BACKGROUND: Zinc is widely used in oral care products due to its antimicrobial and anti-biofilm properties; however, the molecular mechanisms by which zinc influences Streptococcus mutans (S. mutans) physiology remain incompletely understood. The AdcABC system is the primary high-affinity zinc transporter in S. mutans. We investigated whether loss of adcBC is associated with altered physiological responses in S. mutans.
MATERIAL AND METHODS: Wild-type (WT), ΔadcBC mutant, and complemented S. mutans UA159 strains were evaluated under zinc-replete and zinc-limited conditions using zinc sulphate

Microbial homeostasis is maintained by the antagonistic capacity of commensal bacteria against cariogenic pathogens. In the oral cavity, commensal Streptococcus, dominant colonizers of the tooth surface, can produce hydrogen peroxide (H2O2), modulating virulent cross-kingdom biofilm formation. To investigate their ecological role, clinical isolates from dental plaque were compared with reference strains, including Streptococcus oralis ATCC 35037 and S. oralis subsp. tigurinus J22 to determine their H2O2-producing capabilities. The antagonistic potential of S. oralis against Streptococcus mutan

Dental caries is initiated by the adherence of Streptococcus mutans to tooth surfaces, leading to biofilm (plaque) formation and the production of enamel-eroding acids. Sortase A (SrtA) plays a critical role in this process because it anchors many protein adhesins to the bacterial cell wall, making it a promising target for plaque prevention. Our previous research demonstrated that edible polyphenols from maple extracts-nortrachelogenin-8'-O-β-D-glucopyranoside, lariciresinol, isoscopoletin, and (-)-epicatechin gallate (ECG)-inhibit SrtA in Listeria monocytogenes and act as antibiofilm agents.

Dental caries of permanent teeth is a common public health concern and has the second-highest incidence among global diseases. The exopolysaccharides (EPS) synthesized by Streptococcus mutans (S. mutans) are the principal virulence factor for cariogenic etiology. We previously discovered that an endogenous antisense vicR RNA (ASvicR) could significantly inhibit EPS synthesis in S. mutans and reduce its cariogenicity. However, ASvicR cannot be directly applied in the oral environment. An appropriate vector is of great need to protect ASvicR from being degraded by nucleases for effective gene de

The motor impairments of cerebral palsy (CP) are typically accompanied by subsequent musculoskeletal issues, seizures, and abnormalities of sensation, intelligence, communication, and behaviour. These kids have a lower capacity for regulating oral health because of their poor voluntary movements. Poor oral hygiene brought on by insufficient brushing and flossing, increased use of sugary foods, and orally administered drugs puts people at risk for periodontal disorders and dental caries. Poor dental health and rising therapy demands establish a sadistic cycle that affects patient overall health

Despite the cariogenic role of Candida suggested from recent studies, oral Candida acquisition in children at high risk for early childhood caries (ECC) and its association with cariogenic bacteria Streptococcus mutans remain unclear. Although ECC disproportionately afflicts socioeconomically disadvantaged and racial-minority children, microbiological studies focusing on the underserved group are scarce. Our prospective cohort study examined the oral colonization of Candida and S. mutans among 101 infants exclusively from a low-income and racial-minority background in the first year of life. T

In the oral cavity, Candida species form mixed biofilms with Streptococcus mutans, a pathogenic bacterium that can secrete quorum sensing molecules with antifungal activity. In this study, we extracted and fractioned culture filtrate of S. mutans, seeking antifungal agents capable of inhibiting the biofilms, filamentation, and candidiasis by Candida albicans. Active S. mutans UA159 supernatant filtrate components were extracted via liquid-liquid partition and fractionated on a C-18 silica column to resolve S. mutans fraction 1 (SM-F1) and fraction 2 (SM-F2). We found anti-biofilm activity for

Streptococcus mutans is a key cariogenic bacterium responsible for the initiation of tooth decay. Biofilm formation is a crucial virulence property. We discovered a putative glycosyltransferase, SMU_833, in S. mutans capable of modulating dynamic interactions between two key biofilm matrix components, glucan and extracellular DNA (eDNA). The deletion of smu_833 decreases glucan and increases eDNA but maintains the overall biofilm biomass. The decrease in glucan is caused by a reduction in GtfB and GtfC, two key enzymes responsible for the synthesis of glucan. The increase in eDNA was accompani

Non-mechanical procedures for removing caries-infected dentin are warranted in dentistry. We previously demonstrated the marked sterilization effect for direct irradiation of low-temperature plasma using dentin model infected with Streptococcus mutans. However it requires 180 s of intraoral plasma irradiation to eliminate bacteria. We alternatively investigated whether plasma-treated water (PTW), i.e., pure water exposed to plasma in an atmosphere, has a same bactericidal activity with the plasma irradiation. In the infected dentin model, the viable S. mutans counts recovered by bur at depth o

INTRODUCTION: Many essential oils have been advocated for use in complementary medicine for bacterial and fungal infections. However, few of the many claims of therapeutic efficacy have been validated adequately by either in vitro testing or in vivo clinical trials.
OBJECTIVE: To study the antibacterial activity of nine commercially available essential oils against Streptococcus mutans in vitro and to compare the antibacterial activity between each material.
METHODOLOGY: Nine pure essential oils; wintergreen oil, lime oil, cinnamon oil, spearmint oil, peppermint oil, lemongrass oil, cedarwoo

