Enhanced quantification and cell tracking of dual fluorescent labeled extracellular vesicles.

Extracellular Vesicles (EVs) are nanosized particles with significant role in disease pathogenesis and as therapeutic potential. However, the lack of reliable and efficient methods for the characterization, quantification and tracking of EVs, combined with the limitations of detection techniques in differentiating specific EVs subtypes with beneficial properties, makes these process complex and time-consuming. To address this challenge, EVs were engineered using a tricistronic plasmid that encodes fluorescent proteins fused to tetraspanins (eGFP-CD63 and mCherry-CD9), with both fluorophores lo

Diagnostic performances of Schistosoma haematobium and Schistosoma mansoni recombinant proteins, peptides and chimeric proteins antibody based tests. Systematic scoping review.

BACKGROUND: Traditional diagnostic tests for schistosome infections are suboptimal, particularly when the parasite burden is low. In the present review we sought to identify recombinant proteins, peptides, and chimeric proteins with potential to be used as sensitive and specific diagnostic tools for schistosomiasis. METHODS: The review was guided by PRISMA-ScR guidelines, Arksey and O'Malley's framework, and guidelines from the Joanna Briggs Institute. Five databases were searched: Cochrane library, PubMed, EMBASE, PsycInfo and CINAHL, alongside preprints. Identified literature were assessed

Diagnosis of propofol allergy (the innovative role of the basophil activation test in the identification of allergen): a case report.

Drug allergies (DA), particularly to anesthetics, are underexplored in Morocco. Traditional diagnostic methods such as skin tests and specific IgE assays are costly, risky, and often inaccessible. The basophil activation test (BAT) is emerging as a safer and more effective alternative, especially for diagnosing propofol allergy, an under-researched area. This study examines the role of the basophils activation test in diagnosing perioperative anaphylaxis to propofol. A 32-year-old woman experienced two anaphylactic reactions during fibroscopy after propofol administration. The basophil activat

[Basophil activation test--a practical approach to diagnosis of common respiratory allergy].

BACKGROUND: The diagnosis of immediate allergy is based on clinical data, skin prick tests (SPT), and measurements of allergen-specific IgE (sIgE). Basophil activation test (BAT) can supplement these methods and obviate their disadvantages, and possibly replace allergen challenge tests, such as a nasal provocative test (NPT). In this study, we assessed the influence of different storage times on BAT results. Futhermore, we compared the results of SPT, sIgE and NPT against BAT for common aeroallergens. METHODS: BAT was performed in twelve patients with allergic rhinitis sensitized to birch pol

Cooperative role for tetraspanins in adhesin-mediated attachment of bacterial species to human epithelial cells.

The tetraspanins are a superfamily of transmembrane proteins with diverse functions and can form extended microdomains within the plasma membrane in conjunction with partner proteins, which probably includes receptors for bacterial adhesins. Neisseria meningitidis, the causative agent of meningococcal disease, attaches to host nasopharyngeal epithelial cells via type IV pili and opacity (Opa) proteins. We examined the role of tetraspanin function in Neisseria meningitidis adherence to epithelial cells. Tetraspanins CD9, CD63, and CD151 were expressed by HEC-1-B and DETROIT 562 cells. Coincubat

Label-free electrical detection of salivary extracellular vesicles using an anti-CD63-functionalized gold extended-gate field-effect transistor.

Salivary extracellular vesicles (sEVs) are accessible candidates for non-invasive biomarker analysis, but their measurement commonly requires multistep, label-dependent workflows. Here a label-free gold extended-gate field-effect transistor (EG-FET) functionalized with an antibody that binds specifically to the Cluster of Differentiation 63 (anti-CD63), was developed for electrical detection of isolated salivary sEVs. Gold gates were modified with an 11-mercaptoundecanoic acid self-assembled monolayer and [1-Ethyl-3-(3-Dimethylaminopropyl) carbodiimide/N-Hydroxysuccinimide] EDC/NHS-mediated an

Exercise affects systemic basophil responses in humans: Applying an automated and modified basophil activation test (mBAT) in a field study.

Basophils are effector cells in type I hypersensitivity reactions. Upon cross-linking of surface-bound (allergen-specific) IgE, basophils release a variety of mediators. Determining the activation of basophils ex vivo with anti-IgE (a basophil activation test or BAT) can be achieved using flow cytometry. However, this requires an experienced laboratory, hampering penetration of the BAT into general allergen diagnostics. Automation of the BAT could make this requirement redundant. Automation of the BAT enables rapid analysis in a field study setting of the effect of prolonged endurance exercise

[C-terminal lysosome targeting domain of CD63 modifies cellular localization of rabies virus glycoprotein].

The glycoprotein of rabies virus is the central antigen elicited the immune response to infection; therefore, the majority of developing anti-rabies vaccines are based on this protein. In order to increase the efficacy of DNA immunogen encoding rabies virus glycoprotein, the construction of chimeric protein with the CD63 domain has been proposed. The CD63 is a transmembrane protein localized on the cell surface and in lysosomes. The lysosome targeting motif GYEVM is located at its C-terminus. We used the domain that bears this motif (c-CD63) to generate chimeric glycoprotein in order to reloca

Association of the transmembrane 4 superfamily molecule CD53 with a tyrosine phosphatase activity.

Cell surface proteins of the transmembrane 4 superfamily (TM4SF) are a newly characterized family of proteins which are presumed to span the plasma membrane four times. The function of this family of molecules is poorly understood, but based on monoclonal antibody studies there is some evidence that they may be involved in transmembrane signal transduction and regulation of cell proliferation, differentiation, or both, in a number of different cell types. CD53 is a member of this family that is expressed on leukocytes, and transduces activation signals through unknown mechanisms that may invol

The Basophil Activation Test in Allergy Diagnostics: Clinical Applications, Methodological Challenges, and Proposed Clinical Frameworks.

