[Prospective study of the transit time in intestinal neuronal abnormalities].

In a prospective study 106 children with intestinal neuronal malformations underwent intestinal transit-time studies. In only 50% of the children with intestinal neuronal dysplasia type B or immature ganglia was the transit time prolonged. On the contrary, hypoganglionosis and heterotopia of the submucous plexus led to severe transport disorders with subsequent bowel resection.

Sigmoid volvulus is associated with a decrease in enteric plexuses and ganglion cells: a case-control study.

PURPOSE: Although sigmoid volvulus (SV) causes acute obstruction, its pathogenesis and mechanism of torsion are unknown, and few reports have described its pathological findings. Here, we evaluated the clinicopathological characteristics of volvulus and factors contributing to volvulus of the sigmoid colon. METHODS: We compared 14 patients with SV (10 men and 4 women; median age, 78.5 years) with 14 age- and sex-matched control patients for differences in clinical characteristics, focusing on dysmotility (enteric visceral myopathy, neuropathy, and mesenchymopathy). RESULTS: Of the 14 SV pati

Changes in pituitary adenylate cyclase-activating Peptide 27-like immunoreactive nervous structures in the porcine descending colon during selected pathological processes.

This study reports on changes in the pituitary adenylate cyclase-activating peptide 27-like immunoreactive (PACAP-27-LI) nerve structures of the enteric nervous system (ENS) in the porcine descending colon, caused by chemically induced inflammation, nerve injury, and proliferative enteropathy (PE), which is a "natural" inflammation of the porcine digestive tract. The distribution pattern of PACAP-27-LI structures was studied using the immunofluorescence technique in the circular muscle layer, enteric plexuses (i.e., myenteric plexus (MP), outer submucous plexus (OSP), and inner submucous plexu

The development of avian enteric nervous system: distribution of artemin immunoreactivity.

Among the factors that control neural crest cell precursors within the enteric nervous system, the ligands of the glial cell line-derived neurotrophic factor family (GFL) seem to be the most influential. Artemin, a member of the GFLs, was previously described only in the oesophagus and stomach of mouse embryos. In this study, the presence and distribution of artemin is reported in duck embryos and adults. Artemin immunoreactivity was apparent in the intestinal tract at embryonic day 7 (d7), firstly in the myenteric plexus and then in the submucous plexus. Later, artemin immunoreactive nerve fi

Distribution of substance P-like immunoreactive nervous structures in the duck gut during development.

The ontogeny and distribution of nerve cell bodies and fibers which are substance P-like (SPL) immunoreactive have been studied in the duck gastrointestinal tract by means of immunohistochemistry. The appearance of SPL immunoreactivity was observed in cell bodies of the oesophagus, proventriculus, gizzard and antrum region at 6 days of incubation. At 9 days of incubation SPL immunoreactive nervous structures were observed in the whole length of the intestine. In each area of the gut, SPL immunoreactivity was initially observed in cell bodies of the myenteric plexus and then of the submucous pl

Region-dependent effects of diabetes and insulin-replacement on neuronal nitric oxide synthase- and heme oxygenase-immunoreactive submucous neurons.

AIM: To investigate the intestinal segment-specific effects of diabetes and insulin replacement on the density of different subpopulations of submucous neurons. METHODS: Ten weeks after the onset of type 1 diabetes samples were taken from the duodenum, ileum and colon of streptozotocin-induce diabetic, insulin-treated diabetic and sex- and age-matched control rats. Whole-mount preparations of submucous plexus were prepared from the different gut segments for quantitative fluorescent immunohistochemistry. The following double-immunostainings were performed: neuronal nitric oxide synthase (nNOS

Development of neural receptors for serotonin in the murine bowel.

During the ontogeny of the enteric nervous system, the varicosities of mature neurons contact dividing neural precursors, which persist in the developing murine gut for several weeks postnatally. This phenomenon has led to the hypothesis that the release of transmitter from mature neurons may influence the subsequent development of uncommitted neuroblasts. In order to test this hypothesis, it is necessary to know the timing of the expression of postsynaptic receptors for enteric neurotransmitters. Since serotoninergic neurons are among the earliest of enteric neurons to develop (appearing on d

[The hypoganglionic and aganglionic high pressure zone of the anterior esophagus (the esophageal opening) and its special blood supply (angiomuscular sphincter closure].

At the mouth of the oesophagus there is an aganglionic zone similar to that in the anorectal organ of continence. This is part of the system of permanent closure. Since the musculature at the oesophageal entrance is arranged in a screw-like fashion the aganglionic zone lies obliquely to the longitudinal axis of the oesophagus. Closure at the oesophageal entrance is further supported by a kind of corpus cavernosum similar to that in the rectum. In this pharyngeal corpus cavernosum blood is drained between the muscular fibres and their contraction prevents its drainage, thus facilitating the clo

The enteric nervous system undergoes significant chemical and synaptic maturation during adolescence in mice.

Adolescence is a critical period of development. It is very likely that there is significant maturation of the enteric nervous system (ENS) of the gut during this stage of life, especially since there are substantial changes in factors known to influence the ENS including diet and microbiota during this time, but this remains unknown. To examine maturation of the ENS during adolescence, we performed immunohistochemistry using advanced microscopy and analytical methods to compare enteric neurons and glia of the duodenum and colon of mice taken prior to weaning with those of young adult mice. We

Mianserin blocks alpha 2 adrenoceptors in submucous neurones of the guinea-pig caecum.

