Tuft cells in the main excretory duct epithelia of the three major rat salivary glands.

We report the tuft cells in the main excretory duct epithelia of rat salivary glands. These cells exhibit similar fundamental characteristics in the salivary glands and in other organs. However, numerous membrane-bound electron-dense granules are present among the microvilli of the tuft cells in the submandibular gland, but not in other organs. The apical cytoplasm contains numerous vesicles with a filamentous substance that reacts positively for glycoconjugates. The vesicles frequently are close to the apical plasma membrane and seem to open into the lumen. Nerve endings with synaptic vesicle

Significance of Tuft Cells Expressing Hematopoietic Prostaglandin D Synthase During the Pathogenesis of Eosinophilic Chronic Rhinosinusitis.

Background/Objective: Eosinophilic chronic rhinosinusitis (ECRS) is a subtype of chronic rhinosinusitis with nasal polyps that is characterized by abundant eosinophilic infiltration within nasal polyps. Tuft cells are epithelial chemosensory cells that are present in the normal respiratory tract and activate group-2 innate lymphoid cells through their secretion of interleukin-25 and prostaglandin (PG) D2. ECRS is also characterized by the activation of group-2 innate lymphoid cells; however, the involvement of tuft cell-derived PGD2 in this process remains unclear. Methods: We selected consecu

Adhesion receptor ADGRG2/GPR64 is in the GI-tract selectively expressed in mature intestinal tuft cells.

OBJECTIVE: GPR64/ADGRG2 is an orphan Adhesion G protein-coupled receptor (ADGR) known to be mainly expressed in the parathyroid gland and epididymis. This investigation aimed to delineate the cellular expression of GPR64 throughout the body with focus on the gastrointestinal (GI) tract. METHODS: Transgenic Gpr64mCherry reporter mice were histologically examined throughout the body and reporter protein expression in intestinal tuft cells was confirmed by specific cell ablation. The GPCR repertoire of intestinal Gpr64mCherry-positive tuft cells was analyzed by quantitative RT-PCR analysis and i

Fine structure of tuft cells of the main excretory duct epithelium in the rat submandibular gland.

BACKGROUND: Tuft cells, which are characterized by long microvilli with prominent rootlets and by vesicular and tubular profiles, are present in the mucosal epithelium of a number of hollow organs, including the main excretory duct of the rat submandibular gland. Despite their widespread occurrence, little is known of their function. METHODS: Main excretory ducts of the submandibular gland were obtained from 20 male Wistar rats and prepared for electron microscopic examination. Specimens also were subjected to various histochemical procedures, including HRP (horseradish peroxidase) uptake, th

Olfaction in Lamprey Pallium Revisited-Dual Projections of Mitral and Tufted Cells.

The presence of two separate afferent channels from the olfactory glomeruli to different targets in the brain is unravelled in the lamprey. The mitral-like cells send axonal projections directly to the piriform cortex in the ventral part of pallium, whereas the smaller tufted-like cells project separately and exclusively to a relay nucleus called the dorsomedial telencephalic nucleus (dmtn). This nucleus, located at the interface between the olfactory bulb and pallium, in turn projects to a circumscribed area in the anteromedial, ventral part of pallium. The tufted-like cells are activated wit

The Taste Receptor TAS1R3 Regulates Small Intestinal Tuft Cell Homeostasis.

Tuft cells are an epithelial cell type critical for initiating type 2 immune responses to parasites and protozoa in the small intestine. To respond to these stimuli, intestinal tuft cells use taste chemosensory signaling pathways, but the role of taste receptors in type 2 immunity is poorly understood. In this study, we show that the taste receptor TAS1R3, which detects sweet and umami in the tongue, also regulates tuft cell responses in the distal small intestine. BALB/c mice, which have an inactive form of TAS1R3, as well as Tas1r3-deficient C57BL6/J mice both have severely impaired response

Dendritic excitability and calcium signalling in the mitral cell distal glomerular tuft.

