Association of EEF1A1 and linc00993 expression with sperm morphology in teratozoospermia: an integrative in silico and in vitro study.

Teratozoospermia (TZ) represents a significant factor in male infertility, and for many years, evaluation of sperm morphology has been the standard diagnostic method. This work investigates the molecular basis of TZ using a combined strategy that integrates computational analyses with experimental validation. Expression profiles of mRNA and lncRNA from individuals with TZ and from healthy subjects were examined using publicly available datasets from the NCBI Gene Expression Omnibus (GEO). We carried out differential expression analysis through GEO2R, then applied functional enrichment and prot

Bi-allelic mutations of CAMSAP1 result in teratozoospermia with sperm head and flagella defects in humans.

STUDY QUESTION: What is the role of calmodulin-regulated spectrin-associated protein 1 (CAMSAP1) in human spermatogenesis? SUMMARY ANSWER: Biallelic CAMSAP1 variants cause human male infertility with multiple sperm head deformities and MMAF-like phenotypes by disrupting manchette microtubule dynamics and the microtubule-spectrin-actin scaffold essential for sperm head shaping. WHAT IS KNOWN ALREADY: Sperm head shaping requires coordinated acrosome formation, chromatin condensation, and manchette-driven nuclear remodeling. Although several genes have been implicated in globozoospermia, macroz

Serum and Seminal Plasma Zinc Levels and Immunopositivity of the ZIP6 and ZIP14 Transporters in Men with Normo- and Teratozoospermia.

Zinc plays a crucial role in spermatogenesis, sperm function, and fertilisation. Zinc homeostasis is regulated by ZIP and ZnT transporter proteins, which mediate Zn2+ influx and efflux across sperm cell membranes. This study analysed total Zn concentration in seminal plasma and serum of 10 normozoospermic and 32 teratozoospermic men involved in the process of infertility treatment, using inductively coupled plasma mass spectrometry. In addition, the expression of Zn transporters ZIP6 and ZIP14 in the sperm of two normozoospermic and two teratozoospermic men was analysed using immunofluorescenc

Epididymal RNase T2 contributes to astheno-teratozoospermia and intergenerational metabolic disorder through epididymosome-sperm interaction.

BACKGROUND: The epididymis is crucial for post-testicular sperm development which is termed sperm maturation. During this process, fertilizing ability is acquired through the epididymis-sperm communication via exchange of protein and small non-coding RNAs (sncRNAs). More importantly, epididymal-derived exosomes secreted by the epididymal epithelial cells transfer sncRNAs into maturing sperm. These sncRNAs could mediate intergenerational inheritance which further influences the health of their offspring. Recently, the linkage and mechanism involved in regulating sperm function and sncRNAs durin

Pathogenic variant in ACTL7A causes severe teratozoospermia characterized by bubble-shaped acrosomes and male infertility.

Teratozoospermia is a common factor associated with male infertility. However, teratozoospermia characterized by bubble-shaped acrosomes (BSAs) has not yet been identified in men and the causative genes are unknown. The present study is of a patient with severe teratozoospermia characterized by BSA and carrying a variant (c.1204G>A, p.Gly402Ser) of actin-like 7A (ACTL7A). For further verification, we generated an Actl7a-mutated mouse model (p.Gly407Ser) carrying an equivalent variant to that in the patient. We found that homozygous Actl7a-mutated (Actl7aMut/Mut) male mice were sterile, and all

Sperm motility in mice with oligo-astheno-teratozoospermia restored by in vivo injection and electroporation of naked mRNA.

Oligo-astheno-teratozoospermia (OAT), a recurrent cause of male infertility, is the most frequent disorder of spermatogenesis with a predominantly genetic origin. Patients and mice bearing mutations in the ARMC2 gene exhibit reduced sperm concentration, multiple morphological defects, and impaired motility, defining a canonical OAT phenotype. Intracytoplasmic sperm injection (ICSI) is required to treat this condition; however, it is associated with a slightly increased risk of birth defects compared with natural conception, highlighting the need for novel targeted therapies. Here, in vivo test

Preliminary FISH studies on spermatozoa and embryos in patients with variable degrees of teratozoospermia and a history of poor prognosis.

