Chemical receptors of the arytenoid: A comparison of human and mouse.

OBJECTIVES/HYPOTHESIS: The larynx is a highly responsive organ exposed to mechanical, thermal, and chemical stimuli. Chemicals elicit responses both in intraepithelial nerve fibers and in specialized chemosensory cells, including scattered solitary cells as well as taste cells organized into taste buds. Activation of both chemosensory cells and taste buds in the larynx elicit cough, swallow, or apnea with exposure to sour or bitter substances, and even by water or sweet-tasting chemicals. In an effort to begin understanding their function, we sought to compare the distribution, density, and ty

The emerging role of the bitter taste receptor T2R38 in upper respiratory infection and chronic rhinosinusitis.

BACKGROUND: Maintaining a clean upper respiratory tract requires efficient detection of pathogenic bacteria so that the airway mucosa can mount proper defenses to neutralize and clear the offending microbes. Bitter taste receptors (T2Rs) may play a critical role in this process. T2Rs were originally identified in taste cells of the tongue, where they protect against the ingestion of toxic plant and/or bacterial products. However, T2Rs are also expressed in extragustatory tissue including the airways. One specific T2R isoform, T2R38, was recently shown to be expressed in cilia of sinonasal epit

Gene expression analyses of TAS1R taste receptors relevant to the treatment of cardiometabolic disease.

The sweet taste receptor (STR) is a G protein-coupled receptor (GPCR) responsible for mediating cellular responses to sweet stimuli. Early evidence suggests that elements of the STR signaling system are present beyond the tongue in metabolically active tissues, where it may act as an extraoral glucose sensor. This study aimed to delineate expression of the STR in extraoral tissues using publicly available RNA-sequencing repositories. Gene expression data was mined for all genes implicated in the structure and function of the STR, and control genes including highly expressed metabolic genes in

Salty taste deficits in CALHM1 knockout mice.

Genetic ablation of calcium homeostasis modulator 1 (CALHM1), which releases adenosine triphosphate from Type 2 taste cells, severely compromises the behavioral and electrophysiological responses to tastes detected by G protein-coupled receptors, such as sweet and bitter. However, the contribution of CALHM1 to salty taste perception is less clear. Here, we evaluated several salty taste-related phenotypes of CALHM1 knockout (KO) mice and their wild-type (WT) controls: 1) In a conditioned aversion test, CALHM1 WT and KO mice had similar NaCl avoidance thresholds. 2) In two-bottle choice tests, C

Multiscale Elucidation of Taste Interactions in Osmanthus Green Tea: Bitter Masking via EGCG and Phenylethanoid Glycoside Interplay.

This study revealed the molecular mechanism by which Osmanthus fragrans masks the bitterness of summer-autumn green tea infusion. Taste dilution analysis identified catechins such as EGCG in green tea as the primary bitter compounds. Phenylethanoid glycosides (PhGs), such as verbascoside and salidroside in Osmanthus fragrans exhibited significant bitter-masking effects. Molecular docking identified TAS2R31 as the primary bitter receptor for EGCG, with verbascoside or salidroside competitively disrupting the EGCG-TAS2R31 binding. Molecular dynamics simulations showed that PhGs altered the TAS2R

Bitterness-Suppressing Effect of Umami Dipeptides and Their Constituent Amino Acids on Diphenhydramine: Evaluation by Gustatory Sensation and Taste Sensor Testing.

Diphenhydramine, a sedating antihistamine, is an agonist of human bitter taste receptor 14 (hTAS2R14). Diphenhydramine hydrochloride (DPH) was used as a model bitter medicine to evaluate whether the umami dipeptides (Glu-Glu and Asp-Asp) and their constituent amino acids (Glu, Asp) could suppress its bitterness intensity, as measured by human gustatory sensation testing and using the artificial taste sensor. Various concentrated (0.001-5.0 mM) Glu-Glu, Asp-Asp, Glu and Asp significantly suppressed the taste sensor output of 0.5 mM DPH solution in a dose-dependent manner. The effect of umami di

Linking intestinal bitter taste receptors and GSPE-induced long-lasting benefits in ageing rats: an integrative analysis.