The processing of odour information starts at the level of the olfactory glomerulus, where the mitral cell distal dendritic tuft not only receives olfactory nerve sensory input but also generates dendrodendritic output to form complicated glomerular synaptic circuits. Analysing the membrane properties and calcium signalling mechanisms in these tiny dendritic branches is crucial for understanding how the glomerular tuft transmits and processes olfactory signals. With the use of two-photon Ca2+ imaging in rat olfactory bulb slices, we found that these distal dendritic branches displayed a signif

Differential expression of villin and advillin by neuroendocrine and tuft cells in the murine lower airways.

Previous studies identified a rare cell type in the mouse tracheal epithelium with immunoreactivity to the microvillus protein villin (Vil1), which persisted in mice lacking tuft cells due to deletion of the transcription factor Pou2f3. This study aimed to clarify the identity of this ill-defined cell type. Ultrastructurally, all cells with tightly packed microvilli observed in the tracheal epithelium of Pou2f3-/--mice contained basally located dense core vesicles, a characteristic feature of neuroendocrine cells (NEC). Accordingly, immunofluorescence double-labeling utilizing NEC markers reve

Trained immunity of intestinal tuft cells during infancy enhances host defense against enteroviral infections in mice.

Innate immune cells have been acknowledged as trainable in recent years. While intestinal tuft cells are recognized for their crucial roles in the host defense against intestinal pathogens, there remains uncertainty regarding their trainability. Enterovirus 71 (EV71), a prevalent enterovirus that primarily infects children but rarely infects adults. At present, there is a significant expansion of intestinal tuft cells in the EV71-infected mouse model, which is associated with EV71-induced interleukin-25 (IL-25) production. Further, we found that IL-25 pre-treatment at 2 weeks old mouse enabled

Tracheal tuft cell-released leukotrienes promote antibacterial immune responses.

Tuft cells (TCs) act as crucial airway sentinels that detect bacterial metabolites and initiate immune responses, yet the underlying mechanisms remain poorly understood. Here, we identify tracheal TCs as the initial source of leukotrienes (LTs), released during bacterial infection. Tracheal TCs discriminate pathogenic from commensal bacteria by sensing extracellular ATP (eATP) released by pathogens, including Pseudomonas aeruginosa and Rodentibacter pneumotropicus, within 4 h of infection, through the transient receptor potential cation channel subfamily M member 5 (Trpm5). This induces the LT

Pan-cancer analysis and mechanistic insights of tuft cell-like tumors.

Tuft cells are rare, chemosensory epithelial cells present in various tissues, including the respiratory and gastrointestinal tracts. Recent studies have revealed their significant role in cancer biology, particularly through the expression of the transcription factor POU2F3, which serves as a master regulator of tuft cell lineage. In several cancer types, including small cell lung cancer, gastric cancer, and breast cancer, POU2F3 expression defines a distinct molecular subtype termed "tuft cell-like" tumors. These tumors exhibit unique transcriptional programs and altered tumor-immune interac

Basal cell of origin resolves neuroendocrine-tuft lineage plasticity in cancer.

Neuroendocrine and tuft cells are rare chemosensory epithelial lineages defined by the expression of ASCL1 and POU2F3 transcription factors, respectively. Neuroendocrine cancers, including small cell lung cancer (SCLC), frequently display tuft-like subsets, a feature linked to poor patient outcomes1-9. The mechanisms driving neuroendocrine-tuft tumour heterogeneity and the origins of tuft-like cancers are unknown. Using multiple genetically engineered animal models of SCLC, we demonstrate that a basal cell of origin (but not the accepted neuroendocrine origin) generates neuroendocrine-tuft-lik

The Pre-synaptic Landscape of Mitral/Tufted Cells of the Main Olfactory Bulb.