The basophil activation test (BAT) is a modern in vitro functional assay used for the diagnosis of immunoglobulin E-mediated allergic reactions. The method is based on flow cytometric assessment of activation marker expression on peripheral blood basophils after stimulation with specific allergens. BAT has gained increasing clinical relevance in allergology, particularly in patients with anaphylaxis risk, polysensitization, drug hypersensitivity, and inconclusive skin or serological test results. This narrative review summarizes current evidence on the diagnostic performance and clinical appli

Pharmacological Inhibition of Small Extracellular Vesicle Secretion by ALK5i SD-208 via Lysosomal Rerouting of CD63+ Compartments.

Pharmacological tools to selectively modulate extracellular vesicle (EV) secretion are scarce. Here, we identify the ALK5 (TGF-β receptor I) inhibitor SD-208 as a potent suppressor of small EV (sEV) secretion that acts independently of its canonical anti-fibrotic activity. SD-208 not only reversed myofibroblast activation but also markedly inhibited sEV secretion. Strikingly, this inhibitory effect persisted in non-activated cardiac fibroblasts and non-fibrotic HEK293 cells, demonstrating that SD-208 regulates EV secretion through mechanisms uncoupled from TGF-β/Smad signalling. Mechanistic an

CD63 is a diagnostic marker of prostate cancer and a prognostic marker of biochemical progression following radical prostatectomy.

AIMS: We aimed to analyze CD63, a cell surface protein that has been associated with tumor aggressiveness in several cancers, including breast, colorectal, and lung cancer, as well as melanoma, in prostate cancer. METHODS: CD63 expression was analyzed immunohistochemically in a cohort of primary prostate cancers from 281 patients. The results were correlated with clinico-pathologic parameters, including biochemical recurrence. In addition, CD63 expression in 251 of the 281 patients with prostate cancer was compared with CD63 expression in matched benign tissue samples (490 tissue samples). Th

Dual-mode exosome detection leveraging a nanozyme-active artificial receptor: PDA@Fe@Zn-based nucleic acid aptamer sensor.

Exosomes, extracellular vesicles crucial for intercellular communication, are emerging as significant biomarkers for disease diagnosis, especially in cancer. This study presented a dual-mode exosome detection platform using polydopamine microspheres doped with iron and zinc ions (PDA@Fe@Zn). These materials served as both artificial receptors for nucleic acid aptamers and nanozymes with peroxidase-like activity. By integrating colorimetric and fluorescence detection, the platform enables cross-validation of results. PDA@Fe@Zn nanozymes catalyzed the TMB-H2O2 reaction under acidic conditions, p

PAM-assembled CRISPR-Cas12a activation-based fluorescent and colorimetric dual-modal biosensor for detecting prostate cancer exosomes.

Prostate specific membrane antigen (PSMA)-positive exosomes hold significant potential for the diagnosis and risk assessment of prostate cancer. However, accurate detection is severely hindered by their low abundance in blood and interference from similarly sized particles. In this study, we have developed a protospacer adjacent motif (PAM)-assembled clustered regularly interspaced short palindromic repeat (CRISPR)-Cas12a activation-based fluorescent and colorimetric dual-modal biosensor for the highly sensitive detection of PSMA-positive exosomes. In this work, two split strands respectively

Streptococcus pneumoniae invades endothelial host cells via multiple pathways and is killed in a lysosome dependent manner.

Streptococcus pneumoniae is one of the major causative agents of pneumonia, sepsis, meningitis and other morbidities. In spite of its heavy disease burden, surprisingly little is known about the mechanisms involved in the switch of life style, from commensal colonizer of the nasopharynx to invasive pathogen. In vitro experiments, and mouse models have shown that S. pneumoniae can be internalized by host cells, which coupled with intracellular vesicle transport through the cells, i.e. transcytosis, is suggested to be the first step of invasive disease. To further dissect the process of S. pneum

IgE Nonresponders in Basophil Activation Tests: A Narrative Review From Mechanistic Insights to Clinical Implications and Future Directions.

It has been known for about 50 years that a proportion of human basophils does not release histamine or leukotrienes or upregulates activation markers in response to IgE-dependent stimuli. This group of so-called nonresponders with unresponsive basophils is found in up to 20% of tests conducted. There seems to be no relation to clinical symptoms. The nonresponder status is not defined in a standardized manner, but basophils are often considered nonresponsive if they release less than 5% to 10% of their histamine content or show activation of less than or equal to 10% CD63+ basophils after FcεR

Lysosomal Targeting of Cystinosin Requires AP-3.

Cystinosin is a lysosomal cystine transporter defective in cystinosis, an autosomal recessive lysosomal storage disorder. It is composed of seven transmembrane (TM) domains and contains two lysosomal targeting motifs: a tyrosine-based signal (GYDQL) in its C-terminal tail and a non-classical motif in its fifth inter-TM loop. Using the yeast two-hybrid system, we showed that the GYDQL motif specifically interacted with the μ subunit of the adaptor protein complex 3 (AP-3). Moreover, cell surface biotinylation and total internal reflection fluorescence microscopy revealed that cystinosin was par

Nonneural granular cell tumor of the oral cavity: a case report and review of the literature.

Nonneural granular cell tumors (NNGCTs) are rare benign neoplasms originally described in 1991 by Leboit et al. Typical granular cell tumors (GCTs) are commonly encountered in the oral cavity, but NNGCTs, unlike GCTs, are S-100 negative and may display cytologic atypia, allowing for misdiagnosis as a more aggressive lesion. We report a case of a 43-year-old male with a lesion of the mandible that we believe to be the first intraoral example of an NNGCT.
