A Defensin from the Model Beetle Tribolium castaneum Acts Synergistically with Telavancin and Daptomycin against Multidrug Resistant Staphylococcus aureus.

The red flour beetle Tribolium castaneum is a common insect pest and has been established as a model beetle to study insect development and immunity. This study demonstrates that defensin 1 from T. castaneum displays in vitro and in vivo antimicrobial activity against drug resistant Staphylococcus aureus strains. The minimum inhibitory concentration (MIC) of defensin 1 against 11 reference and clinical staphylococcal isolates was between 16-64 μg/ml. The putative mode of action of the defensin peptide is disruption of the bacterial cell membrane. The antibacterial activity of defensin 1 was at

Demographic suppression and oxidative responses of Tribolium beetles to cold-plasma and microwave treatment in wheat flour: efficacy-quality trade-offs.

BACKGROUND: Non-chemical physical treatments may complement stored-product pest management, but acute mortality alone may underestimate their value when surviving adults reproduce poorly and post-treatment population recovery is suppressed. This study compared atmospheric cold-plasma (CP; 10-20 kV) and microwave (MW; 300-900 W) exposure against sex-balanced adult cohorts of Tribolium castaneum and Tribolium confusum in wheat flour. RESULTS: Mortality increased with intensity and exposure duration. At 20 min, Abbott-corrected mortality reached 59.07% and 53.65% under CP-20 kV and 59.59% and 59

Functional reorganization and systemic toxicity induced by microplastics and climate-relevant stressors in Tribolium castaneum: Are we simulating tomorrow's toxic reality?

Microplastic (MP) pollution and the intensification of global climate change raise concerns regarding potential interactions between these stressors and their ecotoxicological effects on terrestrial biota. In this study, we investigated the ecotoxicological effects of polyethylene MPs (PE-MPs) under different climate scenarios in Tribolium castaneum. Adults were exposed for 21 days to two environmentally relevant concentrations of PE-MPs (0.005 % and 0.1 %) under three contrasting temperature and CO2 scenarios (27 °C/1000 ppm, 28.5 °C/1500 ppm, and 30 °C/2000 ppm). Insecticidal and Repellent Effects of Selected Botanicals against Tribolium Castaneum (Herbst) (Coleoptera: Tenebrionidae) with Reference To their Effect on Detoxifying Enzymes.

The excessive application of various synthetic pesticides led to control difficulties, including insect resistance and environmental contamination. This study aimed to evaluate the insecticidal and repellent activities of twelve botanical powders and aqueous extracts against Tribolium castaneum, with a focus on acetylcholinesterase and glutathione-S-transferase detoxification enzyme activities. Toxicity tests revealed that Azadirachta indica dry powder was the most harmful, having the lowest LD50 value of 2.09% w/w, while in aqueous extract A. indica was the most toxic, with an LC50 of 2.20% a

Assessment of the potential of non-thermal atmospheric pressure plasma discharge and microwave energy against Tribolium castaneum and Trogoderma granarium.

This study was carried out to investigate the efficacy of the non-thermal atmospheric pressure plasma produced with dielectric barrier discharge (APPD) using air as a processing gas and microwave energy to control Tribolium castaneum and Trogoderma granarium adults and larvae in wheat grains. Insects' mortality was found to be power and time-dependent. The results indicated that non-thermal APPD and the microwave have enough insecticidal effect on the target pests. From the bioassay, LT50's and LT90's levels were estimated, T. granarium larvae appeared more tolerant to non-thermal APPD and the

Exposure-time optimization of ozone fumigation against two Tribolium flour beetles: integrating insect suppression, oxidative stress, and wheat flour quality.

BACKGROUND: Stored wheat flour is highly susceptible to infestation by Tribolium castaneum and Tribolium confusum. This laboratory-scale study evaluated how ozone exposure duration at 1000 ppm affected insect suppression, reproductive performance, oxidative stress, infestation-associated flour weight loss, and flour quality. RESULTS: Larvae and adults were exposed for 0, 5, 10, or 15 h. After 15 h, mortality reached 94.56% and 90.56% in T. castaneum and T. confusum adults and 81.92% and 77.44% in their larvae, respectively. F₁ progeny suppression reached 97.34%, 95.18%, 91.67%, and 87.91% in

Booklice (Liposcelis spp.), Grain Mites (Acarus siro), and Flour Beetles (Tribolium spp.): 'Other Pests' Occasionally Found in Laboratory Animal Facilities.

Pests that infest stored food products are an important problem worldwide. In addition to causing loss and consumer rejection of products, these pests can elicit allergic reactions and perhaps spread disease-causing microorganisms. Booklice (Liposcelis spp.), grain mites (Acarus siro), and flour beetles (Tribolium spp.) are common stored-product pests that have previously been identified in our laboratory animal facility. These pests traditionally are described as harmless to our animals, but their presence can be cause for concern in some cases. Here we discuss the biology of these species an

Candidate gene screen in the red flour beetle Tribolium reveals six3 as ancient regulator of anterior median head and central complex development.

Several highly conserved genes play a role in anterior neural plate patterning of vertebrates and in head and brain patterning of insects. However, head involution in Drosophila has impeded a systematic identification of genes required for insect head formation. Therefore, we use the red flour beetle Tribolium castaneum in order to comprehensively test the function of orthologs of vertebrate neural plate patterning genes for a function in insect head development. RNAi analysis reveals that most of these genes are indeed required for insect head capsule patterning, and we also identified severa

Extraction, Chemical Composition and Insecticidal Activities of Lantana camara Linn. Leaf Essential Oils against Tribolium castaneum, Lasioderma serricorne and Callosobruchus chinensis.

