T-2 toxin, a mycotoxin commonly found in food, is recognized as one of the most harmful contaminants. Herein, a sensitive photoelectrochemical (PEC) aptasensor based on an "on-off-on" signal response strategy was developed for T-2 detection using WO3/CdIn2S4 as a photoanode. The sensitization of WO3 with CdIn2S4 significantly enhanced the photocurrent, leading to the initial "signal-on" state. As a signal label of probe DNA (pDNA), CoO significantly inhibited the photocurrent response of WO3/CdIn2S4 ("signal-off" state), enhancing the signal recovery space and improving the detection sensitivi

This study investigated the expression of Zip6, a gene predominantly located in the placenta, breast, and prostate tissues, in patients with Kashin-Beck disease (KBD). Environmental risk factor models for KBD were developed using low selenium (Se) feeding (with a Se content of 0.02 mg Se/kg in the feed) and exposure to T-2 toxin (200 ng/g*BW/D). Additionally, the study examined the alterations in Se and Zn2+ levels, along with the mRNA and protein expression levels of Zip6 and KBD related genes, including Mtf1, Mmp3, Mmp13, Adamts5, and Col2a1. Differentially expressed genes (DEGs) were examin

T-2 toxin, a type A trichothecene, is a secondary metabolite produced by Fusarium poae, Fusarium sporotrichioides, and Fusarium tricinctum. As the most toxic trichothecenes, T-2 toxin causes severe damage to multiple organs, especially to liver. However, the contamination of T-2 toxin covers a wide range of plants, including nuts, grains, fruits and herbs globally. And due to chemical stability of T-2 toxin, it is difficult to be completely removed from the food and feeds, which poses a great threat to human and animal health. Liver is the major detoxifying organ which also makes it the main t

T-2 toxin, a trichothecene mycotoxin, was injected iv or ip in the single lethal dose of 2 mg/kg. Vomiting was elicited in normal and decerebrate cats with an average onset time of 26.6 min. Chronic ablation of the area postrema significantly delayed the emetic latency to 304 min. Polygraph recording revealed a steady decline in mean arterial blood pressure and pulse pressure to an extreme shock level resulting in death after 5 to 15 hr. Heart rate varied unremarkably throughout the course of circulatory failure, and the cardiac beat persisted after respiratory arrest. No protection against th

Using sodium [51Cr]chromate as radiolabeled marker, gastrointestinal transit of a milk test meal was determined in mice receiving for 4 days T-2 toxin (1 mg/kg per day per os) alone or with a clay, smectite (2 g/kg per day), given according to four different procedures. Gastric emptying and small intestinal transit were significantly accelerated after the 1st T-2 administration and during the 4 days of treatment. When smectite was given together with the toxin with or without pretreatment by smectite alone for 2 days, the T-2 induced disturbances in gastrointestinal transit remained unchanged.

T-2 toxin, a prevalent mycotoxin, represents a notable global public health risk. Neutrophil extracellular traps (NETs) and ferroptosis are involved in a variety of pathophysiological processes and are implicated in goat immunity. However, the impact of T-2 toxin on NETs release, ferroptosis, and their interplay have not been previously documented. In this study, neutrophils were stimulated with T-2 toxin for 4 h. The structure and mechanism of NETs were analyzed using immunofluorescence and Pico Green staining. The expressions of glutathione peroxidase 4 (GPX4) and ferritin (FT) was quantifie

Kashin-Beck disease (KBD) is an endemic osteoarthropathy, with T-2 toxin recognized as a major pathogenic factor. This study investigates the role of the YAP/NLRP3/GSDMD signaling pathway in T-2 toxin-induced cartilage injury. Differentially expressed genes associated with KBD were retrieved from PubMed and subjected to KEGG pathway enrichment analysis, revealing significant enrichment of the Hippo signaling pathway. Elevated serum levels of IL-1β, IL-18, and LDH were observed in KBD patients. In both in vivo and in vitro models, T-2 toxin inhibited Hippo/YAP signaling and promoted the nuclear

This study aims to investigate the impact of T-2 toxin on the regulation of downstream target genes and signaling pathways through exosome-released miRNA in the development of cartilage damage in Kashin-Beck disease (KBD). Serum samples from KBD patients and supernatant from C28/I2 cells treated with T-2 toxin were collected for the purpose of comparing the differential expression of exosomal miRNA using absolute quantitative miRNA-seq. Target genes of differential exosomal miRNAs were identified using Targetscan and Miranda databases, followed by GO and KEGG enrichment analyses. Validation of

Our goals were to investigate whether environmentally relevant doses of T-2 toxin can affect human ovarian granulosa cells' function and to reveal the potential mechanism of T-2 toxin's action. Results showed that T-2 toxin strongly attenuated luteinizing hormone/choriogonadotropin receptor (LHCGR) mRNA expression in follicle-stimulating hormone (FSH)-stimulated human cumulus granulosa cells. Addition of human chorionic gonadotropin was not able to elicit maximal response of ovulatory genes amphiregulin, epiregulin, and progesterone receptor. T-2 toxin reduced mRNA levels of CYP19A1 and steroi

The present study aimed to reveal the molecular mechanism of T-2 toxin-induced cerebral edema by aquaporin-4 (AQP4) blocking and permeation. AQP4 is a class of aquaporin channels that is mainly expressed in the brain, and its structural changes lead to life-threatening complications such as cardio-respiratory arrest, nephritis, and irreversible brain damage. We employed molecular dynamics simulation, text mining, and in vitro and in vivo analysis to study the structural and functional changes induced by the T-2 toxin on AQP4. The action of the toxin leads to disrupted permeation of water and p

