Differential expression of monocyte chemotactic protein-1 (MCP-1) in transforming rat hepatic stellate cells.

BACKGROUNDS/AIMS: Hepatic stellate cells and infiltrating leukocytes play a key role in the pathogenesis of liver fibrosis. The chronic phase of liver inflammation is characterized by immigrating mononuclear cells. To understand the underlying mechanisms responsible for the attraction of mononuclear cells in the pathogenesis of liver fibrosis, we investigated the inducible production of chemotactic activities in hepatic stellate cells. METHODS: Cultured hepatic stellate cells of different transformation grades and after in vitro transformation to myofibroblast-like cells were stimulated with

Role of microbial activity in Fe(III) hydroxysulfate mineral transformations in an acid mine drainage-impacted site from the Dabaoshan Mine.

Fe(III) hydroxysulfate minerals are secondary minerals commonly found in acid mine drainage (AMD) sites and have a major impact on water and soil quality in these environments. While previous studies showed that the Fe(III) hydroxysulfate mineral transformation could be mediated by some bacterial strains under laboratory conditions, the role of indigenous microbial activity in Fe(III) hydroxysulfate mineral transformation in natural environment has received little attention. In this study, microcosms were constructed with AMD-affected river water and sediment from the Dabaoshan Mine that was e

Basic Characterization of Natural Transformation in Avibacterium paragallinarum.

Avibacterium paragallinarum is the pathogen involved in infectious coryza (IC), an acute infectious upper respiratory disease in chickens. The prevalence of IC has increased in China in recent years. There is a lack of reliable and effective procedures for gene manipulation, which has limited the research on the bacterial genetics and pathogenesis of A. paragallinarum. Natural transformation has been developed as a method of gene manipulation in Pasteurellaceae by the introduction of foreign genes or DNA fragments into bacterial cells, but there has been no report on natural transformation in

Intragenomic conflict between transformation and prophage explains taxon-specific DNA acquisition.

The activation of many chromosomally integrated mobile genetic elements by RecA-DNA nucleoprotein filaments may represent an adaptation to evading deletion through homologous recombination. Consistent with bacterial transformation facilitating such eliminations of inserted viral DNA, cross-taxa comparisons found isolates' prophage content was negatively associated with homologous recombination rates. Assaying the effect of the composition of the extracellular DNA pool on transformation of a lysogen demonstrated that DNA from conspecific donors drove both prophage activation and deletion, where

[Seasonal Response Features of Microbial Community Structure for Nitrogen Transformation in Inland Lakes on the Qinghai-Xizang Plateau].

Studying the nitrogen transformation characteristics of microbial communities in the inland lakes of the Qinghai-Xizang Plateau (QXP) is of great significance for a deeper understanding of the nitrogen budget balance and biogeochemical cycling in the regional lake ecosystems. Based on metagenomic sequencing technology, this study constructed a non-redundant gene library of nitrogen-transforming microorganisms and conducted multivariate statistical analysis to explore the characteristics and assembly mechanisms of nitrogen-transforming microbial communities in inland lakes on the QXP. The resul

Bacterial models for tumor development. Mini-review.

UNLABELLED: The tumor-inducing effects of Agrobacterium, Bartonella and Helicobacter bacterial species are compared step by step. An analogy for the existence of these individual steps is considered in connection with the development of cancer. The transformations of eukaryotic cells occur in particular in the type IV secretion system, i.e. involving the simultaneous transmission of DNA and protein from bacterial cells to eukaryotic cells. Thus, transfected cells facilitate the indefinite growth of tissue cells and additionally produce growth factors, triggering further bacterial multiplicatio

Cross-Genus "Boot-Up" of Synthetic Bacteriophage in Staphylococcus aureus by Using a New and Efficient DNA Transformation Method.

Staphylococcus aureus is an opportunistic pathogen that causes a wide range of infections and food poisoning in humans with antibiotic resistance, specifically to methicillin, compounding the problem. Bacteriophages (phages) provide an alternative treatment strategy, but these only infect a limited number of circulating strains and may quickly become ineffective due to bacterial resistance. To overcome these obstacles, engineered phages have been proposed, but new methods are needed for the efficient transformation of large DNA molecules into S. aureus to "boot-up" (i.e., rescue) infectious ph

Heat shock-mediated transformation is possible in several Gram-negative bacteria.

