[Eleven-Cities skating marathon and the windchill factor].

The weather conditions during the 15th Eleven Cities ice skating marathon (200 kilometers) in Friesland (the Netherlands) on 4 January, 1997, were better than predicted. For measurement of the windchill factor the Steadman scale is preferred to the Siple and Passel scale. For ice skaters the windchill factor is lower than for the spectators, due to the drag effect.

Thermal Perception as a Key Factor for Assessing Effects of Trigeminal Nerve Injury.

AIMS: To conduct a functional examination using multimodal exploration of a sample of patients with iatrogenic trigeminal nerve injury to understand the underlying mechanisms of neuropathic pain following trigeminal nerve injury. METHODS: Subjective and objective symptoms and responses to thermal and electrical quantitative sensory testing (QST) were evaluated in 85 patients with unilateral trigeminal nerve injury. Objective symptoms were measured by seven clinical sensory tests. Thermal QST included cold detection threshold (CDT), warm detection threshold (WDT), and heat pain threshold (HPT)

Perception of temperature on oral and facial skin.

The intensity of sensations of warmth and cold was measured psychophysically at 12 loci on the face and in the mouth in 20 human subjects. Significant differences were found among areas in the relative sensitivity to both cooling and warming, although the range of sensitivities was greater for warming than for cooling. Except for the vermilion lip and the tongue tip, oral regions were significantly less sensitive to warming than were facial regions. No such difference was found for cooling. The most posterior location tested on the hard palate, for example, exhibited a suprathreshold sensitivi

Cold suppresses agonist-induced activation of TRPV1.

Cold therapy is frequently used to reduce pain and edema following acute injury or surgery such as tooth extraction. However, the neurobiological mechanisms of cold therapy are not completely understood. Transient receptor potential vanilloid 1 (TRPV1) is a capsaicin- and heat-gated nociceptive ion channel implicated in thermosensation and pathological pain under conditions of inflammation or injury. Although capsaicin-induced nociception, neuropeptide release, and ionic currents are suppressed by cold, it is not known if cold suppresses agonist-induced activation of recombinant TRPV1. We demo

A Reference Atlas of the Human Dorsal Root Ganglion.

Somatosensory perception largely emerges from diverse peripheral sensory neurons whose cell bodies reside in dorsal root ganglia (DRG). Damage or dysfunction of DRG neurons is a major cause of chronic pain and sensory loss. In mice, deep single-cell transcriptomic profiling and genetically defined models have offered important clues into DRG function, but in humans, the cellular and molecular landscape of DRG neurons remains less understood. Here, we constructed a reference cell atlas of the human DRG by profiling transcriptomes of cells and nuclei from 126 donors sampled across cervical, thor

The TRPM3 ion channel mediates nociception but not itch evoked by endogenous pruritogenic mediators.

During the molecular transduction of itch, the stimulation of pruriceptors on sensory fibers leads to the activation or sensitization of ion channels, which results in a consequent depolarization of the neurons. These ion channels mostly belong to the transient receptor potential (TRP) channels, which are involved in nociception and thermosensation. In particular, TRPV1 and TRPA1 were described in the transduction of both thermal nociception as well as histaminergic and non-histaminergic itch. The thermosensitive TRPM3 plays an indispensable role in heat nociception together with TRPV1 and TRP

Transient receptor potential A1 mediates an osmotically activated ion channel.

Transient receptor potential (TRP)A1 channel has been implicated in various physiological processes, including thermosensation and pain. A recent study of TRPA1 knockout mice demonstrated deficits in sensing mechanical stimuli, suggesting a role for TRPA1 also in somatic mechanosensation. However, direct evidence of TRPA1 activation by mechanical forces has thus far been lacking. Here we show, using an intracellular calcium assay, that hypertonic solution (HTS) activates TRPA1 channels in human embryonic kidney 293 cells transiently expressing rat TRPA1. In contrast, hypotonic solution has no

High-threshold mechanosensitive ion channels blocked by a novel conopeptide mediate pressure-evoked pain.

Little is known about the molecular basis of somatosensory mechanotransduction in mammals. We screened a library of peptide toxins for effects on mechanically activated currents in cultured dorsal root ganglion neurons. One conopeptide analogue, termed NMB-1 for noxious mechanosensation blocker 1, selectively inhibits (IC(50) 1 microM) sustained mechanically activated currents in a subset of sensory neurons. Biotinylated NMB-1 retains activity and binds selectively to peripherin-positive nociceptive sensory neurons. The selectivity of NMB-1 was confirmed by the fact that it has no inhibitory e

A New Paradigm for Precision Medicine in Heatstroke: From Molecular Subtyping to Targeted Prevention and Treatment.

Heatstroke is a life-threatening systemic disorder caused by extreme heat exposure and characterized by thermal toxicity, dysregulated inflammation, endothelial injury, and coagulation abnormalities. Despite timely cooling and supportive care, marked inter-individual heterogeneity in disease onset, dominant pathological processes, and outcomes remains a major barrier to early recognition and effective intervention. Increasing evidence indicates that heatstroke represents a spectrum of mechanistically distinct subtypes rather than a uniform clinical entity. This review summarizes advances relev

A Precise Temperature-Responsive Bistable Switch Controlling Yersinia Virulence.

Different biomolecules have been identified in bacterial pathogens that sense changes in temperature and trigger expression of virulence programs upon host entry. However, the dynamics and quantitative outcome of this response in individual cells of a population, and how this influences pathogenicity are unknown. Here, we address these questions using a thermosensing virulence regulator of an intestinal pathogen (RovA of Yersinia pseudotuberculosis) as a model. We reveal that this regulator is part of a novel thermoresponsive bistable switch, which leads to high- and low-invasive subpopulation

Inhibition of TRPM8 channels reduces pain in the cold pressor test in humans.

