Amphiphilic Sialic Acid Derivatives as Potential Dual-Specific Inhibitors of Influenza Hemagglutinin and Neuraminidase.

In the shadow of SARS-CoV-2, influenza seems to be an innocent virus, although new zoonotic influenza viruses evolved by mutations may lead to severe pandemics. According to WHO, there is an urgent need for better antiviral drugs. Blocking viral hemagglutinin with multivalent N-acetylneuraminic acid derivatives is a promising approach to prevent influenza infection. Moreover, dual inhibition of both hemagglutinin and neuraminidase may result in a more powerful effect. Since both viral glycoproteins can bind to neuraminic acid, we have prepared three series of amphiphilic self-assembling 2-thio

Synthetic and Immunological Studies of Mycobacterial Lipoarabinomannan Oligosaccharides and Their Protein Conjugates.

Lipoarabinomannan (LAM) is one of the major constituents of the Mycobacterium tuberculosis cell wall and an attractive molecular scaffold for antituberculosis drug and vaccine development. In this paper, a convergent strategy was developed for the synthesis of LAM oligosaccharides with an α-1,2-linked dimannopyranose cap at the nonreducing end. The strategy was highlighted by efficient coupling of separately prepared nonreducing end and reducing end oligosaccharides. Glycosylations were mainly achieved with thioglycoside donors, which gave excellent yields and stereoselectivity even for reacti

Visible-light-induced glycosyl radicals from diverse precursors for C-glycosylation.

C-Glycosides are crucial structural motifs in drug discovery and biology due to their enhanced metabolic stability and resistance to enzymatic hydrolysis compared to their O-glycoside counterparts. The synthesis of these molecules often relies on the generation of glycosyl radicals as key intermediates for forging C-C bonds at the anomeric center. In this context, visible-light photoredox catalysis has emerged as a transformative platform, enabling the mild, selective, and sustainable generation of glycosyl radicals from a diverse array of bench-stable precursors. This review provides a compre

A biomimetic phage-inspired supramolecular system based on glucose-conjugated pillar[5]arene with ciprofloxacin hydrochloride.

Self-assembling phage-inspired carriers based on macrocyclic molecules have the potential to overcome antibiotic resistance and extend the antimicrobial effect of drugs. In this study, we have synthesized a new water-soluble pillar[5]arene containing thioglycoside fragments. This compound had an affinity for a model lipid membrane, underwent self-assembly, and associated with the fluoroquinolone antibiotic ciprofloxacin hydrochloride (Cipro) to form biocompatible supramolecular nanostructures. UV-vis and fluorescence spectroscopies were used to assess the ability of the pillar[5]arene/antibiot

Floridoside Phosphotriester Derivatives: Synthesis and Inhibition of Human Neutrophils' Oxidative Burst.

Floridoside (2-O-D-glycerol-α-D-galactopyranoside) is a natural product typically found in red algae. It serves as the algae's carbon reserve and is produced as a protective response against osmotic and heat stress. Both floridoside and its acylated derivatives have been associated with modulating redox homeostasis and inflammatory responses. Therefore, we aimed to evaluate whether the newly synthesized floridoside phosphotriesters (1b-1d, 1f-1h) and acylated floridoside derivative (1e) can modulate the oxidative burst in stimulated human neutrophils. Synthetic strategies included the glycosyl

New 1-C-(5-thio-D-xylopyranosyl) derivatives as potential orally active venous antithrombotics.

In the search for new orally active antithrombotic drugs that are metabolically stable, we explored the synthesis of 1-C-(5-thio-D-xylosyl) derivatives, examining radical and nucleophilic methods. Thus synthesized were aryl, benzyl, alkylcarboxymethylenyl, arylsulfonylmethylenyl and alkylaminocarboxymethylenyl C-linked analogues of 5-thio-D-xylopyranosides.

Lewis-X-Containing Triterpenoid Saponins Inhibit DC-SIGN- and L-SIGN-Mediated Transfer of HIV-1 Infection.

