Quality control assessment for the serological diagnosis of tick borne encephalitis virus infections.

BACKGROUND: The diagnosis of tick borne encephalitis (TBE) is mainly based on the demonstration of specific antibodies in serum when neurological disease is manifested. Improving diagnostics is the most important step in detecting and dealing with these pathogens. Quality control measures are essential for TBE diagnosis. OBJECTIVE: To assess an external quality assurance (EQA) program for the serologic diagnosis of TBE infections. STUDY DESIGN: A panel of 12 serum samples was sent out to be tested for the presence of TBE virus-specific IgM and IgG. This panel contained seven TBE-positive sam

Quality control assessment for the PCR diagnosis of tick-borne encephalitis virus infections.

BACKGROUND: Reverse transcriptase-polymerase chain reaction (RT-PCR) is an efficient method for the early detection of tick-borne encephalitis virus (TBEV) RNA in blood and serum samples taken prior to the appearance of antibodies. Improved diagnostics are critical for optimally detecting and managing TBE infections and quality control measures are therefore essential. OBJECTIVE: To assess the diagnostic quality of laboratories by performing an external quality assurance (EQA) programme for the molecular detection of TBE infections. STUDY DESIGN: A panel of 12 prepared human plasma samples w

Subunit vaccines for the control of tick-borne diseases: implications for the future.

Tick-borne parasites are a major constraint to the improvement of livestock productivity in the developing world. These parasites include Theileria parva, T. annulata, Babesia bigemina, B. bovis, Anaplasma marginale and Cowdria ruminantium. The impact of these diseases is currently limited by the use of acaricides to decrease transmission by the tick vectors and immunization of the host animals using live vaccines. The use of acaricide is hampered by the development of acaricide resistance and live vaccines are dependent on cold chain facilities, which are generally unreliable in developing co

Epidemiology of tick-borne pathogens of cattle and tick control practices among mixed farming and pastoral communities in Gairo and Monduli districts, Tanzania.

Tick-borne diseases including East Coast fever, anaplasmosis and babesiosis constitute a major constraint to improving livestock production worldwide, including Tanzania. Determination of the prevalence and factors associated with the occurrence of pathogens in cattle is important for informed decision making on the control and prevention of these diseases. However, little is known about the epidemiology of these pathogens in cattle in some regions of Tanzania. Therefore, this study aimed at establishing the prevalence of Anaplasma marginale, Babesia bigemina, Babesia bovis and Theileria parva

Tick testing as a method of controlling Rocky Mountain spotted fever.

In South Carolina, 1974-1980, only two matches were found between 536 Rocky Mountain spotted fever (RMSF) cases and 965 individuals who submitted ticks that tested rickettsial antigen positive. In neither case did the positive test prevent RMSF. Tick rickettsial positivity rates varied inversely with human RMSF attack rates in different geographic areas. A physician survey established it as unlikely that RMSF occurred in positive tick submitters (PTS), and that although not recommended, 34 per cent of asymptomatic PTS received prophylactic treatment. Only 18 per cent of positive ticks were eng

The potential effect of Garlium GEM HCTM as a tick control agent in cattle.

As resistance to chemical acaricides increases amongst tick populations, investigations of natural remedies that have acaricidal or tick-repellent effects have become essential in the search for alternatives for sustainable tick control. Garlium GEM HC (Garlium), a natural concentrated garlic product, was supplemented in-feed to beef cattle to determine if it could assist in reducing the number of attached ticks on cattle. Two grams of Garlium was supplemented daily to eight Bonsmara steers over 43 days. A second group of eight steers served as controls. All animals were infested with a simila

Tick-borne diseases in Europe: Current prevention, control tools and the promise of aptamers.

In Europe, tick-borne diseases (TBDs) cause significant morbidity and mortality, affecting both human and animal health. Ticks can transmit a wide variety of pathogens (bacteria, viruses, and parasites) and feed on many vertebrate hosts. The incidence and public health burden of TBDs are tending to intensify in Europe due to various factors, mainly anthropogenic and often combined. Early detection of tick-borne pathogens (TBPs), preventive measures and treatment are of great importance to control TBDs and their expansion. However, there are various limitations in terms of the sensitivity and/o

Availability and Nature of Commercial Tick Control Services in Three Lyme Disease Endemic States.

In an update of earlier surveys conducted in Connecticut and New Jersey in the mid-1990s, an online survey of private commercial pest control firms engaged in residential tick control showed that the application of synthetic acaricides continues to be the primary method of control used. The carbamate and organophospate acaricides, previously the most commonly used against ticks, have given way to synthetic pyrethroids and, to a lesser extent, the use of natural product/organic acaricides. Typical costs for a single acaricide application today ($100-$200 for a 1 acre [0.4 ha] property) remain s

The brown dog tick, Rhipicephalus sanguineus (Latreille, 1806) (Acari: Ixodidae): from taxonomy to control.

