We prepared 3 samples of 19S thyroglobulin (Tg), 1 from a patient with Graves' disease, another from a patient with nontoxic goiter, and the third from a pool of Tg from normal subjects, and used each Tg preparation to produce a polyvalent antiserum in rabbits. The 3 antisera were similar to each other in their reactivity with thyroid Tg samples from 25 patients with various thyroid disorders and from 10 normal subjects. However, the immunoreactivity of the 35 individual Tg samples varied considerably. Decreased reactivity was associated with proteolysis during Tg preparation, iodination in vi

Methods used for estimating in vivo diffusion velocity of thyroglobulin (Tgb), the factors affecting hydrodynamic properties of thyroidal colloid and the effects of changing diffusion properties on follicular function are briefly reviewed. The principal methods, besides pure in vitro techniques, are autoradiography of thyroid sections after in vivo labelling of Tgb and freezing autoradiography for examining colloidal diffusion of ions or other small molecules. The main factors known to affect diffusion of Tgb in the colloid space are concentration and actual physico-chemical properties of the

19 S thyroglobulins from several mammals were prepared under conditions designed to minimize endogenous proteolytic activities. The purified iodoproteins were then fractionated by RbCl isopycnic gradient centrifugation and fractions of varying iodine content analyzed by SDS-polyacrylamide gel electrophoresis. The dependence on the iodination level of molecular dissociability, previously reported in preparations "markedly' differing in their iodination degree, has been now ultimately assessed in all species investigated and demonstrated even for fraction differing from each other only by a few

Thyroglobulin biosynthesis was studied in thyroid glands of rats treated during 30 days with a daily dose of propranolol, a non-selective beta-adrenergic blocking drug. Studies were carried out by extraction of soluble proteins from homogenates after incubation of the glands in presence of [3H]-4,5-L-leucine and [3H]-D-1-galactose. Thyroglobulin 19S, 12S and 4-8S soluble proteins were separated and identified by ultracentrifugation in a saccharose gradient. The glands of rats treated with propranolol showed a decreased amount of soluble proteins as well as a decreased incorporation of [3H] lab

A specific and reproducible double antibody radioimmunoassay for the measurement of thyroglobulin (HTg) in human serum has been developed. Since antithyroglobulin autoantibodies combine with the [(131)I] HTg tracer, antibody-positive sera were rejected for measurement. Specificity is demonstrated in that thyroid analogous such as thyroxine (T(4)), triiodothyronine (T(2)) monoiodotyrosine (MIT) and diiodotyrosine (DIT) did not crossreact. Sera previously reacted with anti-HTg-Sepharose contained no immunoassayable HTg. Finally, sera obtained from patients after total thyroid ablation for thyroi

The influence of antigen (thyroglobulin, Tg) on Tg antibody synthesis has been investigated using cultures of Hashimoto thyroid and peripheral blood lymphocytes. In cultures of thyroid lymphocytes, Tg antibody synthesis was stimulated by a 24h pulse of Tg (10-100 micrograms/ml) and similar results were obtained using spleen lymphocytes from BALB/c mice immunized with human Tg. In contrast, Tg antibody synthesis by Hashimoto peripheral blood lymphocytes was not affected by similar concentrations of Tg (1-240 micrograms/ml) in the presence or absence of pokeweed mitogen (PWM). However, periphera

Thyroglobulin (Tg) is the macromolecular precursor of thyroid hormones and is thought to be uniquely expressed by thyroid epithelial cells. Tg and the thyroid-stimulating hormone receptor (TSHR) are targets for autoantibody generation in the autoimmune disorder Graves disease (GD). Fully expressed GD is characterized by thyroid overactivity and orbital tissue inflammation and remodeling. This process is known as thyroid-associated ophthalmopathy (TAO). Early reports suggested that in TAO, both Tg and TSHR become overexpressed in orbital tissues. Previously, we found that CD34(+) progenitor cel

An enzyme-linked immunosorbent assay was established to detect autoantibodies in canine patients with hypothyroidism. Thyroglobulin, purified from thyroid glands of euthanized, healthy dogs, was used as antigen. Utilizing this test system, sera of 39 patients with hypothyroidism were screened for the presence of thyroglobulin specific autoantibodies. Positive titers were found in 38% of these dogs, whereas 18 out of 72 patients (25%) with other internal diseases showed a positive reaction. In addition 21 healthy dogs were tested for the presence of anti-thyroglobulin autoantibodies. Three dogs

A case of congenital goitre with defective thyroglobulin (Tg) synthesis was studied from the clinical, biochemical and morphological perspectives. The patient, 5.5-year-old boy, who was clinically euthyroid, showed a positive perchlorate discharge test (37.2%). However, the iodination system seemed to be normal since radioiodine uptake into the thyroid was very high, and inspection of the H2O2-generating system using thyroid slices and an assay for peroxidase activity in microsomes showed no abnormalities. On the other hand, virtually no Tg was detected in the serum, and the amount of Tg in th