The aim of this study was to analyse to what extent sperm aneuploidy is associated with sperm morphology and subsequently with embryo aneuploidy. Fifty-nine men with variable degrees of teratozoospermia and previously poor assisted reproduction prognosis were included in the study. Samples from 10 normozoospermic men with proven fertility were used as controls. Individual spermatozoa were scored for chromosomes 13, 21 and for 18, X, Y separately. Compared with controls, 23 out of 59 cases (39.0%) were found to have increased sperm aneuploidy for at least one of the chromosomes analysed in a tr

Loss-of-function variant in TDRD6 cause male infertility with severe oligo-astheno-teratozoospermia in human and mice.

Oligo-astheno-teratozoospermia (OAT) is a common cause of male infertility, but the genetic basis of most OAT cases is still unknown. Here, one homozygous loss-of-function (LOF) variant in TDRD6, c.G1825T/p.Gly609X, was identified in an infertile patient with severe OAT by whole-exome sequencing (WES) and Sanger confirmation. Furthermore, Tdrd6-mutant mice (p.Gly615X; equivalent to p.Gly609X in human TDRD6) were generated. Remarkably, the Tdrd6-mutated mice mimicked the severe OAT symptoms of the patient. In addition, the architecture of chromatoid bodies (CBs) were disrupted in round spermati

Transmission electron microscopy analysis of the origin and incidence of sperm intranuclear cytoplasmic retention in fertile and teratozoospermia men.

The human sperm nucleus contains cytoplasm. However, the origin and incidence of human sperm intranuclear cytoplasmic retention (INCR) remain unknown. The objectives of this study were to observe the morphological origin of INCR within the seminiferous epithelium and investigate the incidence of INCR in fertile and teratozoospermia men using transmission electron microscopy (TEM). By TEM, INCR initially appeared in elongating round spermatid nuclei and varied in size, number, shape, content, location and distribution within sperm nuclei. The teratozoospermia group (n = 16) demonstrated a highe

Differential expression of mRNA aromatase in ejaculated spermatozoa from infertile men in relation to either asthenozoospermia or teratozoospermia.

Oestrogen biosynthesis in ejaculated spermatozoa is an autonomous process, which may influence sperm functions. The purpose of this study was to evaluate the relationship between the expression of aromatase, sperm quality and seminal neutral α-glucosidase marker in semen of Tunisian infertile men: asthenozoospermia (A; n = 16), teratozoospermia (T; n = 12) and asthenoteratozoospermia (AT; n = 11) in comparison with 18 normozoospermic ones. Aromatase mRNA levels estimated by real-time PCR were reduced in groups T (52%) and AT (67%) compared to controls and inversely correlated with the percenta

Sperm preparation techniques affect biological parameters, fertility indices, and DNA fragmentation in patients with oligo-astheno-teratozoospermia.

ABSTRACT: In normozoospermia, the albumin gradient filtration (AGF) and swim-up processing techniques are used to isolate spermatozoa with the highest biological parameters and specific sperm fertility indices (SSFI). In contrast, the highest number of sperm with the best motility rate, velocity, and SSFI, and the lowest DNA damage and nuclear fragmentation are isolated using the density gradient centrifugation (DGC) preparation technique in oligo-astheno-teratozoospermia (OAT) patients. The study aimed to evaluate the effect of DGC, AGF-1, AGF-2, and swim-up preparation techniques on biologic

Unveiling the Genetic Complexity of Teratozoospermia: Integrated Genomic Analysis Reveals Novel Insights into lncRNAs' Role in Male Infertility.

Male infertility is a global health issue, affecting over 20 million men worldwide. Genetic factors are crucial in various male infertility forms, including teratozoospermia. Nonetheless, the genetic causes of male infertility remain largely unexplored. In this study, we employed whole-genome sequencing and RNA expression analysis to detect differentially expressed (DE) long-noncoding RNAs (lncRNAs) in teratozoospermia, along with mutations that are exclusive to teratozoospermic individuals within these DE lncRNAs regions. Bioinformatic tools were used to assess variants' impact on lncRNA stru
