[Tooth germ transplantation or gap closure--a comparative study in the loss of the first molar].

In our study 37 transplanted and 33 mesialized molars by orthodontics were clinically and radiologically compared. The teeth moved by orthodontics showed a better long-term prognosis. As vital teeth with functioning periodontal structures and only slightly shortened roots they were equivalent to healthy teeth. However, the disadvantage of orthodontic space closure is the requirement of a good compliance due to long treatment time. The prognosis of maintaining a transplanted tooth germ is uncertain. One third of the teeth showed an altered periodontium with resorption areas. The danger of a con

Prenatal ultrasound and postmortem histologic evaluation of tooth germs: an observational, transversal study.

INTRODUCTION: Hypodontia is the most frequent developmental anomaly of the orofacial complex, and its detection in prenatal ultrasound may indicate the presence of congenital malformations, genetic syndromes and chromosomal abnormalities. To date, only a few studies have evaluated the histological relationship of human tooth germs identified by two-dimensional (2D) ultrasonography. In order to analyze whether two-dimensional ultrasonography of tooth germs may be successfully used for identifying genetic syndromes, prenatal ultrasound images of fetal tooth germs obtained from a Portuguese popul

[The role of pulp in the root resorption of primary teeth without permanent tooth germs].

OBJECTIVE: To investigate the role of pulp in the root resorption of primary teeth without permanent tooth germs. METHODS: The animal model without permanent tooth germs was established by surgery in Beagle dog. The root resorption was observed by taking periapical radiographs periodically. The samples of mandibular bone and pulp at different resorption stages were collected. The distribution of odontoclasts and the activating factor was analyzed by histological staining and semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR). The role of pulp in the root resorption of p

No developmental failure of cultured tooth germs from osteopetrotic (op/op) mice.

BACKGROUND: Incisor tooth germs of osteopetrotic (op/op) mice are known to fail to erupt, but form odontomas in their root apices instead, due to invasion of alveolar bone trabeculae into the tooth germs. The purpose of this study is to determine if the tooth developmental failures in op/op mice are intrinsic or secondarily arise as a result of the defective bone metabolism due to lack of macrophage colony-stimulating factor (M-CSF). METHODS: We isolated mandibular first molar tooth germs from normal and op/op mice and cultured them under conditions with or without bone tissues which had been

Spontaneous uprighting of permanent tooth germs after elimination of local eruption obstacles.

Four clinical cases are presented to demonstrate the self-correcting potential of aberrant tooth germs after the elimination of eruption obstacles (in 2 cases cysts, in 2 other cases severely infraoccluded primary teeth). In the case of the submerging deciduous teeth, the tilted adjacent teeth were orthodontically uprighted after the surgical procedure. Possible causative mechanisms are discussed.

[Etiology and clinical treatment strategies for second primary molars without permanent tooth germs].

Congenital tooth agenesis is a type of craniofacial developmental anomaly with reduced number of teeth, which is caused by disturbances in tooth germ development. If the number of missing teeth is less than six (excluding the third molars), it is termed as hypodontia. The second premolars are most commonly affected. When the second premolars are missing, the second primary molars are more prone to suffer from retention, infraocclusion, caries, pulpitis, or periapical periodontitis. Without timely prevention and appropriate treatment, congenital loss of second premolars may cause adverse effect

Relationship between congenitally missing lower third molars and late formation of tooth germs.

In this investigation, we used longitudinal panoramic radiographs from 96 subjects (47 boys, 49 girls) who did not have any congenital disease. The subjects were selected based on their age at the initial radiograph and the number and frequency of radiographs. Lower left premolars and second molars were used as parameters to identify factors that influence the age of formation of the third molar. We classified teeth into eight formation stages according to the method of Demirjian. The age at the beginning of formation of the lower third molar was determined with a regression equation of longit

Effects of suramin, a polyanionic drug inducing lysosomal storage disorders on tooth germs in vitro.

Suramin, a potent inhibitor of lysosomal enzymes, is commonly employed as a tool for inducing experimental mucopolysaccharidosis and lipidosis. The effects of the drug on embryonic mouse molars were analysed. Presecretory ameloblasts and odontoblasts were loaded with lysosome-like vacuoles. Staining with MC22-33F, an antibody to choline phospholipids and sphingomyelin, was completely reversed in the suramin-treated germs, in that it stained only presecretory ameloblasts (versus odontoblasts and some pulpal cells in the control group), according to a developmentally regulated pattern. The suram

Ultrastructure of a new generation of odontoblasts in grafted coronal tissues of mouse molar tooth germs.

