Involution of the mammalian thymus, one of the leading regulators of aging.

During the past century of research on the thymus, the fact that every mammalian thymus undergoes marked morphological changes during the complex process of aging has been defined as a basic histogenetical rule. In characterizing the physiological (i.e. chronic) involution of the mammalian thymus, the term "Altersinvolution" referring to age-related involution is used. All other types of thymic involution are associated with an initial trigger and a relatively "acute" mechanism. In all of these factor-dependent cases of thymic involution, we use the term "akzidentelle Involution" (i.e. acute a

Age- and disease-related decline in immune function: an opportunity for "thymus-boosting" therapies.

The thymus is the site of production of mature T lymphocytes and thus is indispensable for the development and maintenance of the T cell-mediated arm of the immune system. Thymic production of mature T cells is critically dependent on an influx of bone marrow-derived progenitor T cells that undergo replication and selection within the thymus. Thymus cellularity and thymic hormone secretion reach a peak during the first year of life and then decline gradually until the age of 50-60 years, a process known as "thymic involution." A rapid reduction of thymus cellularity occurs in young patients fo

[Hyperplasia of the thymus gland in children: physiology or pathology?].

A critical analysis of the literature data and of the results of clinicoroentgenological, hormonal and immunological studies in children with thymomegaly makes it possible to regard hyperplasia of the thymus gland at early childhood as a sign of functional incompetence of the thymus and of the secondary immunodeficient state.

Effect of glutaurine on vitamin A and prednisolone treated thymus cultures.

The hormone of the parathyroid gland, Glutaurine (gamma-L-glutamyl-taurine) exerts on rat thymus cultures an effect synergistic with vitamin A and antagonistic to prednisolone. When applied in itself, it increased the vitality and macrophage reaction of thymus cultures.

[Endocrine function of the thymus in experimental hypothyroidism].

An Endocrine function of the thymus was studied by analyzing the level of thymulin in rats in 1-6 months after extirpation of the thymus. The level of thymulin in rats after thyroidectomy has been shown to be reduced maximally in 6 months after an operation. An inhibitor of thymulin was detected in the blood serum in a month after extirpation, then it disappeared completely from it. After thyroidectomy the mass of animals decreased, as well as an absolute and relative mass of the thymus and its cellular content. Thyroidectomy was followed with a marked decrease in the thyroid hormones content

The isolation of thymopoietin (thymin).

The isolation from bovine thymus of two closely related polypeptides, thymopoietin I and II, is described. These are considered to be thymic hormones, which physiologically induce the differentation of prothymocytes to thymocytes within the thymus. Ths isolation of the thymopoietins was monitored not by their differentiative effects, but by a presumably secondary effect on neuromuscular transmission. This was discerned in experimental studies related to the human disease myasthenia gravis in which it was suggested that autoimmune thymitis was regularly present. In an animal model, experimental

Relation of lymphoid system and hormones to aging.

Congenital deficiencies of developmental hormones, such as growth hormone and thyroxine, are responsible for the thymus-dependent immunodeficiency disease observed in dwarf mice. Such an immunodeficiency state is associated with early aging phenomena. This fact suggests that lymphoid cells and primarily T-derived cells are involved in the control of aging processes.

[The role of the pineal-thymus system in the regulation of autoimmunity, aging and lifespan].

Thymus is an immunoendocrine organ, the hormones of which mainly influence its own lymphatic elements. It has a central role in the immune system, the neonatal removal causes the collapse of immune system and the whole organism. The thymic nurse cells select the bone marrow originated lymphocytes and destroy the autoreactive ones, while thymus originated Treg cells suppress the autoreactive cells in the periphery. The involution of the organ starts after birth, however, this truly happens in the end of puberty only, as before this it is overcompensated by developmental processes. From the end

Effects of sex steroids on thymic epithelium and thymocyte development.

Sex steroid hormones have major effects on the thymus. Age-related increases in androgens and estrogens and pregnancy-induced increases in progestins all cause dramatic thymic atrophy. Atrophy can also be induced by treatment with exogenous sex steroids and reversed by ablation of endogenous sex steroids. Although these observations are frequently touted as evidence of steroid lymphotoxicity, they are often driven by steroid signaling in thymic epithelial cells (TEC), which are highly steroid responsive. Here, we outline the effects of sex steroids on the thymus and T cell development. We focu

The role of zinc in pre- and postnatal mammalian thymic immunohistogenesis.

Mammalian thymic histogenesis can be morphologically divided into three consecutive stages: a) epithelial, b) lymphopoietic or lympho-epithelial, and 3) differentiated cellular microenvironmental, with formation of Hassall's bodies (HBs). Immunomorphological changes characteristic of human thymic involution begin during or soon after the first year after birth, and continue progressively throughout the entire life span. The 3% to 5% annual reduction in the number of cells of the human thymic microenvironment continues until middle age, when it slows down to less than 1% per year. According to

Influence of the thymus on steroidogenesis by rat ovarian cells in vitro.

