Reconsidering remineralization strategies to include nanoparticle hydroxyapatite.

Dental caries is a transmissible biofilm-mediated disease of the teeth that is defined by prolonged periods of low pH resulting in net mineral loss from the teeth. Hydroxyapatite, fluorapatite, and the carbonated forms of calcium phosphate form the main mineral content of dental hard tissues: enamel, dentin, and cementum. Active dental caries results when the biofilm pH on the tooth surface drops below the dissolution threshold for hydroxyapatite and fluorapatite. The clinical evidence of this net mineral loss is porosity, whitespot lesions, caries lesions, and/or cavitation. The potential to

Comparative Evaluation of Remineralization Efficacy of a Novel Proanthocyanidin Biovarnish with Fluoride Varnish on Primary Teeth: An In Vitro Study.

AIMS AND BACKGROUND: Early dental caries are caused by subsurface demineralization of enamel and manifests 0.0015 as noncavitated, white lesions. Remineralization is a process that replaces lost mineral ions in demineralized enamel. Saliva naturally aids in remineralization. However, this process is slow and can be hampered under cariogenic conditions. Fluoride has been a significant caries-preventive agent, promoting remineralization, and preventing demineralization. However, interest in non-fluoridated treatments has increased due to concerns about dental fluorosis. Proanthocyanidins (PACs),

The Potential of QP3VH-Chitosan Peptide as Biomimetic Remineralization in Early Dental Caries Treatment: An In Vitro Study.

OBJECTIVES: The development of remineralization biomimetics using organic peptide molecules is expected to resemble the hydroxyapatite (HA) mineralization process in tooth enamel. The development of an amelogenin derivative peptide combined with antimicrobial peptide was designed, resulting in QP3VH. This combination then was mixed with chitosan as a carrier. This study aimed to evaluate the biomimetic efficacy of QP3VH as a remineralizing agent combined with chitosan. MATERIALS AND METHODS: Fifty deciduous mandibular incisor enamel samples were used in this study. The artificial enamel lesio

Enamel Remineralizing Agents: State of the Art.

Dental caries remains the most prevalent chronic disease worldwide, yet early enamel lesions are reversible if managed with appropriate remineralizing agents. This narrative review synthesizes current evidence on remineralizing agents, their mechanisms of action, and clinical applications, with a focus on dental materials used in preventive and minimally invasive dentistry. Traditional fluoride-based approaches enhance remineralization through fluorapatite formation; however, their effectiveness is limited when calcium and phosphate bioavailability is insufficient. Biomimetic agents, including

Remineralization of root surfaces demineralized in solutions of differing fluoride levels.

The beneficial effects of fluoride on enamel have been well documented. However, limited data are available concerning the amount of fluoride required for beneficial effects on tooth root. Although studies have shown that fluoride inhibits root demineralization, the aim of this study was to investigate the location, extent and amount of remineralization on root dentin substrates after demineralization has occurred. The root surfaces of extracted human teeth were demineralized in a pure chemical buffer containing varying concentrations of sodium fluoride. After this lesion initiation, the same

In Vivo and In Vitro Response to a Regenerative Dental Scaffold.

As dental pulp contains the stem cells necessary for regeneration, the tooth should hold the intrinsic capacity for self-repair. A triphasic hybrid dental biocomposite (3HB) composed of biocompatible biopolymers to provide strength, antibacterial properties and protein-based cell support could provide a conducive microenvironment for the regeneration of dental structures. 3HB was incorporated into Mineral Trioxide Aggregate (ProRoot MTA) to construct a malleable injectable implant. Human tooth pulp cells (hDPCs) significantly increased proliferation in the presence of 3HB+MTA compared to 3HB o

Yellow light and ultrasound Dual-responsive strontium-doped zinc oxide composites for dental caries prevention and remineralization.

Dental caries is one of the most prevalent chronic infectious diseases, mainly due to acid production by bacteria in the plaque biofilm, leading to enamel demineralization, tooth defects and pulpitis. Most of the current treatments are invasive and do not combine restoration of dental tissues, resolution of tooth staining or prevention of dental caries. In this study, a polydopamine (PDA)-encapsulated strontium-doped zinc oxide composite (Sr-ZnO@PDA, denoted as SZ@PDA) has been developed for caries prevention and remineralization. The PDA wrapping endows SZ@PDA with a narrow energy band gap (1

Targeted antimicrobial self-assembly peptide hydrogel with in situ bio-mimic remineralization for caries management.

