Archaerhodopsin Selectively and Reversibly Silences Synaptic Transmission through Altered pH.

Tools that allow acute and selective silencing of synaptic transmission in vivo would be invaluable for understanding the synaptic basis of specific behaviors. Here, we show that presynaptic expression of the proton pump archaerhodopsin enables robust, selective, and reversible optogenetic synaptic silencing with rapid onset and offset. Two-photon fluorescence imaging revealed that this effect is accompanied by a transient increase in pH restricted to archaerhodopsin-expressing boutons. Crucially, clamping intracellular pH abolished synaptic silencing without affecting the archaerhodopsin-medi

Muscarinic receptor regulation of synaptic transmission in airway parasympathetic ganglia.

Muscarinic receptor regulation of synaptic transmission in guinea pig bronchial parasympathetic ganglia was evaluated with the use of intracellular recording of the intrinsic ganglion neurons. Methacholine (1 microM) decreased the amplitude of vagus nerve-stimulated fast excitatory postsynaptic potentials (fEPSP) by 33 and 46% (at 0.8 and 8.0 Hz, respectively) but had no effect on the amplitude of the depolarizations evoked by a bath-applied nicotinic receptor agonist. Methoctramine (1 microM) inhibited methacholine's effect on fEPSP but alone did not influence the magnitude of the fEPSP evoke

Synaptic transmission in submucosal ganglia of guinea pig distal colon.

Intracellular electrical recording was used to investigate synaptic behavior of ganglion cells in the neural networks of the submucosal plexus of the guinea pig distal colon. Fast excitatory postsynaptic potentials (EPSPs), mediated by nicotinic receptors, were found in all S/type 1 neurons, 70% of AH/type 2, 75% of type 3, and 95% of type 4 neurons. Slow EPSPs were characterized by membrane depolarization, increased input resistance, enhanced action potential discharge, and suppression of hyperpolarizing afterpotentials in 64% of the S/type 1 neurons, 74% of AH/type 2, 31% of type 3, and 70%

Impact of vesicular glutamate leakage on synaptic transmission at the calyx of Held.

KEY POINTS: It is controversial whether glutamate can leak out of vesicles in the nerve terminal. To address this issue, we abolished vesicular glutamate uptake by washing out presynaptic cytosolic glutamate or by blocking vacuolar ATPase activity using bafilomycin A1. In the absence of vesicular glutamate uptake, both spontaneous and nerve-evoked EPSCs underwent a rundown, suggesting that vesicular glutamate can leak out of vesicles. However, the rundown of evoked EPSCs was caused mainly by accumulation of unfilled vesicles after exocytic release of glutamate, suggesting a minor influence of

Kv1.3 channels regulate synaptic transmission in the nucleus of solitary tract.

The voltage-gated K(+) channel Kv1.3 has been reported to regulate transmitter release in select central and peripheral neurons. In this study, we evaluated its role at the synapse between visceral sensory afferents and secondary neurons in the nucleus of the solitary tract (NTS). We identified mRNA and protein for Kv1.3 in rat nodose ganglia using RT-PCR and Western blot analysis. In immunohistochemical experiments, anti-Kv1.3 immunoreactivity was very strong in internal organelles in the soma of nodose neurons with a weaker distribution near the plasma membrane. Anti-Kv1.3 was also identifie

Nyctalopin is essential for synaptic transmission in the cone dominated zebrafish retina.

The first synapse in the vertebrate visual system is the photoreceptor synapse between rod and cone photoreceptors and the second-order bipolar cells. Although mutations in the nyctalopin gene (NYX) in humans lead to congenital stationary night blindness (CSNB1), affecting synaptic transmission between both types of photoreceptors and ON-bipolar cells, the function of nyctalopin in cone-dominant animal models has not been studied. Because the larval zebrafish retina is cone-dominant, we isolated the zebrafish nyx ortholog and raised a polyclonal antibody against the protein. Nyctalopin is expr

Cholinergic modulation of excitatory synaptic transmission in the CA3 area of the hippocampus.

