[Pseudohypoparathyroidism and variants: A translational medicine success story].

Pseudohypoparathyroidism (PHP) is an uncommon disorder which is characterized by end-organ PTH resistance. The genetic defect is located at the GNAS locus that encodes the alpha-subunit of the stimulatory G protein (Gαs) and several splice variants thereof. This complex locus undergoes parental specific methylation changes that result in tissue-specific silencing of the paternal allele. Heterozygous inactivating mutations that disrupt Gαs function or epigenetics changes that impair Gαs expression contribute to a wide clinical spectrum of the disease: PHP1A, PHP1B, osseous heteroplasia, osteody

Emerging in vitro models of corneal stromal fibrosis: Toward human-relevant platforms for anti-scarring therapeutics.

Corneal stromal fibrosis, characterized by the aberrant transformation of keratocytes into contractile, extracellular matrix-secreting myofibroblasts, is a major cause of irreversible visual impairment worldwide. Despite extensive studies in animal models, interspecies differences in immune regulation, stromal architecture, and wound healing limit their translational relevance. This review highlights recent advances in human-relevant in vitro models that aim to overcome these challenges. In vitro models increasingly mimic critical features of corneal stromal fibrosis, including keratocyte-myof

Translating biotechnology to knowledge-based innovation, peace, and development? Deploy a Science Peace Corps--an open letter to world leaders.

Scholarship knows no geographical boundaries. This science diplomacy and biotechnology journalism article introduces an original concept and policy petition to innovate the global translational science, a Science Peace Corps. Service at the new Corps could entail volunteer work for a minimum of 6 weeks, and up to a maximum of 2 years, for translational research in any region of the world to build capacity manifestly for development and peace, instead of the narrow bench-to-bedside model of life science translation. Topics for translational research are envisioned to include all fields of life

Hypertension and Atrial Fibrillation: Insight From Basic to Translational Science Into the Mechanisms and Management.

There is a parallel epidemic of atrial fibrillation (AF) and hypertension (HTN) occurring globally. Both AF and HTN are no longer confined to the older population. The pathophysiology of AF related to HTN is complex with many inter-related factors. Hypertension causes hypertrophy of the ventricular musculature, dilatation of atria, elevated intracardiac pressures, atrial and ventricular mechanical dysfunction, abnormal expression of ion channels, dysregulation of the renin-angiotensin-aldosterone system, and the autonomic function. The pathophysiological alterations and structural changes that

Ubrogepant: Mechanism of action, clinical and translational science.

In recent years, the treatment of migraine has experienced a breakthrough in the development of drugs that target the calcitonin gene-related peptide (CGRP) signaling pathway. Monoclonal antibodies against the receptor or ligand have been developed for the preventive treatment of migraine; whereas, orally administered small molecule CGRP receptor antagonists, called gepants, have been developed for both acute and/or preventive treatment. Both modalities have demonstrated safe and effective treatment of migraine, reducing the number of migraine days for patients as well as reducing symptoms and

Immunobiology of pregnancy: from basic science to translational medicine.

Embryo implantation failure and spontaneous abortions represent the main causes of infertility in developed countries. Unfortunately, incomplete knowledge of the multiple factors involved in implantation and fetal development keeps the success rate of medically assisted procreation techniques relatively low. According to recent literature, cellular and molecular mechanisms of 'immunogenic tolerance' towards the embryo are crucial to establish an 'anti-inflammatory' state permissive of a healthy pregnancy. In this review we dissect the role played by the immune system in the endometrial-embryo

Do Sustainable Palliative Single Fraction Radiotherapy Practices Proliferate or Perish 2 Years after a Knowledge Translation Campaign?

In early 2017, the Canadian Partnership Against Cancer and CancerCare Manitoba undertook a comprehensive knowledge translation (KT) campaign to improve the utilization of single fraction radiotherapy (SFRT) over multiple fraction radiotherapy (MFRT) for palliative management of bone metastases. The campaign significantly increased short-term SFRT utilization. We assess the time-dependent effects of KT-derived SFRT utilization 12-24 months removed from the KT campaign in a Provincial Cancer Program. This study identified patients receiving palliative radiotherapy for bone metastases in Manitoba

Opportunities and challenges for the application of single-cell RNA sequencing and spatial transcriptomics in ischemic stroke.

Ischemic stroke (IS) is characterized by high rates of morbidity, disability and mortality. The pathological process underlying IS involves spatiotemporal dynamic responses of multiple cell types and marked cellular heterogeneity which cannot be fully elucidated using traditional bulk sequencing technologies. Single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) have provided high-resolution tools for investigating pathological mechanisms and developing precise diagnostic and therapeutic strategies for IS. In this review, we systematically summarize the core principles, mains

Exploring how microbiome signatures change across inflammatory bowel disease conditions and disease locations.

Understanding the variables that influence microbiome studies is critical for successful translational research. Inflammatory bowel disease (IBD) is a complex group of diseases that can present at multiple locations within the Gastrointestinal tract. Here, using the FAMISHED study cohort, we aimed to investigate the relationship between IBD condition, IBD disease location, and the microbiome. Signatures of the microbiome, including measures of diversity, taxonomy, and functionality, all significantly differed across the three different IBD conditions, Crohn's disease (CD), ulcerative colitis (

Trophic effects of multiple administration of mesenchymal stem cells in children with osteogenesis imperfecta.

The safety of mesenchymal stem cell therapy for osteogenesis imperfecta has been demonstrated previously. However, it is unknown how the trophic effects are mediated by stem cells. In the present commentary, we bring to the attention of readers the recent report by Infante et al in the journal of clinical and translational medicine. The TERCELOI clinical trial presented the possible paracrine effect of transplanted MSCs in vitro and in vivo using proteomics and transcriptomic analysis. This novel finding adds new knowledge in the field of regenerative medicine. However, the scarcity of solid e

The future of research in craniofacial biology and what this will mean for oral health professional education and clinical practice.

Today, and looking to the future, scientific discoveries from cellular, developmental and molecular biology inform our understanding of cell, tissue and organ morphogenesis as exemplified in skin, bone, cartilage, dentine, enamel, muscle, nerve and many organs such as salivary glands and teeth. Present day biomedical science yields principles for the biomimetic design and fabrication of cells, tissues and organs. Bioengineering has become a strategy that can 'mimic' biological processes, and inform clinical procedures for tissue and organ replacements. The future of regenerative craniofacial b

Diabetes and Atrial Fibrillation: Insight From Basic to Translational Science Into the Mechanisms and Management.

In spite of significant progress made in the management in recent decades, atrial fibrillation (AF) continues to cause increased mortality and significant morbidities, including heart failure and stroke. Diabetes mellitus (DM) is an independent risk factor for AF, and adds risks to increased mortality and hospitalizations when present along with AF. The pathophysiology of AF related to DM is complex with many inter-related factors. Atrial cardiomyopathy (atriopathy) related to structural changes from subcellular abnormalities and fibrosis, coupled with cardiac mechanical dysfunction, abnormal

The Rare Knowledge Mining Methodological Framework for the Development of Practice Guidelines and Knowledge Translation Tools for Rare Diseases.

Rare diseases bring on a heavy health, social and economic burden that impacts patients' lives and puts pressure on the healthcare system. Furthermore, they are often associated with limited published studies to inform multidisciplinary clinical practice thus limiting evidence-based practice. Moreover, the development of knowledge translation products including clinical care guidelines are often very challenging based on the current available methodological frameworks relying mostly on critical appraisal of the published research evidence where randomized clinical trial design is considered as
