Strontium: new drug. Postmenopausal osteoporosis: too many unknowns.

(1) Strontium ranelate is marketed in the European Union for the treatment of postmenopausal osteoporosis. Strontium, a cation closely related to calcium, was already used for this purpose in the 1950s but was abandoned because it caused bone mineralization disorders (mainly due to the high doses used at the time). (2) Strontium has only been compared with placebo: there are no clinical trials versus a diphosphonate. (3) On the basis of bone mineral density, two dose-finding studies suggested that, in women who are also taking calcium and vitamin D, the effective minimal dose of strontium is 1

Strontium causes osteomalacia in chronic renal failure rats.

BACKGROUND: We recently reported an association between increased bone strontium (Sr) levels and osteomalacia in dialysis patients. METHODS: To delineate whether or not Sr acts as a causal factor in the development of osteomalacia, we devised the following study: four groups of chronic renal failure (CRF) rats were given Sr, aluminum (Al), both of these compounds or none of the elements (controls). RESULTS: Administration of Sr and/or A1 resulted in increased bone levels of the respective elements. Histological examination revealed impairment of mineralization in the Sr group and to a lesser

Harnessing strontium in regenerative dentistry: progress and perspectives.

Strontium (Sr), an alkaline earth element characterized by its unique bioactivity, has garnered significant interest in the field of oral regenerative medicine. This interest is primarily due to its inherent presence in bone tissues, particularly in growth plates and trabeculae, and its ability to influence bone metabolism through the calcium-sensing receptor (CaSR). Various signaling pathways, including MAPK/ERK1/2 and Wnt/NFATc, are involved in the mechanisms behind these effects. Strontium-doped biomaterials progressively release Sr2+ ions, which impart a variety of biological effects, amon

Advances in strontium-releasing biomaterials for bone repair.

Strontium-containing biomaterials offer promising avenues for enhancing bone repair by leveraging the ion's reported dual anabolic and anti-catabolic effects on bone cells. These biomaterials aim to encourage site-specific bone growth, heal fractures, and improve metallic implant fixation through controlled release of strontium ions. Here, we provide an overview of strontium's use as a treatment for osteoporosis, and report current advances in biomaterials designed to deliver strontium locally to the skeleton from bioactive glasses, calcium phosphates, bone cements, polymers, and implant coati

Antiadipogenesis and Osseointegration of Strontium-Doped Implant Surfaces.

The decreased bone density and increased marrow adiposity that occur with aging may influence the outcome of dental implants. Strontium (Sr), an anabolic agent for the treatment of osteoporosis, has an inhibitory effect on adipogenesis but favors osteogenesis of bone marrow-derived mesenchymal stem cells (BMSCs). However, little is known about the effects and mechanisms of local Sr release on adipogenesis during bone formation in aged bone. In this study, a potential dental implant material, Sr-doped titanium, was developed via a sandblasted, large-grit, and acid-etched (SLA) method combined w

Setd2 is associated with strontium-induced bone regeneration.

UNLABELLED: Strontium Ranelate has been utilized as a preventative treatment option for osteoporosis with the release of Sr ions having a direct effect on preventing osteoclast activation and promoting osteoblast differentiation. Previously our group has prepared and characterized a porous Sr-mesoporous bioactive glass (Sr-MBG) scaffold demonstrating its ability to enhance new bone formation when compared to MBG alone. The goal of the present study was to elucidate the bone-inducing properties of Sr by utilizing RNA-seq on in vivo tissue samples to investigate potential target genes responsibl

The position of strontium ranelate in today's management of osteoporosis.

Osteoporosis accounts for about 3 % of total European health-care spending. The low proportion of costs for the pharmacological prevention of osteoporotic fracture means that it is highly cost saving, especially in patient with severe osteoporosis or patients who cannot take certain osteoporosis medications due to issues of contraindications or tolerability. Following recent regulatory changes, strontium ranelate is now indicated in patients with severe osteoporosis for whom treatment with other osteoporosis treatments is not possible, and without contraindications including uncontrolled hyper

Strontium ranelate--a promising therapeutic principle in osteoporosis.

Strontium ranelate (2g/day) appears to be a safe and efficient treatment of osteoporosis (OP), reducing the risks of both vertebral and non-vertebral fractures (including hip) in a wide variety of patients. Thus, the agent can now be considered as a first-line option to treat women at risk of OP fractures, whatever their age and the severity of the disease. A long-term treatment with strontium ranelate in OP women leads to a continued increase in bone mineral density at spine and hip levels, and a sustained antifracture efficacy. The mode of action of strontium ranelate involves a dissociation

Strontium ranelate: a novel treatment in postmenopausal osteoporosis.

Strontium ranelate (SR) is a novel antiosteoporotic agent, electively concentrated in positions of active bone formation, and especially onto the crystal surface that allows permanent exchanges with extracellular fluid. Although the mechanism(s) of action is still under rigorous research, SR appears to reduce bone resorption by decreasing osteoclast differentiation and activity and to stimulate bone formation by increasing replication of preosteoblast cells, leading to increased matrix synthesis. In the placebo-controlled, phase III trial spinal osteoporosis therapeutic intervention (SOTI) (no

[Strontium ranelate: a novel concept for the treatment of osteoporosis].

The di-strontium salt strontium ranelate, a novel orally active agent consisting of two atoms of stable strontium and the organic moiety ranelic acid, has been developed for the treatment of osteoporosis. It has been shown to enhance osteoblastic cell replication and increase collagen synthesis while decreasing pre-osteoclast differentiation and bone-resorbing activity of mature osteoclasts in vitro. Studies performed in healthy animals have shown that strontium ranelate not only increases bone mass at various skeletal sites but also improves mechanical properties of femoral, humeral and verte

[Therapy of osteoporosis: bisphosphonates, SERM's, teriparatide and strontium].

The therapy of osteoporosis is mostly based upon the use of drugs which inhibit bone resorption. Among these, the bisphosphonate family is the best known and mostly used by clinicians. Both second and third generation bisphosphonates, like alendronate and risedronate, are now available as weekly tablets which have facilitated the patient compliance to treatment together with a decreased occurrence of gastrointestinal side effects. These compounds are used efficiently to treat postmenopausal osteoporosis and osteoporosis of men as well. Their use did provide good evidence of increased bone mine

Strontium is a potent and selective inhibitor of sensory irritation.

BACKGROUND: Irritant contact dermatitis occurs following topical exposure to many chemicals found in cosmetics, personal care products, drugs, and during occupational exposure. Signs and symptoms may include sensory irritation (sting, burn, and/or itch), erythema, edema, and vesiculation. OBJECTIVE: In an attempt to discover new classes of anti-irritant compounds without anesthetic properties, I observed that topical application of strontium salts to intact skin produced potent suppression of sting, burn, and itch caused by many irritant chemicals. METHODS: Chemically and biologically unrela

End-Stage Tibiotalar Osteoarthritis and Chronic Strontium Toxicity.

Metal toxicity due to environmental sources or orthopedic implants has been a primary focus in recent literature. Specifically, in orthopedics, total joint arthroplasty regarding metal-on-metal articulation with cobalt-chromium articulation has adverse local and systemic effects. In particular, strontium toxicity is less known metal toxicity that can cause many systemic effects such as severe osteoporosis. This is the first reported case of strontium toxicity and end-stage tibiotalar osteoarthritis. We present a case of a 68-year-old female with bilateral ankle pain and deformity that were ref
