Soluble inflammatory mediators of synoviocytes stimulated by monosodium urate crystals induce the production of oxidative stress, pain, and inflammation mediators in chondrocytes : Secretome of synoviocytes induces chondrocyte damage.

We hypothesized that the secretion of inflammatory mediators from synoviocytes affects the chondrocyte homeostasis of articular cartilage. This study was a preliminary attempt to elucidate the molecular mechanisms by which soluble mediators obtained from activated synoviocytes induce oxidative stress and inflammation in chondrocytes. We measured the concentrations of interleukin-6 (IL-6), interleukin-8 (IL-8), monocyte chemoattractant protein-1 (MCP-1), nerve growth factor (NGF), superoxide anion (O2•-), hydrogen peroxide (H2O2), and nitric oxide (NO•) from articular human cells. First, we cre

Regulation of fibroblast-like synoviocyte function by cadherin 6 in rheumatoid arthritis.

BACKGROUND: Cadherins (CDH), such as CDH11, are glycoprotein adhesion molecules contributing to cell-cell interactions in health and disease. CDH11 has demonstrated important functions in rheumatoid arthritis (RA) fibroblast-like synoviocytes (FLS). In transcriptome expression studies, we observed that Cadherin 6 (CDH6) expression was higher in RA compared to osteoarthritis (OA). CDH6 is associated with cancer progression, but little information is known on the role of CDH6 in RA. The present study investigates CDH6 expression, regulation, function in FLS, and distribution in RA synovia. METH

Endogenous production of hyaluronan, PRG4, and cytokines is sensitive to cyclic loading in synoviocytes.

Synovial fluid is composed of hyaluronan and proteoglycan-4 (PRG4 or lubricin), which work synergistically to maintain joint lubrication. In diseases like osteoarthritis, hyaluronan and PRG4 concentrations can be altered, resulting in lowered synovial fluid viscosity, and pro-inflammatory cytokine concentrations within the synovial fluid increase. Synovial fibroblasts within the synovium are responsible for contributing to synovial fluid and can be targeted to improve endogenous production of hyaluronan and PRG4 and to alter the cytokine profile. We cyclically loaded SW982 synoviocytes to 0%,

Paclitaxel Inhibits Synoviocyte Migration and Inflammatory Mediator Production in Rheumatoid Arthritis.

Activated fibroblast-like synoviocytes (FLSs) play a crucial role in the pathogenesis and progression of rheumatoid arthritis (RA). It is urgent to develop new drugs that can effectively inhibit the abnormal activation of RA-FLS. In our study, the RA-FLS cell line, MH7A, and mice with collagen-induced arthritis (CIA) were used to evaluate the effect of paclitaxel (PTX). Based on the results, PTX inhibited the migration of RA-FLS in a dose-dependent manner and significantly reduced the spontaneous expression of IL-6, IL-8, and RANKL mRNA and TNF-α-induced transcription of the IL-1 β, IL-8, MMP-

The emerging role of fibroblast-like synoviocytes-mediated synovitis in osteoarthritis: An update.

Osteoarthritis (OA), the most ubiquitous degenerative disease affecting the entire joint, is characterized by cartilage degradation and synovial inflammation. Although the pathogenesis of OA remains poorly understood, synovial inflammation is known to play an important role in OA development. However, studies on OA pathophysiology have focused more on cartilage degeneration and osteophytes, rather than on the inflamed and thickened synovium. Fibroblast-like synoviocytes (FLS) produce a series of pro-inflammatory regulators, such as inflammatory cytokines, nitric oxide (NO) and prostaglandin E2

Autoantibodies from rheumatoid arthritis patients recognize antigens on the synoviocyte surface.

We have found autoantibodies in the sera from rheumatoid arthritis (RA) patients which recognize two cell surface antigens of approximately 70 kDa and 28 kDa from synoviocyte extracts as detected by immunoprecipitation analysis. These polypeptides were immunoprecipitated from extracts containing mainly macrophage-like synoviocytes (type A) but not from extracts of homogeneous fibroblast-like synoviocytes (type B). These autoantigens are not selectively expressed by RA synoviocytes, since both RA and non-rheumatoid synovia were reactive for RA sera. From the panel of different RA sera tested, 6

Morphologic, quantitative and cytoenzymologic studies of synoviocytic and monocytic cells in synovial fluid.

