Plant-parasitic nematodes are one of the major threats to crop protection. However, only limited nematicides are currently available and are confronted with a growing resistance problem, which necessitates the development of novel nematicides. In this study, a series of trifluorobutene amide derivatives was synthesized through the strategy of amide bond reversal, and their nematicidal activity against Meloidogyne incognita was evaluated. The bioassay showed that compounds C2, C10, and C18 and some analogues thereof exhibited good nematicidal activity. Among them, the derivatives of compound C2

Root-knot nematode is one of the major problems that face the agricultural production of several vegetable crops. Chemical nematicides have been banned because of their healthy and environmental undesirable attributes. So, this study aimed to evaluate the potential use of sweet annie (Artimisia annua) and garden cress (Lepidium sativum) as green routes for the development of effective and eco-friendly alternative nematicides. Nematicidal activity of sweet annie and garden cress aqueous extracts (500 g/L) in the original and nano-forms were evaluated against Meloidogyne incognita in tomato plan

Root-knot nematodes (RKNs; Meloidogyne spp.) are obligate endoparasites that infect many crops and cause severe yield losses. In this research, we studied the effect of Welsh onion, grown as a companion plant, on the resistance of cucumber plants to RKN infection and analyzed the most abundant components of Welsh onion root exudates. The results showed that, when grown with Welsh onion as a companion plant, cucumber roots had 77.0% fewer root knots and egg masses than the control cucumber roots. Welsh onion root exudates were collected and extracted with chloroform, ethyl ether, n-butanol and

The sedentary plant-parasitic nematodes are considered among the most economically damaging pathogens of plants. Following infection and the establishment of a feeding site, sedentary nematodes become immobile. Loss of mobility is reversed in adult males while females never regain mobility. The structural basis for this change in mobility is unknown. We used a combination of light and transmission electron microscopy to demonstrate cell-specific muscle atrophy and sex-specific renewal of neuromuscular tissue in the sedentary nematode Heterodera glycines. We found that both females and males un

Plant-parasitic nematodes cause annual global crop losses, driving the urgent demand for sustainable alternatives to synthetic nematicides to control them. Among biological control agents, Purpureocillium lilacinum and Pochonia chlamydosporia have emerged as the most promising egg-parasitic fungi. However, their comparative biology and deployment contexts remain incompletely understood. This review synthesizes the current knowledge on their taxonomy, ecology, molecular genomics, parasitism mechanisms, biocontrol efficacy and commercial formulations. Both fungi parasitize eggs of economically d

Pea (Pisum sativum L.) is a cool-season annual legume crop cultivated globally. It is increasingly integrated into cereal-based rotations across Mediterranean agroecosystems, raising concerns about plant-parasitic nematodes (PPNs) that affect both legumes and subsequent crops. This study establishes the prevalence, diversity, and abundance of PPNs in a novel cropping system in southern Spain, where pea is introduced into cereal rotations. Nematode community structure in the surveyed fields is strongly influenced by intensive tillage practices and the desiccation tolerance of certain species, w

Citrus nematode, Tylenchulus semipenetrans Cobb, 1913 is a major economically important plant-parasitic nematode responsible for citrus slow decline worldwide. This study aimed to evaluate the efficacy of controlled mild water stress on priming defences in navel orange (Citrus sinensis “Navel”) seedlings and to model nematode-host interactions under stress. Five greenhouse treatments were applied: negative control (C-, 100% field capacity), positive control (C+, 100%), pre-infection water stress (T1, 40%), post-infection water stress (T2, 40%), and continuous water stress (T3, 70%). Enolase (2-phospho-D-glycerate hydrolase, EC 4.2.1.11) is a conserved glycolytic enzyme that catalyzes the reversible dehydration of 2-phosphoglycerate to phosphoenolpyruvate. In animal- and entomopathogenic nematodes, enolase has been shown to interact with host organisms and activate immune responses. In the plant-parasitic cyst nematode Heterodera schachtii, an established model species, enolase is hypothesized to be similarly exposed to host plant tissues during infection, based on its identification in the secretomes of related plant-parasitic nematodes. Unlike animals, plants lack an ada

The increase of plant parasitic nematode resistance poses a great threat to agricultural and forestry production. To search for nematicides with novel mechanism of action, nematicidal activity of the 29 pyrazine compounds were tested. Compound C2 showed excellent nematicidal activity against Bursaphelenchus xylophilus, Meloidogyne incongnita, and Aphelenchoides besseyi, with LC50 values were 2.3, 3.3, and 10.6 mg/L, respectively, and the pot experiment showed that C2 reduced the number of tomato roots knots to a certain extent. In addition, C2 significantly inhibited egg hatching, locomotion a