The transient receptor potential (subfamily M, member 8; TRPM8) is a nonselective cation channel localized in primary sensory neurons, and is a candidate for cold thermosensing, mediation of cold pain, and bladder overactivity. Studies with TRPM8 knockout mice and selective TRPM8 channel blockers demonstrate a lack of cold sensitivity and reduced cold pain in various rodent models. Furthermore, TRPM8 blockers significantly lower body temperature. We have identified a moderately potent (IC50 = 103 nM), selective TRPM8 antagonist, PF-05105679 [(R)-3-[(1-(4-fluorophenyl)ethyl)(quinolin-3-ylcarbon

High-density thermal sensitivity maps of the body of people with multiple sclerosis: Implications for inclusive personal comfort systems.

Inclusive thermal comfort solutions should accommodate the need of clinical groups such as people with Multiple Sclerosis (pwMS), who experience abnormal thermal sensitivity. The aim of this study was to develop high-density body maps of temperature sensitivity in pwMS to inform the design of patient-centred personal comfort systems. Fourteen pwMS (6 M/8 F; 48.6 ± 10.0 y) and 13 healthy individuals (CTR; 5 M/8 F; 47.8 ± 10.4) underwent a quantitative sensory test in a thermoneutral environment, during which they rated their local thermal sensations arising from the application of warm (39°C) a

A 43-year of human thermal comfort in Central Africa.

In this study, the human thermal stress was quantified across Central Africa (C.A.) using the Universal Thermal Climate Index (UTCI). Although many of the documented studies on the use of UTCI in relation to human health are currently restricted to countries in the northern hemisphere, this contribution constitutes a prerequisite of information for future research in the region. To mitigate the problem of lack of field data in the Central African sub-region, we downloaded UTCI data via the ERA5 reanalysis portal. Based on this data source, we have explored the spatiotemporal characteristics an

The posterior insular-opercular cortex: An access to the brain networks of thermosensory and nociceptive processes?

In spite of systematic investigations, the existence of a specific cortex that could encode for the intensities of somatosensory stimuli, including within nociceptive ranges, is still a matter of debate. The present consensus is that pain is expressed in a distributed network made of thalamus, SII, insula, ACC, and, less consistently, SI. Here we argument that there must be an entrance to this network. The common denominator to every functional imaging study is that the subjects can distinguish between noxious and non-noxious stimuli, or between two different intensities of noxious stimuli. Th

Influence of view factors on outdoor thermal comfort of residential areas in hot-humid regions.

Sky View Factor (SVF) is commonly used to describe the impact of urban geometry on the urban thermal environment. Shading effects from plants and buildings also exert a considerable influence. To investigate the influence of view factors on outdoor thermal comfort in residential areas, we employed the Physiological Equivalent Temperature (PET) and view factors (SVF, TVF, BVF) as indicators to determine outdoor thermal comfort and the quantity of shaded spaces. Thermal measurements collected from 13 points in Guangzhou, China, Our findings revealed that high TVF points exhibited more stable air

Analytical and subjective interpretation of thermal comfort in hospitals: A case study in two sterilization services.

Hospital facilities are normally very complex, which combined with patient requirements promote conditions for potential development of uncomfortable working conditions. Thermal discomfort is one such example. This study aimed to determine levels of thermal comfort, sensations, and preferences, from a field investigation conducted in two sterilization services (SS) of two hospitals from Porto and Aveiro, Portugal. The analytical determination and interpretation of thermal comfort was based upon assumptions of ISO 7726:1998 and ISO 7730:2005. The predicted mean vote (PMV) and predicted percenta

Contribution of skin thermal sensitivities of large body areas to sweating response.

The thermal sensitivity of different parts of the body was investigated by heating large areas of the body surface while the mean skin temperature calculated from Hardy and DuBois ' formula (1938) was always kept constant. The right arm sweating responses recorded under a local thermal clamp were related to changes in segmental skin temperatures of the different parts of the body. The results show that: 1) the various local peripheral signals are projected into integrating structures in the central nervous system; 2) the thermal sensitivity is greater for the head-and-trunk area in comparison

TMC6 functions as a GPCR-like receptor to sense noxious heat via Gαq signaling.

Thermosensation is vital for the survival, propagation, and adaption of all organisms, but its mechanism is not fully understood yet. Here, we find that TMC6, a membrane protein of unknown function, is highly expressed in dorsal root ganglion (DRG) neurons and functions as a Gαq-coupled G protein-coupled receptor (GPCR)-like receptor to sense noxious heat. TMC6-deficient mice display a substantial impairment in noxious heat sensation while maintaining normal perception of cold, warmth, touch, and mechanical pain. Further studies show that TMC6 interacts with Gαq via its intracellular C-termina

The role of transient receptor potential polycystin channels in bone diseases.

Transient receptor potential (TRP) channels are cation channels which act as molecular sensors that enable cells to detect and respond to a plethora of mechanical and environmental cues. TRPs are involved in various physiological processes, such as mechanosensation, non-inception and thermosensation, while mutations in genes encoding them can lead to pathological conditions, called "channelopathies". The subfamily of transient receptor potential polycystins (TRPPs), Polycystin 1 (PC1, TRPP1) and Polycystin 2 (PC2, TRPP2), act as mechanoreceptors, sensing external mechanical forces, including s
