Gefitinib modulates stress fibers and tubular-like structure formation and attenuates angiogenesis in an in vivo chicken model of chorioallantoic membrane angiogenesis.

Gefitinib is an ATP-competitive inhibitor of receptor tyrosine kinases known to repress the progression of various types of cancers. Emerging evidence has shown that this molecule modulates endothelial cells to inhibit angiogenesis. However, the biological effects of gefitinib have not been comprehensively investigated in endothelial cells. In this study, gefitinib-mediated regulation of cell proliferation, migration, cell attachment, cytoskeletal actin filament reorganization, tubular-like structure formation and angiogenesis in vivo was examined along with the corresponding mechanisms. Combined glutamine, dietary fiber, and oligosaccharide supplementation attenuates fasting-associated intestinal dysbiosis and stress responses in rats.

BACKGROUND: Prolonged preoperative fasting is associated with intestinal dysbiosis, impaired mucosal homeostasis, and delayed postoperative recovery. Combined nutritional supplementation with glutamine, dietary fiber, and oligosaccharides has the potential to protect intestinal function during nutritional stress. In this study, we investigated the effects of this nutritional combination in a rat model of prolonged fasting. METHODS: Male Sprague-Dawley rats received oral supplementation with a combination of glutamine, dietary fiber, and oligosaccharides (GFO) or a 15% glucose solution twice d

Methyl 3,4-dihydrobenzoate attenuates muscle fiber necroptosis and macrophage pyroptosis by regulating oxidative stress in inflammatory myopathies.

Inflammatory cell infiltration and myofiber necrosis are pathological hallmarks of idiopathic inflammatory myopathies (IIM). Methyl 3,4-dihydroxybenzoate (MDHB) is a natural phenolic acid compound, renowned for its anti-inflammatory and antioxidant effects. In this study, we investigated its protective mechanisms targeting muscle fiber necrosis and macrophage pyroptosis by regulating oxidative stress in IIM. In the present study, we found that increased reactive oxygen species (ROS) level and decreased nuclear factor erythroid 2 related factor 2 (Nrf2) protein expression were shown on the musc

Aberrant white matter microstructure evaluation by automated fiber quantification in typhoon-related post-traumatic stress disorder.

Super typhoons can lead to post-traumatic stress disorder (PTSD), which can adversely affect a person's mental health after a disaster. Neuroimaging studies suggest that patients with PTSD may have post-exposure abnormalities of the white matter. However, little is known about these defects, if they are localized to specific regions of the white matter fibers, or whether they may be potential biomarkers for PTSD. Typhoon survivors with PTSD (n = 27), trauma-exposed controls (TEC) (n = 33), and healthy controls (HCs) (n = 30) were enrolled. We used automated fiber quantification (AFQ) to proces

Stress Analysis of PDL and Apical Foramen in Traumatized Teeth With Incisal/Cervical Fiber Splinting: A Finite Element Study.

BACKGROUND/AIM: Healing of the periodontal ligament (PDL) and pulp vitality is critical for successful management of traumatic dental injuries (TDI), which could be compromised due to adverse stress distributions caused by occlusal forces. As a common treatment to stabilize traumatized teeth, the impact of fiber splinting locations on the masticatory stress distribution in the PDL and dental pulp has not reached a consensus. The biomechanical effects of incisal and cervical fiber splinting on stress distribution in traumatized teeth were investigated in this study. MATERIAL AND METHODS: A con

Mechanism of skeletal muscle atrophy by muscle fiber types in male rats under long-term fasting stress.

Fasting induces metabolic changes in muscles, which are differentiated by muscle fiber type. In this study, the mechanism of fasting-induced muscle atrophy in rats was examined to determine the differences between muscle fiber types in energy production. Fasting for 96 h did not alter the weight of the soleus (SOL), a fiber type I muscle, but did significantly reduce the weight of gastrocnemius (GM), a fiber type II muscle. GM, SOL and blood pregnenolone and testosterone levels decreased under fasting, which induced energy deprivation, whereas corticosterone (CORT) levels significantly increas

Analysis of the Effect of Fiber Orientation on Mechanical and Elastic Characteristics at Axial Stresses of GFRP Used in Wind Turbine Blades.