This study assesses salivary conditions of 20 children with cardiac disease comparing with a control group of 15 healthy children. The results showed that there was no difference between the groups on salivary flow, buffer capacity and the level of Streptococcus mutans (Sm). The test group i.e., children with cardiac disease, showed a lower level of Lactobacillus sp. The association between the usage of antibiotics and the risk of developing caries, measuring the level of Streptococcus mutans and Lactobacillus sp., showed that children taking antibiotics frequently had a significant lower leve

The salivary lactobacilli and Streptococcus mutans counts in 217 university students of 25.4-yr-old were studied with regard to the state of eruption of their third molar teeth. Statistically significantly higher counts of both bacterial species were found in students who had one or more third molars partly erupted, compared with those whose third molars were lacking or fully erupted. No other parameters studied such as salivary flow rate or buffering capacity of the saliva were found to explain the high bacterial counts. These findings should be taken into consideration when interpreting sali

From its discovery in the late 1800s to the present day, Streptococcus mutans research has been at the forefront of dental caries investigations, providing the foundation for our current understanding of the microbiological determinants of this disease. In addition, research on S. mutans has greatly advanced key areas of microbiology, establishing the organism as a notably useful model for understanding the biology of other streptococcal species, as well as Gram-positive bacteria more broadly. In this piece, we provide a retrospective of how S. mutans has served this dual role, highlighting sp

OBJECTIVE: This research aimed to determine whether capsaicin (CAP), a phytochemical in chilli peppers, inhibits cariogenic Streptococcus mutans-Candida albicans cross-kingdom biofilms and their key virulence factors, and to explore underlying mechanisms.
DESIGN: The anti-caries potential of CAP was assessed in S. mutans-C. albicans co-cultures by quantifying viable cell counts, biomass and biofilm metabolic activity. Biofilm microstructure was visualized by confocal laser scanning microscopy with COMSTAT quantification, and CAP-induced transcriptional changes were profiled by qRT-PCR. Matche

BACKGROUND: Streptococcus mutans (S. mutans) contributes to caries. The biofilm formed by S. mutans exhibits greater resistance to drugs and host immune defenses than the planktonic form of the bacteria. The objective of this study was to evaluate the anti-biofilm effect of cordycepin from the perspective of metabolomics.
METHODS: The minimum inhibitory concentration (MIC) was determined to evaluate the antimicrobial effect of cordycepin on planktonic S. mutans. The 24-h biofilm was treated with 128 µg/mL of cordycepin for 10 min at the 8- or 20-h time points. Biofilm biomass and metabolism w

Houttuynia cordata is an herbal plant distributed throughout Asia. H. cordata has many bioactive properties, including antibacterial properties. The antibacterial effects of H. cordata on S. mutans remain unknown. Therefore, we treated S. mutans with 1, 3, 5, 10, 20, 30, or 40 mg/mL H. cordata extract at 37°C for 24 h. The antibacterial effect of H. cordata against S. mutans was confirmed using colony forming unit assay and disk diffusion assays. The results of the cell concentration assay demonstrated that H. cordata inhibited the growth of S. mutans in a dose-dependent manner. Prominent grow

Streptococcus mutans (S. mutans) is the main pathogenic bacterium causing dental caries, and the modes in which its traits, such as acid production, acid tolerance, and adhesion that contribute to the dental caries process, has been clarified. However, a growing number of animal experiments and clinical revelations signify that these traits of S. mutans are not restricted to the detriment of dental tissues. These traits can assist S. mutans in evading the immune system within body fluids; they empower S. mutans to adhere not merely to the surface of teeth but also to other tissues such as vasc

Large septic pulmonary embolus is a rare finding in right-sided endocarditis. The entity represents a challenging diagnosis due to its variable and nonspecific clinical and radiological presentation and similarities with other conditions. We present a case of a 41 year-old woman who developed a large main pulmonary artery embolus and bilateral cavitary lung nodules in the setting of severe sepsis. Pulmonary artery exploration and clot retrieval ultimately revealed a large septic embolus from Streptococcus mutans native pulmonary valve endocarditis. The diagnosis of septic pulmonary emboli from

Stroke is the fifth leading cause of death and is responsible for approximately nine percent of all deaths worldwide. Cases of Streptococcus mutans (S. mutans)-induced intracerebral hemorrhage as a result of bloodstream infections have seldom been reported. New reports show that bacteria with specific collagen binding proteins (CBPs), such as the Cnm type produced by S. mutans, may inhibit platelet aggregation and cause bleeding. In this article, we report on a 62-year-old man with a recent history of left frontal intracerebral hemorrhage (ICH) who presented to the emergency department after a

An antigen that cross reacts with mammalian heart tissue has been shown to be present in several strains of S mutans. Inoculation of S mutans into rabbits elicited heart-reactive antibody as part of the immunologic response. This heart-reactive antibody was demonstrated to be tissue-specific in that it only bound heart and smooth muscle tissue. Similar results have been previously reported using group A streptococci. Adsorption studies using sarcolemmal sheaths, group A streptoccal membranes, and fractions of cariogenic streptococci demonstrated the presence of a similar antigen. The aforement
