Tau hyperphosphorylation in synaptosomes and neuroinflammation are associated with canine cognitive impairment.

Canine cognitive impairment syndrome (CDS) represents a group of symptoms related to the aging of the canine brain. These changes ultimately lead to a decline of memory function and learning abilities, alteration of social interaction, impairment of normal housetraining, and changes in sleep-wake cycle and general activity. We have clinically examined 215 dogs, 28 of which underwent autopsy. With canine brains, we performed extensive analysis of pathological abnormalities characteristic of human Alzheimer's disease and frontotemporal lobar degeneration, including β-amyloid senile plaques, tau

[Phosphorylation of rat brain synaptosomal proteins stimulated by alpha-latrotroxin].

alpha-Latrotoxin (LTX) is a presynaptic neurotoxin from black widow spider (Latrodectus mactans tredecimguttatus) venom; it causes massive Ca(2+)-independent neurotransmitter release. The effect of LTX on phosphorylation of synaptosomal proteins was studied including synapsin I, synaptotagmin, and dynamin. Experiments were performed in Ca(2+)-supplemented medium containing 1 mM CaCl2 or nominally Ca(2+)-free medium (without added Ca2+ and EGTA) using intact synaptosomes isolated from rat brain and prelabeled with 32P(i). The data indicate the exposure of synaptosomes to 10 nM LTX for 10 sec st

Effect of tityustoxin and ouabain on protein phosphorylation in a crude synaptosome fraction.

1. The effect of tityustoxin (TsTX) and ouabain on the incorporation of 32P into a protein of the same apparent molecular weight as synapsin I is described. 2. Tityustoxin-stimulated protein phosphorylation in a crude synaptosome fraction increased up to a concentration of 3.0 microM at an incubation time of 15 s. 3. Trifluoperazine (100 microM) inhibited, while trifluoperazine sulphoxide (100 microM) did not alter the effect on the protein phosphorylation induced by tityustoxin. 4. Unlike tityustoxin, ouabain (100 microM) had no effect on protein phosphorylation even after incubation up to 20

[Cytoskeletal regulation of the sensitivity of brain synaptosomes to the depolarizing action of veratrine].

Using a radioactive permeant cation 3H-tetraphenylphosphonium, the sensitivity of rat brain synaptosomes to depolarizing action of veratrine, which specifically opens the sodium channels, was compared before and after destruction of microtubules and microfilaments. Depolymerization of microtubules with colchicin and vinblastine decreased an apparent affinity of veratrine to its receptor in the channel, while destruction of microfilaments with cytochalasin B had the opposite effect. Colchicine did not change allosteric interactions between the receptor for veratrine and that for scorpion venom

Effects of 1,1,1-trichloroethane on synaptosomal calcium accumulation in mouse brain.

Addition of 1,1,1-trichloroethane (TCE), in vitro to synaptosomes isolated from the mouse cerebellum and cerebral cortex inhibited the slow phase of K+-stimulated 45Ca2+ influx and the net 45Ca2+ influx (delta k). In the brain stem, however, TCE increased the fast and slow phases of calcium uptake under depolarizing conditions and also delta k. The non-depolarized calcium influx was not altered by TCE added in vitro. Two hours after injection of TCE (2.4 g/kg) the calcium accumulation in the presence of high K+ was lowered in the cerebellar synaptosomes, while it was increased in brain stem sy

TPP+ accumulation in rat brain synaptosomes as a probe for Na+ channels.

Tetraphenylphosphonium (TPP+) accumulation in rat brain synaptosomes was measured in the presence of various stimuli. TPP+ accumulation was sensitive to an increase of extracellular K+ or to the presence of Na+ channel neurotoxins. Examination of the effects of drugs with different physico-chemical properties on TPP+ accumulation showed that there was no effect under low and high K+ conditions; pimozide, flunarizine and buterizine were active in the presence of veratridine or scorpion venom. These results were compared to data obtained for the specific binding of [3H]BTX-B, a selective marker

The actions of barbiturates on release of noradrenaline from rat hippocampal synaptosomes.