Transformation of foreign DNA, subjecting Escherichia coli to CaCl2 treatment followed by heat shock exposure, is a regular approach in recombinant DNA technology. However, in spite of its popularity in E. coli, heat shock transformation is rarely reported in other Gram-negative bacteria. The objective of this research is to demonstrate that the heat shock transformation method can also be performed to introduce genes in different Gram-negative bacteria, such as Ralstonia pseudosolanacearum, Pseudomonas aeruginosa, Pseudomonas putida, and Enterobacter roggenkampii. A green fluorescent plasmid,

Biotechnological transformation of Miscanthus x giganteus: Technological and environmental aspects.

Miscanthus is recognized as a priority feedstock for sustainable bioeconomy. However, the technological and environmental aspects of biotechnological transformation of Miscanthus remain understudied. In this work, a complete cycle of transformation of Miscanthus into bacterial nanocellulose, bioethanol and lactic acid was carried out, and the yields of the bio-based products from 1 ton of the feedstock and sustainability factors of production were estimated for the first time. Miscanthus giganteus from the Russian selection (the Kamis variety) was used as the feedstock. The complete cycle of b

Electroactive bacteria-driven electron transfer accelerates molecular transformation of tetracycline in iron-carbon coupled wetland microbial fuel cells.

Conventional constructed wetlands (CWs) often suffer from adsorption saturation and incomplete degradation when treating antibiotic wastewater, posing risks of pollutant re-release. An iron-carbon substrate was coupled with a microbial fuel cell in a vertical subsurface flow CW (FeC-MFC) based on the hypothesis that electroactive microbial enrichment and strengthened electron transfer would promote TC transformation and treatment stability under chronic TC stress (800 μg/L). FeC-MFC achieved 99.97% TC removal, reducing residual parent TC to 0.28 μg/L, while maintaining stable COD (86.95%) and

Dichotomous Role of Humic Substances in Modulating Transformation of Antibiotic Resistance Genes in Mineral Systems.

Widespread antibiotic resistance genes (ARGs) have emerged as a focus of attention for public health. Transformation is essential for ARGs dissemination in soils and associated environments; however, the mechanisms of how soil components contribute to the transformation of ARGs remain elusive. Here we demonstrate that three representative mineral-humic acid (HA) composites exert contrasting influence on the transformation of plasmid-borne ARGs in Bacillus subtilis. Mineral surface-bound HA facilitated transformation in kaolinite and montmorillonite systems, while an inhibitory effect of HA was

Agrobacterium-mediated genetic transformation of chia (Salvia hispanica L.), a rich source of omega-3 fatty acid.

Chia (Salvia hispanica L.), a nutritionally valuable crop, is the richest source of α-linolenic acid, a key omega-3 fatty acid. Despite its nutritional benefits, a stable genetic transformation method for chia is not available. This study presents a sonication-assisted Agrobacterium-mediated transformation protocol optimized for high-efficiency transformation of chia seedlings. Key parameters including bacterial cell density, acetosyringone concentration, sonication duration, vacuum infiltration, and infection time were optimized. Results demonstrated that an optical density at 600 nm (OD600)

[Hemolytic-uremic syndrome in pneumococcal meningitis and infection. Importance of T-transformation].

A 4 month old girl developed a severe hemolytic uremic syndrome (HUS) following pneumococcal sepsis and meningitis. As a result of the hemolytic anemia and thrombocytopenia with associated gastrointestinal bleeding several red blood cell and thrombocyte transfusions became necessary. No plasma was transfused to the patient and all cellular blood components to be transfused were washed thoroughly in order to avoid the administration of eventually dangerous donor anti-T antibodies. Continuous peritoneal dialysis was performed for 23 days until sufficient spontaneous urine production was resumed.

Shwachman-Diamond syndrome: early bone marrow transplantation in a high risk patient and new clues to pathogenesis.

