Hydrogen sulfide (H2S) has been regarded as a small gasotransmitter associated with physiological and biochemical regulation in plant responses to environmental stresses. However, the regulatory mechanisms of H2S in photosynthesis under adverse conditions remains poorly understood in plants. Here, the role of H2S in the regulation of photosystem I (PSI) and photosystem II (PSII) was investigated in rice seedlings subjected to salt stress. Our results showed that NaHS (H2S donor) pretreatment significantly enhanced photosynthetic pigment content, gas exchange parameters, and the photochemical c

Earlier work suggested that Ca2+ ions in the chloroplast thylakoid lumen interact with thylakoid membrane proteins to produce a proton flux gating structure which functions to regulate the expression of the energy-coupling H+ gradient between localized and delocalized modes [Chiang, G., & Dilley, R. A. (1987) Biochemistry 26, 4911-4916]. In this work, one of the phenothiazine Ca2+ antagonists, chlorpromazine, was used as a photoaffinity probe to test for Ca(2+)-dependent binding of the probe to thylakoid proteins. [3H]Chlorpromazine photoaffinity-labels thylakoid polypeptides of Mr 8K and 6K,

Thylakoid membranes (TMs), the photosynthetic membrane systems in chloroplasts, are multifunctional bioactive systems capable of light-driven ATP production. Their capacity of ATP production has inspired strategies for cross-species bioenergetic support and treatment of diseases. However, their oral application against intestinal diseases requires stabilization throughout the gastrointestinal tract. To address this, we developed an enteric delivery system by fabricating TMs-loaded sodium alginate (SA) microspheres (TMs-SPMs) via microfluidics, followed by coating with a chitosan-sodium tripoly

We describe the construction and characterization of a laser-line-scanning microscope capable of detection of broad fluorescence spectra with a resolution of 1 nm. A near-infrared femtosecond pulse train at 800 nm was illuminated on a line (one lateral axis, denoted as X axis) in a specimen by a resonant scanning mirror oscillating at 7.9 kHz, and total multi-photon-induced fluorescence from the linear region was focused on the slit of an imaging polychromator. An electron-multiplying CCD camera was used to resolve fluorescence of different colours at different horizontal pixels and fluorescen

STN7 and STN8 kinases phosphorylate several proteins of the photosynthetic apparatus in response to changes in environmental conditions. Phosphorylation is linked to the dynamic arrangement of both the 3D structure of the thylakoid membrane and thylakoid protein complexes, thereby controlling excitation energy transfer between photosystems and the repair cycle of photosystem II (PSII). However, mutants with phosphorylation of PSII core proteins show only minor changes compared with the wild-type (WT) under conditions tested so far, and thus the physiological significance of reversible thylakoi

Protein cross-linking studies with a thylakoid membrane translocation intermediate were used to demonstrate that chloroplast SecA functions as a membrane-associated component of the Sec-like ATP-dependent protein translocase of pea chloroplasts. In assays with isolated thylakoids, it was observed that translocation of the 33-kDa protein of the oxygen-evolving complex of photosystem II (OE33) decreased when the ATP concentration was low, and that the protein accumulated as a bound precursor. The bound precursor was able to be translocated into the lumen when the ATP concentration was raised, in

Thylakoid membranes are highly organized yet dynamic systems in which Photosystem II (PSII) complexes undergo continual assembly, repair and lateral redistribution. Here a hypothesis is proposed that local lumenal pH, generated by the trans-thylakoidal proton gradient and potentially exhibiting transient spatial heterogeneity, may contribute to the regulation of protein movement and PSII maturation within the thylakoid membrane. Five mechanistically distinct but potentially coupled routes by which ΔpH could influence protein dynamics are outlined: (i) pH-dependent modulation of lumenal protein

Plant chloroplasts have complex membrane systems. Among these, thylakoids serve as the sites for photosynthesis and photosynthesis-related adaptation. In addition to the photosynthetic membrane complexes and associated molecules, lipids in the thylakoid membranes, are predominantly composed of MGDG (monogalactosyldiacylglycerol), DGDG (digalactosyldiacylglycerol), SQDG (sulfoquinovosyldiacylglycerol) and PG (phosphatidylglycerol), play essential roles in shaping the thylakoid architecture, electron transfer, and photoregulation. In this review, we discuss the effect of abiotic stress on chloro

Phosphatidylglycerol (PG) ubiquitous in thylakoid membranes of photosynthetic organisms was previously shown to contribute to accumulation of chlorophyll through analysis of the cdsA- mutant of a cyanobacterium Synechocystis sp. PCC6803 defective in PG synthesis (SNC1). Here, we characterized effects of manipulation of the PG content in thylakoid membranes of Synechocystis sp. PCC6803 on the photosystem complexes to specify roles of PG in biogenesis of thylakoid membranes. SNC1 cells with PG deprivation in vivo, together with the chlorophyll decrease, exhibited a decline not in PSII, but in PS