Intracellular recordings were made from submucous plexus neurones of the guinea-pig caecum in vitro. The peak amplitude of the adrenergic inhibitory postsynaptic potential (IPSP) was depressed by mianserin in a dose-dependent manner (300 nM-100 microM). This was due to a direct blockade of postsynaptic alpha 2 adrenoceptors. The nicotinic excitatory postsynaptic potential (EPSP) and the non-cholinergic EPSP were not affected by mianserin (100 microM). The presynaptic inhibition of the release of acetylcholine, mediated by presynaptic alpha 2 receptors, was also blocked by mianserin (30 microM)

Isolation of Myenteric and Submucosal Plexus from Mouse Gastrointestinal Tract and Subsequent Co-Culture with Small Intestinal Organoids.

Intestinal homeostasis results from the proper interplay among epithelial cells, the enteric nervous system (ENS), interstitial cells of Cajal (ICCs), smooth muscle cells, the immune system, and the microbiota. The disruption of this balance underpins the onset of gastrointestinal-related diseases. The scarcity of models replicating the intricate interplay between the ENS and the intestinal epithelium highlights the imperative for developing novel methods. We have pioneered a sophisticated tridimensional in vitro technique, coculturing small intestinal organoids with myenteric and submucosal n

Bugs, guts, and glia: how microbiota influence enteric gliogenesis and migration.

Enteric glia are neural crest derivatives. Kabouridis et al. (2015) now show in adult animals that new glia arise within the ganglia of enteric plexuses and then migrate centripetally to colonize the mucosa. Remarkably, enteric microbiota regulate this critical migration.

Distribution, quantification, and characterization of substance P enteric neurons in the submucosal and myenteric plexuses of the porcine colon.

The pig is an important translational model for studying intestinal physiology and disorders for its many homologies with humans, including the organization of the enteric nervous system (ENS), the major regulator of gastrointestinal functions. This study focused on the quantification and neurochemical characterization of substance P (SP) neurons in the pig ascending (AC) and descending colon (DC) in wholemount preparations of the inner submucosal plexus (ISP), outer submucosal plexus (OSP), and myenteric plexus (MP). We used antibodies for the pan-neuronal marker HuCD, and choline acetyltrans

[Quantitative and qualitative studies of the Auerbach and Meissner plexuses of the esophagus in dogs inoculated with Trypanosoma cruzi].

A quantitative and qualitative study was conducted on the Auerbach and Meissner plexuses of the esophagus of four chagasic dogs sacrificed during the acute phase of infection. Ganglionitis and periganglionitis of the Auerbach plexus ranged from mild to moderate and induced significant neuronal lesions, especially in two animals. The ganglions of the Meissner plexus were observed in small number which did not permit any analysis. Mild or moderate myositis was observed mainly in the lower third of the esophagus and was rarely associated with amastigote nests. Ganglion and neuron counts did not d

The neuropathological diagnosis of neuronal intestinal dysplasia (NID B).

Between 1986 and 1991 773 infants were investigated by biopsy. 209 children suffered from a neuronal dysplasia of the submucous plexus (NID B). 64 of these 209 cases had concomitant Hirschsprung's disease with NID. The combination of Hirschsprung's disease with NID was established at biopsy not earlier than at 12 +/- 6 months of age. The classical form of an isolated aganglionosis had a median age at diagnosis of 4 +/- 2 months. The preconditions for a reliable diagnosis of NID are mucosal biopsies with submucosa taken 1, 3 and 9 cm above the pectinate line, the preparation of 15 microns thick

Expression of Cocaine and Amphetamine Regulated Transcript (CART) in the Porcine Intramural Neurons of Stomach in the Course of Experimentally Induced Diabetes Mellitus.

In the present study, the effect of streptozotocin-induced diabetes on the cocaine- and amphetamine-regulated transcript-like immunoreactive (CART-LI) enteric nervous structures was investigated within the porcine stomach. To induce diabetes, the pigs were administered intravenously streptozotocin at a dose of 150 mg/kg of body weight. A significant decrease of the number of CART-LI perikarya was observed in the myenteric plexus of the gastric antrum, corpus, and pylorus in the experimental group. In contrast, submucous plexus was devoid of CART-positive neuronal cells both in control and expe

Protocol to culture enteric glial cells from the submucosal and myenteric plexi of neonatal and juvenile pig colons.

Here, we present our protocol to culture enteric glial cells from the submucosal and myenteric plexus of neonatal and juvenile pig colons. We describe steps for colon isolation, microdissection, and enzymatic and mechanical dissociation. We include procedures for passaging and analyzing cell yield, freeze/thaw efficiency, and purity. This protocol allows for the generation of primary cultures of enteric glial cells from single-cell suspensions of microdissected layers of the colon wall and can be used to culture enteric glia from human colon specimens. 5xFAD mice do not have myenteric amyloidosis, dysregulation of neuromuscular transmission or gastrointestinal dysmotility.

BACKGROUND: Alterations in gastrointestinal (GI) function and the gut-brain axis are associated with progression and pathology of Alzheimer's Disease (AD). Studies in AD animal models show that changes in the gut microbiome and inflammatory markers can contribute to AD development in the central nervous system (CNS). Amyloid-beta (Aβ) accumulation is a major AD pathology causing synaptic dysfunction and neuronal death. Current knowledge of the pathophysiology of AD in enteric neurons is limited, and whether Aβ accumulation directly disrupts enteric neuron function is unknown. METHODS: In 6-mo