Due to its physical and mechanical properties, glass-fiber-reinforced polymer (GFRP) is utilized in wind turbine blades. The loads given to the blades of wind turbines, particularly those operating offshore, are relatively significant. In addition to the typical static stresses, there are also large dynamic stresses, which are mostly induced by wind-direction changes. When the maximum stresses resulting from fatigue loading change direction, the reinforcing directions of the material used to manufacture the wind turbine blades must also be considered. In this study, sandwich-reinforced GFRP ma

Stress Concentration of Endodontically Treated Molars Restored with Transfixed Glass Fiber Post: 3D-Finite Element Analysis.

The loss of dental structure caused by endodontic treatment is responsible for a decrease in tooth resistance, which increases susceptibility to fracture. Therefore, it is important that minimally invasive treatments be performed to preserve the dental structure and increase the resistance to fracture of endodontically treated posterior teeth. To evaluate under axial loads, using the finite element method, the stress distribution in endodontically treated molars restored with both transfixed or vertical glass fiber posts (GFP) and resin composite. An endodontically treated molar 3D-model was a

Impact of feeding a high fiber diet and roughage on stress and clinical welfare indicators in fast-growing broiler breeder pullets.

Qualitative feed restriction has been proposed as a means to alleviate hunger, frustration, and stress associated with feed restriction in broiler breeder chickens. The aim of this study was to investigate the effect of a fiber-rich diet containing oat-hulls and a daily allocation of roughage (EXP) compared to a standard commercial diet (CON) on broiler breeder welfare. A total of 600 day-old female Ross 308 breeder chicks were allocated to 12 pens of 50 birds each, with each pen receiving one of the two dietary treatments. At the conclusion of the study, a welfare assessment was performed whi

Oxytocin and lysophosphatidic acid induce stress fiber formation in human myometrial cells via a pathway involving Rho-kinase.

The actin cytoskeleton is important for stress fiber formation and contributes to the initiation and maintenance of smooth muscle contraction. To determine if oxytocin and lysophosphatidic acid (LPA) induce stress fiber formation, cultured human myometrial cells were exposed to oxytocin (10(-5) M) or LPA (10(-6) M), and filamentous (F) and globular (G) actin pools were stained with fluorescein isothiocyanate-phalloidin and Texas red DNase I, respectively. The F- to G-actin fluorescent-staining ratio was measured by fluorescence microscopy. Oxytocin and LPA increased stress fiber formation, as

Ninety-day experimental hyperthyroidism in male rats: Effects on muscle contractility, fiber composition, oxidative stress, and nerve conduction.

Hyperthyroidism generates a hypermetabolic state that promotes adaptations in the skeletal muscle and peripheral nerves leading to deleterious effects. However, there is a paucity of data and conflicting evidence on the influence of thyroid hormones on muscles with predominantly glycolytic fibers and on peripheral nerves. We assessed the contractility, oxidative stress, and muscle fiber composition in the extensor digitorum longus (EDL) and diaphragm, in addition to assessing the nerve conduction in isolated sciatic nerve in a Wistar rat model of chronic hyperthyroidism, induced by levothyroxi

Effect of carbon-fiber-reinforced polyetheretherketone on stress distribution in a redesigned tumor-type knee prosthesis: a finite element analysis.

Background: Surgery for bone tumors around the knee often involves extensive resection, making the subsequent prosthetic reconstruction challenging. While carbon fiber-reinforced polyetheretherketone (CF-PEEK) has been widely used in orthopedic implants, its application in tumor-type prosthesis is limited. This study aims to evaluate the feasibility of using 30wt% and 60wt% carbon fiber-reinforced polyetheretherketone (CF30-PEEK and CF60-PEEK) as materials for a redesigned tumor-type knee prosthesis through numerical analysis. Methods: A knee joint model based on CT data was created, and the r

The Involvement of Endothelin Pathway in Chronic Psychological Stress-Induced Bladder Hyperalgesia Through Capsaicin-Sensitive C-Fiber Afferents.

INTRODUCTIONS: Interstitial cystitis/bladder pain syndrome (IC/BPS) is a poorly understood chronic disorder characterized by bladder-related pain. Chronic psychological stress plays a key role in the exacerbation and development of IC/BPS via unclear mechanisms. This study aimed to investigate the role of endothelin 1 (ET-1) and its receptors in the development of chronic stress-induced bladder dysfunction. METHODS: Wistar-Kyoto rats were exposed to chronic (10 days) water avoidance stress (WAS) or sham stress, with subgroups receiving capsaicin pretreatment to desensitize C-fiber afferents.

