Co-dependencies in the tumor immune microenvironment.

Activated oncogenes and disrupted tumor suppressor genes (TSGs) not only endow aspiring cancer cells with new biological capabilities but also influence the composition and function of host cells in the tumor microenvironment (TME). These non-cancer host cells can in turn provide cancer cells with growth support and protection from the anti-tumor immune response. In this ecosystem, geospatially heterogenous "subTME" adds to the complexity of the "global" TME which bestows tumors with increased tumorigenic ability and resistance to therapy. This review highlights how specific genetic alteration

[Immune cell contexture in the tumor microenvironment].

Antitumor immune response is generally suppressed in different ways in many types of tumors. In fact, a variety of immunosuppressive cells, such as regulatory T cells, myeloid-derived suppressor cells, and tumor-associated macrophages, surround the tumor modulate antigen-presenting cells and effector T cells. The strategy to abreact the immunosuppressive conditions is necessary for a successful immunotherapy against cancers. Particularly, the improvement of the tumor microenvironment (TME) from this point is important for cancer immunotherapy. The checkpoint blockade as the representative succ

Cholangiocarcinoma pathogenesis: Role of the tumor microenvironment.

Cholangiocarcinoma is a tumor that originates from the neoplastic transformation of the epithelial cells of the intrahepatic or extrahepatic bile ducts. This type of cancer is difficult to diagnose, extremely aggressive, and has very poor prognosis. It is also relatively resistant to chemotherapy and radiation therapy. Its pathogenesis is poorly understood, however it is known that the tumor microenvironment is a very important factor in the regulation of tumor angiogenesis, invasion, and metastasis. The current knowledge about the mechanisms by which these events are regulated as well as the

Tumor-derived DLK1 restrains tumor progression through remodeling of the tumor immune microenvironment.

The tumor immune microenvironment (TIME) plays a pivotal role in tumor progression and therapeutic responsiveness, yet the tumor-derived factors governing immune remodeling remain incompletely understood. Here, we identified delta-like homolog 1 (DLK1), a noncanonical Notch ligand, as a candidate regulator of the TIME through comparative transcriptomic analyses of murine tumors. DLK1 expression progressively declined during tumor progression and was inversely associated with tumor hypoxia. The hypoxia-responsive microRNA miR-338-5p contributed, at least in part, to the downregulation of DLK1 e

Environmental chemicals and the tumor immune microenvironment.

Impact of representative environmental chemicals on tumor immune microenvironment (TIME). Representative environmental chemicals are ubiquitously distributed across air, soil, aquatic systems, and biota. These toxicants enter the human body via multiple exposure routes, including inhalation, dietary intake, and dermal absorption, subsequently accumulating in vital organs (such as the liver, lungs, kidneys, and intestines) as well as within tumor tissues. By disrupting physiological homeostasis, they impair the functionality of critical biological systems, including the metabolic, nervous, circ

Metabolic Cross Talk in the Tumor Microenvironment.

The tumor microenvironment (TME) is a dynamic network of diverse cellular and noncellular components that interact with malignant cells to influence cancer progression, immune evasion, metastasis, and the development of therapeutic resistance. The major constituents of the TME include malignant cells, immune cells, stromal cells, endothelial cells (ECs), pericytes, adipocytes, nerve cells, and the extracellular matrix (ECM). Findings revealed that a dynamic metabolic crosstalk occurs continuously within the tumor niche, leading to metabolic rewiring in the tumor resident cells that supports ca

The electrical tumor microenvironment: abnormalities and opportunities.

Abnormal electrical properties in cancerous tumors have been observed for more than a century. Modern technological advances have enabled rapid improvements in our understanding of these electrical facets and our ability to leverage this information for diagnosis and treatment. Here, we outline the current knowledge of the electrical tumor microenvironment (eTME), including electrical abnormalities at the tissue, cell, and molecular scales. Of particular interest to solid tumors and immunotherapy, the innate immune system responds profoundly to these abnormalities with altered mobility and eff

Inflammasome-autophagy crosstalk in the tumor microenvironment.

The NLRP3 inflammasome has emerged as a critical sensor of cellular stress within the tumor microenvironment (TME), where its context-dependent activation can exert paradoxical effects on tumor progression and antitumor immunity. Diverse tumor-associated stressors, including mitochondrial dysfunction, reactive oxygen species (ROS), mitochondrial DNA (mtDNA) release, and cGAS-STING signaling, converge on NLRP3 activation, promoting caspase-1-dependent cytokine maturation and gasdermin D (GSDMD)-mediated pyroptosis. However, persistent or dysregulated inflammasome signaling may instead establish

Tumor microenvironment activatable immunomodulator for cancer immunotherapy.

The tumor microenvironment (TME) represents a sophisticated ecosystem wherein resident biomaterials orchestrate therapeutic resistance, and immune evasion, making it a critical target for cancer immunotherapy. This review underscores the central role of the TME in immunomodulation and the potential of TME-activatable immunomodulators to revolutionize cancer immunotherapy. We first provide an overview of the characteristics and components within the TME that drive tumor growth and metastasis, including immune cells, stromal cells, and metabolic factors. Next, we summarize the design principles

Targeting cytokines: Reshaping the pancreatic tumor microenvironment.

