Bovine tuberculosis is a disease of worldwide importance yet comparatively little is known about chemokine responses to infection. We report on the levels of chemokine expression within lymph nodes of cattle infected with Mycobacterium bovis when infection would be well established. Expression levels of a number of chemokines were increased in infected cattle and could be correlated to levels of respective chemokine receptors. Several chemokines were significantly correlated to pathology within the lymph node, indicating a direct relationship between chemokine expression and disease. Vaccinate

Possums are a wildlife vector of bovine tuberculosis in New Zealand. Vaccination of possums with BCG is being considered as a measure to control the spread of bovine tuberculosis to cattle and deer. Delivery via oral bait is feasible but BCG is degraded in the stomach. The aim was to determine whether ranitidine (Zantac) would reduce gastric acidity and enhance the efficacy of intragastrically administered BCG. A dose of 75 mg reduced gastric acidity for at least 4 h. Thus, possums were vaccinated intragastrically with BCG after receiving 75 mg ranitidine or ranitidine or BCG alone, as control

Biological assays carried out in the Republic of Ireland in naturally sensitized (M. bovis infected) tuberculous cattle have shown that Rotterdam and Weybridge bovine PPD tuberculins issued for routine use in 1981 and 1982 were equipotent with an estimated potency of about 24,000 Community Tuberculin Units (C.T.U.) per mg of PPD. However, previously it was shown that Weybridge bovine PPD produced in 1978 had only a potency of about 4,000 C.T.U./mg in M. bovis infected cattle. The clinical significance of the latter finding is seen in the results of field trials carried out in the Republic of I

BACKGROUND: Bovine tuberculosis (BTB) has a significant impact on both the cattle industry and human health. Understanding its transmission, clinical signs, risk factors, and control measures is essential for prevention. This study aimed to evaluate the knowledge, attitudes, and practices regarding BTB in high-burden areas of Malawi.
METHODS: A stratified sampling design was employed to collect data from 463 butchers, dairy farmers and cattle farmers in different locations and settings in Malawi. Aggregate scores on knowledge, attitudes, and practices were taken as multivariate measurements a

Developing evidence-based approaches to combat infectious diseases is essential in resource-limited settings to enable prioritisation of interventions where they are most effective in reducing the burden of disease. Bovine tuberculosis (bTB) is a major disease in cattle, but its impact remains poorly characterised in many contexts and production systems. To support the development of policies for accelerated bTB control, we conducted a systematic review to collate the available evidence on the impact of bTB on animal health, welfare and production. We identified 91 eligible studies providing i

An immunochromatographic test system has been developed for the simultaneous rapid multiplex serodiagnostics of bovine brucellosis, tuberculosis, and leukemia. The test system is based on the use of a conjugate of gold nanoparticles with the chimeric protein Cysteine-A/G and three analytical zones with immobilized pathogen antigens: Brucella abortus lipolysaccharide, recombinant proteins MPB64 and MPB83-MPB63 of Mycobacterium bovis, and recombinant protein p24 of the bovine leukemia virus. Prototypes of the test system were tested on 98 samples of sera from healthy and infected animals. The di

Bovine tuberculosis (BTB) and brucellosis are major endemic zoonoses in ruminants in Morocco that impact on both animal and human health. This study presents an assessment of the epidemiological and socioeconomic burden of bacterial zoonoses in Sidi Kacem Province in Northern Morocco from a cross-sectional survey of 125 cattle and/or small ruminant-owning households. In total, 1082 sheep and goats were examined from 81 households. The single intradermal comparative cervical test to screen for bovine tuberculosis was undertaken on 1194 cattle from 123 households and all cattle were blood sample

Wild ruminants have an important role in the epidemiology of bovine tuberculosis (bTB). This study describes an outbreak of bovine tuberculosis occurring in a fallow deer herd in Sicily. In 2012 a Sicilian herd of 47 animals was referred for cachexia. Pathological examination of 2 dead animals revealed disseminated granulomas predominantly involving the skin and subcutaneous tissues. Tissue samples were submitted for histological analysis, bacteriological culture, and biomolecular assay. PCR analysis identified Mycobacterium strains. Genotyping by spoligotyping and MIRU-VNTR profiles identifie