Polarization Maintaining Fiber Temperature and Stress Gradient Sensitization Sensor Based on Semiconductor-Metal-Polymer Three-Layer Film Coating.

Increasing sensitivity, measuring points, and stability have always been the pursuit of sensors. ZnSe9:CO1 and Ag composite nano films were coated on polarization maintaining fiber (PMF). Then, the coated PMF was nested in capillary and hose which was encapsulated with polydimethylsiloxane (PDMS) and epoxy resin. The integrated capillary sensor and thermoplastic hose sensor were prepared. The gradient sensitization of various measurement parameters such as temperature, stress, and micro bending is realized. The temperature sensitivity is 1.49 nm/°C, the micro bending sensitivity is 1.72 nm/102

Relationship between cell stiffness and stress fiber amount, assessed by simultaneous atomic force microscopy and live-cell fluorescence imaging.

Actomyosin stress fibers, one of the main components of the cell's cytoskeleton, provide mechanical stability to adherent cells by applying and transmitting tensile forces onto the extracellular matrix (ECM) at the sites of cell-ECM adhesion. While it is widely accepted that changes in spatial and temporal distribution of stress fibers affect the cell's mechanical properties, there is no quantitative knowledge on how stress fiber amount and organization directly modulate cell stiffness. We address this key open question by combining atomic force microscopy with simultaneous fluorescence imagin

Expression of the proteoglycan syndecan-4 and the mechanism by which it mediates stress fiber formation in folliculostellate cells in the rat anterior pituitary gland.

Folliculostellate (FS) cells in the anterior pituitary gland appear to have multifunctional properties. FS cells connect to each other at gap junctions and thereby form a histological and functional network. We have performed a series of studies on network formation in FS cells and recently reported that FS cells markedly prolong their cytoplasmic processes and form numerous interconnections with neighboring FS cells in the presence of laminin, an extracellular matrix (ECM) component of the basement membrane. In this study, we investigated the mechanism of this extension of FS cell cytoplasmic

Dates, Polyunsaturated Fatty Acids, Dietary Fiber, and Probiotics as Potential Complementary Modulators of Gut Microbiota and Oxidative Stress: An Integrative Review.

This integrative review examines the potential of dates, omega-3 (ω-3) and omega-6 (ω-6) polyunsaturated fatty acids (PUFAs), dietary fibers (DFs), and probiotic bacteria to support gut health through complementary nutritional pathways. The literature was searched in PubMed, Web of Science, Scopus, and Google Scholar for English-language studies published through June 2026. Dates contain fermentable substrates, including DFs and non-digestible oligosaccharides, that can serve as growth substrates for beneficial gut microbes. However, the whole fruit has not been formally established as a prebi

Acute stress impairs hippocampal mossy fiber-CA3 long-term potentiation by enhancing cAMP-specific phosphodiesterase 4 activity.

The mossy fiber synapses onto hippocampal CA3 neurons show unique molecular features and a wide dynamic range of plasticity. Although acute stress has been well recognized to alter bidirectional long-term synaptic plasticity in the hippocampal CA1 region and dentate gyrus, it remains unclear whether the same effect may also occur at the mossy fiber-CA3 synapses. Here, we report that hippocampal slices prepared from adult mice that had experienced an acute unpredictable and inescapable restraint tail-shock stress showed a marked impairment of long-term potentiation (LTP) induced by high-frequen

A clinicoanatomical study of the novel nerve fibers linked to stress urinary incontinence: the first morphological description of a nerve descending properly along the anterior vaginal wall.

When performing anterior colporrhaphy for cystocele, most pelvic surgeons have not considered the neuroanatomy that contributes to urethral function. The aim of the study was to anatomically identify nerve fibers located in the anterior vagina associated with the pathogenesis of incontinence and pelvic organ prolapse. Anterior vaginal specimens were obtained from 17 female cadavers and 33 cases of clinical cystocele by anterior vaginal resection. The specimens were step-sectioned and stained with hematoxylin-eosin, S100 antibody, and tyrosine hydroxylase antibody. As a result, descending nerve
