Drug resistance is one of the emerging and re-emerging epidemics affecting both veterinary and public health sectors. Buparvaquone provides the most satisfactory means in the treatment of bovine tropical theileriosis. However, recently there has been widespread reports of development of resistance of Theileria annulata to buparvaquone. To investigate the situation in Sudan where bovine tropical theileriosis is endemic, fifty blood samples from T. annulata-positive cattle. were used for DNA extraction, PCR and cytochrome b gene nucleotide sequencing. Analysis of the two buparvaquone binding sit

Tropical theileriosis, caused by the protozoan parasite Theileria annulata and transmitted by several species of ixodid ticks of the genus Hyalomma, is an economically important disease of bovines. Concerningly, studies conducted in recent years have shown an increase in the rate of failure when using the primary drug of treatment, buparvaquone (BPQ), particularly in infection caused by T. annulata populations bearing V135A and P253S mutations on the Cytochrome b (Cyto b) gene of the parasite mitochondrial genome. The aim of this study was to demonstrate the relationship between BPQ-resistance

Babesia bigemina and Theileria annulata are tick-borne protozoans that cause piroplasmosis in cattle, resulting in huge damages to the livestock industry. The prevalence of these infections depends on various intrinsic and extrinsic risk factors. In Pakistan, there is no information regarding the molecular characterization of Babesia bigemina and the risk factors associated with piroplasmosis. This study aimed to molecularly characterize Babesia spp. and Theileria spp. infecting various cattle breeds in Khyber Pakhtunkhwa, Pakistan, and to shed light on risk factors associated with these infec

Tropical theileriosis is a disease caused by infection with an apicomplexan parasite, Theileria annulata, and giving rise to huge economic losses. In recent years, parasite resistance has been reported against the most effective antitheilerial drug used for the treatment of this disease. This emphasizes the need for alternative methods of treatment. Enolase is a key glycolytic enzyme and can be selected as a macromolecular target of therapy of tropical theileriosis. In this study, an intron sequence present in T. annulata enolase gene was removed by PCR-directed mutagenesis, and the gene was f

Understanding the progression of Theileria annulata infection is essential for effective treatment and control of theileriosis transmission. A PCR method was used to detect the parasite's sporozoite surface protein gene (TaSP), and the results were compared with two quantitative indirect ELISAs targeting stage-specific antibodies. The early-stage ELISA focused on sporozoite-stage antibodies, while the late-stage ELISA targeted merozoite-stage antibodies based on the TaSP linear B cell peptide epitope and rTaMS protein, respectively, to identify life stages (sporozoite and merozoite) and diseas

BACKGROUND: Theileria annulata, a tick-borne protozoan that causes tropical theileriosis, poses a serious threat to livestock production in endemic regions. The emergence of resistance to buparvaquone, the primary chemotherapeutic treatment, has been attributed to acquired mutations in the cytochrome b (Cytb) gene, with identical resistance-associated polymorphisms observed in both laboratory-adapted strains and field isolates from China.
METHODS: A dual probe-specific real-time polymerase chain reaction (PCR) assay was developed to detect point mutations in the Cytb gene. The specificity, se

Ticks are economically important obligatory blood feeding arthropods that have a pivotal role in transmission of infection. The present study was conducted in ixodid ticks collected from four districts of coastal Odisha, India to investigate the prevalence of Theileria annulata. Adult semi engorged Hyalomma anatolicum ticks (n = 178) were dissected, the salivary gland was isolated and DNA was extracted. A nested PCR targeting the Tams1 gene of T. annulata, utilizing two sets of primers (N516F, N517R, and Ta14136iF, Ta249R) was utilized for detection of the parasite. The PCR products were then

The endogenous development of Theileria annulata in the vector tick--Hyalomma anatolicum anatolicum has been described. Parasites within and outside the erythrocytes were seen in the gut contents of larvae and nymphs immediately after their removal from the infected cattle but not after 24 hours. Two bodies resembling the intra-erythrocytic stage of the parasite, lying close to each other were seen near the gut epithelium of a larva on the 4th day of its dropping off the infected host. Neither multiplication nor union of the parasite resulting in zygote formation in the gut of the tick, was no