Cholinergic innervation of the hippocampus has been implicated in memory formation and retrieval. Here we study cholinergic modulation of excitatory transmission in the CA3 area of the rat hippocampus. We used a combination of optical measurements of presynaptic calcium and electrophysiological measurements of synaptic currents to study associational-commissural (A/C) and mossy fiber (MF) synapses in brain slices. Direct synaptic modulation mediated by ACh receptors is only evident at the A/C synapse, where synaptic inhibition primarily reflects presynaptic calcium channel inhibition mediated

Long-term depression of excitatory synaptic transmission in the rat amygdala.

In view of the fact that both kindling and fear-potentiated startle are expressed by long-term enhancement of synaptic transmission in the amygdala, synaptic plasticity in this area of the brain is of particular importance. Here, we show for the first time that low-frequency stimulation of the lateral nucleus at 1 Hz for 15 min elicited a long-term depression (LTD) in the basolateral amygdala (BLA) neurons. LTD is expressed specifically at the lateral-BLA synapses but not at ventral endopyriform nucleus-BLA synapses. The induction of LTD requires activation of both NMDA and metabotropic glutam

A model of graded synaptic transmission for use in dynamic network simulations.

1. The heartbeat central pattern-generating network of the medicinal leech contains elemental neural oscillators, comprising reciprocally inhibitory pairs of segmental heart interneurons, that use graded as well as spike-mediated synaptic transmission. We are in the process of developing a general computer model of this pattern generator. Our modeling goal is to explore the interaction of membrane currents and synaptic transmission that promote oscillation in heart interneurons. As a first step toward this goal, we have developed a computer model of graded synaptic transmission between recipro

Biochemical plasticity of synaptic transmission: a critical review of Dale's Principle.

"Dale's Principle" states that each neuron releases one and only one synaptic transmitter. Mental disorders and behavioral drug effects are attributed to activation or blockade of one or more of these specific transmitters. A series of biochemical, electrophysiological, and behavioral studies suggests the alternative view that at each monoaminergic synapse the action of the transmitter is modulated by several metabolically related substances: amine analogs (2-phenylethylamine [PEA], p-tyramine, etc.), deaminated products (aldehydes, acids, and alcohols), and possibly also amino acid precursors

Perinatal nutritional iron deficiency reduces hippocampal synaptic transmission but does not impair short- or long-term synaptic plasticity.

Studies show that perinatal nutritional iron deficiency (ID) produces learning and memory impairments in humans and animals. This suggests that the functional physiology of learning and cognitive centers in the brain, such as the hippocampus, may be compromised by developmental ID. The present study used electrophysiological brain slice methods to examine multiple measures of hippocampal synaptic efficacy from rats that were subjected to perinatal ID diets or control (CN) diets. Measures of synaptic efficacy were obtained from the first and last synaptic regions of the hippocampal tri-synaptic

Impact of Elovl5 Deficiency on Cerebellar Excitatory Synaptic Transmission in Mice.

The neurotransmitter release and the synaptic vesicle cycle require a specific lipidic composition of presynaptic and vesicle membranes. Phospholipids with long-chain acyl groups are necessary to confer to membranes the physical properties necessary for synaptic transmission. Elovl5 is crucial for the elongation of polyunsaturated fatty acids (PUFAs) beyond 18-carbon atoms, and its deletion or mutation causes cerebellar motor deficits in humans and mice. In the mouse cerebellum of both sexes, deletion of Elovl5 increased 18- and 20-carbon atoms PUFAs, decreased long-chain PUFAs, and increased

Characterization of excitatory synaptic transmission in the retrosplenial cortex of adult mice.

The retrosplenial cortex (RSC) plays an important role in navigation, memory and pain. However, there are few studies on excitatory synaptic transmission in the RSC. Here, we used a multi-electrode array recording system (MED64) to study the characteristics of excitatory synaptic transmission in the RSC and the contribution of different types of voltage-gated Ca2+ channels (VGCCs) in excitatory synaptic transmission. We found that glutamate is the major excitatory transmitter for RSC, and postsynaptic alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors contribute to excit

Role of LGI1 protein in synaptic transmission: From physiology to pathology.