Storage pests and the food spoilage they cause are problems of great concern. Using essential oil obtained from different plants as an insecticide against these storage pests can be considered an environmentally friendly pest management option. Lantana camara Linn. (family Verbenaceae) is a flowering species, and is also a noxious weed that can proliferate well in nearly all geographical habitats. A biopesticide derived from the essential oil extracted from this plant can offer an effective solution for controlling storage pests. The goal of this study is to extract and analyse the chemical co

Unveiling the molecular architecture of TcJHAMT: structural basis for catalytic efficiency and targeted inhibitor screening in the red flour beetle, Tribolium castaneum.

BACKGROUND: The red flour beetle, Tribolium castaneum, is a holometabolous insect regulated by juvenile hormone (JH), and juvenile hormone acid methyltransferase (JHAMT) serves as a rate-limiting enzyme involved in JH biosynthesis. JHAMT thus represents a promising molecular target for pest control, although the structural properties of canonical JHAMT remain uncharacterized. RESULTS: Herein, we present the canonical JHAMT of T. castaneum, TcJHAMT3, and report its three-dimensional structure at 2.50 Å resolution. TcJHAMT3 exhibits a conserved structural pattern with insect JHAMT homologs, con

Carbonic anhydrase metabolism is a key factor in the toxicity of CO2 and COS but not CS2 toward the flour beetle Tribolium castaneum [Coleoptera: Tenebrionidae].

The analogues carbon dioxide (CO(2)), carbonyl sulfide (COS) and carbon disulfide (CS(2)) have been useful as substrate probes for enzyme activities. Here we explored the affinity of the enzyme carbonic anhydrase for its natural substrate CO(2), as well as COS and CS(2) (1) by in vitro kinetic metabolism studies using pure enzyme and (2) through mortality bioassay of insects exposed to toxic levels of each of the gases during carbonic anhydrase inhibition. Hydrolysis of COS to form hydrogen sulfide was catalysed rapidly showing parameters K(m) 1.86 mM and K(cat) 41 s(-1) at 25 degrees C; howev

Real-time cell analysis and heat shock protein gene expression in the TcA Tribolium castaneum cell line in response to environmental stress conditions.

The rust red flour beetle, Tribolium castaneum (Herbst, 1797) (Coleoptera: Tenebrionidae), is a pest of stored grain and one of the most studied insect model species. Some of the previous studies involved heat response studies in terms of survival and heat shock protein expression, which are regulated to protect other proteins against environmental stress conditions. In the present study, we characterize the impedance profile with the xCELLigence Real-Time Cell Analyzer and study the effect of increased temperature in cell growth and viability in the cell line BCIRL-TcA-CLG1 (TcA) of T. castan

Performance of a Low-Cost Acoustic Insect Detector System with Sitophilus oryzae (Coleoptera: Curculionidae) in Stored Grain and Tribolium castaneum (Coleoptera: Tenebrionidae) in Flour.

Reduction of postharvest losses is gaining increased priority in warm regions where insect infestation may cause rapid deterioration of staple commodities. Acoustic detection can be used to assess the likelihood of insect infestations in bags of grain, flour, and other commodities that are stored in small holdings in developing countries, enabling rapid targeting of treatments. A portable postharvest insect detection system was developed with the goal to provide low-cost capability to acoustically assess infestations in small-scale storage facilities. The effects of daily fluctuating low-oxygen conditions on life table parameters of Tribolium castaneum (Coleoptera: Tenebrionidae) and their subsequent generations in controlled atmospheres.

Understanding pest insect survival and development under conditions of fluctuating low oxygen levels (less than 4.4%) is of practical significance in the control of nitrogen atmospheres for grain storage. Egg hatching, sixth instar survival, adult emergence rate, and adult survival rates of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) were determined following 8-d exposure periods of eggs, sixth-instar larvae, pupae, and adults to daily fluctuating oxygen concentrations within ranges of 1.9% to 4.4%, 1.9% to 3.9%, 1.9% to 3.5%, 1.9% to 3.2%, 1.9% to 2.7%, 1.9% to 2.5%, and 1.9% to

Distinguishing Infested Flour from Uninfested Flour through Chemometric Processing of DART-HRMS Data─Revealing the Presence of Tribolium castaneum, the Red Flour Beetle.

Insect infestation of agricultural stored products is a significant challenge to food security across the globe. One common pest is Tribolium castaneum (red flour beetle). In a new approach to addressing the threat of these beetles, Direct Analysis in Real Time-High-Resolution Mass Spectrometry was used to examine infested and uninfested flour samples. These samples were then distinguished through statistical analysis techniques, including EDR-MCR, in order to highlight the important m/z values contributing to the differences in the flour profiles. A subset of these values responsible for the

Comparison of the effects of multiple variables on the levels of infection of the rat tapeworm, Hymenolepis diminuta, in its intermediate host, the confused flour beetle Tribolium confusum.

The interaction of the rat tapeworm, Hymenolepis diminuta, with its intermediate beetle host, Tribolium confusum, is a well-studied model system. However, there is so much variation in the methods and designs of the studies that it is difficult to draw comparisons. This study simultaneously compared several aspects of the infection protocol including beetle age, sex, density and mating status; parasite egg condition, infection environment humidity and the times for the three steps of infection: starvation, feeding and post-feeding development. Although statistically limited by low prevalence a