Blocking dendritic cell-specific intercellular adhesion molecule-3-grabbing nonintegrin (DC-SIGN)- and liver/lymph node-specific intercellular adhesion molecule-3-grabbing integrin (L-SIGN)-mediated human immunodeficiency virus 1 (HIV-1) attachment to immune cells represents a promising strategy for developing antiretroviral agents effective during the early stages of sexual transmission. Although mannose- and fucose-based ligands have received considerable attention, Lewis-based inhibitors remain relatively underexplored. In this study, we report the first synthesis of Lewis-X-containing trit

New synthesis of oligosaccharides modelling the M epitope of the Brucella O-polysaccharide.

Brucellosis is a dangerous zoonotic disease caused by bacteria of the genus Brucella. Diagnosis of brucellosis is based on the detection in animal and human sera of antibodies to the O-polysaccharide of Brucella lipopolysaccharide. The currently employed serodiagnosis of brucellosis relies on the use of the Brucella O-polysaccharide as a diagnostic antigen. However, the existence of bacterial species, which also express O-polysaccharides structurally similar to that of Brucella, may decrease the specificity of the brucellosis detection due to false-positive test results. It has been shown that

Synthesis and Bioactivity of Ancorinoside B, a Marine Diglycosyl Tetramic Acid.

The sponge metabolite ancorinoside B was prepared for the first time in 16 steps and 4% yield. It features a β-d-galactopyranosyl-(1→4)-β-d-glucuronic acid tethered to a d-aspartic acid-derived tetramic acid. Key steps were the synthesis of a fully protected d-lactose derived thioglycoside, its attachment to a C20-aldehyde spacer, functionalization of the latter with a terminal N-(β-ketoacyl)-d-aspartate, and a basic Dieckmann cyclization to close the pyrrolidin-2,4-dione ring with concomitant global deprotection. Ancorinoside B exhibited multiple biological effects of medicinal interest. It i

Structure-activity relationship for calanthoside, a potential hair-growth stimulant with an indole 2-S-,3-O-bis-glucoside structure. Part 1: role of two glucoside moieties in promoting HFDPC proliferation.

The development of new and effective drugs for treating androgenetic alopecia (AGA) remains challenging. Minoxidil and finasteride, two drugs currently used to treat AGA, were originally developed for treating hypertension and prostate disease, respectively. Therefore, the pharmacological effects of these drugs may be double-edged in patients with AGA. Calanthoside (1) is an indole 2-S-,3-O-bis-glucoside isolated from the Calanthe genus. Compared to minoxidil, 1 induces significantly higher proliferation of human hair follicle dermal papilla cells (HFDPCs). This study aimed to identify the key

Photolysis of the lysosomal neuraminidase in cultured human skin fibroblasts in the presence of a photoreactive competitive inhibitor.

Photolysis of the lysosomal neuraminidase in crude homogenates of cultured human skin fibroblasts was carried out using the potent competitive enzyme inhibitor, 9-S-(4-azido-2-nitrophenyl)-5-acetamido-2,6-anhydro-2,3,5,9-tetradeoxy-9 -thio-D - glycero-D-galacto-non-2-enonic acid (9-PANP-2,3-D-NANA). Irradiation of the homogenate and the inhibitor (2 min, pH 4.3, 10 degrees C) with a medium pressure mercury lamp resulted in about a 24% reduction of enzyme activity compared to irradiated controls that did not contain additives. No significant loss of activity was observed with homogenate that co

Clinical applications of the naturally occurring or synthetic glycosylated low molecular weight drugs.

Four major types of biomacromolecules are nucleic acids (DNAs and RNAs), proteins, lipids, and glycans. Only carbohydrates or glycans are a universal existence in all types of the biomacromolecules. The monosaccharides deoxyribose and ribose are an integral part of DNA and RNA structures. Glycosylated proteins and glycosylated lipids are common in all life forms. As expected, many well-known glycosylated low molecular weight drugs (GLMWDs) that target biomacromolecules, such as antibiotics, anticancer, steroids, and antidiabetic therapeutics, are naturally made glycosylated products. A drug us

Stereochemistry of intramolecular cyclization of tetra-β-(1→6)-D-glucosamines and related tetrasaccharides: the role of the conformational stereocontrol and the neighboring group participation.