Ageing is associated with attenuated type-2 bitter-taste receptor (TAS2R) signalling and contributes to metabolic, inflammatory and barrier decline, but its system-wide impact along the gut remains undefined. We combined transcription analysis, physiology, metabolomics and microbiota profiling to test whether a brief grape-seed proanthocyanidin extract (GSPE) intervention can counter age-related dysfunction by long-term modulation of intestinal Tas2r expression. Female Wistar rats were distributed into young (2 months, n = 10) or aged (21 months, n = 24) groups; eleven aged animals received GS

The inhibitory effect of mannoproteins on bitter aftertaste: From the salivary layer to the tongue surface to the receptor surface.

To investigate the mechanisms by which mannoproteins (MPs, CECA, and 2082) with different chain conformations and concentrations inhibit the bitter aftertaste of monomeric and oligomeric proanthocyanidins (MOPC), a multilayer penetration approach was employed to reveal their mechanisms of action across different levels. At the salivary level, MPs formed a dense network with artificial saliva (AS) through key hydrophobic interactions. Fluorescence spectroscopy results indicated that the primary interaction force between CECA and AS were electrostatic interaction. However, as the concentration o

An Artemisia-derived natural product-based fluorescent probe for the bitter taste receptor hTAS2R38.

The discovery of taste receptors hTAS2Rs expression in extra oral tissue, especially in the gastrointestinal tract and in the respiratory system, has endowed bitter receptors of functionalities that exceed the simple perception of taste and flavour. In particular, stimulation of hTAS2Rs by bitter agents in the airway smooth muscle triggers bronchodilation of possible pharmacological relevance. To study the receptor localization in pulmonary smooth muscle cells and to investigate their biological response to hTAS2R38 activation, we have developed a fluorescent probe for hTAS2R38 starting from t

Sensory nutrition of sweet taste. Physiological, pathophysiological and pharmacological implications of T1R2/T1R3 signalling pathways on energy (glucose) and body weight homeostasis.

Taste perception plays a key role in food chose. Both taste receptors type 1 member 2 (T1R2) and member 3 (T1R3) are responsible for detecting mono- and disaccharides, and sweet taste perception is dependent of the recognition threshold (RT) of these molecules. Differences in sweet RT are mainly attributed to polymorphisms of T1R2 and T1R3 as well as their expression in taste buds, which are regulated by several hormones including glucagon, ghrelins, glucagon-like peptide, leptins and endocannabinoids. Interestingly, aging, diabetes and obesity are characterized by significant changes in sweet

[Gustatory disorders in COVID-19].

Taste disturbances are present in 20.3-88.0% of COVID-19 cases and are the first signs of infection in 11.0-18.1% of cases. They often manifested 3-7 days after the onset of general respiratory symptoms and last 2 to 16 days, followed by recovery. There are also prolonged disturbances of taste sensation (up to 61-76 days or more), which is associated with damage to various types of receptor cells of the mucous membrane of the tongue. More severe taste disturbances are recorded in the elderly. In women, changes in taste sensation are noted more often than in men, and with a longer recovery peri

Parthenolide promotes glucagon-like peptide-1 secreting in human Caco-2 cells via regulation of bitter taste receptor-induced of calcium signaling.

Bitter taste receptors (TAS2Rs), in addition to being expressed in oral tissues, are also present in the gastrointestinal tract and are promising targets for inducing glucagon-like peptide-1 (GLP-1) secretion and treating type 2 diabetes mellitus (T2DM). However, natural bitter agonists capable of enhancing endogenous GLP-1 secretion remain scarce. In this study, we aimed to identify natural bitter agonists with GLP-1-inducing potential through comprehensive screening of the BitterDB and BitterX databases, and to evaluate their effects and underlying mechanisms in human enteroendocrine Caco-2

Activation of human bitter taste receptor (TAS2R) subtypes and TAS2R14 genotypes by Bacillus amyloliquefaciens exopolysaccharides.

