In vivo study of schistosomicidal action of 1-benzyl-4-[(4-fluoro-phenyl)-hydrazono]-5-thioxo-imidazolidin-2-one.

Praziquantel has been the drug most widely used therapy against different forms of schistosomiasis around the world. However, this treatment has shown ineffective in humans and in experimental models of Schistosoma mansoni. New therapeutic alternatives have been tested, including the imidazolidine derivative LPSF/PT-09, which has shown high therapeutic potential in vitro. In this work, we tested the schistosomal activity of this derivative in doses of 250mg/kg and 200mg/kg in mice experimentally infected with a high parasite load of S. mansoni. Parasitological evaluations related to the number

[AGRANULOCYTOSIS CAUSED BY APALUTAMIDE : A CASE REPORT].

We report a case of agranulocytosis associated with apalutamide treatment in a patient with non-metastatic castration-resistant prostate cancer (nmCRPC). An 89-year-old man was diagnosed with prostate cancer (cT2cN0M0) four years earlier and treated with surgical castration and anti-androgen therapy. Subsequently, his PSA levels increased. Imaging studies showed no obvious metastases, and he was diagnosed with nmCRPC. Darolutamide was initiated but discontinued because of liver dysfunction, and treatment was switched to apalutamide. Forty-five days after the initiation of apalutamide, the pati

Androgen deprivation therapy with apalutamide in the treatment of metastatic castration-sensitive prostate cancer: the systematic review of its effectiveness and applicability for Asian men in the targeted investigational treatment analysis of novel anti-androgen (TITAN) trial.

BACKGROUND: The TITAN trial established survival benefits of adding apalutamide to androgen deprivation therapy (ADT) in metastatic castration‑sensitive prostate cancer (mCSPC). Whether efficacy and safety are comparable in Asian men remains insufficiently characterized. METHODS: We conducted a PRISMA-guided review (PROSPERO CRD420251054772) and adhered to the SWiM framework for narrative synthesis. Databases searched in MEDLINE, Embase, Cochrane Library, and CINAHL from inception to 15 December 2024. Eligible studies presented Asian subpopulation outcomes for apalutamide + ADT in mCSPC from

Maculopapular Drug Eruption Caused by Apalutamide: Case Report and Review of the Literature.

Apalutamide, an oral androgen receptor signaling inhibitor, is approved for the treatment of non-metastatic castration-resistant prostate cancer and metastatic prostate cancer. In the international randomized placebo-controlled clinical trials, apalutamide was associated with a higher rate of rash than placebo. However, given that reports from a dermatological perspective are limited, the skin manifestations and histopathology of the skin lesions caused by apalutamide are largely unknown. Here, we report a case of apalutamide-induced drug eruption. A 66-year-old man developed itchy maculopapul

Predicting in vivo performance of piroxicam and apalutamide drug nanocrystals from in vitro flux measurements combined with in silico modelling.

The development and optimization of pharmaceutical nanocrystal formulations is often complex and resource-intensive, as particle properties depend on composition, manufacturing parameters, and batch variability, all of which influence in vivo performance. Predictive in vitro-in silico approaches can streamline formulation development by reducing the need for extensive animal and clinical testing. This study reports on a methodological evaluation of an integrated workflow combining dissolution-permeation data with a mechanistic absorption model based on the gastrointestinal unified theoretical

Digital Pathology-Based Multimodal Artificial Intelligence Scores and Outcomes in a Randomized Phase III Trial in Men With Nonmetastatic Castration-Resistant Prostate Cancer.

PURPOSE: The SPARTAN trial demonstrated that the addition of apalutamide to androgen deprivation therapy improves outcomes among patients with nonmetastatic castration-resistant prostate cancer (nmCRPC). We applied a previously reported digital histopathology-based multimodal artificial intelligence (MMAI) algorithm to estimate clinical outcomes in SPARTAN. METHODS: Patients with available hematoxylin and eosin-stained slides from the primary tumor were included. Histopathology slides were digitized. MMAI scores ranging from 0 to 1 were generated from digital histopathology and baseline clini

Prior Local Therapy and First-Line Apalutamide in Patients With Nonmetastatic Castration-Resistant Prostate Cancer: A Secondary Analysis of the SPARTAN Randomized Clinical Trial.

IMPORTANCE: Preclinical studies suggest that exposure to prostate-directed local therapy (LT) may influence the efficacy of subsequent systemic therapy including androgen receptor pathway inhibitors. However, there is insufficient clinical evidence to support this premise in patients with nonmetastatic castrate-resistant prostate cancer (nmCRPC). OBJECTIVE: To determine whether exposure to prior prostate-directed LT (radical prostatectomy [RP], radiation therapy [RT], or both) played any effect-modifying role in the treatment effect of apalutamide on metastasis-free survival (MFS) and overall

Radioprotective effects of thiomethylhydantoin derivatives on Escherichia coli and mice.

Protection of Escherichia coli NIHJ and C57BL mice from the effects of 60Co gamma-rays provided by S-alk(en)yl-L-cysteines and their hydantoin derivatives was examined. E. coli (10(6) cells/ml) suspended in a 20 mM aqueous solution of one of the drugs was irradiated with 60 Gy of gamma-rays. Five week-old male mice were exposed to 5.0-9.5 Gy of gamma-rays after a single intraperitoneal injection of 0.75 mmol/kg body weight of each compound. In both E. coli and mice, S-allyl compounds afforded more effective radioprotection than S-propyl compounds. The replacement of the alpha-hydrogen of S-sub

Integral Lipophilicity Studies with in Silico ADMET Parameter Analysis of Novel Pseudothiohydantoin Derivatives.

