Newly synthesized and preformed acetylcholine are released from Torpedo synaptosomes by different pathways.

In this study, we investigated the mechanisms underlying the release of preformed and of newly synthesized acetylcholine (ACh) from isolated Torpedo nerve terminals (synaptosomes). This was pursued by examining and comparing the effects of anticytoskeletal and anticalmodulin drugs and of activating the presynaptic muscarinic ACh receptors on the release of preformed endogenous ACh and of newly synthesized radiolabeled ACh. The anticytoskeletal drugs vinblastine, cytochalasin B, and colchicine inhibit the Ca2+-dependent K+-mediated release of newly synthesized radiolabeled ACh, but have no effe

A Plastic Torpedo-Shaped Firecracker That Caused Injury to the Parotid Gland: A Case Report.

Penetrating injuries affecting the parotid gland are relatively rare compared to injuries affecting other head and neck structures. Likewise, foreign bodies impacting the parotid gland are extremely rare. These foreign bodies can be secondary to penetrating injury or may reach the parotid gland by retrograde migration through Stensen's duct. Management of parotid gland foreign bodies is a challenging clinical situation for otolaryngologists due to the course of the facial nerve through the gland. In this article, a patient with penetrating parotid injury as a result of the explosion of a torpe

Torpedo-like lesions in the ocular fundi of Gardner syndrome: hiding in plain view.

Background: The coexistence of fundus torpedoes and Gardner syndrome was initially reported in 1989 and 1992 by Gass and Roseman. Gardner syndrome is a dominantly inherited disease characterized by a multitude of potentially lethal gastrointestinal polyps. Over the ensuing quarter century, several publications demonstrated uncommon, but easily recognized, fundus lesions called Torpedo Maculopathy in apparently normal individuals. These fundus abnormalities were thought to be single and isolated without systemic associations. During the same period of time, a variety of nonspecific fundus abnor

Tacrine and physostigmine block nicotinic receptors in Xenopus oocytes injected with Torpedo electroplaque membranes.

Tacrine and physostigmine were tested for direct nicotinic actions on Xenopus oocytes microinjected with Torpedo electroplaque membranes. In this preparation, responses to acetylcholine arise 6-8 h after microinjection, due to the incorporation of nicotinic receptors into the plasma membrane by a process not involving protein synthesis. Currents elicited by acetylcholine (100-1000 microM) were recorded by two-electrode voltage clamping. Tacrine (1-1000 microM) and physostigmine (1-100 microM) exerted a potent, reversible block of the nicotinic receptors. The concentration-dependence curves fit

Agelenopsis aperta venom and FTX, a purified toxin, inhibit acetylcholine release in Torpedo synaptosomes.

The presence of P-type calcium channels in synaptosomes prepared from electric organ of Torpedo marmorata was investigated by using the venom of Agelenopsis aperta, a toxin purified from it, FTX, and its synthetic analog. We analysed the action of these agents on acetylcholine release which was continuously followed using a chemiluminescent assay. Agelenopsis aperta venom, FTX and synthetic FTX inhibit acetylcholine release from synaptosomes induced by a presynaptic membrane depolarization with 60 mM KCl. A stronger inhibition of acetylcholine release was observed with the venom than with FTX

Interactions of piperidine derivatives with the nicotinic cholinergic receptor complex from Torpedo electric organ.

The interactions of eight piperidine derivatives with nicotinic receptor complexes from Torpedo californica electric organ were studied using [125I] alpha-bungarotoxin [( 125I]BGT) as a probe for the acetylcholine binding site and [3H]perhydrohistrionicotoxin [( 3H]H12-HTX) as a probe for a site associated with the receptor-gated ion channel. Cis- and trans-2-methyl-6-n-undecanyl piperidines (MUP), major constituents of fire ant venom, had a high-affinity for [3H]H12-HTX binding sites (Ki = 0.08-0.24 microM), but had no affect on receptor binding. MUP affinity for [3H]H12-HTX binding sites was

Ultraviolet light-induced labeling by noncompetitive blockers of the acetylcholine receptor from Torpedo marmorata.

Reversible ligands were attached covalently to membrane-bound acetylcholine receptor from Torpedo marmorata by a method which is generally applicable and does not require the synthesis of specially designed molecules. UV irradiation of the receptor in the presence of [3H]trimethisoquin, [3H]phencyclidine, or [3H]perhydrohistrionicotoxin resulted in the labeling of the binding site(s) for these noncompetitive blockers of the permeability response. The labeling of the delta chain was enhanced by carbamoylcholine, and this increase was blocked by snake alpha-toxins. The effect of carbamoylcholine

Bioaccumulation of organotin compounds in the marbled electric ray (Torpedo marmorata) in the northern Adriatic Sea.

Organotin compounds (OTC), tri-, di- and monobutyl tin, were determined in the tissues of marbled electric ray (Torpedo marmorata) in the Adriatic Sea. Marbled electric ray specimens were provided by local fishermen from three localities in the northern Adriatic: area close to the shipyard in Seča, the natural protected area Strunjan Nature Reserve and along the west Istrian coast. To assess the concentration of OTC in the environment, sediment samples were also analysed. After an adequate extraction of OTC from both matrices, their concentrations were determined by GC-ICP-MS. The results indi

Coexistence of torpedo maculopathy and retinoblastoma: Differentiation the lesions with hand-held optical coherence tomography.