In the rice agroecosystems of Southeast Asia, rice root knot nematode (Meloidogyne graminicola) significantly impairs yield, representing a major species within the 'graminis-group' known for its morphological similarities with other root knot nematodes (RKNs). This study delves into the variations in reproductive potential, morphology, morphometrics, and genetic diversity among thirty RKN populations in rice across three distinct agroecological zones in Jharkhand, India. Despite notable differences in reproductive potential among the populations, morphological and morphometric correlations to

Gall-inducing insects and nematodes engage in sophisticated interactions with their host plants. These parasites can induce major morphological and physiological changes in host roots, leaves, and other tissues. Sedentary endoparasitic nematodes, root-knot and cyst nematodes in particular, as well as gall-inducing and leaf-mining insects, manipulate plant development to form unique organs that provide them with food from feeding cells. Sometimes, infected tissues may undergo a developmental switch resulting in the formation of aberrant and spectacular structures (clubs or galls). We describe h

Root-knot nematode causes severe yield losses in cucumber and other cucurbits. The wild relative Cucumis metuliferus exhibits strong resistance, but the metabolic basis of this resistance and the potential of resistance-associated metabolites to protect susceptible crops remains poorly understood. Here, we compared host status, phenolic content, and untargeted metabolomes of roots and rhizosphere soils of C. metuliferus and C. sativus upon Meloidogyne incognita infection. C. metuliferus showed delayed nematode development, fewer galls, and a lower reproduction factor compared with C. sativus,

Eclepins are root-secreted compounds that induce the hatching of cyst nematodes. Solanoeclepin A (SEA) and B (SEB) have been isolated as potent hatching factors for potato cyst nematodes (PCNs). SEB is biosynthesized in roots, released into the rhizosphere, and converted into SEA by soil microorganisms. However, given that SEB and SEA exhibit comparable hatching-inducing activity toward PCNs, the ecological significance of microbial solanoeclepin metabolism in eclepin-mediated communication remains unclear. In this study, we identified solanoeclepin C (SEC), a previously unrecognized solanoecl

The soybean cyst nematode (SCN) is a devastating pathogen for economic and food security considerations. Although the SCN genome has recently been sequenced, the presence of any miRNA has not been systematically explored and reported. This paper describes the development of a species-specific SCN miRNA discovery pipeline and its application to the SCN genome. Experiments on well-documented model nematodes (Caenorhabditis elegans and Pristionchus pacificus) are used to tune the pipeline's hyperparameters and confirm its recall and precision. Application to the SCN genome identifies 3342 high-co

BAK1-INTERACTING RECEPTOR LIKE KINASE1 (BIR1) is a negative regulator of various aspects of disease resistance and immune responses. Here, we investigated the functional role of soybean (Glycine max) BIR1 (GmBIR1) during soybean interaction with soybean cyst nematode (SCN, Heterodera glycines) and the molecular mechanism through which GmBIR1 regulates plant immunity. Overexpression of wild-type variant of GmBIR1 (WT-GmBIR1) using transgenic soybean hairy roots significantly increased soybean susceptibility to SCN, whereas overexpression of kinase-dead variant (KD-GmBIR1) significantly increase

Meta-analysis was used to compare yield protection and nematode suppression provided by two seed-applied and two soil-applied nematicides against Meloidogyne incognita and Rotylenchulus reniformis on cotton across 3 years and several trial locations in the U.S. Cotton Belt. Nematicides consisted of thiodicarb- and fluopyram-treated seed, aldicarb and fluopyram applied in furrow, and combinations of the seed treatments and soil-applied fluopyram. The nematicides had no effect on nematode reproduction or root infection but had a significant impact on seed cotton yield response ([Formula: see tex

BACKGROUND: Plant-parasitic nematodes pose a severe threat to crops, causing over $175 billion in economic losses to global agriculture.
RESULTS: To discover novel nematicides, a database of more than 2000 molecules was constructed, and several active scaffold structures were obtained through rapid in vitro nematicidal activity screening. Eight pyrimidine derivatives containing 1,2,4-oxadiazole moieties were designed and synthesized using scaffold hopping strategies that grafted tioxazafen onto the active pyrimidine structure. Among the 38 pyrimidine compounds screened or synthesized, the maj