Pancreatic ductal adenocarcinoma (PDAC) contains various cell types scattered by desmoplastic stroma and communicate through small molecular weight signaling molecules called cytokines. Cytokines include interleukins, chemokines, interferons, tumor necrosis, and growth factors (TGF-β, VEGF, and EGF) promote cancer cell proliferation, differentiation, modulation of tumor microenvironment (TME), and metastasis. Targeting cytokines/ cytokine receptor is a potential approach for modulating immune responses, increasing therapeutic efficacy, and overcoming therapy resistance. This review summarizes

T-cell Senescence in the Tumor Microenvironment.

T-cell senescence occurs in the tumor microenvironment (TME) and influences cancer outcomes as well as the effectiveness of immunotherapies. The TME triggers this T-cell senescence via multiple pathways, including persistent stimulation with tumor-associated antigens, altered metabolic pathways, and activation of chronic inflammatory responses. Senescent T cells exhibit characteristics such as genomic instability, loss of protein homeostasis, metabolic dysregulation, and epigenetic alterations. Direct cross-talk between senescent T cells and other immune cells further exacerbates the immunosup

Pathology and tumor microenvironment of vestibular schwannoma.

Vestibular schwannomas (VS) are WHO Grade 1 tumors arising from the vestibule-cochlear nerve and account for 8% of all intracranial neoplasms. In recent years, there has been a dramatic evolution in how the pathology of these tumors is regarded, and there is emerging interest in the role of the tumor microenvironment. In this chapter, we provide a comprehensive description of the pathology and microenvironment of VS, with a particular focus on the interlinked processes of immune cell infiltration and angiogenesis. Improved understanding of the pathophysiology of these tumors has already yielde

Small-molecule modulators of tumor immune microenvironment.

In recent years, tumor immunotherapy, aimed at increasing the activity of immune cells and reducing immunosuppressive effects, has attracted wide attention. Among them, immune checkpoint blocking (ICB) is the most commonly explored therapeutic approach. All approved immune checkpoint inhibitors (ICIs) are clinically effective monoclonal antibodies (mAbs). Compared with biological agents, small-molecule drugs have many unique advantages in tumor immunotherapy. Therefore, they also play an important role. Immunosuppressive signals such as PD-L1, IDO1, and TGF-β, etc. overexpressed in tumor cells

Therapeutic targeting of the pituitary tumor microenvironment.

The tumor microenvironment (TME), the complex environment in which tumors develop, has been increasingly targeted for cancer treatment in recent years. Aggressive pituitary tumors and pituitary carcinomas have been so far targeted with immune-checkpoint inhibitors (28 cases, including a large cohort), and anti-angiogenic drugs (34 cases), specifically bevacizumab (30 cases), sunitinib (three cases), and apatinib (one case). Here, we reviewed all these cases, reporting tumor response, potential predictors of response, as well as adverse events. Given that the histological type could potentially

cGAS-STING signaling in the tumor microenvironment.

The cGAS-STING signaling is an important pathway involved in the regulation of tumor microenvironment, which affects many cellular functions including immune activation. Its role in combating tumor progression is widely recognized, especially with its function in inducing innate and adaptive immune responses, on which many immunotherapies have been developed. However, a growing number of findings also suggest a diversity of its roles in shaping tumor microenvironment, including functions that promote tumor progression. Here, we summarize the functions of the cGAS-STING signaling in tumor micro

Targeting Tumor Physical Microenvironment for Improved Radiotherapy.

Radiotherapy has led to important clinical advances; existing cancer radiotherapy resistance is one remaining major challenge. Recently, biophysical cues in the tumor microenvironment (TME) have been regarded as the new hallmarks of cancer, playing pivotal roles in various cancer behaviors and treatment responses, including radiotherapy resistance. With recent advances in micro/nanotechnologies and functional biomaterials, radiotherapy exerts great influence on biophysical cues in TME, which, in turn, significantly affect the response to radiotherapy. Besides, various strategies have emerged t

The tumor immune microenvironment in peritoneal carcinomatosis.

One in four patients with colorectal cancer, 40% of gastric cancer patients, and 60% of ovarian cancer patients will develop peritoneal metastases (PM) in the course of their disease. The outcome of patients with widespread PM remains poor with currently available treatments. Despite the relatively common occurrence of PM, little is known on the pathophysiology that drives the peritoneal metastatic cascade. It is increasingly recognized that the stromal components of the peritoneal microenvironment play an essential role in tumor progression. However, little is known about the specific interac

Immune Tumor Microenvironment in Ovarian Cancer Ascites.

Ovarian cancer (OC) has a specific type of metastasis, via transcoelomic, and most of the patients are diagnosed at advanced stages with multiple tumors spread within the peritoneal cavity. The role of Malignant Ascites (MA) is to serve as a transporter of tumor cells from the primary location to the peritoneal wall or to the surface of the peritoneal organs. MA comprise cellular components with tumor and non-tumor cells and acellular components, creating a unique microenvironment capable of modifying the tumor behavior. These microenvironment factors influence tumor cell proliferation, progre
