Multitarget Transcranial Ultrasound Therapy in Small Animals Based on Phase-Only Acoustic Holographic Lens.

Transcranial ultrasound therapy has become a noninvasive method for treating neurological and psychiatric disorders, and studies have further demonstrated that multitarget transcranial ultrasound therapy is a better solution. At present, multitarget transcranial ultrasound therapy in small animals can only be achieved by the multitransducer or phased array. However, multiple transducers may cause spatial interference, and the phased array system is complicated, expensive, and especially unsuitable for small animals. This study is the first to design and fabricate a miniature acoustic holograph

Unaltered neuronal and glial counts in animal models of magnetic seizure therapy and electroconvulsive therapy.

Anatomical evidence of brain damage from electroconvulsive therapy (ECT) is lacking; but there are no modern stereological studies in primates documenting its safety. Magnetic seizure therapy (MST) is under development as a less invasive form of convulsive therapy, and there is only one prior report on its anatomical effects. We discerned no histological lesions in the brains of higher mammals subjected to electroconvulsive shock (ECS) or MST, under conditions that model closely those used in humans. We sought to extend these findings by determining whether these interventions affected the num

Veterinary Spinal Manipulative Therapy or Animal Chiropractic in Veterinary Rehabilitation.

Veterinary rehabilitation is a multimodal diagnostic and treatment approach that is recommended and provided to patients daily. One therapeutic modality that may be beneficial (diagnostically and therapeutically) is veterinary spinal manipulative therapy or animal chiropractic (AC). AC is a receptor-based health-care modality being provided more frequently in veterinary practices. All clinicians should strive to understand the mode of action, indications, contraindications, how it affects the patient from the neuro-anatomical and biomechanical point of view, and most importantly, when not to p

Animal models for researching approaches to therapy of Duchenne muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a relatively widespread genetic disease which develops as a result of a mutation in the gene DMD encoding dystrophin. In this review, animal models of DMD are described. These models are used in preclinical studies to elucidate the pathogenesis of the disease or to develop effective treatments; each animal model has its own advantages and disadvantages. For instance, Caenorhabditis elegans, Drosophila melanogaster, and zebrafish (sapje) are suitable for large-scale chemical screening of large numbers of small molecules, but their disease phenotype differs f

Transgenic animal models for study of the pathogenesis of Huntington's disease and therapy.

Huntington's disease (HD) is caused by a genetic mutation that results in polyglutamine expansion in the N-terminal regions of huntingtin. As a result, this polyQ expansion leads to the misfolding and aggregation of mutant huntingtin as well as age-dependent neurodegeneration. The genetic mutation in HD allows for generating a variety of animal models that express different forms of mutant huntingtin and show differential pathology. Studies of these animal models have provided an important insight into the pathogenesis of HD. Mouse models of HD include transgenic mice, which express N-terminal

Large animal models and new therapies for glycogen storage disease.

Glycogen storage diseases (GSD), a unique category of inherited metabolic disorders, were first described early in the twentieth century. Since then, the biochemical and genetic bases of these disorders have been determined, and an increasing number of animal models for GSD have become available. At least seven large mammalian models have been developed for laboratory research on GSDs. These models have facilitated the development of new therapies, including gene therapy, which are undergoing clinical translation. For example, gene therapy prolonged survival and prevented hypoglycemia during f

Novel oral drug administration in an animal model of neuroleptic therapy.

A novel method of oral drug administration was used in a neuroleptic animal study. Seventy male Sprague-Dawley rats were randomly subdivided into four groups, which were treated with clozapine, haloperidol, diazepam or a vehicle solution (5% sucrose solution). Oral drug treatment was achieved by training the rats to drink the drug of choice mixed with five percent sucrose or vehicle solution from a syringe. Within 3-4 weeks the haloperidol group developed vacuous chewing movement, which did not disappear with discontinuation of the drug. Significant weight gain was observed for all drug groups

Valvular heart disease: medical therapy and experimental and animal models.

Vasodilators have been advocated for the treatment of regurgitant valvular heart diseases for more than 12 years, and new information on their mechanisms of action is available. Anticoagulants continue as a mainstay of therapy for patients with prosthetic heart valves, and the most desirable levels of anticoagulation for different conditions have been examined. Clinical and animal research studies of cardiac adaptation to mitral and aortic valve diseases are discussed.

Immunoglobulin therapy for neonatal sepsis: an overview of animal and clinical studies.