The single-function agents with wide-spectrum activity which tend to disturb the ecological balance of oral cavity cannot satisfy dental treatment need. A multi-functional agent with specifically targeted killing property and in situ remineralization is warranted for caries management. A novel multi-functional agent (8DSS-C8-P-113) consisting of three domains, i.e., a non-specific antimicrobial peptide (AMP) (P-113), a competence stimulating peptide (C8), and an enhancing remineralization domain (8DSS), is fabricated and evaluated in this study. The findings demonstrates that 2 μM mL-1 of 8DSS

The Remineralization of Enamel from Saliva: A Chemical Perspective.

The natural remineralization of enamel is of major importance for oral health. In principle, early erosions (demineralization) induced by acidic beverages and foods as well as initial caries lesions can be covered and remineralized by the deposition of calcium phosphate, i.e., tooth mineral. This remineralization effect is characterized by the presence of calcium and phosphate ions in saliva that form hydroxyapatite on the enamel surface. Although it is apparently a simple crystallization, it turns out that remineralization under in vivo conditions is actually a very complex process. Calcium p

Engineered antibacterial and remineralization strategies for efficient dental caries management.

Dental caries, one of the most prevalent oral diseases among adults and children, is caused by oral microorganisms and diet and involves a complex, multi-step process of biofilm formation and tooth decay. The existing clinical treatment modalities are not satisfactory due to the disadvantages of incomplete eradication of pathogens, poor material compatibility and susceptibility to secondary caries. In recent years, there has been a growing interest in treating dental caries, with various strategies emerging to combat these bacterial films and promote tooth remineralization. However, comprehens

A multifunctional polymeric coating with self-adsorbed, antifouling and in situ remineralization properties for caries management.

Dental caries is a biofilm-induced bacterial infectious oral disease, where the early attachment of proteins and pathogenic bacteria to tooth surfaces has been known as the main cause of biofilm formation. Typically, dental caries is commonly accompanied by mineral depletion of enamels, thus causing dental demineralization. Multifunctional materials are highly attractive candidates for treating dental caries. Herein, we successfully synthesized diblock copolymers poly(ethylene glycol)-b-poly(aspartic acid) (PEG-PAsp) and modified them with alendronate sodium (ALN) to serve as bioactive bifunct

Layer-by-layer coated probiotics with tannic acid-Ca2+ and casein phosphopeptide complexes for caries prevention and enamel remineralization.

Dental caries is a common disease resulting from tooth demineralization caused by bacterial plaque. Probiotics have shown great potential against caries by regulating the balance of oral flora. However, obstacles such as poor colonization and lysozyme sensitivity in oral cavity hinder their further application. In this study, an efficient layer-by-layer surface coating of tannic acid (TA)-Ca2+ and casein phosphopeptide (CPP) was applied to the probiotic Bifidobacterium infantis (BI) with potential anti-caries activity. Multi-functionalized probiotics thus prepared (called BI@TA-Ca2+@CPP) exhib

Remineralizing efficacy of Calcarea Fluorica tablets on the artificial carious enamel lesions using scanning electron microscope and surface microhardness testing: in vivo study.

CONTEXT: Remineralization is defined as the process whereby calcium and phosphate ions are supplied from a source external to tooth to promote ion deposition into crystal voids in demineralized enamel to produce net mineral gain. The remineralization produced by saliva is less and also a slow process, therefore remineralizing agents are required. AIMS: The study was planned to evaluate the effectiveness of homeopathic Calcarea Fluorica (calc-f) tablets as remineralizing agents on artificial carious lesions using scanning electron microscope (SEM) and surface microhardness (SMH) testing. An engineered dual-functional peptide with high affinity to demineralized dentin enhanced remineralization efficacy in vitro and in vivo.

Dental caries continues to be a major global public health problem. Remineralization of demineralized dentin is regarded as one of the hotspots in the current study in the treatment of dental caries. However, traditional remineralization agents, which usually lack the ability to bind to demineralized dentin collagen, are easily removed by the fluids in the oral cavity, thus decreasing the remineralization efficacy. Non-collagenous proteins (NCPs) have significant effects on the biomineralization of dentin due to their dual high binding capacity to the collagen fibers and minerals. But NCPs are

Triple Function of Amelogenin Peptide-Chitosan Hydrogel for Dentin Repair.

Biomimetic strategies like peptide-guided collagen mineralization promise to enhance the effectiveness of dentin remineralization. We recently reported that rationally designed amelogenin-derived peptides P26 and P32 promoted apatite nucleation, mineralized collagen, and showed potential in enamel regrowth and dentin remineralization. To facilitate the clinical application of amelogenin-derived peptides and to uncover their effectiveness in repairing dentin, we have now implemented a chitosan (CS) hydrogel for peptide delivery and have investigated the effects of P26-CS and P32-CS hydrogels on