In olfaction, all volatile odor information is tunneled through the main olfactory bulb (OB). Odor information is then processed before it is transferred to higher brain centers. Odor processing in the OB is carried out by numerous local inhibitory circuits and modulated by top-down input. Top-down modulation of OB function has been shown to act via interneurons but evidence also exists for its direct impact onto the principle mitral and tufted cells (M/Ts). Here, we used monosynaptic rabies trans-synaptic tracing from the OB to map and quantify the local and top-down pre-synaptic landscape of

Olfactory bulb external tufted cells are synchronized by multiple intraglomerular mechanisms.

In rat olfactory bulb slices, external tufted (ET) cells spontaneously generate spike bursts. Only ET cells affiliated with the same glomerulus exhibit significant synchronous activity, suggesting that synchrony results mainly from intraglomerular interactions. The intraglomerular mechanisms underlying their synchrony are unknown. Using dual extracellular and patch-clamp recordings from ET cell pairs of the same glomerulus, we found that the bursting of ET cells is synchronized by several mechanisms. First, ET cell pairs of the same glomerulus receive spontaneous synchronous fast excitatory sy

External tufted cells: a major excitatory element that coordinates glomerular activity.

The glomeruli of the olfactory bulb are the first site of synaptic processing in the olfactory system. The glomeruli contain three types of neurons that are referred to collectively as juxtaglomerular (JG) cells: external tufted (ET), periglomerular (PG), and short axon (SA) cells. JG cells are thought to interact synaptically, but little is known about the circuitry linking these neurons or their functional roles in olfactory processing. Single and paired whole-cell recordings were performed to investigate these questions. ET cells spontaneously fired rhythmic spike bursts in the theta freque

Some properties of receptive fields of olfactory mitral/tufted cells in the frog.

1. Different regions of the frog's olfactory epithelium were stimulated with nine glass micropipettes either individually or simultaneously in different combinations. The stimulus was a positive electrical pulse (4 s) consisting of a progressive increase, a plateau, and a progressive decrease in current intensity. Extra- and intracellular recordings were made from olfactory bulb mitral/tufted cells. Some of these cells were identified by intracellular injection of Lucifer yellow. 2. The action potential response patterns of mitral/tufted cells during the different phases of the stimulation wer

A nasal cell atlas reveals heterogeneity of tuft cells and their role in directing olfactory stem cell proliferation.

The olfactory neuroepithelium serves as a sensory organ for odors and forms part of the nasal mucosal barrier. Olfactory sensory neurons are surrounded and supported by epithelial cells. Among them, microvillous cells (MVCs) are strategically positioned at the apical surface, but their specific functions are enigmatic, and their relationship to the other specialized epithelial cells is unclear. Here, we establish that the family of MVCs comprises tuft cells and ionocytes in both mice and humans. Integrating analysis of the respiratory and olfactory epithelia, we define the distinct receptor ex

Generation and functional characterization of tuft cells in non-human primate pancreatic ducts through organoid culture systems.

The pancreatic duct plays a key role in collecting pancreatic juice, which is rich in digestive enzymes. The fluid flows unidirectionally into the duodenum, where it mixes with partially digested food to further facilitate digestion. In this study, we report the generation of pancreatic ductal organoids from non-human primates for the first time, aimed at investigating the role of tuft cells that reside in the pancreatic duct since no studies have addressed the role of tuft cells in the pancreas. The organoids were maintained in a medium supplemented with Wnt3a, Noggin, R-spondin, and other fa

De novo formation of tuft and goblet cells protects the intrapancreatic biliary duct system from inflammatory injury.

The biliary tree is susceptible to inflammatory injury that can result in severe liver damage. However, the intrapancreatic biliary duct system is largely refractory to damage caused by pancreatitis; the mechanisms that protect these ducts from injury remain unknown. Here, we demonstrate that, although serine/threonine-protein kinase DCLK1 (DCLK1) + tuft cells and mucin-2 (MUC2) + goblet cells were largely restricted to the ampulla of non-injured mice, they exhibited a marked increase in the ampulla and a progressive appearance in the intrapancreatic biliary duct system after injury. The incre
