Talaromyces marneffei infection in a lung cancer patient: a rare case report.

BACKGROUND: Talaromyces marneffei is an invasive, and thermal dimorphic pathogenic fungus, whose infection is life threatening in human. Although immunocompromised patients, such as patients with human immunodeficiency virus infection and recipients of organ transplant, are susceptible hosts, infections have been recently reported in people with normal immune function. Patients with cancer may also be susceptible hosts but no case of T. marneffei infection has been reported in patients with lung cancer. In this case, we describe T. marneffei infection coexisting with primary pulmonary lymphoep

Development and characterization of an immunochromatographic test for the rapid diagnosis of Talaromyces (Penicillium) marneffei.

Talaromyces (Penicillium) marneffei is a thermally dimorphic fungus that can cause opportunistic systemic mycoses in patients infected with the human immunodeficiency virus (HIV). It has also been reported among patients with other causes of immunodeficiency, such as systemic lupus erythematosus, cancer, organ transplanted patients receiving immunosuppressive drug and adult onset immunodeficiency syndromes. Recent studies indicate that the clinical manifestations, laboratory findings and treatment strategies of talaromycosis (penicilliosis) marneffei are different between patients with and wit

Nitric oxide inhibitory and cytotoxic secondary metabolites from Talaromyces sp. BHM8-7.

Two previously undescribed sesquiterpenes (1 and 2), one meroterpenoid (3), and one cyclic peptide (4), together with four known metabolites (5-8) were isolated from Talaromyces sp. BHM8-7. Their structures were elucidated by comprehensive spectroscopic techniques, such as 1D and 2D NMR, HRESIMS, ECD calculations, and the advanced Marfey's method following acid hydrolysis. Among the isolates, compound 5 exhibited significant inhibitory activity against nitric oxide (NO) production in LPS-stimulated RAW 264.7 cells without observable cytotoxicity. In addition, compounds 1 and 5 displayed potent

Disseminated Talaromyces marneffei infection presenting as pulmonary cavitary nodules: a case report.

BACKGROUND: Talaromycosis is an invasive fungal infection with high mortality in patients with advanced human immunodeficiency virus (HIV) disease in the tropical and subtropical regions of Asia. The lungs are the most commonly involved organ, and chest computed tomographic (CT) typically reveals diffuse pulmonary reticular and ground-glass changes. However, cavitary nodules are rare in HIV-positive patients, often leading to delay diagnosis. CASE PRESENTATION: A 46-year-old female with a 3-year history of untreated HIV infection was referred to our hospital due to recurrent fever lasting for

Aspergillus calidoustus and Talaromyces columbinus infections in chronic graft-versus-host disease.

Advancements in allogeneic haematopoietic stem cell transplant (alloHSCT) procedures have improved patient outcomes over the last two decades, though invasive fungal infections (IFIs) remain a significant risk. The incidence of IFIs in alloHSCT recipients is estimated at 6%, with a mortality rate of 13%, and Aspergillus species are the most common pathogens involved. Posaconazole is effective in preventing IFIs post-transplant and is standard care during neutropenia or when managing graft-versus-host disease (GvHD) with high-dose steroids. However, azole prophylaxis may cause resistant Aspergi

Antibody screening reveals antigenic proteins involved in Talaromyces marneffei and human interaction.

Talaromycosis is a fungal infection that generally affects immunocompromised hosts and is one of the most frequent systemic mycoses in HIV patients, especially in endemic areas such as Southeast Asia. Talaromyces marneffei, the causative agent of talaromycosis, grows as a mold in the environment but adapts to the human body and host niches by transitioning from conidia to yeast-like cells. Knowledge of the human host and T. marneffei interaction has a direct impact on the diagnosis, yet studies are still lacking. The morbidity and mortality rates are high in taloromycosis patients if the diagn

Eco-phylogenetic diversity and relative pigment production in environmental Talaromyces purpurogenus isolates.

Talaromyces purpurogenus is a filamentous fungus capable of producing red polyketide-like pigments with potential as natural alternatives to synthetic colorants. This study investigated the ecological distribution, phylogenetic diversity, and relative pigment-producing capacity of T. purpurogenus isolates recovered from agricultural soils, decaying fruits, spoiled grains/cereals, and stored feed/food materials. A total of 35 isolates were obtained, with the highest isolation frequencies observed in agricultural soils and spoiled grains/cereals. Molecular identification based on ITS rDNA and β-

Dihydro-α-Pyrone Antibiotics from a Soil-Derived Fungus Talaromyces sp. G23.

Nine novel dihydro-α-pyrone derivatives, talaromypyrones A-I (1-9), were isolated from the soil-derived fungus Talaromyces sp. G23. These compounds share a conserved 3'R-configuration within the 4-hydroxy-5,6-dihydro-2-pyrone core but display notable variations in their flanking polyketide chains. Talaromypyrones A-H (1-8) demonstrate potent antibacterial activity against Gram-positive bacteria, including drug-resistant strains, while exhibiting excellent safety profile by showing no cytotoxicity toward normal human LO2 cells. However, they exhibit no activity against Gram-negative bacteria (l

Talaromyces spp. are tunable producers of light-activated secondary metabolites with anticancer potential.

Photodynamic therapy (PDT) is a promising treatment strategy for resistant or non-surgical tumors. To develop photosensitizers tailored to specific cancer types, new chemical scaffolds are needed. Macrofungi have recently been identified as a potent source of such compounds, particularly dimeric anthraquinones. However, the sources previously described (i.e., dermocyboid Cortinarii) cannot be cultivated in a biotechnologically applicable way. Thus, in this proof-of-concept study, we explored Talaromyces species (Ascomycota) for their potential to produce similar light-activated metabolites. Si

Amphotericin B deoxycholate (DAmB) is a first-line agent for the initial treatment of talaromycosis. However, little is known about the population pharmacokinetics and pharmacodynamics of DAmB for talaromycosis. Pharmacokinetic data were obtained from 78 patients; among them, 55 patients had serial fungal CFU counts in blood also available for analysis. A population pharmacokinetic-pharmacodynamic model was fitted to the data. The relationships between the area under the concentration-time curve (AUC)/MIC and the time to blood culture sterilization and the time to death were investigated. Ther

Unexpected talaroenamine derivatives and an undescribed polyester from the fungus Talaromyces stipitatus ATCC10500.

Chemical investigation of the fungus Talaromyces stipitatus ATCC 10500, whose genome has been sequenced, led to the isolation of four undescribed talaroenamines B-E along with the known talaroenamine A. Their structures were elucidated on the basis of spectroscopic studies including mass spectrometry, extensive 2D NMR and electronic circular dichroism (ECD). Interestingly, talaroenamine A had previously been isolated from the strain of T. stipitatus Δtrop C, a strain knocked out for the gene encoding a non-heme Fe(II)-dependent dioxygenase catalyzing the oxidative ring expansion leading to the