Thymic hormones and factors have been shown to modulate the function of other endocrine glands including the gonads. Absence of the thymus during development results in ovarian dysgenesis characterized by a decrease in the number of follicles and corpora lutea, bringing about severe changes in reproductive function. To examine whether thymic secretions might affect ovarian activity, whole dispersed ovarian cells obtained from immature rats pretreated with pregnant mare serum gonadotrophin were exposed to a thymus fraction of approximately 28 kDa and also to the media from incubated thymuses (T

Role of thymic peptides as transmitters between the neuroendocrine and immune systems.

Thymic peptides, a heterogenous family of polypeptidic hormones synthesized within the thymus, not only exert important regulatory effects within both the immune and neuroendocrine systems but are also themselves subject to control by hormones derived from the hypothalamic-pituitary-adrenal axis (HPA) and other endocrine glands. Regarding thymic hormonal function, thymulin production is up-regulated by several hormones, including prolactin, growth hormone and thyroid hormones. Other aspects of the physiology of thymic epithelial cells can also be modulated by hormones and neuropeptides, partic

Thymus Gland: A Double Edge Sword for Coronaviruses.

The thymus is the main lymphoid organ that regulates the immune and endocrine systems by controlling thymic cell proliferation and differentiation. The gland is a primary lymphoid organ responsible for generating mature T cells into CD4+ or CD8+ single-positive (SP) T cells, contributing to cellular immunity. Regarding humoral immunity, the thymic plasma cells almost exclusively secrete IgG1 and IgG3, the two main complement-fixing effector IgG subclasses. Deformity in the thymus can lead to inflammatory diseases. Hassall's corpuscles' epithelial lining produces thymic stromal lymphopoietin, w

Male BXSB mice develop a thymic hormonal dysfunction with presence of intraepithelial crystalline inclusions.

The thymus is morphologically abnormal in male BXSB mice with cortical involution densification of the epithelium and the presence of intraepithelial crystals. The thymic endocrine function in BXSB mice was appraised using a biological assay to measure the level of the zinc-dependent thymic hormone, thymulin, and an indirect immunofluorescence technique to evaluate the number of cells synthesizing the hormone within the thymus. Unlike the dramatically accelerated age-linked decline of thymulin production reported in females of other autoimmune strains (measured as early as 6 weeks of age), onl

Shen-Qi-Jie-Yu-Fang has antidepressant effects in a rodent model of postpartum depression by regulating the immune organs and subsets of T lymphocytes.

BACKGROUND: Shen-Qi-Jie-Yu-Fang (SJ Fang) is a herbal preparation used in traditional Chinese medicine, and is a potentially important new therapeutic agent in postpartum depression (PPD). Previously, we have elucidated the effects of SJ Fang on hormone receptors and monoamine neurotransmitters involved in the hypothalamic-pituitary-adrenal and hypothalamic-pituitary-gonadal axes in PPD rats. However, the immune-modulating effects of SJ Fang in PPD are still unknown. In this study, we explored the effects of SJ Fang on the immune organs and subsets of T lymphocytes in PPD rats. METHODS: PPD w

The functions of the thymus.

In rodents the thymus performs at least two functions. It is a major site of lymphopoiesis in the embryo and newborn, with the resulting lymphocytes migrating from the thymus to seed the spleen, lymph nodes and other lymphoid organs. In addition, the thymus produces a hormone which has an immunotrophic effect, i.e. it endows cells having immunological potential with immunological competence. In some animals other organs, in addition to the thymus, are responsible for directing the normal development of the immunological system. These are the bursa of Fabricius in birds and the appendix in rabb

Have we underestimated the importance of the thymus in man?

Recent immunological research has concentrated on the complex and subtle interactions between T cells, B cells and accessory cells. In these studies, little attention has been given to the adult thymus gland. Modern textbooks of disease and anatomy all stress that the gland undergoes fatty involution with age in man but omit reference to the statements here and there in the literature that the gland is active and produces lymphocytes throughout life. To suggest that the bone marrow, which also builds up fat throughout life, is atrophic and not important to adult man would deny all modern hemat

Hormonal function of nonendocrine cells: role of new biological phenomenon in the regulation of homeostasis.

Recent studies revealed a new biological phenomenon: hormone synthesis in nonendocrine cells. Here we review hormone production by 4 types of nonendocrine cells of different origins, localizations, and functions and the role of this biological phenomenon in the maintenance of homeostasis. Our results and published data suggest that hormonal function is a general biological property not specific for only neuroendocrine cells, but rather typical of all living cells independently of their origin and role in the body.
