Simultaneous ocean acidification and warming do not alter the lipid-associated biochemistry but induce enzyme activities in an asterinid starfish.

Ocean acidification and warming affect marine ecosystems from the molecular scale in organismal physiology to broad alterations of ecosystem functions. However, knowledge of their combined effects on tropical-subtropical intertidal species remains limited. Pushing the environmental range of marine species away from the optimum initiates stress impacting biochemical metabolic characteristics, with consequences on lipid-associated and enzyme biochemistry. This study investigates lipid-associated fatty acids (FAs) and enzyme activities involved in biomineralization of the tropical-subtropical sta

Developing in a warming intertidal, negative carry over effects of heatwave conditions in development to the pentameral starfish in Parvulastra exigua.

Ocean warming and increasing incidence of marine heat waves (MHW) are having far-reaching impacts on coastal ecosystems. The small intertidal asterinid starfish, Parvulastra exigua, in south-eastern Australia, occurs in a global warming hotspot. Development occurs in the intertidal as this species lays eggs and has benthic larvae. The impact of temperature on development to the juvenile was determined over a broad temperature range (12-28 °C) encompassing temperatures experienced during the breeding season (16-20 °C) and cool (- 4 °C) and warm (+10 °C) extremes with the higher temperatures (24

Inhibition of soluble epoxide hydrolase by steroidal metabolites from the starfish Patiria pectinifera: Integrated enzyme kinetic and computational studies.

Soluble epoxide hydrolase (sEH) is a promising therapeutic target for inflammation-related disorders; however, current synthetic inhibitors are limited due to pharmacokinetic constraints. Marine steroidal metabolites offer a potential source of natural sEH inhibitors. Therefore, this study aims to investigate the sEH inhibitory activity of compounds isolated from the starfish Patiria pectinifera. Six steroidal compounds (1-6) were isolated and evaluated using enzymatic assays, kinetic analysis, molecular docking, and molecular dynamics simulations. Compounds 1 and 2 demonstrated potent sEH inh

Identification of the Active EPA/AA-Binding Ether-Type Phosphatidylcholine Derived from the Starfish Patiria pectinifera for C2C12 Myotube Growth.

Concerns about nutritional approaches for promoting skeletal muscle mass and function have increased. This study assessed the effects of starfish-derived glycerophospholipids (PLs) (SPL), characterized by unique ether-linked subclasses, alkylacyl (Alk)- and alkenylacyl (Pls)-PL, on skeletal muscle function, focusing on myotube formation in C2C12 myoblasts. SPL was prepared via chloroform/methanol extraction from Patiria pectinifera, followed by silica gel chromatography fractionation. Myoblasts were induced to differentiate with or without SPL treatment. On day 7 of differentiation, 50 μg/mL o

Being a Better Starfish Thrower, Carpal Tunnel Syndrome, and Cardiac Amyloidosis: The Hand Surgeon's Opportunity to Make a Profound Difference.

In recent years, there is increasing literature in cardiac and hand surgery journals demonstrating a stronger association between seemingly idiopathic carpal tunnel and amyloidosis. Despite this, it can be difficult for hand surgeons to identify who need biopsies, and this is further complicated by the cost of a biopsy and the low likelihood that a patient has cardiac amyloidosis. In patients with cardiac amyloidosis and carpal tunnel syndrome (CTS), CTS is typically diagnosed 5-10 years prior. Early diagnosis of cardiac amyloidosis is crucial, as current medications work to slow disease progr

Protocol for a multicentre randomised controlled trial of STeroid Administration Routes For Idiopathic Sudden sensorineural Hearing loss: The STARFISH trial.

Idiopathic sudden sensorineural hearing loss (ISSNHL) is the rapid onset of reduced hearing due to loss of function of the inner ear or hearing nerve of unknown aetiology. Evidence supports improved hearing recovery with early steroid treatment, via oral, intravenous, intratympanic or a combination of routes. The STARFISH trial aims to identify the most clinically and cost-effective route of administration of steroids as first-line treatment for ISSNHL. STARFISH is a pragmatic, multicentre, assessor-blinded, three-arm intervention, superiority randomised controlled trial (1:1:1) with an intern

Starfish-Inspired Synergistic Reinforced Hydrogel Wound Dressing: Dual Responsiveness and Enhanced Bioactive Compound Delivery for Advanced Skin Regeneration and Management.