The findings are presented of a morphologic, quantitative, cytochemical and cytoenzymologic study of the mononucleated nonlymphoid cells in knee synovial fluids from osteoarthritis and various inflammatory diseases. The morphologic criteria allowed the identification of subtypes, including phagocytic subtypes, among synoviocytic and monocytic cells in the fluids. The quantitative study showed an important afflux of monocytes and a hyperexfoliation of synoviocytes in the inflammatory diseases. In fluids with intermediate cellularity, the ratio of monocytes to synoviocytes allowed the differenti

Artemisinin increases susceptibility to ferroptosis of fibroblast-like synoviocytes in rheumatoid arthritis.

OBJECTIVE: Rheumatoid arthritis (RA) is a chronic inflammatory disease characterized by extensive synovial hyperplasia. Artemisinin (ART), a compound extracted from the traditional Chinese herb artemisia annua, has been proven to have anti-malarial and anti-cancer effects. Recently, the ability of artemisinin to induce ferroptosis has shown inhibitory effects on tumor cell proliferation. This study intends to examine whether artemisinin can inhibit the proliferation of RA synovial cells by sensitizing them to ferroptosis. METHOD: The effect of artemisinin on fibroblast-like synoviocytes (FLSs

Antifibrotic effect of brown algae-derived fucoidans on osteoarthritic fibroblast-like synoviocytes.

Synovial fibrosis is a pathological process which contributes to joint pain and stiffness in several musculoskeletal disorders. Fucoidans, sulfated polysaccharides found in brown algae, have recently emerged as promising therapeutic agents. Despite the increasing amount of evidence suggesting the protective role of fucoidans in different experimental approaches of human fibrotic disorders, the effect of these sulfated polysaccharides on synovial fibrosis has not been investigated yet. By an in vitro experimental approach in fibroblast-like synoviocytes, we detected that fucoidans inhibit their

Rhein inhibits ATP-triggered inflammatory responses in rheumatoid rat fibroblast-like synoviocytes.

Rheumatoid arthritis (RA) is a chronic and systemic inflammatory disorder, which may lead to joint disabilities. So far the pathogenesis of RA remains largely undetermined, and there are still no potent drugs for clinical treatment. Rhein, a natural bioactive anthraquinone derivative, exhibited significant anti-inflammatory activities demonstrated by previous studies. Here we aimed to investigate the effects of rhein on ATP-induced inflammation responses in fibroblast-like synoviocytes isolated from a rat model of collagen induced arthritis (CIA). Our results showed that ATP triggered rapid cy

GACAT3 promoted proliferation of osteoarthritis synoviocytes by IL-6/STAT3 signaling pathway.

OBJECTIVE: Osteoarthritis (OA) is one of the most common chronic joint diseases, caused by lesions in articular cartilage and synovial membranes. Synovitis is a major characteristic of OA, due to the proliferation of synoviocytes. Long noncoding RNAs (lncRNAs) have implicated to play an important role in many different diseases. The aim of this study was to verify the role of lncRNA gastric cancer-associated transcript 3 (GACAT3) in osteoarthritis. MATERIALS AND METHODS: We utilized the qRT-PCR to detect the expression of lncRNA GACAT3 in osteoarthritis synoviocytes (OAS) and normal synoviocy

PUMA gene delivery to synoviocytes reduces inflammation and degeneration of arthritic joints.

In rheumatoid arthritis (RA), the proliferation of fibroblast-like synoviocytes (FLS) is the cause of chronic inflammation in joints and of joint damage. Delivery of the pro-apoptotic gene PUMA to FLS via human adenovirus type 5 (HAdV5) vectors has been tested as a therapeutic approach, but efficiency is hampered by low transduction, as FLS do not express HAdV5 receptors on the cell surface. Here we show that efficient transduction of PUMA in FLS can be achieved by conjugating HAdV5 to a baculovirus, which binds to the cell surface via the envelope glycoprotein Gp64. Intra-articular injection

Abundant lubricin expression suggests a link between synoviocytes, synovial tumors, and myxomas.