Rhipicephalus sanguineus, commonly known as the brown dog tick, is a three-host tick that feeds primarily on dogs and occasionally on other hosts, including humans. R. sanguineus ticks are widely distributed around the world and they are known vectors of pathogens, such as Babesia canis, Ehrlichia canis, and Rickettsia conorii. The increasing number of cases of human parasitism by R. sanguineus ticks reported in the literature indicates that the interaction between humans and R. sanguineus ticks may be more common than it is actually recognized. The indiscriminate use of acaricides is an emerg

Cellular control of the tick-borne virus antigen production in persistently infected cell culture.

The influence of inhibition or stimulation of cellular DNA synthesis on tick-borne virus antigen production in persistently infected cell culture was studied. Either mitomycin C or cytosine-arabinoside caused cessation of antigen-containing cell number increase. Stimulation of cellular DNA synthesis by growth medium change increased the level of antigen-containing cells. When HEp-2-Sof culture was synchronized, a correlation was observed between the entrance of cells into DNA synthesis phase and the increase of proportion of antigen-containing cells.

Unveiling the tick-borne pathogens from domestic ruminant ticks in Malawi and the emergence of the brown ear tick in the southern region: implications for East Coast fever control.

Ticks transmit a wide range of protozoan, bacterial, and viral pathogens to humans and animals globally. However, data on ticks infesting domestic ruminants and the pathogens they carry are scarce in Malawi. In this study, we examined ticks collected from domestic ruminants and screened them for selected veterinary and medically important protozoan and bacterial pathogens. A total of 964 ticks were collected from 202 cattle, 63 goats, and 16 sheep across eleven districts in Malawi. Ticks were morphologically identified to species level using taxonomical keys, with molecular confirmation by PCR

Control of ixodid ticks and prevention of tick-borne diseases in the United States: The prospect of a new Lyme disease vaccine and the continuing problem with tick exposure on residential properties.

In the United States, exposure to human-biting ixodid ticks can occur while spending time on residential properties or in neighborhood green spaces as well as during recreational or occupational activities on public lands. Human-biting tick species collectively transmit >15 species of pathogenic microorganisms and the national burden of tick-borne diseases is increasing. The prospect of a new Lyme disease vaccine for use in humans provides hope for substantial reduction in the >450,000 estimated annual cases of Lyme disease but this breakthrough would not reduce cases of other tick-borne disea

Updating the application of Metarhizium anisopliae to control cattle tick Rhipicephalus microplus (Acari: Ixodidae).

The bovine tick, Rhipicephalus microplus, is the main ectoparasite of cattle and causes loss of billions of dollars worldwide in lost meat, milk, and leather production, as well as control expenses. In addition to systemically impacting the host during the parasitic act, this parasite is also an important disease vector. Traditionally, the main commercial control of the tick is achieved through application of chemical acaricides, which can leave residues in the meat and milk. Moreover, ticks can become resistant to these chemicals due to their massive and incorrect use. Many alternative method

Does fluralaner (Exzolt® 5%) affect the enzootic stability of tick fever in beef calves subjected to strategic tick control in a tropical region?

BACKGROUND: Little is known about the impact of fluralaner on herd enzootic stability regarding tick fever pathogens (TFPs). This study aimed to assess whether implementing strategic control of Rhipicephalus microplus using fluralaner could influence the enzootic stability of beef herds regarding TFPs on a farm where this compound had never been previously used. METHODS: One hundred Bos taurus indicus calves were divided into two groups of 50 animals each. One group underwent strategic tick control with pour-on fluralaner at 2.5 mg/kg (FLU), while the control group received a pour-on combinat

Solar tick trap with a pheromone lure - A stand-in approach for off-host control of Rhipicephalus sanguineus sensu lato ticks.

Rhipicephalus sanguineus sensu lato (s.l.), the brown dog tick, transmits several important haemo-protozoan and rickettsial pathogens to dogs. Effective control of these ticks is entirely by the use of acaricidal preparations both on and off the host. Indiscriminate use of acaricides can cause environmental pollution and toxicity to humans as well as development of acaricidal resistance in ticks. As an alternative control strategy, a novel, eco-friendly and economical solar tick trap, which attracts ticks using pheromones and kills them by electrocution, was fabricated for use in kennels to co

Photosensitizers in the fight against ticks: safranin as a novel photodynamic fluorescent acaricide to control the camel tick Hyalomma dromedarii (Ixodidae).

Ticks transmit more pathogen species than any other group of blood-feeding arthropods worldwide, affecting humans, livestock, and companion animals. Hyalomma dromedarii is the predominant tick species infesting camels, and its effective control is of pivotal importance. In this research, we compared the phytoefficacy of safranin (SF), a fluorescent dye applied as an acaricide for the first time, to that of tetramethrin (TM) against engorged females of H. dromedarii through in vitro immersion bioassays. Furthermore, the effect of SF exposure was evaluated on the reproductive potential of surviv