Bacteria such as Escherichia coli and group B streptococci are common neonatal pathogens. Neonates infected perinatally often develop overwhelming sepsis which may rapidly progress to shock and death in just a few hours. This fulminant course suggests that immunity to these bacteria is deficient. Studies from several laboratories have shown that antibody is important in immunity to group B streptococcus. In vitro studies have demonstrated that efficient phagocytosis and killing of these bacteria by neutrophils or monocytes requires opsonic antibody. Many premature babies have decreased amounts

Stem cell-derived exosomes as a potential therapy for schistosomal hepatic fibrosis in experimental animals.

Schistosomiasis is a neglected tropical disease. Egg-induced granuloma formation and tissue fibrosis are the main causes of the high morbidity and mortality of schistosomiasis. Mesenchymal stem cells (MSCs)-derived exosomes play an important role with a superior safety profile than MSCs in the treatment of liver fibrosis. Therefore, the aim of this study was to investigate the potential therapeutic effect of MSCs-derived exosomes on schistosomal hepatic fibrosis. Exosomes were isolated from bone marrow MSCs and characterized. A total of 85 mice were divided into four groups: group I (control g

Enhanced Bone Regeneration by Diabetic Cell-Based Adenoviral BMP-2 Gene Therapy in Diabetic Animals.

The application of bone morphogenetic protein 2 (BMP-2) has been extensively investigated to improve diabetes-impaired bone healing; however, the delivery of BMP-2 by gene therapy for bone regeneration has rarely been investigated in diabetic animals. In this study, we aimed to evaluate which cells induce more new bone formation in diabetic animals when cell-based BMP2 gene therapy is applied. For this purpose, we harvested bone marrow stromal cells (BMSCs) twice in the same animal before (non-diabetic BMSCs; nBMSCs) and after diabetes induction (diabetic BMSCs; dBMSCs) using modified bone mar

Epididymal phenotype in luteinizing hormone receptor knockout animals and its response to testosterone replacement therapy.

Previous studies reported that epididymis contains functional LH receptors. The LH receptor knockout mice, which have epididymal phenotypes, gave us an opportunity to test the hypothesis that testosterone replacement alone may not be sufficient to reverse phenotypes to wild-type epididymis. The morphological phenotype in knockout animals includes a decrease in luminal diameter of the proximal and distal caput and cauda epididymis, the absence of clear and halo cells in the epithelial lining, a decrease in the height of principal cells and the number of cells containing cilia, a decrease in cil

[Relation between slow virus infection of animals and man, their pathogenicity and resistance to therapy].

Besides conventional slow viruses, a category of pathogens exist, called "unconventional slow viruses". The latter evoke laughing death syndrome, Creutzfeldt-Jakob disease (resp. its special form of Gerstmann-Sträussler's syndrome) in man, in animal scrapie, infectious encephalopathy of the mink, and chronic wasting disease. They all are relatives, and their characteristics are longtime incubation, lacking immune reactions, high chemical and physical resistance, no proof of nucleic acids till now, a lacking proof in electron microscopy as well as a ceasing change of infectivity by immunologic

Use of human and animal amniotic membranes in local antibiotic therapy.

OBJECTIVE: The antibacterial properties of amniotic membranes are the reason for their wide clinical use. Amniotic membrane soaked in antibiotics can be used in local antibiotic therapy, creating new options for the treatment of infections. The aim of this study was to analyse the inhibiting effect of both human and porcine amniotic membranes soaked in antibiotics on the growth of microorganisms. METHOD: Human and porcine placentas were collected during natural births, under aseptic conditions. Each amnion was divided into three parts: intravital; cryopreserved; and radio-sterilised. Gene therapy with AAV9-SGPL1 in an animal model of lung fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive scarring disease of the lung that leads rapidly to respiratory failure. Novel approaches to treatment are urgently needed. The bioactive lipid sphingosine-1-phosphate (S1P) is increased in IPF lungs and promotes proinflammatory and profibrotic TGF-β signaling. Hence, decreasing lung S1P represents a potential therapeutic strategy for IPF. S1P is degraded by the intracellular enzyme S1P lyase (SPL). Here we find that a knock-in mouse with a missense SPL mutation mimicking human disease resulted in reduced SPL activity, increased S1P, increase

Animal assisted therapy in pediatric mental health conditions: A review.

Animal assisted therapy (AAT) has been explored as a treatment option for children with mental health disorders and associated symptoms. AAT can be implemented in inpatient, outpatient, and residential care settings. Some studies demonstrate significant improvement in symptom severity while others report only modest results. In this review, we summarize previous studies that have investigated the applications of AAT for pediatric mental health treatment, focusing on management of symptoms related to post-traumatic stress disorder, depression, anxiety, attention-deficit/hyperactivity disorder,