Effective wound management demands advanced dressings that protect while actively supporting healing. Traditional wound dressings often fall short of meeting the complex needs of skin repair. Inspired by the regenerative abilities of starfish, we developed a bionically engineered hydrogel designed to enhance wound healing. The hydrogel is synthesized through the coassembly of dopamine-modified cellulose nanofibers, chitosan, (3-aminobenzeneboronic acid)-grafted oxidized dextran, and poly(vinyl alcohol), utilizing dynamic Schiff base and boronic ester linkages. This innovative design imparts mu

An RPA-assisted homogeneous electrochemical DNA sensor for on-site eDNA detection toward early warning of crown-of-thorns starfish outbreaks.

Crown-of-thorns starfish (COTS) outbreaks seriously threaten coral reef ecosystems, while conventional monitoring approaches are time-consuming and often lack sufficient sensitivity for early warning. Existing electrochemical DNA sensors usually require complex electrode-surface immobilization procedures, which can lead to uneven probe distribution, significant steric hindrance, and poor stability. Meanwhile, the low concentration of environmental DNA (eDNA) in marine environments further complicates detection. To overcome these challenges, this study developed a homogeneous electrochemical DN

A tungsten-based solar absorber with a strip-combined starfish structure with high photothermal conversion efficiency and low radiative heat loss.

We propose a tungsten-based broadband resonator capable of achieving near-perfect solar absorption with suppressed radiative heat loss. Across the entire solar radiation spectrum, the absorber exhibits a spectrally integrated total solar absorptance of 95%. Taking into account the thermal emission under an operating temperature of 100 °C, the photothermal conversion efficiency remains as high as 92-93%. Using particle swarm optimization, the initial square-based structure is transformed into adjacent and closely packed strip structures with multiple lengths to achieve enhanced solar absorption

Disulfated Ophiuroid Type Steroids from the Far Eastern Starfish Pteraster marsippus and Their Cytotoxic Activity on the Models of 2D and 3D Cultures.

New steroidal 3β,21-disulfates (2-4), steroidal 3β,22-disulfate (5), and the previously known related steroidal 3β,21-disulfate (1) were isolated from the ethanolic extract of the Far Eastern starfish Pteraster marsippus, collected off Urup Island in the Sea of Okhotsk. The structures of these compounds were determined by intensive NMR and HRESIMS techniques as well as by chemical transformations. Steroids 2 and 3 have an oxo-group in the tetracyclic nucleus at position C-7 and differ from each other by the presence of the 5(6)-double bond. The Δ24-22-sulfoxycholestane side chain of the steroi

Activated M-phase-promoting factor (MPF) is exported from the nucleus of starfish oocytes to increase the sensitivity of the Ins(1,4,5)P3 receptors.

Starfish oocytes that are extracted from the ovaries are arrested at the prophase of the first meiotic division. At this stage of maturation, they are characterized by a large nucleus called the germinal vesicle. Meiosis resumption (maturation) can be induced in vitro by adding the hormone 1-methyladenine (1-MA) to the seawater in which the oocytes are suspended. Earlier work in our laboratory had detected Ca(2+) increases in both the cytoplasm and the nucleus of the oocytes approx. 2 min after the 1-MA challenge. The nuclear Ca(2+) increase was found to be essential for the continuation of th

Anticancer Effect of Pacificusoside D from the Starfish Solaster pacificus in Combination with 2-Deoxy-D-glucose on Oxidative Phosphorylation in Triple-Negative Breast Cancer Cells MDA-MB-231.

Triple-negative breast cancer (TNBC) represents significant therapeutic challenges due to its aggressive behavior, metabolic plasticity, and lack of targeted treatments, prompting investigation of biologically active triterpene glycosides from the starfish Solaster pacificus. This study evaluated the ability of pacificusoside D (SpD) to synergistically enhance the anticancer efficacy of the glycolytic inhibitor 2-deoxy-D-glucose (2-DG) in TNBC MDA-MB-231 cells by targeting mitochondrial oxidative phosphorylation (OXPHOS). Methods included metabolic profiling via glucose uptake, lactate, and gl

Localization and electrical activity of the distance chemoreceptors that mediate predator avoidance behaviour in Acmaea limatula and Acmaea scutum (gastropoda, prosobranchia).