Barbiturates are believed to work both by augmenting GABA action and independently perhaps by decreasing neurotransmitter release. By studying the effect of pentobarbitates on the release of 3H noradrenaline from rat hippocampal synaptosomes it was found that pentobarbitone (10(-6)M and 10(-5)M) augmented the GABA evoked release of 3H noradrenaline but 10(-4)M depressed this GABA effect. This latter concentration of pentobarbitone also depressed the K+-evoked release of 3H noradrenaline by a Ca++ dependent but picrotoxin insensitive mechanism.

[Effect of cytotoxic hypoxia on the metabolism and ultrastructure of cortical synaptosomes and mitochondria].

The study is concerned with morphochemical changes in the metabolism and ultrastructure of the mitochondria and synaptical endings from the cortex of the hemispheres in rats under the influence of cyanide (7mg/kg) following 30 min, 2 hours and 24 hours after its administration. Subfractions of the light and heavy synaptosomes, as well as in the mitochondria of the cell bodies showed changes in the activity of the enzyme systems (cytochromoxidase, K+, Na+-ATPase and Mg2+-ATPase) and disturbances of the ultrastructure of the eliminated organella. After 24 hours the metabolism normalized, which m

Plantainoside D Reduces Depolarization-Evoked Glutamate Release from Rat Cerebral Cortical Synaptosomes.

Inhibiting the excessive release of glutamate in the brain is emerging as a promising therapeutic option and is efficient for treating neurodegenerative disorders. The aim of this study is to investigate the effect and mechanism of plantainoside D (PD), a phenylenthanoid glycoside isolated from Plantago asiatica L., on glutamate release in rat cerebral cortical nerve terminals (synaptosomes). We observed that PD inhibited the potassium channel blocker 4-aminopyridine (4-AP)-evoked release of glutamate and elevated concentration of cytosolic Ca2+. Using bafilomycin A1 to block glutamate uptake

Inhibition of glutamate release by cilnidipine in rat cerebrocortical nerve terminals (synaptosomes).

Cilnidipine is an antihypertensive drug that was reported to have a neuroprotective profile. The present study aimed to investigate the effect of cilnidipine on the 4-aminopyridine (4-AP)-induced glutamate release in the rat cerebral cortex using isolated nerve terminals (synaptosomes). Cilnidipine reduced the release of glutamate release induced by 4-AP in a concentration-dependent manner. This inhibitory effect was associated with a reduction in the 4-AP-induced intrasynaptosomal Ca concentration elevation and was not because of an alteration of the synaptosomal membrane potential. The inhib

Co-localization of amyloid beta and tau pathology in Alzheimer's disease synaptosomes.

The amyloid cascade hypothesis proposes that amyloid beta (Abeta) pathology precedes and induces tau pathology, but the neuropathological connection between these two lesions has not been demonstrated. We examined the regional distribution and co-localization of Abeta and phosphorylated tau (p-tau) in synaptic terminals of Alzheimer's disease brains. To quantitatively examine large populations of individual synaptic terminals, flow cytometry was used to analyze synaptosomes prepared from cryopreserved Alzheimer's disease tissue. An average 68.4% of synaptic terminals in the Alzheimer's disease

Omega-agatoxins differentially block calcium channels in locust, chick and rat synaptosomes.

Three toxins (omega-Agatoxins IA, IIA and IIIA) isolated from the venom of the funnel web spider, Agelenopsis aperta, differentially block depolarization-induced calcium influx in chick, rat and locust synaptosomes. In chick, this block of calcium influx is observed with omega-Agatoxins IIA and IIIA but not with omega-Agatoxin IA. Block by omega-Agatoxin IIA and IIIA is maximal at 70 and 82% respectively of the total depolarization-induced calcium influx; maximal suppression of calcium influx by omega-Conotoxin GVIA (omega-CgTx) is 100%. The IC50 for block with omega-Agatoxin IIA is ca 3 nM as

Microwave induced stimulation of 32Pi incorporation into phosphoinositides of rat brain synaptosomes.

