Learning-induced synaptic potentiation in implanted neural precursor cell-derived neurons.

Neuronal loss caused by neurodegenerative diseases, traumatic brain injury and stroke results in cognitive dysfunctioning. Implantation of neural stem/precursor cells (NPCs) can improve the brain function by replacing lost neurons. Proper synaptic integration following neuronal differentiation of implanted cells is believed to be a prerequisite for the functional recovery. In the present study, we characterized the functional properties of immortalized neural progenitor HiB5 cells implanted into the rat hippocampus with chemically induced lesion. The implanted HiB5 cells migrated toward CA1 py

Reversing cocaine-induced synaptic potentiation provides enduring protection from relapse.

Cocaine addiction remains without an effective pharmacotherapy and is characterized by an inability of addicts to inhibit relapse to drug use. Vulnerability to relapse arises from an enduring impairment in cognitive control of motivated behavior, manifested in part by dysregulated synaptic potentiation and extracellular glutamate homeostasis in the projection from the prefrontal cortex to the nucleus accumbens. Here we show in rats trained to self-administer cocaine that the enduring cocaine-induced changes in synaptic potentiation and glutamate homeostasis are mechanistically linked through g

Long-lasting facilitation of a synaptic potential following tetanization in the in vitro hippocampal slice.

Field potentials evoked by stimulation of afferent fibers in stratum radiatum were recorded in the CA1 region of the hippocampal slice maintained in vitro. Stimulation rates of 3-50/sec produced a large increase in amplitude of the population spike in CA1. This increase was maintained for several hours after the tetanization. The facilitation phenomenon appeared to be specific to the synapse of stratum radiatum afferents onto CA1 pyramidal cells since: (1) stimulation outside the radiatum layer did not produce the effect, (2) antidromic field potentials recorded in CA3 were unchanged, (3) EPSP

Mechanisms of gastric rhythm generation in the isolated stomatogastric ganglion of spiny lobsters: bursting pacemaker potentials, synaptic interactions, and muscarinic modulation.

1. The gastric central pattern generator (CPG), located in the stomatogastric ganglion (STG) of the spiny lobster (Panulirus interruptus), is nonrhythmic when deprived of neuromodulatory inputs from anterior ganglia. Leaving these inputs intact in vitro can sustain a gastric rhythm but also introduces numerous, uncontrolled and largely unknown modulatory and synaptic influences that greatly complicate analysis of this CPG. 2. Here we induced gastric rhythms in the isolated STG, by superfusing a specific modulator, the muscarinic agonist, pilocarpine. Muscarinic agents sustain vigorous gastric

Auditory brain-stem (ABP) and somatosensory evoked potentials (SEP) in an animal model of a synaptic lesion: elevated plasma barbiturate levels.

Experiments were conducted to determine whether a consistent pattern of auditory nerve brain-stem evoked synaptic lesion model in cats (elevated levels of the lesion model in cats (elevated levels of the barbiturate thiopental). The ABP in response to low (10/sec) and high (80/sec) stimulus rates was recorded. In order to differentiate between the effects of the elevated drug levels on axonal propagation and on synaptic transmission, the early components of the somatosensory evoked potential (SEP) were also recorded, with particular attention to the first SEP wave, which is solely an axonal ev

PTPRO Governs Synaptic Density, Long-Term Potentiation and Cognitive Function In Vivo.

Protein Tyrosine Phosphatase Receptor Type O (PTPRO) is a synaptic cell adhesion molecule that is essential for synapse formation in vitro; however, its in vivo role remains unclear. Using PTPRO-knockout mice, we found that PTPRO deletion reduced the number of synapses and impaired both excitatory and inhibitory synaptic transmission. PTPRO knockout also disrupted hippocampal long-term potentiation and caused learning and memory deficits in the water maze as well as in fear conditioning. Notably, overexpression of the N-terminal extracellular domain of PTPRO rescued the frequencies of miniatur

The potential role of aberrant microglial synaptic pruning in the neurodevelopmental pathogenesis of tourette syndrome.