Third molar tooth germs were removed from 14-day-old mice and freed from the enamel organ and follicle. After section of the apical tissues, including Hertwig's sheath, they were transplanted in 1-day-old newborn mice of the same lineage. Electron microscopy of grafts removed 7, 14 and 21 days later showed that, following the disappearance of the initial layer of odontoblasts and a period of adaptation, 14 days after transplantation newly differentiated odontoblasts deposited tubular dentine. The dentine matrix production was increased over that of controls, demonstrating that synthesis was ac

Spatial organization of the epithelium and the role of neural crest cells in the initiation of the mammalian tooth germ.

Teeth develop from composite organ rudiments that are formed through the interaction of oral epithelium and mesenchyme of the first branchial arch; cells of the former differentiate into enamel-secreting ameloblasts whereas those of the latter differentiate into dentine-secreting odontoblasts. Experimental analysis of odontogenic tissue interactions in mammalian embryos has focused on the late developmental stages of morphogenesis and cytodifferentiation; little is known about initial pattern-forming events, during which presumptive tooth-forming cells are specified and the sites of tooth init

Immunolocalization of gamma-carboxyglutamic acid-containing proteins in developing molar tooth germs of the rat.

The localization of dentin gamma-carboxyglutamic acid containing proteins (DGP's) was examined in rat molar tooth germs at various stages of development by immunostaining with the peroxidase-antiperoxidase method. Odontoblasts and their processes stained positive, whereas all other cells were negative. No staining was found in predentin. Dentin stained only after a mild demineralization. The results support the concept that DGP's are deposited by odontoblasts directly into the mineralizing dentin.

A Case of Pediatric Garré's Osteomyelitis Caused by Germ Infection in the Lower Impacted Wisdom Tooth.

Garré's osteomyelitis, first described by Carl Garré in 1893, is a type of chronic osteomyelitis accompanied by hyperplastic periostitis. This condition affects relatively young patients and occurs in the fibula, femur, and other long bones as chronic non-purulent sclerosing osteomyelitis. Further, reactive periosteal bone formation develops due to chronic irritation or infection. In the maxillofacial region, it often occurs in the first molar region of the mandible due to caries and other similar causes, and it is rarely associated with impacted teeth. Herein, we present a 12-year-old female

[In vitro effect of chlorpromazine on the mineralization of tooth germ in mice--comparison with that of retinoic acid and HEBP].

Effects of chlorpromazine on the mineralization and alkaline phosphatase activity (ALP) in the tooth germ were examined and compared with those of retinoic acid and HEBP (1-hydroxyethylidene-1, 1-bisphosphonate). Mandibular first molars from 17-day-old mouse embryos were cultured with or without drugs. Calcium content and ALP in the tooth germ increased gradually from 0 to 7 days in culture, the increase of calcium being preceded by that of ALP. Retinoic acid suppressed increases of calcium and ALP in the tooth germ but not in the specimens precultured for 2 days, suggesting that retinoic acid

Ultrastructural distribution of sulfated glycosaminoglycans in epithelial-mesenchymal interface of developing rat tooth germs.

We investigated the ultrastructural distribution of sulfated glycosaminoglycans in the epithelial-mesenchymal interface of tooth germs by use of the high-iron diamine thiocarbohydrazide silver proteinate (HID-TCH-SP) staining and enzymatic digestion method. At an early stage in odontoblast differentiation, HID-TCH-SP stain deposits were sparsely distributed in the basement membrane and in the intercellular spaces. Subsequently, as formation of the initial predentin matrix began, HID-TCH-SP stain deposits were densely distributed in the interfibrillar spaces and the basement membrane. Testicula

Effect of ascorbic acid deficiency on mouse second molar tooth germs cultivated in vitro.

Mandibular second molar tooth germs from two-day old mice were cultured in vitro, on millipore membranes, for periods of up to 20 days in liquid medium with or without added ascorbic acid. Tooth germs grown in ascorbate medium were characterized by relatively normal growth, differentiation, morphology and histology. Cuspation patterns were maintained. The epithelial root sheath continued to grow along the millipore membrane. Tooth germs cultured in ascorbate-deficient medium manifested a consistent and striking failure in maintenance of differentiated odontoblastic and ameloblastic tissue with