The effect of reaction conditions, the nature of a leaving group, and a substituent at C-2 in the glycosylating monosaccharide on the stereochemical outcome of cyclization of linear tetra-β-(1→6)-d-glucosamines and some 'mixed' tetrasaccharides comprising glucose and glucosamine residues has been examined. Toluene and nitrile solvents improved the β-stereoselectivity of cyclization, however, the overall efficiency of the formation of cyclic products in these solvents was lower than that in dichloromethane. The use of bromide or pentenyl glycoside as leaving groups instead of the thioglycoside

Comparative study between extraction techniques and column separation for the quantification of sinigrin and total isothiocyanates in mustard seed.

Glucosinolates are β-thioglycosides which are found naturally in Cruciferae including the genus Brassica. When enzymatically hydrolysed, glucosinolates yield isothiocyanates and give a pungent taste. Both glucosinolates and isothiocyanates have been linked with anticancer activity as well as antifungal and antibacterial properties and therefore the quantification of these compounds is scientifically important. A wide range of literature exists on glucosinolates, however the extraction and quantification procedures differ greatly resulting in discrepancies between studies. The aim of this study

Development of new glycosylation methodologies for the synthesis of archaeal-derived glycolipid adjuvants.

To commercialize the production of glycolipid adjuvants, their synthesis needs to be both robust and inexpensive. Herein we describe a semi-synthetic approach where the lipid acceptor is derived from the biomass of the archaeon Halobacterium salinarum, and the glycosyl donors are chemically synthesized. This work presents some preliminary results using the promoter system N-iodosuccinimide (NIS) and a stable 0.25 M solution of boron trifluoride-trifluoroethanol (BF(3) x TFE(2)) in dichloromethane. This promoter system allows for the use of peracetyl alkyl(aryl)thioglycosides that are available

Reduction of intimal hyperplasia by naroparcil, a 4-methylumbelliferyl beta-D-xyloside analogue, after arterial injury in the hypercholesterolemic rabbit.

4-Methylumbelliferyl beta-D-xylosides (beta-D-xylosides) inhibit proteoglycan synthesis, and this is associated with reduced proliferation and extracellular matrix production by vascular smooth muscle cells. This study evaluated whether treatment with naroparcil, a beta-D-xyloside analogue, reduced intimal hyperplasia after arterial injury in the hypercholesterolemic rabbit. Forty-two rabbits were assigned to three groups that received either a 1% cholesterol-enriched diet (group 1, n = 15) or the same diet with added 100 mg.kg-1 naroparcil (group 2, n = 15) or 300 mg.kg-1 naroparcil (group 3,

Characterization of the UDP-glucuronosyltransferases involved in the glucuronidation of an antithrombotic thioxyloside in rat and humans.

To investigate the glucuronidation on the hydroxyl group of carbohydrate-containing drugs, the in vitro formation of glucuronides on the thioxyloside ring of the antithrombotic drug, LF 4.0212, was followed in rat and human liver microsomes and with recombinant UDP-glucuronosyltransferases (UGT). The reaction revealed a marked regioselectivity in rat and humans. Human liver microsomes glucuronidated the compound mainly on the 2-hydroxyl position of the thioxyloside ring, whereas rat was able to form glucuronide on either the 2-, 3-, or 4- hydroxyl group of the molecule, although to a lower ext

Cytotoxic activity of mouse macrophages studied by various inhibitors.

The influence of various inhibitors on the cytolytic potential of mouse macrophages against syngeneic erythrocytes has been investigated in vitro by isotope techniques. Intact macrophage membrane and cell metabolism was essential for full cytotoxic activity. The process was completely blocked by anaerobiosis and cold. ATP from both mitochondrial respiration and glycolysis seems to be the high energy intermediate which is utilized during the cytotoxic activity of macrophages leading to target cell lysis. The process did not depend on concomitant DNA transcription, translation, or protein synthe