Tourette syndrome is a neurodevelopmental disorder traditionally attributed to dopamine system hyperactivity within the cortico-striato-thalamo-cortical circuitry. However, classical neurotransmitter hypotheses fail to fully explain the spatiotemporal and developmental specificities of the disorder. Consequently, research focus has shifted toward the neuroimmune microenvironment, specifically the role of microglia. This review aims to comprehensively explore the potential mechanisms of microglia-mediated aberrant synaptic pruning in the pathogenesis of tourette syndrome and to evaluate emergin

Ceftriaxone Treatment Weakens Long-Term Synaptic Potentiation in the Hippocampus of Young Rats.

Disrupted glutamate clearance in the synaptic cleft leads to synaptic dysfunction and neurological diseases. Decreased glutamate removal from the synaptic cleft is known to cause excitotoxicity. Data on the physiological effects of increased glutamate clearance are contradictory. This study investigated the consequences of ceftriaxone (CTX), an enhancer of glutamate transporter 1 expression, treatment on long-term synaptic potentiation (LTP) in the hippocampus of young rats. In this study, 5-day administration of CTX (200 mg/kg) significantly weakened LTP in CA3-CA1 synapses. As shown by elect

Apolipoprotein E in synaptic plasticity and Alzheimer's disease: potential cellular and molecular mechanisms.

Alzheimer's disease (AD) is clinically characterized with progressive memory loss and cognitive decline. Synaptic dysfunction is an early pathological feature that occurs prior to neurodegeneration and memory dysfunction. Mounting evidence suggests that aggregation of amyloid-β (Aβ) and hyperphosphorylated tau leads to synaptic deficits and neurodegeneration, thereby to memory loss. Among the established genetic risk factors for AD, the ɛ4 allele of apolipoprotein E (APOE) is the strongest genetic risk factor. We and others previously demonstrated that apoE regulates Aβ aggregation and clearan

Bilirubin potentiates inhibitory synaptic transmission in lateral superior olive neurons of the rat.

Bilirubin is a well-known neurotoxin that can result in multiple neurologic deficits. Previous studies have suggested that bilirubin affects aspects of synaptic transmission; however the acute effects of bilirubin on synaptic transmission have not been examined in real-time. In this study, using whole-cell voltage-clamp recordings, we observed the effect of bilirubin on inhibitory postsynaptic currents (IPSC) in postnatal 13-15-day-old neurons dissociated from lateral superior olive nuclei (LSO), one of the brainstem auditory nucleus that are highly vulnerable to bilirubin. The results showed

Long-term potentiation of synaptic transmission in the hippocampus induced by a bee venom peptide.

Several neurotoxins have been isolated from bee venom. One of these, the mast cell degranulating peptide (MCD), releases histamine from mast cells and on central administration produces arousal at low concentrations and convulsions at higher doses. These effects are mediated through specific high-affinity binding sites which are concentrated in cortical structures, notably the hippocampus. This structure appears to be the source of changes in the electrocorticogram that follow injections of MCD into the cerebral ventricle, and which induce a quasi-permanent hippocampal theta rhythm in the moti

A systematic review of ketamine and esketamine-induced long-term potentiation and synaptic scaling: Do the molecular and synaptic plasticity effects inform dosing intervals?

Ketamine and esketamine exhibit rapid antidepressant effects in persons with treatment-resistant depression (TRD) and bipolar depression (TRBD). However, the synaptic mechanisms governing dose, frequency, and durability of response remain unclear. This review aims to evaluate the dosing parameters, including minimum dose and optimal dosing intervals, that reliably induce or maintain long-term potentiation (LTP) and/or synaptic scaling in humans and preclinical models. A systematic search (database inception to June 2025) was conducted on OVID and PubMed to identify preclinical and clinical stu

Boosting of synaptic potentials and spine Ca transients by the peptide toxin SNX-482 requires alpha-1E-encoded voltage-gated Ca channels.