INTRODUCTION: Free-ranging white-tailed deer (Odocoileus virginianus) are a self-sustaining reservoir for bovine tuberculosis (bTB) in northeastern lower Michigan, (United States) continually putting the area's cattle industry at risk. Liberal recreational deer harvest, baiting bans, and mitigation measures on farms have reduced but not eliminated bTB in deer nor have they eliminated transmission to cattle. With apparent prevalence in deer being low (1-2%) but constant, vaccination could be an additional tool to aid in addressing the problem and merits investigation. Mycobacterium tuberculosis

Bovine tuberculosis is a persistent and significant challenge for cattle farmers in Zambia, especially among rural farmers. As livestock farming constitutes a crucial component of the country's agricultural economy, the prevalence of bTB threatens not only animal health and productivity but also poses serious risks to human health, food safety, and economic stability within rural communities. A mixed-methods study was conducted in Lundazi and Monze districts of Zambia between December 2021 and June 2022, combining a cross-sectional survey of 280 respondents with qualitative insights from five

Animal tuberculosis in cattle (TB) has been controlled in many European countries through long-standing eradication programs, yet sporadic breakdowns continue to occur in officially tuberculosis-free (OTF) areas, challenging the sustainability of disease freedom. This study aimed to identify and quantify herd-level and area-level risk factors associated with TB occurrence in Italian OTF regions in order to support risk-based surveillance strategies. A national longitudinal open-cohort study was conducted using data from the Italian Veterinary Information System, including approximately 300,000

Although several brands of tuberculin purified protein derivatives (PPDs) are available for diagnosing bovine tuberculosis (bTB), comparative studies to determine their diagnostic accuracy are infrequent. In Ecuador we compared two different PPD brands for bTB diagnosis using skin testing and measuring skin thickness increase. Additionally, we evaluated four PPD brands, including those used for skin testing, in the Bovine Tuberculosis Interferon Gamma Test (IFN-γ test) measuring IFN-γ induction in whole blood. The study included 17 naturally tuberculosis-infected PPD and IFN-γ test positive bo

This contribution describes the development of a simple, fast, cost-effective, and sensitive impedimetric immunosensor for quantifying bovine tuberculosis (TB) in bovine serum samples. The construction of the immunosensor involved immobilizing the purified protein derivative (PPD) of M. bovis onto a screen-printed electrode that was modified with gold nanoparticles (AuNPs) and a polypyrrole (pPy) film synthesized electrochemically. The immunosensor exhibited a linear range from 0.5 μg mL-1 to 100 μg mL-1 and achieved a limit of detection (LD) of 100 ng mL-1 for the detection of anti-M. bovis a

The dynamics of Mycobacterium bovis infection in cattle can influence the proportion of infected animals that are diagnosed by ante-mortem tests in routine bovine tuberculosis (bTB) surveillance and monitoring programmes. Although the current diagnostic tests based on cell-mediated or serological responses are imperfect, they are effective in diagnosing the majority of infected animals. However, the lack of perfect sensitivity and specificity also leads to failure to diagnose all infected animals leading to persistence of infection in herds. The terms residual, subclinical, latent and anergy h

Bovine tuberculosis (bTB), a global zoonotic disease, causes negative effects on human and animal health. PhoP protein is a key regulator of pathogenic phenotypes in members of the Mycobacterium tuberculosis complex, which includes the causative agent of bTB. Despite extensive research on this protein focused in deciphering its regulatory role, little was explored about it as a diagnostic antigen. In humans, a novel role of anti-PhoP antibodies as a possible marker for the diagnosis of TB was demonstrated. However, this issue was not addressed in bovines. In this study, antigenic properties of

Bacille Calmette-Guerin (BCG) is the world's most widely used vaccine, but there are concerns that it provides little protection against pulmonary tuberculosis of humans in countries that have a high prevalence of environmental mycobacteria. Experiments in cattle provide a model to investigate this situation and to develop an improved tuberculosis vaccine. In the third of a series of BCG vaccination trials, calves had high interferon-gamma (IFN-gamma) responses to purified protein derivative (PPD) from Mycobacterium avium prior to vaccination, indicating that infection with environmental mycob

This study shows that gamma interferon (IFN-gamma) and interleukin-4 (IL-4) cytokine responses are produced by peripheral blood cells in cattle infected with Mycobacterium bovis. The different kinetics of the IFN-gamma and IL-4 responses to bovine tuberculin and to ESAT-6 following experimental intratracheal infection with M. bovis are described. An early increase in IFN-gamma was observed that was maintained throughout the period studied. In contrast, the IL-4 response was delayed and confined to a peak of activity lasting 6 to 8 weeks. Interestingly, an experimental challenge of cattle with