Exposure of synaptosomes to microwave radiation at a power density of 10 mW/sq cm or more produced stimulation of the 32Pi-incorporation into phosphoinositides. The extent of 32Pi incorporation was found to be much more pronounced in phosphatidylinositol-4-phosphate (PIP), and phosphatidylinositol-4,5-bisphosphate (PIP2) as compared to phosphatidylinositol (PI) and phosphatidic acid (PA). Other lipids were also found to incorporate 32Pi but no significant changes in their labeling were seen after exposure to microwave radiation. Inclusion of 10 mM lithium in the medium reduced the basal labeli

A spider toxin (JSTX) inhibits L-glutamate uptake by rat brain synaptosomes.

Joro spider toxin (JSTX), a specific blocker of glutamate receptors, was found to exert a prominent suppressive action on the Na+-dependent binding of L-glutamate to synaptic membranes and on glutamate uptake by synaptosomes in a dose-dependent manner. In contrast, the synthesized 2,4-dihydroxyphenylacetylasparagine (2,4-DHPA-ASN), a common moiety of spider toxins, which has been shown to exhibit almost the same activity as intact JSTX with respect to the inhibition of Na+-independent glutamate binding to its synaptic membrane receptors, shows lower potency in inhibiting Na+-dependent binding

Phenothiazines and haloperidol block Ca-activated K channels in rat forebrain synaptosomes.

Ca-dependent, K-stimulated 86Rb efflux, a measure of Ca-activated K conductance in rat brain synaptosomes, was blocked by phenothiazines and haloperidol. Micromolar concentrations of the phenothiazines, fluphenazine and trifluoperazine, and haloperidol, a non-phenothiazine antipsychotic and calmodulin antagonist, selectively inhibited the Ca-activated K channels. The IC50 values of all three agents for inhibition of the Ca-activated K channels was on the order of 0.5-1 microM. Measurements of K-stimulated 45Ca uptake indicated that the effects of these agents on Ca-activated K channels was not

Ionizing radiation alters the properties of sodium channels in rat brain synaptosomes.

The effect of ionizing radiation on neuronal membrane function was assessed by measurement of neurotoxin-stimulated 22Na+ uptake by rat brain synaptosomes. High-energy electrons and gamma photons were equally effective in reducing the maximal uptake of 22Na+ with no significant change in the affinity of veratridine for its binding site in the channel. Ionizing radiation reduced the veratridine-stimulated uptake at the earliest times measured (3 and 5 s), when the rate of uptake was greatest. Batrachotoxin-stimulated 22Na+ uptake was less sensitive to inhibition by radiation. The binding of [3H

Inhibition by neuroleptics of uptake of 3H-GABA into rat brain synaptosomes.

Neuroleptics from various chemical groups and some amino acids were examined for GABA uptake-inhibiting properties in rat brain synaptosomes. The phenothiazine and thioxanthene neuroleptics inhibited GABA uptake by 50% in the concentration range of 10-30 micron. No difference in potency was found between the cis and the trans isomers of the thioxanthenes. Clozapine and particularly sulpiride were weak inhibitors of the GABA uptake process, whereas some of the butyrophenones examined exhibited rather potent effect. Secific GABA uptake inhibitors (nipecotic acid and guvacine) were as active as t

Distribution and properties of NADPH-linked aldehyde reductases from rat brain synaptosomes.

Studies on the subcellular distribution of NADPH-linked aldehyde reductase from rat brain showed that 10% of the total reductase activity is located in the mitochondrial-synaptosomal fraction. There are differences in the percentages of reductase activity found in the synaptosomes compared to cytosol in various regions of the brain. The NADPH-linked aldehyde reductase from the synaptosomal fraction exhibited a nonlinear Lineweaver-Burk plot. This nonlinearity is due to the presence of two distinct aldehyde reductases, which can be distinguished by Michealis constants forp-nitrobenzaldehyde of
