The developmentally dynamic microRNA transcriptome of Glossina pallidipes tsetse flies, vectors of animal trypanosomiasis.

SUMMARY: MicroRNAs (miRNAs) are single stranded gene regulators of 18-25 bp in length. They play a crucial role in regulating several biological processes in insects. However, the functions of miRNA in Glossina pallidipes, one of the biological vectors of African animal trypanosomosis in sub-Saharan Africa, remain poorly characterized. We used a combination of both molecular biology and bioinformatics techniques to identify miRNA genes at different developmental stages (larvae, pupae, teneral and reproductive unmated adults, gravid females) and sexes of G. pallidipes. We identified 157 mature

Recirculation of Trypanosoma brucei brucei in cattle after T. congolense challenge by tsetse flies.

The effect of challenging cattle, chronically infected with Trypanosoma brucei brucei, with T. congolense on the development of the T. b. brucei infection was investigated. For this purpose, nine experimental animals were first infected with T. b. brucei through the bites of infected tsetse flies. Once the T. b. brucei had developed into a chronic infection, that was difficult to detect using routine parasitological diagnostic tools, seven of the experimental animals were challenged by tsetse flies infected with T. congolense. Two of the animals infected with T. b. brucei were kept as control.

Feeding behaviour of tsetse flies (Glossina pallidipes Austen) on Trypanosoma-infected oxen in Kenya.

An incomplete ring of electric nets was placed around oxen which were either uninfected, infected with Trypanosoma vivax, or infected with T. congolense. The numbers of fed and unfed Glossina pallidipes caught on the nets were used to estimate the attractiveness of the oxen to tsetse, and the feeding success of the tsetse on the oxen. Oxen infected with T. congolense attracted more G. pallidipes than the other groups of oxen. Taking into consideration daily variation in the abundance or activity of the flies, oxen infected with T. congolense were about 70% more attractive to G. pallidipes than

Population structure of G. pallidipes tsetse flies in Northwestern Kenya - Towards effective vector control.

BACKGROUND: Tsetse flies are vectors of trypanosome parasites responsible for Human and Animal African Trypanosomiasis (HAT and AAT), diseases that greatly affect health and economic growth across sub-Saharan Africa. Vector control, one of the possible strategies to tackle these diseases, is most effective on genetically isolated tsetse populations. While the population genetic structure of Glossina pallidipes tsetse flies has been well studied in southern Kenya, little is known about populations in northwestern Kenya. This study investigated the presence, genetic diversity, and population str

Characterization of a composite with enhanced attraction to savannah tsetse flies from constituents or analogues of tsetse refractory waterbuck (Kobus defassa) body odor.

Savannah tsetse flies avoid flying toward tsetse fly-refractory waterbuck (Kobus defassa) mediated by a repellent blend of volatile compounds in their body odor comprised of δ-octalactone, geranyl acetone, phenols (guaiacol and carvacrol), and homologues of carboxylic acids (C5-C10) and 2-alkanones (C8-C13). However, although the blends of carboxylic acids and that of 2-alkanones contributed incrementally to the repellency of the waterbuck odor to savannah tsetse flies, some waterbuck constituents (particularly, nonanoic acid and 2-nonanone) showed significant attractive properties. In another

Identification of a Tsal152-75 salivary synthetic peptide to monitor cattle exposure to tsetse flies.

BACKGROUND: The saliva of tsetse flies contains a cocktail of bioactive molecules inducing specific antibody responses in hosts exposed to bites. We have previously shown that an indirect-ELISA test using whole salivary extracts from Glossina morsitans submorsitans was able to discriminate between (i) cattle from tsetse infested and tsetse free areas and (ii) animals experimentally exposed to low or high numbers of tsetse flies. In the present study, our aim was to identify specific salivary synthetic peptides that could be used to develop simple immunoassays to measure cattle exposure to tset

Shifting from wild to domestic hosts: the effect on the transmission of Trypanosoma congolense to tsetse flies.

The epidemiology and impact of animal African trypanosomosis are influenced by the transmissibility and the pathogenicity of the circulating trypanosome strains in a particular biotope. The transmissibility of 22 Trypanosoma congolense strains isolated from domestic and wild animals was evaluated in a total of 1213 flies. Multivariate mixed models were used to compare infection and maturation rates in function of trypanosome origin (domestic or sylvatic) and pathogenicity. Both trypanosome pathogenicity and origin significantly affected the ability to establish a midgut infection in tsetse fli

Diversity of tsetse flies and trypanosome species circulating in the area of Lake Iro in southeastern Chad.

BACKGROUND: African trypanosomiases are vector-borne diseases that affect humans and livestock in sub-Saharan Africa. Although data have been collected on tsetse fauna as well as trypanosome infections in tsetse flies and mammals in foci of sleeping sickness in Chad, the situation of tsetse fly-transmitted trypanosomes remains unknown in several tsetse-infested areas of Chad. This study was designed to fill this epidemiological knowledge gap by determining the tsetse fauna as well as the trypanosomes infecting tsetse flies in the area of Lake Iro in southeastern Chad. Molecular prevalence of trypanosome infections in cattle and tsetse flies in the Maasai Steppe, northern Tanzania.

BACKGROUND: African trypanosomosis is a disease of public health and economic importance that poses a major threat to the livelihoods of people living in the Maasai Steppe, where there is a significant interaction between people, livestock and wildlife. The vulnerability of the Maasai people to the disease is enhanced by the interaction of their cattle, which act as vehicles for trypanosomes, and tsetse flies close to wildlife in protected areas. This study was aimed at identification of trypanosome infections circulating in cattle and tsetse flies in order to understand their distribution and

Improving Sterile Insect Technique (SIT) for tsetse flies through research on their symbionts and pathogens.

