Biochemical and electrophysiological studies of mammalian CNS indicate that the amino-acid, glycine, is a major inhibitory neurotransmitter whose location is, for the large part, confined to the spinal cord and brain stem. In this study, autoradiographs of C57BL/6J mouse brain sections labeled with [3H] strychnine, a potent antagonist of glycine, were used to map the distribution of glycine receptors in the CNS. Autoradiographs showed highly localized areas of grain density, which confirmed the gross distributions described in homogenate binding studies and gave a more precise regional localiz

We have investigated in mice the effects of several drugs which may be administered as part of an anaesthetic technique on the convulsive threshold to laudanosine and to strychnine, which is reported to have a similar mechanism of action. I.v. administered propofol, thiopentone and midazolam increased the dose of convulsant necessary to produce seizure when administered 2 min before the convulsive stimulus. In contrast, methohexitone and etomidate exhibited a proconvulsant effect, although with the latter this was significant only in laudanosine-treated mice. Pethidine was proconvulsant in bot

Averaged evoked potentials to sciatic nerve stimulation and to direct stimulation of the cervical spinal cord were recorded from the dura and skin over the spinal cord and cauda equina in bipolar and common reference leads in cats and compared. Response waveform and conduction characteristics are described. The effects of increasing stimulus intensity, asphyxia and strychnine on these potentials are related. Alterations in these potentials produced by spinal cord at one or two levels are also described. These potentials are compared to similar potentials which have been recorded in man and the

A rapid, simple, and sensitive method has been developed for the identification and quantitation of strychnine in human blood. The sample cleanup procedure involved solid-phase extraction with Oasis(R) HLB cartridges. The extracts were analyzed by gas chromatography-electron impact ionization-mass spectrometry. Limits of detection (LOD) and quantitation (LOQ) were 0.03 and 0.10 microg/mL, respectively, and the method was found to be linear between the LOQ and 2.5 microg/mL, with a correlation coefficient of 0.9994. Intra- and interday precision and accuracy were determined at both low and high

The NMDA receptor (NMDAR) is a Ca (2+)-permeable cation channel that plays a critical role in neural network formation during brain development. Since it is blocked in a voltage-dependent manner by extracellular Mg(2+), in order for the NMDA to be activated, the membrane must be strongly depolarized. Immature neurons in the developing neocortex can be depolarized by ligand-gated Cl(-) channels, such as the glycine receptor (GlyR) or GABA(A) receptor (GABA(A) R). We here assess the contribution of GlyRs to Ca(2+) influx via NMDARs in neonatal mouse cortical neurons. The GlyR antagonist, strychn

While occasional myoclonic jerks are prevalent in cancer patients receiving opioids, severe myoclonic jerks and seizures due to opioids are uncommon. In this retrospective case series, we describe five cancer patients with refractory cancer pain and severe neuroexcitatory toxicity associated with extremely high-dose opioid therapy to characterize better the syndrome, its treatment, and its outcome. Two patients died following seizures, but three patients recovered following prompt treatment with parenteral midazolam infusions and rotation to alternative opioids. Possible mechanisms and treatme

The present study investigated if blockade of the modulatory glycine receptor of the NMDA receptor complex influences the expression of behavior (sniffing stereotypy and locomotion) and dopamine metabolism in rats as it has been shown for NMDA receptor antagonists. The glycine receptor antagonist, 7-chlorokynurenate (7-chloro-4-hydroxyquinoline-2-carboxylic acid), induced a dose-dependent sniffing stereotypy but had no effect on locomotion when it was given i.c.v. The glycine receptor agonist, D-cycloserine (D-4-amino-3-isoxazolidinone), antagonized the sniffing stereotypy. 7-Chlorokynurenate