T-2 toxin (T-2) and HT-2 toxin (HT-2) are trichothecene mycotoxins frequently detected in cereals. While exposure has primarily been evaluated using dietary assessment methods, human biomonitoring provides an alternative approach to assess internal exposure. This follow-up research aimed to further characterise the toxicokinetics of the mycotoxins T-2 and HT-2 and to subsequently develop a physiologically based kinetic (PBK) model, enabling prediction of urinary biomarker concentrations based on external exposure and vice versa. To this end, urine samples from 40 Norwegian adults were analysed

Plant materials used in the production of pig feed are frequently contaminated with mycotoxins. T-2 toxin is a secondary metabolite of selected Fusarium species, and it can exert a harmful influence on living organisms. Most mycotoxins enter the body via the gastrointestinal tract, and they can modulate the gut-associated lymphoid tissue (GALT) function. However, little is known about the influence of low T-2 toxin doses on GALT. Therefore, the aim of this study was to evaluate the effect of T-2 toxin administered at 50% of the lowest-observed-adverse-effect level (LOAEL) on the percentage of

T-2 toxin, a potent Class A trichothecene mycotoxin, is a global food contaminant with poorly understood cardiotoxic mechanisms. Epidemiological evidence links T-2 toxin exposure to cardiac injury and selenium deficiency, yet the molecular basis of this interaction remains unclear. Selenoprotein S (SelS), an endoplasmic reticulum (ER)-resident selenoprotein, is critically involved in ER stress regulation and its expression correlates positively with selenium intake. This study investigates the role of SelS in T-2 toxin-induced myocardial damage and its mechanistic link to ER stress. Using in v

T-2 toxin is a typical mycotoxin that seriously threatens human and animal health. Liver is the major target organ of T-2 toxin. To elucidate the precise hepatotoxicity mechanism and discover a natural antagonist of T-2 toxin. T-2 toxin (0, 0.5, 1, 2 mg/kg BW)-induced liver injury model, Ferrostatin-1 (1 mg/kg·BW) interference model, Parkin-/- mice model, Nrf2-activating model (tBHQ, 20 mg/kg·BW) and lycopene (5 mg/kg·BW) treatment model were constructed. Proteomics revealed that ferroptosis is a critical hepatotoxicity mechanism of T-2 toxin. Blocking ferroptosis alleviated the liver damage a

T-2 toxin, a potent immunosuppressive mycotoxin produced by Fusarium species, has not yet been fully elucidated regarding its immunotoxic effects under chronic exposure during developmental stages. To address the limitations of previous studies that primarily focused on single-organ assessments or acute toxicity evaluations in adult animals, this study established a 28-day subchronic T-2 toxin exposure model in Wistar juvenile rats (0-0.8 mg/kg body weight). We systematically evaluated its developmental immunotoxicity through three integrated dimensions: 1) immune organs (thymus, spleen), 2) i

Despite being one of the most common contaminants of poultry feed, the molecular effects of T-2 toxin on the liver of the exposed animals are still not fully elucidated. To gain more accurate understanding, the effects of T-2 toxin were investigated in the present study in chicken-derived three-dimensional (3D) primary hepatic cell cultures. 3D spheroids were treated with three concentrations (100, 500, 1000 nM) of T-2 toxin for 24 h. Cellular metabolic activity declined in all treated groups as reflected by the Cell Counting Kit-8 assay, while extracellular lactate dehydrogenase activity was

T-2 toxin is a trichothecene mycotoxin and is considered as an extremely inevitable pollutant with potent hepatotoxicity. However, the approach to alleviation of T-2 toxin-triggered hepatotoxicity has been recognized as a serious challenge. Resveratrol (Res) is a polyphenol natural product isolated from various plant species, but its protective effect against T-2 toxin hepatotoxicity and detailed mechanism remains obscure. In the present study, the effect of Res against the hepatotoxicity was evaluated, and the underlying mechanisms were further revealed in mice. Functionally, Res inhibited li

T-2 toxin can induce bone and cartilage development disorder, and oxidative stress plays an important role in it. It is well known that selenomethionine (Se-Met) has antioxidative stress properties and promotes the repair of cartilage lesion, but it remains unclear whether Se-Met can relieve damaged cartilage exposure to T-2 toxin. Here, the oxidative stress and ferroptosis of chondrocytes exposure to T-2 toxin were observed. Mechanistically, T-2 toxin increased ROS, lipid ROS, MDA and Fe2+ contents in chondrocytes, decreased GSH and GPX4 activity, and inhibited the system Xc-/GSH/GPX4 antioxi

T-2 toxin has been reported to cause severe oral lesions and neural disturbances in young broiler chickens. T-2 toxin, when added at a level of 20 mug per g of feed, caused oral lesions but no abnormal neural disturbances in young broiler chickens. T-2 toxin, when added at a level of 20 mug per g of feed, caused oral lesions but no abnormal neural symptoms in laying hens. T-2 toxin had no effect on either hemoglobin, hematocrit values, erythrocyte count, plasma glucose, prothrombin times, or the sizes of the liver, spleen, pancreas, and heart. Lipid content of the liver was not altered. Feed c

T-2 toxin is a typical fusarium mycotoxin that induces reproductive injury. As a male reproductive toxicant, impaired testosterone biosynthesis is the primary reproductive toxic effect induced by T-2 toxin. Therefore, clarifying how T-2 toxin disrupts testosterone synthesis at the molecular level and screening natural plant extracts that can alleviate this reproductive toxicity are of great importance. Lycopene (LYC) is a natural carotenoid known for its reproductive protective properties. Hence, this work assessed the protective efficacy and molecular mechanisms of LYC against T-2 toxin-cause