Torpedo maculopathy (TM) is a rare congenital defect of the retina pigment epithelium, which always localizes to the temporal fovea. It is most commonly unilateral and often occurs in patients without any relevant disease. Classic finding is a flat, hypopigmented, oval lesion, resembling a 'torpedo,' with a wedge-shaped tail extending outward and directed toward the foveola. Visual acuity is generally unaffected due to the central fovea is not involved and this lesions remain stable. Therefore, when it accompanied with a posterior segment disease, we should make differential diagnosis to avoid

Inhibition by trimethyl-tin, probenecid and phenylglyoxal of depolarization-induced acetylcholine release in Torpedo synaptosomes.

The effects of trimethyl-tin (anion-hydroxyde ionophore, inhibiting oxydative phosphorylation and H(+)-ATPase) probenecid (inhibitor of anion transport in neural cells) and phenylglyoxal (arginine-specific reagent, inhibiting chloride exchanges in erythrocytes) were examined in Torpedo synaptosomes prepared from electric organ. All drugs significantly reduced the stimulated release of acetylcholine triggered by depolarization of nerve endings with high-K(+) and/or gramicidin D. In contrast, trimethyl-tin, probenecid and phenylglyoxal did not affect the ionophore A23187-induced release of acety

Torpedo Maculopathy in a 6-Month-Old Infant: Early Clinical and Optical Coherence Tomography Findings.

A 6-month-old male infant presented for routine ophthalmologic examination. Indirect ophthalmoscopy revealed a flat, oval, hypopigmented lesion located in the temporal macula in the right eye with the tip pointing toward the fovea, which was compatible with torpedo maculopathy. Optical coherence tomography (OCT) was performed at the time of diagnosis. OCT scans of the lesion revealed slight retinal pigment epithelium hyperreflectivity. This case serves as the earliest OCT finding of the youngest patient diagnosed as having torpedo maculopathy in the literature. [J Pediatr Ophthalmol Strabismus

Spontaneous release of acetylcholine from Torpedo synaptosomes: effect of cetiedil and its analogue MR 16728.

The effects of cetiedil and its analogue MR 16728 were examined on spontaneous acetylcholine release measured with a chemiluminescent assay using choline oxidase in a synaptosomal suspension obtained from Torpedo marmorata electric organ. Evoked acetylcholine release is inhibited by cetiedil, whereas this drug enhances spontaneous extracellular Ca(2+)-independent acetylcholine release (up to 340%). This effect was examined as a function of cetiedil concentration and incubation time. On the other hand, the analogue MR 16728, which enhances A23187-evoked acetylcholine release, also enhances spon

A 3-YEAR-OLD PEDIATRIC CASE OF TORPEDO MACULOPATHY ASSOCIATED WITH A SEROUS RETINAL DETACHMENT.

PURPOSE: To report an atypical retinal finding of torpedo maculopathy in a 3-year-old boy. METHOD: A retrospective case report. RESULTS: Optical coherence tomography examination of the retinal lesion showed a hyper-reflectivity of the outer layers associated with a serous retinal detachment with an extension of the outer segments of the photoreceptors. Furthermore, the external limiting membrane and the retinal pigment epithelium are intact. Although the reflectance of the ellipsoid zone seems diminished, no disruptions were observed, suggesting that the ellipsoid zone is likely undamaged.

Acetylcholine receptor and ionic channel of Torpedo electroplax: binding of perhydrohistrionicotoxin to membrane and solubilized preparations.

The electric organ of the ray, Torpedo ocellata, can serve as a source for both the acetylcholine (ACh) receptor and its ionic channel. The two entities were identified by their specific binding of [3H]ACh and [3H]perhydrohistrionicotoxin ([3H]H12-HTX), respectively. Binding of [3H]H12-HTX was inhibited by certain drugs and toxins, e.g., histrionicotoxin (HTX), amantadine, and tetraethylammonium (TEA) ions at concentrations that did not inhibit [3H]ACh binding. However, the specific carbamoylcholine-induced 22Na efflux from microsacs from the electric organ membranes was blocked by inhibitors

T cell receptor transgenic mice recognizing the immunodominant epitope of the Torpedo californica acetylcholine receptor.

Myasthenia gravis (MG) is an autoimmune disease caused by T cell-dependent antibody-mediated reduction of acetylcholine receptors (AChR) at the neuromuscular junction. Immunization of animals with Torpedo californica AChR (TAChR) results in an experimental model of MG. We used the variable regions of alpha and beta T cell receptor (TCR) genes recognizing an immunodominant peptide containing amino acids 146-162 from the alpha subunit of TAChR presented in the context of I-A(b) to generate TCR-transgenic mice. We found that the transgenic TCR was strongly positively selected and that transgenic

Distribution patterns of torpedo maculopathy: Further evidence of a congenital retinal nerve fiber layer-driven etiology.

With fewer than 100 peer-reviewed cases reported in the world to date, the underlying etiology of torpedo maculopathy has remained elusive. In this literature review, we provide new evidence to better support, reject and unify claims regarding cause, diagnosis, and proper clinical management of this disease. We reviewed 44 case reports and case series, which included 77 patients (after exclusions). We additionally introduced 3 new cases from our clinical practice for a total of 80 cases. Ages at presentation ranged from 6 months old to 73 years old (mean: 24.2 years old). The nasal aspects of
