BACKGROUND: Agarwood is derived from Aquilaria trees, the trade of which has come under strict control with a listing in Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora. Many secondary metabolites of agarwood are known to have medicinal value to humans, including compounds that have been shown to elicit sedative effects and exhibit anti-cancer properties. However, little is known about the genome, transcriptome, and the biosynthetic pathways responsible for producing such secondary metabolites in agarwood.
RESULTS: In this study, we present a

Genkwa Flos (GF), the dried flower bud from Daphne genkwa Sieb. et Zucc. (Thymelaeaceae), is a well-known and widely used traditional Chinese medicine. However, we know little about the in vivo mechanism of GF toxicity. Nematode Caenorhabditis elegans has been considered as a useful toxicity assay system by offering a system best suited for asking the in vivo questions. In the present study, we employed the prolonged exposure assay system of C. elegans to perform the full in vivo toxicity assessment of raw-processed GF. Our data show that GF exposure could induce the toxicity on lifespan, deve

BACKGROUND: Combination antiretroviral therapy (cART) and latency-reversing agents (LRAs) have not achieved a successful cure of HIV infection, due to latent HIV reservoir for the former, and insufficient efficacy and adverse side effects for the latter. Therefore, candidate LRAs with high antiviral efficacy and low cytotoxicity are critical for achieving a functional cure of HIV-1 infection.
PURPOSE: To identify promising flavonoids with dual functions in reactivating latent HIV-1 and inhibiting viral infection, and to explore the mechanisms involved therein.
METHODS: We screened 21 flavono

Fibromyalgia is a common, chronic, and generalized pain syndrome that is often associated with comorbid depression. The etiology of fibromyalgia is complex; most researchers have documented that the hallmark symptoms are due to the central nervous system's abnormal functioning. Neurotransmitters such as serotonin, norepinephrine, and glutamate, have been reported to be key regulators of fibromyalgia syndrome. Daphnetin is a 7, 8 dihydroxy coumarin widely distributed in Thymelaeaceae family plants, possessing various activities such as anti-arthritic, anti-tumor, anti-malarial, and anti-parasit

Two previously undescribed sesquiterpenoid-benzaldehyde hybrids, aquisinoids A (1) and B (2), were isolated from agarwood of Aquilaria sinensis. Their structures were elucidated by comprehensive spectroscopic data analysis, single-crystal x-ray diffraction, and electronic circular dichroism calculations. Structurally, both compounds feature a polycyclic core formed by connecting a prezizaane-type sesquiterpenoid unit with a p-hydroxybenzaldehyde moiety via a 1,3-dioxolane bridge. Compounds 1 and 2 exhibited moderate inhibition on nitric oxide (NO) production in lipopolysaccharide (LPS)-induced

Although Edgeworthia gardneri (Wall.) Meisn and its main component tiliroside (TIL) show good bioactivity, its intestinal absorption data supporting its low bioavailability have not been reported. The evaluation results of three absorption models in vitro and in vivo indicated that the results of the Ussing chamber model were basically consistent with the results of in vivo experiments. It was thus applied to investigate the characteristics of TIL across various intestinal regions and the interaction between TIL and adenosine triphosphate (ATP)-binding cassette family proteins (ABC) including,

Emerging multidrug-resistant (MDR) strains are the main challenges to the progression of new drug discovery. To diminish infectious disease-causing pathogens, new antibiotics are required while the drying pipeline of potent antibiotics is adding to the severity. Plant secondary metabolites or phytochemicals including alkaloids, phenols, flavonoids, and terpenes have successfully demonstrated their inhibitory potential against the drug-resistant pathogens. In quest of potential phytochemicals, we selected tea (Camellia sinensis) and agarwood (Aquilaria malaccensis) leaves for antimicrobial acti
