The mononuclear cell infiltrate in benign and malignant breast tissue was studied using monoclonal antibodies with an immunoenzymatic technique. Infiltrating duct carcinomas contained a mononuclear cell infiltrate made up mostly of T lymphocytes, with minor B-cell and macrophage populations. The most preponderant T-cell subset was the T8, cytotoxic-suppressor cell. Some tumors contained T6-positive cells, possibly immature T cells. In contrast, stromal lymphocytes in benign fibrous mastopathy were largely T cells, but the T4, helper-inducer cell was the most preponderant, and T6 cells were not

The distribution of T lymphocyte subsets was assessed using monoclonal antibodies (MoAbs) in 44 untreated patients with multiple myeloma (MM) subdivided according to the clinical stage of the disease. A significant reduction (P less than 0.001) of T lymphocytes was observed only in stage II and III patients. Direct immunofluorescence analysis of circulating blood lymphocytes from patients with all stages of Type I diabetes revealed marked phenotypic abnormalities among all major T-lymphocyte subpopulations. The alterations were most pronounced in the prediabetics and comprised a stimulation of the T4+2H4+ and T8+2H4+ suppressor inducer subpopulations and corresponding depression of the T4+4B4+ helper inducers.

Previously, we reported the short-term immunological effects of postoperative adjuvant interferon-gamma (IFN-gamma) administration to renal cell carcinoma patients as determined by three-color flow cytometry. Adult T-cell leukemia (ATL) cells usually express the helper/inducer associated antigen OKT 4 with lack of OKT8. However, there are a few case reports indicating that there are atypical cell phenotypes in ATL including OKT4+/OKT8+. Use of monoclonal antibodies directed against T cell antigens for the phenotypic characterization of neoplastic lymphoid cells in peripheral T cell lymphomas is described. Studies of cryostat sections revealed the distribution of T cell subsets in nodal and extranodal infiltrates, and immuno-ultrastructural techniques demonstrated discrete localization of T cell antigens to the cytoplasmic membranes of neoplastic cells. Although histologically similar, the tumors appeared heterogeneous as to their immunologic phenotype, with the majority demonstrating markers for T helper/inducer lymphocytes.

Recent epidemiologic studies have shown that human T-lymphotropic virus type I (HTLV-I) is associated with polymyositis. We studied the cellular immune responses in 10 patients with systemic form of juvenile rheumatoid arthritis. The numbers of peripheral T lymphocytes and their helper/inducer and cytotoxic/suppressor subpopulation were within normal levels. There is a significant decrease in the ratio of inducer (helper) to suppressor T lymphocytes in the peripheral blood of patients with primary biliary cirrhosis (PBC). These changes are not seen in other forms of cholestatic liver disease, but are similar to those described in chronic graft-versus-host disease. Cyclosporin A treatment increased the proportion of suppressor A lymphocytes and improved liver function in 6 patients with PBC. This indicates that cyclosporin A is an immunoregulatory drug. Unfortunately nephrotoxicity precluded long-term use.

The function of peripheral blood T-lymphocytes was tested in a series of untreated patients with hairy-cell leukaemia (HCL).