Progenitor cell differentiation into fibroblast-like synoviocytes (FLSs) and their ensuing phenotypic changes are incompletely explored. Synovial lining is composed of intimal macrophages and FLSs. FLSs have epithelioid morphology and directionally secrete components of synovial fluid, including lubricin. We stained human tissues and tumors using two anti-lubricin antibodies. Lubricin was found in FLSs in synovium and in tenosynovial giant cell tumors (TSGCTs) and not in the associated monocyte/macrophage cells, which were identified by double immunostaining for CD163. In TSGCTs, giant cells,

CP690,550 inhibits oncostatin M-induced JAK/STAT signaling pathway in rheumatoid synoviocytes.

INTRODUCTION: Interleukin (IL)-6-type cytokines exert their effects through activation of the Janus kinase/signal transducers and activators of transcription (JAK/STAT) signaling cascade. The JAK/STAT pathways play an important role in rheumatoid arthritis, since JAK inhibitors have exhibited dramatic effects on rheumatoid arthritis (RA) in clinical trials. In this study, we investigated the molecular effects of a small molecule JAK inhibitor, CP690,550 on the JAK/STAT signaling pathways and examined the role of JAK kinases in rheumatoid synovitis. METHODS: Fibroblast-like synoviocytes (FLS)

Comparison of taurine chloramine and taurine bromamine effects on rheumatoid arthritis synoviocytes.

Fibroblast-like synoviocytes (FLS) participate in rheumatoid arthritis (RA) chronic synovitis by producing pro-inflammatory cytokines (IL-6, IL-8), growth factors (VEGF) and other inflammatory mediators (PGE2, NO). We have previously reported that Tau-Cl, generated by neutrophils, inhibits in vitro some of these pathogenic RA FLS functions. Taurine bromamine (Tau-Br) originates from eosinophils and neutrophils, and its immunoregulatory activities are poorly known. Therefore, we investigated the effects of Tau-Br on RA FLS functions and compared it to Tau-Cl anti-inflammatory action. When appli

Transformation of fibroblast-like synoviocytes in rheumatoid arthritis; from a friend to foe.

Swelling and the progressive destruction of articular cartilage are major characteristics of rheumatoid arthritis (RA), a systemic autoimmune disease that directly affects the synovial joints and often causes severe disability in the affected positions. Recent studies have shown that type B synoviocytes, which are also called fibroblast-like synoviocytes (FLSs), as the most commonly and chiefly resident cells, play a crucial role in early-onset and disease progression by producing various mediators. During the pathogenesis of RA, the FLSs' phenotype is altered, and represent invasive behavior

Inhibition of fibroblast activation protein partially restores the homeostatic phenotype of mechanoactivated synoviocytes.

OBJECTIVE: Synovial fibrosis, characterized by mechanoactivation of fibroblast-like synoviocytes (FLS), is thought to be a driver of osteoarthritis (OA). Fibroblast activation protein (FAP) is expressed by activated FLS and contributes to OA progression, but the mechanisms by which it does so are undefined. The objective of this study was to evaluate FAP expression in OA across species, assess FAP regulation in FLS, and determine how FAP inhibition alters the phenotype of mechanoactivated FLS. METHODS: Synovium from human patients (n = 9) and animal models of OA (murine n = 5; porcine n = 8)

NETs promote invasive behavior of fibroblast-like synoviocytes through GPIbα in rheumatoid arthritis.

OBJECTIVE: Neutrophil extracellular traps (NETs) have been implicated in rheumatoid arthritis (RA) pathogenesis, yet their effects on fibroblast-like synoviocytes (FLS) remain unclear. This study aims to investigate the role of NETs in RA FLS migration, proliferation, and invasion, as well as the underlying molecular mechanisms. METHODS: NETs formation was assessed in neutrophils isolated from RA patients and healthy controls (HC) using immunofluorescence staining. RA FLS were stimulated by RA or HC derived NETs, and migration was evaluated via wound healing assays. RNA sequencing (RNA-seq) i

Andrographolide Ameliorates Inflammatory Changes Induced by D-Lactate in Bovine Fibroblast-like Synoviocytes.

During acute ruminal acidosis, the manifestation of aseptic polysynovitis and lameness in cattle has been observed. Evidence suggests that joint inflammation can be attributed to the metabolic alterations induced by D-lactate in fibroblast-like synoviocytes (FLSs). We aimed to investigate whether andrographolide could mitigate the inflammation and metabolic alterations induced by D-lactate in bovine fibroblast-like synoviocytes (bFLSs). To assess this, bFLSs were cultured in the presence or absence of andrographolide. We evaluated its potential interference with the expression of proinflammato