Exposure of Synechococcus leopoliensis to selenite in the light resulted in orange-colored granules associated with the cells. No such particles were made in dark grown cells or when selenite was replaced by selenate. Light and scanning electron microscopy revealed that the particles formed inside the cells. Furthermore, these were easily extracted and shown to be composed of selenium as determined by energy-dispersive X-ray spectroscopy. During selenium particle synthesis there was a concurrent loss of organic pigments in the cyanobacteria. Cells also become heavier as they produced and accum

Soluble and thylakoid membrane polypeptides from senescing leaf tissue of Rossa, a normal yellowing Festuca pratensis genotype, were fractionated by sodium dodecyl sulphate polyacrylamide gel electrophoresis and compared with those of the non-yellowing mutant Bf 993. Subunits of ribulose-1,5-bisphosphate carboxylase were the major soluble polypeptides and declined to low levels in senescing leaves of both genotypes. The major thylakoid polypeptides were those associated with the chlorophyllprotein complexes CPI and CPII. The levels of all thylakoid polypeptide species fell during senescence of

Efficient solar energy conversion is ensured by the organization, physical association, and physiological coordination of various protein complexes in photosynthetic membranes. Here, we visualize the native architecture and interactions of photosynthetic complexes within the thylakoid membranes from a fast-growing cyanobacterium Synechococcus elongatus UTEX 2973 (Syn2973) using high-resolution atomic force microscopy. In the Syn2973 thylakoid membranes, both photosystem I (PSI)-enriched domains and crystalline photosystem II (PSII) dimer arrays were observed, providing favorable membrane envir

The effects of DDT, some of its analogs, and selected cyclodiene insecticides on isolated spinach (Spinacea oleracea L.) thylakoids were identified, characterized, and compared to responses induced by selected herbicides. Except for endrin, the insecticides inhibited light-induced electron transport, altered chlorophyll fluorescence transients, and competitively displaced [14C]atrazine [2-chloro-4-(ethylamino)-6-(isopropylamino)-s-triazine], a known photosystem II inhibitor, from the membranes. The insecticides appeared to act at, or near B, the secondary electron acceptor of photo-system II.

Revealing the spatial organization and stress-induced rearrangements of the photosynthetic machinery in thylakoids of diatoms is crucial for understanding their metabolic responses and/or adaptation. Using a multimodal framework that combines coherent anti-Stokes Raman scattering (CARS), two-photon excited fluorescence, and confocal fluorescence microscopy with machine learning, we investigated the spatial pigment distribution in thylakoids of Cyclotella meneghiniana. Under physiological conditions, we observed dense central packing of fucoxanthins (Fx's). In contrast, the peripheral girdle la

Chlamydomonas reinhardtii mutants defective in the chloroplast ATP synthase are highly sensitive to light. The ac46 mutant is affected in the MDH1 gene, required for production or stability of the monocistronic atpH mRNA encoding CF(O)-III. In this and other ATP synthase mutants, we show that short-term exposure to moderate light intensities-a few minutes-induces an inhibition of electron transfer after the primary quinone acceptor of photosystem II (PSII), whereas longer exposure-several hours-leads to a progressive loss of PSII cores. An extensive swelling of thylakoids accompanies the initi

Programmable artificial photosynthetic cell is the ultimate goal for mimicking natural photosynthesis, offering tunable product selectivity via reductase selection toward device integration. However, this concept is limited by the capacity of regenerating the multiple cofactors that hold the key to various reductases. Here, we report the design of artificial photosynthetic cells using biotic-abiotic thylakoid-CdTe as hybrid energy modules. The rational integration of thylakoid with CdTe quantum dots substantially enhances the regeneration of bioactive NADPH, NADH and ATP cofactors without exte

Photosynthesis impairment and the accompanying leaf yellowing represent a prominent and physiologically critical symptom of phytoplasma infection. However, the underlying molecular mechanisms remain poorly understood. In this study, we identified the Jujube Witches' Broom pathogenic effector PHYL1JWB, which was previously reported to induce phyllody in jujube, as a key inducer of photosynthesis impairment. Overexpression of PHYL1JWB in sour jujube and Arabidopsis thaliana disrupted photosynthesis by causing severe disorganization of chloroplast ultrastructure. PHYL1JWB directly interacts with

This is a brief overview focused on the electron paramagnetic resonance applications to the study of the proton transport processes in chloroplasts. After brief description of structural and functional organization of the chloroplast electron transport chain, our attention is focused on the measurements of trans-thylakoid pH difference (ΔpH) with pH-sensitive spin-probes. The use of spin-probes is based either (i) on measuring the ΔpH-partitioning of spin-probes between the thylakoid lumen and external volume, or (ii) on monitoring changes in the electron paramagnetic resonance spectra of pH-s