This article describes the assessment of the lipophilicity of 18 new pseudothiohydantoin (2-aminothiazol-4(5H)-one) derivatives using experimental and computational methods. Previous studies have shown that the tested derivatives are inhibitors of 11β-hydroxysteroid dehydrogenase type 1 and inhibit the metabolic activity of selected cancer cell lines. Lipophilicity, as log kw and RM0, was determined using reversed-phase high-performance liquid chromatography (RP-HPLC) and reversed-phase thin-layer chromatography (RP-TLC), respectively. In both chromatographic methods, methanol served as a modi

Pharmacist Intervention in Outpatients With Prostate Cancer Prevents Apalutamide-induced Skin Adverse Events.

BACKGROUND/AIM: Apalutamide induces severe skin adverse events (sAEs) in 14.7% of Japanese patients, leading to treatment discontinuation. To maximize the management of sAEs in patients taking apalutamide for prostate cancer, we conducted pharmacist outpatient clinics for patients receiving apalutamide in the outpatient setting. During these sessions, patients were informed about skin care management, including the application of moisturizers to prevent sAEs. This study aimed to evaluate the usefulness of pharmacist-led outpatient services in managing sAEs. PATIENTS AND METHODS: Patients with

Apalutamide use in metastatic hormone-sensitive prostate cancer patients diagnosed by conventional and next-generation imaging. Real world data from 772 patients.

INTRODUCTION: Apalutamide has shown significantly increases in radiographic progression-free survival (rPFS) and overall survival (OS) in metastatic hormone-sensitive prostate cancer (mHSPC) patients diagnosed by conventional imaging (CI). However, there is scarce knowledge on the use of apalutamide in mHSPC population diagnosed by NGI. METHODS: Retrospective multicenter study of mHSPC patients treated with apalutamide from May 2018 to September 2023 registered in the Real-World Evidence APA (RWE-APA). CI and NGI group were defined, according to the diagnostic tool of metastatic disease. Prim

Pharmacokinetic Interactions Between Abiraterone, Apalutamide, Darolutamide or Enzalutamide and Antithrombotic Drugs: Prediction of Clinical Events and Review of Pharmacological Information.

PURPOSE: Abiraterone, apalutamide, darolutamide and enzalutamide are second-generation hormone therapies used for advanced prostate cancer; the majority of patients receiving these treatments are elderly, poly-medicated patients. Since their first market authorizations, their pharmacokinetic (PK) characteristics are increasingly well known. A potential risk of drug-drug interaction (DDI), especially with cardiovascular drugs, needs to be considered. In the case of antithrombotics, treatment imbalance can lead to severe consequences. OBJECTIVES: To describe PK profiles of hormone therapies and

Evaluating Clinical Tools to Monitor Cardiovascular Risk in Men With Prostate Cancer Receiving Hormone Therapy.

PURPOSE: Men with prostate cancer face considerable cardiovascular (CV) comorbidity. Androgen deprivation therapy (ADT) and androgen receptor pathway inhibitors (ARPIs) elevate this risk, yet no prostate-specific CV monitoring tools exist. Clinicians rely on general calculators such as the American Heart Association (AHA)/American College of Cardiology (ACC) atherosclerotic cardiovascular disease (ASCVD) score, which remain unvalidated in this setting. The purpose of this study is to evaluate changes in the estimated CV risk of men with prostate cancer after 6 months of hormone therapy using c

A high eosinophil proportion increases the risk of skin-related adverse events induced by apalutamide in patients with prostate cancer.

BACKGROUND: Skin-related adverse events (AEs) induced by apalutamide occur frequently in Japanese patients with prostate cancer. However, biomarkers for predicting these skin-related AEs have not yet been identified. Therefore, this study investigated whether the proportion of eosinophils could serve as a predictive biomarker for skin-related AEs in Japanese patients with prostate cancer treated with apalutamide. METHODS: A total of 109 patients were enrolled in this study. Among them, 79 patients with prostate cancer who received apalutamide were categorized into two groups: the skin AE grou

Metformin for patients with metastatic prostate cancer starting androgen deprivation therapy: a randomised phase 3 trial of the STAMPEDE platform protocol.

BACKGROUND: Metformin is a widely used anti-diabetic drug. Several studies have suggested that metformin has anticancer activity in some malignancies, including prostate cancer. Metformin might also mitigate the adverse metabolic effects of androgen-deprivation therapy (ADT). We hypothesised that metformin might improve survival in patients with metastatic hormone-sensitive prostate cancer and reduce metabolic complications associated with ADT. METHODS: The STAMPEDE multi-arm, multi-stage, randomised phase 3 trial recruited patients with high-risk locally advanced or metastatic adenocarcinoma

Apalutamide in combination with autophagy inhibitors improves treatment effects in prostate cancer cells.

BACKGROUND: ARN-509 (Apalutamide) is a unique androgen receptor (AR) antagonist for the treatment of castration-resistant (CR) prostate cancer (PC). It inhibits AR nuclear translocation, DNA binding and transcription of AR gene targets. As dysregulation of autophagy has been detected in PC, the targeting of autophagy is a potential approach to overcome early therapeutic resistance. Therefore, we investigated the characteristics of autophagic response to ARN-509 treatment and evaluated the potential effect of a combination with autophagy inhibition. METHODS: Human prostate cancer cells (LNCaP)
