The role of the thalamus and basal ganglia in parkinsonian tremor.

The mechanism of parkinsonian tremor may involve a central oscillator, peripheral feedback to the central nervous system (CNS), or both. The thalamus or the globus pallidus is the most likely site for a central oscillator and would be predicted to generate thalamic tremor-related activity characterized, respectively, by calcium spike-associated bursts and by maximal tremor-related activity in the pallidal relay nucleus of thalamus. Thalamic spike trains demonstrate neither of these characteristics. However, cross-correlation, latency, and transfer function analysis indicate that sensory feedba…

Adrenergic innervation of the monkey thalamus: an immunohistochemical study.

The distribution and function of the neurotransmitter adrenaline in the primate brain are poorly understood. Biochemical studies have shown the presence of adrenaline or its biosynthetic enzyme, phenylethanolamine-N-methyltransferase, in the rat and human thalamus. However, the distribution of the adrenergic fibres in the thalamus has only been demonstrated in rats. We study the adrenergic innervation of the macaque monkey thalamus using immunohistochemistry against phenyletanolamine-N-methyltransferase. The distribution of phenyletanolamine-N-methyltransferase-immunoreactive fibres is markedl…

Apoptosis is restricted to the thalamus in thiamine-deficient rats.

Thiamine deficiency (TD) produces lesions in the thalamus, mamillary and medial geniculate nuclei, and inferior colliculus. To clarify the pathogenesis of these lesions, we examined the occurrence of hallmarks of apoptosis following TD in rat brain. Histological assessment showed apoptotic cells in the thalamus and medial geniculate nucleus but not in the inferior colliculus. We used terminal deoxynucleotidyl transferase-mediated deoxyuridine (dUPT)-biotin nick-end labelling (TUNEL) and gel electrophoresis to demonstrate that TD is associated with apoptotic cell death. In the thalamus, DNA fra…

Plasticity in human somatosensory thalamus as a result of deafferentation.

Experimental studies indicate that deafferentation results in reorganization of the somatosensory map at various levels of the CNS, such that the representation of a body part adjacent to a region that is denervated expands into the deafferented area. Recent data suggest that in the human this occurs at the cortical level, but subcortical structures have not been systematically investigated. To test the hypothesis that the human thalamus is capable of significant reorganization as a result of changes in afferent input, microelectrode recording and stimulating techniques were used to define tha…

Hypoperfusion and vasoreactivity in the thalamus and cerebellum after stroke.

BACKGROUND AND PURPOSE: Cerebellar hypoperfusion in the contralateral hemisphere after stroke is well studied and termed crossed cerebellar diaschisis. However, studies of hypoperfusion in the ipsilateral thalamus have been few. The purpose of this study was to investigate the prevalence of hypoperfusion and vasoreactivity to acetazolamide in the thalamus and cerebellum after stroke. METHODS: We studied cerebral blood perfusion in the thalamus and cerebellum of 14 patients with unilateral cerebral infarction using [123I]isopropyliodoamphetamine single-photon emission computed tomography (123I…

Magnetic resonance angiography of arteriovenous malformation in the thalamus.

A comparative study of magnetic resonance angiography and conventional angiography of arteriovenous malformation in the thalamus showed that both methods clearly visualized the feeding arteries: perforating branches of the posterior cerebral artery, posterior choroidal artery and lenticulostriate artery. Draining veins such as the internal cerebral vein were also demonstrated well.

Movement disorders in astrocytomas of the basal ganglia and the thalamus.

In a series of 225 patients with astrocytomas (grades I-IV) of the basal ganglia and the thalamus, 20 had a movement disorder. In all patients the histological diagnosis was verified by stereotactic biopsy. Tremor was observed in twelve patients, dystonia in eight, chorea in three, and chorea/ballismus and myoclonus in one. The tumour involved the thalamus in 16 patients. Corticospinal tract dysfunction was evident in 70% of the patients with movement disorders and in 73% of those without. Demographic, clinical, histological and neuroradiological data of the patients with a movement disorder w…

Noradrenergic innervation of somatosensory thalamus and spinal cord.

Monoamine systems have been shown to be an important part of an endogenous analgesic system of the central nervous system. Some aspects of the anatomical basis of monoamine modulation of nociceptive input were investigated in these studies. Two sites examined where monoamine systems are known to impinge on the pain transmission system included the grey matter of the somatosensory thalamus and the spinal cord. In particular, the connections of noradrenergic systems with these regions were emphasized. In the ventral posterolateral nucleus of the thalamus the presence of a sparse innervation by b…

Characterization of GABAergic seizure regulation in the midline thalamus.

