Transcriptome analysis of a parasitic clade V nematode: comparative analysis of potential molecular anthelmintic targets in Cylicostephanus goldi.

Clade V nematodes comprise several parasitic species that include the cyathostomins, primary helminth pathogens of horses. Next generation transcriptome datasets are available for eight parasitic clade V nematodes, although no equine parasites are included in this group. Here, we report next generation transcriptome sequencing analysis for the common cyathostomin species, Cylicostephanus goldi. A cDNA library was generated from RNA extracted from 17 C. goldi male and female adult parasites. Following sequencing using a 454 GS FLX pyrosequencer, a total of 475,215 sequencing reads were generate

The distribution of inhibited early third stage Cyathostominae larvae in the large intestine of the horse.

The distribution of inhibited early third stage Cyathostominae larvae in different parts of the large intestine of the horse was studied in 20 Shetland ponies necropsied in autumn 1982, 1983 and 1984. The location of the larvae in the large intestinal wall was studied by histological examination of the intestines of the eight ponies from 1984. Inhibited larvae were located predominantly and more or less equally in the caecum and the ventral colon. Generally fewer early L3 were in the dorsal colon. In 1984 a considerable proportion (mean 17%, range 9.7-36.9%) of the inhibited larvae was found i

Verminous (Strongylus vulgaris) myelitis in a donkey.

A fifth stage Strongylus vulgaris migrated through the spinal cord of a 2-year-old, male donkey resulting in progressive paraparesis and then tetraplegia. A profound neutrophilic pleocytosis was detected on analysis of cerebrospinal fluid. The parasite appeared to have entered the mid-lumbar spinal cord, migrated to the cranial thoracic segments, exited, then re-entered the spinal cord a few segments craniad. It then traveled further cranially and was found in the third cervical spinal cord segment. Some parts of the lesion were remarkably free from tissue necrosis, hemorrhage and inflammation

Control of Filaroides hirthi in commercially reared beagle dogs.

Two commercial research Beagle breeding colonies began a control program for Filaroides hirthi. All stud dogs and all nonpregnant, nonlactating brood bitches were given two courses, 2-4 weeks apart, of 25 mg/kg body weight albendazole per os twice daily for 5 days. Puppies of treated dams were isolated from puppies of nontreated dams. The proportions of infected puppies of puppies killed in 1979 were 45% and 78% from the two colonies. After 1979, the proportions were 0.2% and 24%. One colony may have had a problem with albendazole toxicity; the problem was stopped by changing the diet.

Procyanidin A2 in the Australian plant Alectryon oleifolius has anthelmintic activity against equine cyathostomins in vitro.

There is a need to investigate new methods of controlling cyathostomins in horses due to increasing anthelmintic resistance amongst these parasites. In a previous study we identified the Australian plant Alectryon oleifolius as having anthelmintic activity towards cyathostomins. This study aimed to isolate and identify the bioactive compound(s) responsible for all or part of this anthelmintic activity and quantify its activity in vitro. The condensed tannin procyanidin A2 was isolated from the plant through a process of bioassay guided fractionation and identified using 1D and 2D nuclear magne

The influence of flotation solution, sample dilution and the choice of McMaster slide area (volume) on the reliability of the McMaster technique in estimating the faecal egg counts of gastrointestinal strongyles and Dicrocoelium dendriticum in sheep.

The present study was aimed to evaluate the influence of flotation solution, sample dilution, and the choice of McMaster slide area (volume) on the reliability of the McMaster technique in estimating the faecal egg counts of gastrointestinal (GI) strongyles and Dicrocoelium dendriticum in a composite sample of faeces from naturally infected sheep. Fourteen flotation solutions having densities between 1.200 and 1.450, and six sample dilutions, 1:10, 1:15, 1:20, 1:30, 1:40 and 1:50 were used. Each of the six dilutions was divided into 70 aliquots in order to have five replicates of each of the 1

A national survey of anthelmintic resistance in ascarid and strongylid nematodes in Australian Thoroughbred horses.

This study quantified the extent of anthelmintic resistance (AR) in ascarid and strongylid nematodes against commonly used anthelmintics in Australian Thoroughbred horses. Faecal egg count reduction tests (FECRTs, n = 86) and egg reappearance period (ERP) tests were conducted on 22 farms across Australia. Faecal egg counts (FECs) were determined using the modified McMaster technique, and percent faecal egg count reduction (%FECR) was calculated using the Bayesian hierarchical model and hybrid Frequentist/Bayesian analysis method. The results were interpreted using old (published in 1992) and n

Outbreak of acute larval cyathostominosis - A "perfect storm" of inflammation and dysbiosis.

BACKGROUND: Cyathostomins are prevalent and pathogenic intestinal helminths of horses, causing acute and chronic disease, including acute larval cyathostominosis, which has a mortality rate of 50%. Factors determining individual susceptibility to acute larval cyathostominosis are unknown. Investigation of these factors could lead to novel treatment and prevention strategies. OBJECTIVES: To investigate clinicopathological and faecal microbiota changes associated with disease in individual horses in an acute larval cyathostominosis outbreak. STUDY DESIGN: Case series. METHODS: The study popul

Spatial niche competition among helminth parasites in horse's large intestine.

The spatial distribution of large intestinal helminth parasites of 50 horses was studied. Both adult and mucosal larval stages were examined and counted within the ventral colon, dorsal colon and caecum. Thirty-three species of adult parasites were identified and their distribution in the three large intestinal regions reported, together with the localization of cyathostome encysted larvae and of empty mucosal cysts. In order to highlight interspecific interactions, both Spearman pairwise correlation (among all parasites) and multivariable negative binomial regression (among the most abundant

Seasonal changes in the level of infective strongylate nematode larvae on pasture in the coastal savanna regions of Ghana.

The level and type of infective strongylate nematode larvae on pasture were followed from March 1994 to April 1995 in the coastal savanna regions of Ghana. The number of infective larvae on pasture was high, reaching 2458 kg-1 dry matter of grass during the period of and soon after the rains, and very low or none in the absence of rainfall. The number of infective larvae on pasture was directly related to the pattern of rainfall, but it was also influenced by the number of raindays in the period. The following genera were found in order of prevalence: Haemonchus, Oesophagostomum, Trichostrongy

The Imaginal Disc Growth Factors 2 and 3 participate in the Drosophila response to nematode infection.

The Drosophila imaginal disc growth factors (IDGFs) induce the proliferation of imaginal disc cells and terminate cell proliferation at the end of larval development. However, the participation of Idgf-encoding genes in other physiological processes of Drosophila including the immune response to infection is not fully understood. Here, we show the contribution of Idgf2 and Idgf3 in the Drosophila response to infection with Steinernema carpocapsae nematodes carrying or lacking their mutualistic Xenorhabdus nematophila bacteria (symbiotic or axenic nematodes, respectively). We find that Idgf2 an

Effect of ivermectin treatment on eosinophilic pneumonia and other extravascular lesions of late Strongylus vulgaris larval migration in foals.

Eighteen parasite-free pony foals were infected orally with 500 third stage larvae of Strongylus vulgaris. At 56 days after infection, six ponies were treated with intramuscular ivermectin (22, 23-dihydroavermectin B1); six were treated with oral ivermectin; and six were not treated. Necropsy was done 91 days after infection to study the pathologic effects of migrating S. vulgaris larvae and to determine the efficacy of ivermectin in attenuation of S. vulgaris-induced lesions. Larval migration induced eosinophilic inflammation of the liver, spleen, mesenteric, colic and cecal lymph nodes, and