A specific double antibody radioimmunoassay for human serum thyroglobulin was established. Serum thyroglobulin concentrations were measured in euthyroid non-goitrous subjects and in patients with various specific pathologies of the thyroid gland. Thyroglobulin was detectable in the serum of most euthyroid non-goitrous subjects studied. The serum thyroglobulin concentrations obtained in patients with hyper or hypofunctioning thyroid glands seem to mirror gland activity in terms of hormone production, being high in thyrotoxicosis and low in hypothyroidism. Elevation of serum thyroglobulin concen

Background: Thyroglobulin (Tg), stimulated or unstimulated by recombinant human thyrotropin (TSH), is a static marker of recurrent or persistent disease, and the Tg-doubling rate (Tg-DR) is a dynamic prognostic factor. This study evaluated the prognostic value of an unstimulated Tg (uTg) and Tg-DR papillary thyroid carcinoma (PTC). Methods: This retrospective study included 1818 Tg antibody (Tg-Ab)-negative patients who underwent curative intent total thyroidectomy for PTC without distant metastasis. The uTg was measured 1-3 months post-surgery under TSH suppression (<0.1 mIU/mL). We calculate

CONTEXT: Thyroglobulin (Tg) epitopes of serum Tg autoantibodies (TgAb) have been characterized using inhibition of Tg binding by human monoclonal TgAb in autoimmune thyroid diseases (AITD) [Hashimoto's thyroiditis (HT) and Graves' disease (GD)] but not in non-AITD [nontoxic multinodular goiter (NTMG) and papillary thyroid carcinoma (PTC)].
OBJECTIVE: Our objective was to compare Tg epitopes of serum TgAb from patients with AITD, non-AITD, and PTC associated with histological thyroiditis (PTC-T) using inhibition of Tg binding by four recombinant human TgAb-Fab (epitopic regions A-D).

CONTEXT: Most patients with defective synthesis and/or secretion of thyroglobulin (Tg) present relatively high serum free T3 (FT3) concentrations with disproportionately low free T4 (FT4) resulting in a high FT3/FT4 ratio. The mechanism of this change in FT3/FT4 ratio remains unknown.
OBJECTIVE: We hypothesize that increased type 2 iodothyronine deiodinase (D2) activity in the thyroid gland may explain the higher FT3/FT4 ratio that is frequently observed in patients with abnormal Tg synthesis.
DESIGN: We recently identified a compound heterozygous patient (patient A) with a Tg G2356R mutatio

Inbred Lewis rats regularly develop experimental allergic thyroiditis, delayed skin hypersensitivity and Arthus reactivity, and produce anti-thyroglobulin antibodies 14–21 days after immunization with bovine thyroglobulin in adjuvant. Attempts have been made to transfer thyroiditis to normal rats by means of thymus, spleen or lymph node cells, as well as by serum containing anti-thyroid antibodies. The following types of lymphoid cells were used: viable or non-viable cells from donors immunized with bovine thyroglobulin in adjuvant, cells from donors sensitized only with adjuvant, cells from d

Thyroglobulin (Tg) is a large protein secreted exclusively by the thyroid gland. In a clinical setting, it is measured for the purpose of follow-up of thyroidectomy patients. However, Tg measurements are often impeded by the presence of Tg autoantibodies and/or heterophylic antibodies that interfere with most measuring platforms. This presents a global problem in thyroid cancer patients who need to be postoperatively monitored for recurrent or residual disease. Therefore, in this paper we offer an overview of the existing methodologies and alternative approaches for Tg measurements that are a

Thyroglobulin (TG) plays a main role in the biosynthesis of thyroid hormones (TH), and, thus, it is involved in a wide range of vital functions throughout the life cycle of all vertebrates. Deficiency of TH production due to TG genetic variants causes congenital hypothyroidism (CH), with devastating consequences such as intellectual disability and impaired growth if untreated. To this day, 229 variations in the human TG gene have been identified while the 3D structure of TG has recently appeared. Although TG deficiency is thought to be of autosomal recessive inheritance, the introduction of ma

Canine thyroglobulin (cTg) is one of the thyroid autoantigens associated with hypothyroidism caused by autoimmune thyroiditis in dog. To identify canine-specific areas in cTg, we cloned, by reverse transcriptase PCR, and sequenced the complete cDNA of cTg. It revealed an open reading frame of 8289 nucleotides, which encode a polypeptide of 2762 amino acids that is 78.9 and 78.1% identical to bovine and human thyroglobulin, respectively. This complete cTg sequence may be useful to promote the understanding of the primary structure of cTg and, it will be informative data in the further search ab

Last year marked the 40th anniversary of the discovery that thyroglobulin (Tg) is a major autoantigen in autoimmune thyroiditis. The Tg molecule presents unique challenges for epitope mapping owing to its large size and extensive iodination. Consequently, pathogenic determinants have only recently been identified. Here, George Carayanniotis and Varada Rao summarize the approaches used to determine pathogenic Tg T-cell epitopes and discuss caveats in this unusual quest.