Bacillus amyloliquefaciens amy-1 is recognized as a probiotic for animals, and its exopolysaccharides (EPS) can lower blood sugar by activating specific human bitter-taste receptor type 2 (TAS2R) subtypes, particularly TAS2R14 and TAS2R38. Humans have 26 TAS2R subtypes, with some exhibiting different activities depending on genotype. However, differences in activity among TAS2R14 genotypes remain poorly studied. This research used THP-1 monocytes to identify TAS2Rs with high expression levels and analyzed how different TAS2R14 genotypes and other highly expressed subtypes respond to EPS. Accor

Characterization of the sweetness of natural plant-derived sweeteners mogroside IV and V and their interactions with human sweet taste receptor.

Mogrosides are an important type of natural plant-derived sweeteners, which are used in foods and medicines. However, their sweetness and structure-activity relationship, especially those for the representative members mogrosides IV and V, have been rarely reported. In the present study, we characterized the sweet taste properties (threshold values, etc) of mogrosides IV and V with a cell-based calcium mobilization assay, showing that mogrosides represent a category of primitive sweet compounds which are edible by primates as well as other mammals. Furthermore, it was demonstrated that the swe

Anti-cancer stemness and anti-invasive activity of bitter taste receptors, TAS2R8 and TAS2R10, in human neuroblastoma cells.

Neuroblastoma (NB) originates from immature neuronal cells and currently has a poor clinical outcome. NB cells possess cancer stem cells (CSCs) characteristics that facilitate the initiation of a tumor, as well as its metastasis. Human bitter taste receptors, referred to as TAS2Rs, are one of five types of basic taste receptors and they belong to a family of G-protein coupled receptors. The recent finding that taste receptors are expressed in non-gustatory tissues suggest that they mediate additional functions distinct from taste perception. While it is generally admitted that the recognition

Contribution of Underlying Connective Tissue Cells to Taste Buds in Mouse Tongue and Soft Palate.

Taste buds, the sensory organs for taste, have been described as arising solely from the surrounding epithelium, which is in distinction from other sensory receptors that are known to originate from neural precursors, i.e., neural ectoderm that includes neural crest (NC). Our previous study suggested a potential contribution of NC derived cells to early immature fungiform taste buds in late embryonic (E18.5) and young postnatal (P1-10) mice. In the present study we demonstrated the contribution of the underlying connective tissue (CT) to mature taste buds in mouse tongue and soft palate. Three

Selective taste impairment in relapsing-remitting Multiple Sclerosis and Supervised Learning - driven diagnostic classification.

Taste disturbances have been largely overlooked in Multiple Sclerosis (MS), despite mounting evidence that neuroinflammation and demyelination may disrupt chemosensory pathways. This study investigated taste perception in People with MS (PwMS) compared with healthy controls (HCs), explored whether specific taste-related features could classify MS using Supervised Learning (SL), and examined associations with taste receptor gene polymorphisms. Using validated taste-strip tests, we found that PwMS exhibit marked impairments in bitter, salty, and sour identification, accompanied by a selective in

Dietary soybean or seaweed (Kappaphycus sp.) modulates taste-related gene (tas1r1 and tas1r2.2) expression in Nile tilapia (Oreochromis niloticus).

Taste perception plays a central role in fish feeding behaviour by influencing feed recognition, intake, and nutrient sensing. As aquaculture increasingly adopts plant-based ingredients to replace fishmeal, understanding how these diets affect gustatory mechanisms is critical. This study evaluated TAS-family taste receptor genes in Nile tilapia (Oreochromis niloticus) fed nutritionally formulated diets containing fishmeal (TFM), soybean meal (TSB), red seaweed (Kappaphycus sp., TSW), alongside a natural meal (TNM) reference. Fish were reared for 62 days under controlled conditions, after which

Identification of 4'-Demethyl-3,9-dihydroeucomin as a Bitter-Masking Compound from the Resin of Daemonorops draco.

Masking the bitter taste of foods is one of the key strategies to improve their taste and palatability, particularly in the context of clean labeling, where natural compounds are preferred. Despite the demand, the availability of natural bitter-masking compounds remains limited. Here, we identified the bitter-masking compound 4'-demethyl-3,9-dihydroeucomin (DMDHE) isolated from the resin of Daemonorops draco by means of an activity-guided in vivo (sensory bitterness rating of quinine) and in vitro (cell-based bitter response assays) approach.