Shwachman-Diamond syndrome (SDS) is an autosomal recessive disorder characterised by exocrine pancreas insufficiency, metaphyseal dysostosis and bone marrow dysfunction. Recurrent severe bacterial infections and susceptibility to leukaemia are the major causes of morbidity and mortality occurring preferentially in patients with pancytopenia and features of myelodysplasia. Here we report a patient with SDS leading to recurrent bacterial infections and a deteriorating condition since early infancy. Extensive investigations disclosed severe pancytopenia, myelodysplasia and a clonal cytogenetic ab

Identification of Bacterial Metabolites Modulating Breast Cancer Cell Proliferation and Epithelial-Mesenchymal Transition.

Breast cancer patients are characterized by the oncobiotic transformation of multiple microbiome communities, including the gut microbiome. Oncobiotic transformation of the gut microbiome impairs the production of antineoplastic bacterial metabolites. The goal of this study was to identify bacterial metabolites with antineoplastic properties. We constructed a 30-member bacterial metabolite library and screened the library compounds for effects on cell proliferation and epithelial-mesenchymal transition. The metabolites were applied to 4T1 murine breast cancer cells in concentrations correspond

High levels of genetic recombination during nasopharyngeal carriage and biofilm formation in Streptococcus pneumoniae.

UNLABELLED: Transformation of genetic material between bacteria was first observed in the 1920s using Streptococcus pneumoniae as a model organism. Since then, the mechanism of competence induction and transformation has been well characterized, mainly using planktonic bacteria or septic infection models. However, epidemiological evidence suggests that genetic exchange occurs primarily during pneumococcal nasopharyngeal carriage, which we have recently shown is associated with biofilm growth, and is associated with cocolonization with multiple strains. However, no studies to date have comprehe

Genotoxic Effect of Salmonella Paratyphi A Infection on Human Primary Gallbladder Cells.

Carcinoma of the gallbladder (GBC) is the most frequent tumor of the biliary tract. Despite epidemiological studies showing a correlation between chronic infection with Salmonella enterica Typhi/Paratyphi A and GBC, the underlying molecular mechanisms of this fatal connection are still uncertain. The murine serovar Salmonella Typhimurium has been shown to promote transformation of genetically predisposed cells by driving mitogenic signaling. However, insights from this strain remain limited as it lacks the typhoid toxin produced by the human serovars Typhi and Paratyphi A. In particular, the C

Fe/S Redox-Coupled Mercury Transformation Mediated by Acidithiobacillus ferrooxidans ATCC 23270 under Aerobic and/or Anaerobic Conditions.

Bioleaching processes or microbially mediated iron/sulfur redox processes in acid mine drainage (AMD) result in mineral dissolution and transformation, the release of mercury and other heavy metal ions, and changes in the occurrence forms and concentration of mercury. However, pertinent studies on these processes are scarce. Therefore, in this work, the Fe/S redox-coupled mercury transformation mediated by Acidithiobacillus ferrooxidans ATCC 23270 under aerobic and/or anaerobic conditions was studied by combining analyses of solution behavior (pH, redox potential, and Fe/S/Hg ion concentration

The antihistamine diphenhydramine is demethylated by anaerobic wastewater microorganisms.

While emerging pharmaceutical contaminants are monitored in wastewater treatment and the environment, there is little information concerning their microbial metabolites. The transformation of diphenhydramine by microorganisms in anaerobic digester sludge was investigated using anaerobic cultures amended with 1 mM diphenhydramine as the sole carbon source. Complete transformation of the parent compound to a persistent metabolite occurred within 191 days. Using GC/MS analysis, the metabolite was identified as N-desmethyl diphenhydramine. Loss of the parent compound diphenhydramine followed a fir

Microbiota-mediated enhancement of the chondroprotective effects of epimedium polysaccharides: Insights from fermentation and zebrafish models.

The health benefits of dietary polysaccharides as functional macromolecules have attracted increasing attention, yet their digestive fate, fermentability, and biological relevance after microbial transformation remain incompletely understood. In the present study, we systematically investigated the gastrointestinal digestion resistance, gut microbial fermentation behavior, and chondroprotective potential of Epimedium polysaccharides (EPs). Simulated salivary-gastrointestinal digestion demonstrated that EPs exhibited high resistance to enzymatic hydrolysis, indicating that a substantial proport