Tsetse flies (Diptera: Glossinidae) are the cyclical vectors of the trypanosomes, which cause human African trypanosomosis (HAT) or sleeping sickness in humans and African animal trypanosomosis (AAT) or nagana in animals. Due to the lack of effective vaccines and inexpensive drugs for HAT, and the development of resistance of the trypanosomes against the available trypanocidal drugs, vector control remains the most efficient strategy for sustainable management of these diseases. Among the control methods used for tsetse flies, Sterile Insect Technique (SIT), in the frame of area-wide integrate

Responses of tsetse flies (Glossina spp.) to compounds on the skin surface of an ox: a laboratory study.

The behaviour of male Glossina morsitans morsitans Westwood and Glossina pallidipes Austen (Diptera: Glossinidae) alighting on targets with or without ox sebum was compared. The presence of ox sebum did not increase significantly the number of flies alighting on the target in either species. However, after contact with the sebum coated target, both species showed an increase in flight activity, and G. m.morsitans showed a greater tendency to return to the target. This behaviour resulted in a number of short flights which may reflect the search for a feeding site on a host. The duration of each

Adult blood-feeding tsetse flies, trypanosomes, microbiota and the fluctuating environment in sub-Saharan Africa.

The tsetse fly vector transmits the protozoan Trypanosoma brucei, responsible for Human African Trypanosomiasis, one of the most neglected tropical diseases. Despite a recent decline in new cases, it is still crucial to develop alternative strategies to combat this disease. Here, we review the literature on the factors that influence trypanosome transmission from the fly vector to its vertebrate host (particularly humans). These factors include climate change effects to pathogen and vector development (in particular climate warming), as well as the distribution of host reservoirs. Finally, we

Trypanosoma congolense: drug resistance during cyclical transmissions in tsetse flies and syringe passages in mice.

A drug-resistant Trypanosoma congolense strain with predetermined curative doses (CD50 and CD90) of samorin at 13.9 +/- 1.02 and 20.3 +/- 1.13 mg/kg body weight, respectively, was cyclically transmitted through tsetse flies and by syringe passages in mice in the absence of drug pressure. The changing levels of drug sensitivity were determined after every 3rd cyclic and 5th syringe passage intervals. It was noted that when the strain was maintained in tsetse flies through 12 cyclical transmissions, the CD50 and CD90 dropped slightly from 13.9 to 11.9 +/- 1.06 and from 20.3 to 18.0 +/- 1.08 mg/k

African sleeping sickness: new evidence that mature tsetse flies (Glossina morsitans) can become potent vectors.

Starved mature male tsetse flies (21 to 25 days old) are capable of developing salivary gland (SG) infections of Trypanosoma brucei rhodesiense at rates nearly comparable to teneral males less than 24 hours old when given an infective meal containing parasites, horse red cells and culture medium. Although the over-all SG infection rate for mature males starved for three, four or five days before infection was about half that for teneral males less than 48 hours old (8.0% v. 15.6%), males starved for four days developed infection rates (12.3%) that were comparable to those of teneral flies less

Trypanosoma (Nannomonas) godfreyi sp. nov. from tsetse flies in The Gambia: biological and biochemical characterization.

We provide evidence from isoenzyme analysis, hybridization with repetitive DNA probes, behavioural studies and morphometrics that 4 trypanosome isolates from Glossina morsitans submorsitans in The Gambia constitute a new species now named Trypanosoma (Nannomonas) godfreyi. The bloodstream trypomastigotes of T. (N.) godfreyi are relatively small with a mean length of 13.7 microns (range: 9.1-21.8 microns) and a mean width of 1.65 microns (range: 0.65-2.69 microns). There is no free flagellum and the marginal kinetoplast is subterminal to a rounded posterior end; the undulating membrane is usual

A new automated chilled adult release system for the aerial distribution of sterile male tsetse flies.

Tsetse eradication continues to be a top priority for African governments including that of Senegal, which embarked on a project to eliminate Glossina palpalis gambiensis from the Niayes area, following an area-wide integrated pest management approach with an SIT component. A successful SIT programme requires competitive sterile males of high biological quality. This may be hampered by handling processes including irradiation and the release mechanisms, necessitating continued improvement of these processes, to maintain the quality of flies. A new prototype of an automated chilled adult releas

PCR analysis and spatial repartition of trypanosomes infecting tsetse flies in Sidéradougou area of Burkina Faso.

A parasitological and entomological survey was conducted in the Sideradougou area (south of Bobo Dioulasso, Burkina Faso) in order to identify transmission factors of African trypanosomosis. A total of 3600 tsetse flies (Glossina tachinoides, Glossina palpalis gambiensis) were captured along 120 km of linear gallery forest and half of them were dissected. PCR analysis was undertaken on parasitologically positive flies (161 G. tachinoides, 92 G. palpalis gambiensis) to characterize the different trypanosomes. All the results were integrated in a GIS (Geographical Information System). Spatial re

Suitability of Raycell MK2 Blood X-ray Irradiator for the Use in the Sterile Insect Technique: Dose Response in Fruit Flies, Tsetse Flies and Mosquitoes.

The sterile insect technique (SIT) is based on the inundatory field release of a target pest following their reproductive sterilization via exposure to radiation. Until recently, gamma irradiation from isotopic sources has been the most widely used in SIT programs. As isotopic sources are becoming increasingly expensive, especially for small programs, and regulations surrounding their procurement and shipment increasingly strict, irradiation capacity is one of the limiting factors in smaller or newly developing SIT projects. For this reason, the possibility of using X-ray irradiators has been