1. The marine gastropods Acmaea (Collisella) limatula and Acmaea (Notoacmea) scutum respond to distant predatory starfish (i.e. to starfish scent) by moving up a vertical surface. 2. The distance chemoreceptors that mediate this avoidance behaviour are located on the mantle margin. Heat cauterization of the limpets' mantle margin eliminates their responsiveness to Pisaster ochraceus scent, while a similar cauterization of the ctenidium and the osphradia does not diminish the avoidance behaviour. 3. Primary afferent electrical activity can be recorded from the chemoreceptors on the mantle margi

Effect of Relaxin-Like Gonad-Stimulating Peptide on Ovaries of the Sea Cucumber Holothuria leucospilota.

Relaxin-like gonad-stimulating peptide (RGP), identified in starfish as a gonadotropin-like active peptide, has been shown to have a similar effect in sea cucumbers. Chemically synthesized RGP of the black sea cucumber, Holothuria leucospilota, (Hle-RGP) induced oocyte maturation and ex vivo spawning in ovarian fragments of the same species in the reproductive season. Therefore, this study established ovarian fragments of H. leucospilota as a bioassay model to determine the RGP concentration (EC50) required for 50% ex vivo spawning. The EC50 for Hle-RGP was approximately 7 nM. However, Hle-RGP

Ca2+ spikes in the flagellum control chemotactic behavior of sperm.

The events that occur during chemotaxis of sperm are only partly known. As an essential step toward determining the underlying mechanism, we have recorded Ca2+ dynamics in swimming sperm of marine invertebrates. Stimulation of the sea urchin Arbacia punctulata by the chemoattractant or by intracellular cGMP evokes Ca2+ spikes in the flagellum. A Ca2+ spike elicits a turn in the trajectory followed by a period of straight swimming ('turn-and-run'). The train of Ca2+ spikes gives rise to repetitive loop-like movements. When sperm swim in a concentration gradient of the attractant, the Ca2+ spike

Injection of Acanthaster planci with thiosulfate-citrate-bile-sucrose agar (TCBS). I. Disease induction.

This is the first report of the successful induction of a transmissible disease in the coral-eating crown-of-thorns starfish Acanthaster planci (COTS). Injection of thiosulfate-citrate-bile-sucrose agar (TCBS) culture medium into COTS induced a disease characterized by discoloured and necrotic skin, ulcerations, loss of body turgor, accumulation of colourless mucus on many spines especially at their tip, and loss of spines. Blisters on the dorsal integument broke through the skin surface and resulted in large, open sores that exposed the internal organs. Oedema and reddened digestive tissues a

Sea star wasting disease pathology in Pisaster ochraceus shows a basal-to-surface process affecting color phenotypes differently.

Sea star wasting disease (SSWD) refers to a suite of poorly described non-specific clinical signs including abnormal posture, epidermal ulceration, and limb autotomy (sloughing) causing mortalities of over 20 species of sea stars and subsequent ecological shifts throughout the northeastern Pacific. While SSWD is widely assumed to be infectious, with environmental conditions facilitating disease progression, few data exist on cellular changes associated with the disease. This is unfortunate, because such observations could inform mechanisms of disease pathogenesis and host susceptibility. Here,

The first records of sea star wasting disease in Crossaster papposus in Europe.

Sea star wasting disease (SSWD) refers to a suite of gross pathological signs observed in Asteroidea species. It presents to varying degrees as abnormal posture, epidermal ulceration, arm autotomy and eversion of viscera. We report observations of SSWD in the sunstar Crossaster papposus, the first observations of its kind in Europe. While the exact cause of SSWD remains unknown, studies have proposed pathogenic and environmental-stress pathways for disease outbreaks. Although the present observations do not support a precise aetiology, the presence of SSWD in a keystone predator may have wide