The effects of glycine and other inhibitory amino acid neurotransmitters on strychnine convulsive threshold were studied in mice. The mean intravenous threshold dose for strychnine to produce its convulsive effects in briefly restrained mice was determined to be 1.386 +/- 0.035 mg/kg. The dose of strychnine produced 100% postconvulsive mortality in all the mice tested. Intraperitoneal administration of various doses (100-500 mg/kg) of glycine, beta-alanine and L-threonine, 15-20 minutes prior to strychnine infusion produced an increase of 13.92%, 25.73% and 17.15% respectively in strychnine co

BACKGROUND: Strychnine is a highly toxic, plant-derived alkaloid. It has historically been used in western Canada as a rodenticide for control of Richardson's ground squirrels; and in Alberta, as a predacide against wolves, coyotes, black bears, and skunks. However, nontarget species poisonings with strychnine have been widely documented in Canada and elsewhere. Poisonings manifest as acute neuroexcitation and are usually fatal. In March 2023, strychnine was banned in Canada for control of Richardson's ground squirrels, with concerns over wildlife poisonings cited. However, fatal poisonings ha

INTRODUCTION: Acute promyelocytic leukemia (APL) is defined by the PML-RARα fusion gene, which serves as a key biomarker for pathogenesis, diagnosis, and measurable residual disease (MRD) monitoring. The incorporation of all-trans retinoic acid (ATRA) and arsenic trioxide (ATO) has markedly transformed treatment outcomes. However, treatment-related toxicities frequently lead to treatment interruptions that may lead to persistent MRD post-induction therapy. This study investigates whether variations in doses are associated with persistent MRD. Additionally, statistical modelling is utilized to

1. Drugs were applied to the cerebral cortex of rats anaesthetized with pentobarbitone and changes measured in the somatosensory primary evoked response.2. Computer-derived averages of thirty-two consecutive responses yielded stable and consistent measurements of the potential changes comprising the evoked response, and comparable records from the opposite (non-drug treated) cortex provided an essential control for systemic actions of the drug.3. The modifications produced by curare and strychnine were indistinguishable. The first positive wave (peak latency 7 msec) was unaltered; the second p

A simple, sensitive and selective liquid chromatography-electrospray mass spectrometric (LC-ESI-MS) method was developed and validated for simultaneous determination of strychnine and brucine in rat plasma, using tacrine as the internal standard (IS). Sample preparation involved a liquid-liquid extraction of the analytes with n-hexane, dichloromethane and isopropanol (65:30:5, v/v/v) from 0.1mL of plasma. Chromatographic separation was carried out on a Waters C(18) column using a mobile phase of methanol-20mM ammonium formate-formic acid (32:68:0.68, v/v/v). Positive selected ion monitoring mo

INTRODUCTION: Plants of the Strychnos genus, which include about 200 species, are used for multiple traditional purposes as hunting poison, for example, and have shown interesting pharmacological properties, especially curarizing and tetanizing, but also against malaria. Many monoterpene indole alkaloids have already been isolated and identified. Among them, there is strychnine, a famous alkaloid that can cause death by asphyxiation.
OBJECTIVE: Investigate alkaloidic molecular diversity from Strychnos genus using molecular networking technique and study the Strychnos genus from a chemotaxonom

The ketogenic diet (KD) is a cheap and effective alternative therapy for most epilepsy. There are paucity of experimental data in Nigeria on the usefulness of KD in epilepsy models. This is likely to be responsible for the poor clinical acceptability of the diet in the country. This study therefore aimed at providing experimental data on usefulness of KD on seizure models. The study used 64 Wistar rats that were divided into two dietary groups [normal diet (ND) and ketogenic diet (KD)]. Animal in each group were fed for 35days. Medium chain triglyceride ketogenic diet (MCT-KD) was used and it

Strychnine is a neurotoxin and an active ingredient in some rodenticides which are placed in burrows to suppress pocket gopher (Thomomys talpoides) populations in range and crop land in western North America. The population level impact was modelled of the use of strychnine-based rodenticides on a non-target snake species, the Great Basin Gophersnake (Pituophis catenifer deserticola), which is a predator of pocket gopher and a Species at Risk in Canada. Using information on population density, demographics, and movement and habitat suitability for the Gophersnake living in an agricultural vall