Leucine-Rich Glioma Inactivated protein 1 (LGI1) is a secreted neuronal protein highly expressed in the central nervous system and high amount are found in the hippocampus. An alteration of its function has been described in few families of patients with autosomal dominant temporal lobe epilepsy (ADLTE) or with autoimmune limbic encephalitis (LE), both characterized by epileptic seizures. Studies have shown that LGI1 plays an essential role during development, but also in neuronal excitability through an action on voltage-gated potassium Kv1.1 channels, and in synaptic transmission by regulati

Synaptic transmission in the auditory brainstem of normal and congenitally deaf mice.

The deafness (dn/dn) mutant mouse provides a valuable model of human congenital deafness. We investigated the properties of synaptic transmission in the anteroventral cochlear nucleus (AVCN) of normal and congenitally deaf dn/dn mice. Excitatory postsynaptic currents (EPSCs) were evoked by focal stimulation of single auditory nerve fibres, and measured by whole-cell recordings from neurones in AVCN slices (mean postnatal age = P13). Absolute amplitudes of both AMPA- and NMDA-mediated components of evoked EPSCs were greater (170 %) in deaf versus control animals. Enhanced transmission in deaf m

Nitric oxide in the afferent synaptic transmission of the axolotl vestibular system.

This study was performed using intracellular and multiunit extracellular recording techniques in order to characterize the role of nitric oxide in the afferent synaptic transmission of the vestibular system of the axolotl (Ambystoma tigrinum). Bath application of nitric oxide synthase inhibitors N(G)-nitro-L-arginine (0.01microM to 10microM) and N-nitro-L-arginine methyl ester hydrochloride (0.1microM to 1000microM) elicited a dose-dependent decrease in the basal discharge of the semicircular canal afferent fibers. N(G)-Nitro-L-arginine also diminished the response to mechanical stimuli. Moreo

[Electrophysiological description of synaptic transmission and its modulation by pharmacologic factors].

Synaptic transmission between neurons plays a key role in signal transduction in the central nervous system (CNS). Postsynaptic currents can be routinely recorded using the patch-clamp technique but mechanisms underlying their kinetics are not entirely clear. Model simulations and experimental evidence indicate that the time course of synaptic agonist is extremely fast and short lasting implying that activation of postsynaptic receptors takes place in conditions of extreme non-equilibrium. For this reason, studies of postsynaptic receptors' kinetics requires ultrafast agonist application techn

Enhancement of peptidergic synaptic transmission by catecholamines in the bullfrog sympathetic ganglion.

Extracellular recordings were made from postganglionic branches or ganglion cells of the 9th or 10th ganglia of the paravertebral sympathetic chain of the bullfrog in vitro. Tetanic preganglionic stimulation, applied to nerve fibers between the 7th and 8th ganglia, elicited cholinergic (muscarinic) and non-cholinergic synaptic responses in nicotinized preparations. These were an early after-discharge (EAD) and a late after-discharge (LAD), respectively. Brief application (3-10 min) of catecholamines augmented the LAD and this augmentation continued for 90-120 min after catecholamine withdrawal

Effects of several aminopyridines and analogs on the calcium dependence of synaptic transmission.

The effects of 4-aminopyridine (4-AP), 4-AP methiodide (4-APMI), 3,4-diaminopyridine (3,4-DAP), 4,5-diaminopyrimidine (4,5-DAPM) and 4-dimethylaminopyridine (4-DMAP) on synaptic transmission in isolated frog sympathetic ganglia were examined by means of extracellular recording. Ganglia were exposed to graded reductions in extracellular Ca++, from the normal 1.8 mM in the absence and in the presence of drugs while recording amplitudes of the transmitted postganglionic compound action potential. The amplitudes of compound action potential relative to those in normal Ca++ were then plotted as a f

Electrophysiologic evidence that retinotectal synaptic transmission in the goldfish is nicotinic cholinergic.

A previous study identified, by conduction velocity following optic nerve shock, 3 classes of retinal fibers which project to 3 distinct laminae of the goldfish optic tectum. In the present study, the effect of various pharmacological agents on the synaptic efficacy of each of the 3 classes of retinal fibers was assessed by the use of current source-density analysis. All 3 classes of optic fibers appear to be nicotinic cholinergic. Six different nicotinic antagonists were tested. All 6 were effective in decrementing the responses of all 3 classes to a criterion level: alpha-bungarotoxin (10-8