This study characterized the role of GABA in the central medial intralaminar nucleus on seizures induced by pentylenetetrazol given systemically. Injections of the direct selective GABAA agonist, piperidine-4-sulfonic acid or the indirect GABAA agonists, flurazepam and pentobarbital, in this region depressed arousal and facilitated myoclonic and clonic seizures induced by pentylenetetrazol but only caused slight inhibition of tonic seizures. In contrast the GABAB agonist (-)baclofen facilitated all three types of seizures. Recording after injection of piperidine-4-sulfonic acid and (-)baclofen

Fastigial nucleus projections in the midbrain and thalamus in dogs.

Efferent connections to midbrain and thalamus from portions of the cerebellar fastigial nucleus were investigated using autoradiographic techniques. Bipolar stimulating electrodes were placed in the fastigial nucleus of anesthetized beagles and the area which produced maximal increases in blood pressure and heart rate was localized in each dog. A mixture of [3H]leucine and [3H]proline (4:1) was injected into that area and autoradiograms were prepared. Injections filled the rostral and various parts of the caudal fastigial nucleus. The rostral-caudal extent of injection sites were mapped in the…

System degeneration of the thalamus. A clinico-neuropathological study.

A case of essential degeneration of the thalamus is reported. The patient was a 43-year-old Japanese male, who, a few weeks after mild head trauma, suffered from forgetfulness, psychomotor slowing, and Korsakoff's syndrome. Four to five months later, there were optical hallucinations and delirium and he died 19 months after the onset of symptoms. Neuropathological examination revealed symmetrical thalamic degeneration, whose distribution corresponded to phylogenetically younger subunits of the thalamus. In addition, there was olivovermian degeneration. These findings are identical to those of …

Unveiling Functional Architecture of Human Thalamus: AStereoelectroencephalography Study.

As a critical hub in the thalamo-cortical circuit, the human thalamus engages in a spectrum of fundamental and advanced brain functions through widely distributed circuits. Clinically, dysfunction of thalamo-cortical circuits is shown to be profoundly implicated in a wide range of neurological and psychiatric diseases. Nevertheless, the neuroanatomical substrates governing these functions of the thalamus have rarely been directly mapped in humans. Here, we overviewed the acute responses of direct electrical stimulation (DES) delivered to the distributed thalamus sites in 52 epilepsy patients a…

Plant Material in the Thalamus and Mesencephalon of a Young Dog.

An unusual case of neurological deficits due to intracranial plant material in the thalamus is presented. The case was a young dog with pyrexia, neck pain, and neurological deficits that were consistent with an intracranial multifocal lesion. Based on the imaging findings and the result of the cerebrospinal fluid analysis, a presumptive diagnosis of immune-mediated meningoencephalitis was made. The dog initially responded to immunosuppressive treatment but then deteriorated and was euthanized. At autopsy, plant material was found in the thalamus and mesencephalon. Intracranial foreign bodies a…

From our prior observations, it remains unclear whether the disuse plasticity affects the thalamus and striatum. We extended our analysis to include these subcortical regions and found that both exhibit strengthened cortical FC and spontaneous fMRI signal pulses induced by limb disuse. Network Architecture of Verticality Processing in the Human Thalamus.

OBJECTIVE: Thalamic dysfunction in lesions or neurodegeneration may alter verticality perception and lead to postural imbalance and falls. The aim of the current study was to delineate the structural and functional connectivity network architecture of the vestibular representations in the thalamus by multimodal magnetic resonance imaging. METHODS: Seventy-four patients with acute unilateral isolated thalamic infarcts were studied prospectively with emphasis on the perception of verticality (tilts of the subjective visual vertical [SVV]). We used multivariate lesion-symptom mapping based on su…

[Association between the Thalamus and Social Function in Schizophrenia].

Patients with schizophrenia face several difficulties in their daily social life owing to the impairment of social cognitive ability. However, the neural basis of this impairment has not been fully clarified. We attempted to identify the neuronal basis of social dysfunction of schizophrenia by focusing on the thalamus, which is a subcortical structure. We have shown that right thalamic volume, as well as white matter integrity between the right thalamus and right frontal lobe, is associated with the social function of patients with schizophrenia, by using T1-weighted magnetic resonance imaging…

Evaluation of functional connectivity in subdivisions of the thalamus in schizophrenia.

BACKGROUND: Previous studies in schizophrenia revealed abnormalities in the cortico-cerebellar-thalamo-cortical circuit (CCTCC) pathway, suggesting the necessity for defining thalamic subdivisions in understanding alterations of brain connectivity.AimsTo parcellate the thalamus into several subdivisions using a data-driven method, and to evaluate the role of each subdivision in the alterations of CCTCC functional connectivity in patients with schizophrenia. METHOD: There were 54 patients with schizophrenia and 42 healthy controls included in this study. First, the thalamic structural and func…