Tropical theileriosis, caused by Theileria annulata, continues to be the most significant tick-borne disease affecting cattle, particularly in endemic regions where it leads to significant production losses. Prompt and reliable diagnosis is critically important for timely treatment and disease control; however, conventional diagnostic methods have important limitations. Microscopic examination lacks sensitivity, particularly in carrier animals, while PCR requires specialized laboratory infrastructure. To overcome these limitations, a fluorescence-based RPA-CRISPR/Cas12a assay targeting the mul

Exophthalmia is an important but poorly documented complication of tropical theileriosis caused by Theileria annulata, especially in neonatal and young calves. It involves forward displacement of the eyeball, periorbital swelling, conjunctival congestion, excessive tearing, and enlarged regional lymph nodes. Systemic signs like fever, anaemia, depression, and weakness often occur. The condition arises through complex pathogenetic mechanisms linked to the schizont stage of the parasite, where infected leukocytes transform into rapidly dividing, invasive cells. These cells infiltrate retro-orbit

Ixodid ticks are vectors of pathogens that cause dangerous infectious and parasitic diseases in animals, leading to reduced productivity and, in some cases, mass mortality. In Kazakhstan, information on tick fauna and their epizootological role in the transmission of hemoparasites is limited. This study aimed to determine the species composition of ixodid ticks parasitizing cattle in the Almaty, Zhambyl, and Turkistan oblasts, and to assess their potential role in the transmission of Theileria and Anaplasma spp. A total of 3121 ixodid ticks were collected from cattle, belonging to the genera H

Hyalomma anatolicum is a tick of significant one-health importance due to its role as a vector for various pathogens affecting humans, animals and the environment, such as Theileria annulata, which causes tropical theileriosis in cattle, leading to severe economic losses. When infected with pathogens like T. annulata, the salivary glands of H. anatolicum undergo gene expression changes, secrete modified proteins and activate immune responses, all of which facilitate pathogen survival and transmission by modulating the host immune response and optimizing conditions for pathogen development. BACKGROUND AND AIM: Babesia divergens causes human babesiosis in Europe where the parasite utilizes cattle as animal reservoir and Ixodes ricinus as tick vector. Importation of infected animals and passive carriage of infected ticks through migratory birds can lead to tick/pathogen geographic expansion and emergence of diseases in naïve land. Given the information that Saudi Arabia imports cattle from the European countries and that two global bird flyways pass through the country geographic coordinates, we speculate that B. divergens might be introduced into the Kingdom. Therefore, the aim of

Tropical theileriosis, bovine babesiosis and anaplasmosis are tick-borne protozoan diseases that impose serious constraints on the health and productivity of domestic cattle in tropical and sub-tropical regions of the world. A common feature of these diseases is that, following recovery from primary infection, animals become persistent carriers of the pathogen and continue to play a critical role in disease epidemiology, acting as reservoirs of infection. This study describes development and evaluation of multiplex and single PCR assays for simultaneous detection of Theileria annulata, Babesia

This study analyzed the molecular and phylogenetic profiles of Theileria annulata, the causative agent of tropical theileriosis in cattle, and its tick vector Hyalomma scupense in Algeria. Forty H. scupense ticks were collected in Medea, with 5 testing positive for Theileria spp. based on partial COXIII gene sequences. Positive ticks were further analyzed using COX1 and 12S rRNA genes. Two novel H. scupense 12S rRNA haplotypes and one novel COX1 haplotype were identified. One T. annulata haplotype previously reported in Algerian cattle was detected. This represents the first molecular characte

BACKGROUND: Bovine theileriosis caused by the eukaryotic parasite Theileria annulata is an economically important tick-borne disease. If it is not treated promptly, this lymphoproliferative disease has a significant fatality rate. Buparvaquone (BPQ) is the only chemotherapy-based treatment available right now. However, with the emergence of BPQ resistance on the rise and no backup therapy available, it is critical to identify imperative drugs and new targets against Theileria parasites.