The majority of glutamatergic synapses formed onto principal neurons of the mammalian central nervous system are associated with dendritic spines. Spines are tiny protuberances that house the proteins that mediate the response of the postsynaptic cell to the presynaptic release of glutamate. Postsynaptic signals are regulated by an ion channel signaling cascade that is active in individual dendritic spines and involves voltage-gated calcium (Ca) channels, small conductance (SK)-type Ca-activated potassium channels, and NMDA-type glutamate receptors. Pharmacological studies using the toxin SNX-

Contribution of a calcium-activated non-specific conductance to NMDA receptor-mediated synaptic potentials in granule cells of the frog olfactory bulb.

We studied granule cells (GCs) in the intact frog olfactory bulb (OB) by combining whole-cell recordings and functional two-photon Ca(2+) imaging in an in vitro nose-brain preparation. GCs are local interneurones that shape OB output via distributed dendrodendritic inhibition of OB projection neurones, the mitral-tufted cells (MTCs). In contrast to MTCs, GCs exhibited a Ca(2+)-activated non-specific cation conductance (I(CAN)) that could be evoked through strong synaptic stimulation or suprathreshold current injection. Photolysis of the caged Ca(2+) chelator o-nitrophenol-EGTA resulted in acti

[The generation of nonadrenergic inhibiting synaptic potentials in the smooth muscles of the gastrointestinal tract by substituting cesium ions for potassium ions].

Nonadrenergic inhibitory junction potentials (IJPs) evoked by intramural stimulation were investigated in smooth muscle of guinea pig stomach, caecum and colon by means of sucrose-gap method. IJPs disappeared in the smooth muscle preexposed to K-free Krebs solution for 4-9 h and restored by addition 6mM Cs+. The amplitude of IJPs was half as much as one in normal conditions but the latency and duration were significantly prolonged. In most cases apamin blocks IJPs in these muscles. The results presented suggest that IJPs generation is due to Cs+ ions permeated through Ca(2+)-activated apamin-s

[AMPA receptors: molecular mechanisms of synaptic plasticity and the potential of Ampasse in addressing cognitive impairment].

Dysfunction of the glutamatergic system, specifically involving AMPA receptors, is recognized as a significant pathogenic mechanism underlying cognitive impairment. AMPA glutamate receptors mediate fast excitatory postsynaptic currents in the majority of brain synapses and play a crucial role in the processes associated with long-term potentiation, which is integral to memory function. Additional mechanisms of synaptic plasticity linked to AMPA receptors include long-term depression of synaptic transmission, homeostatic plasticity, and AMPA receptor trafficking. Impaired synaptic transmission and long-term potentiation in severe combined immunodeficient (SCID) mice.

DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is one of the key enzymes involved in DNA double-strand break (DSB) repair. However, recent studies using DNA-PKcs knockout mice revealed that DNA-PKcs plays an important role in neuronal plasticity. The aim of this study was to examine the role of DNA-PKcs on synaptic plasticity in severe combined immunodeficiency disease (SCID) mice that carry a mutation resulting in a DNA-PKcs protein devoid of kinase activity but still expressed in cells, although with a small COOH-terminal truncation. To this aim, we carried out electrophysiologica

Potential role of Bcl2 in lipid metabolism and synaptic dysfunction of age-related hearing loss.

Age-related hearing loss (ARHL) is a prevalent condition affecting millions of individuals globally. This study investigated the role of the cell survival regulator Bcl2 in ARHL through in vitro and in vivo experiments and metabolomics analysis. The results showed that the lack of Bcl2 in the auditory cortex affects lipid metabolism, resulting in reduced synaptic function and neurodegeneration. Immunohistochemical analysis demonstrated enrichment of Bcl2 in specific areas of the auditory cortex, including the secondary auditory cortex, dorsal and ventral areas, and primary somatosensory cortex
