Circulating fatty acids and risk of gastrointestinal cancer in the UK Biobank.

BACKGROUND AND AIMS: Gastrointestinal(GI) cancer poses a significant threat to human health and safety, with studies suggesting a potential correlation between fatty acids(FAs) and GI diseases. We aim to comprehensively explore the association between plasma FAs and the risk of GI cancer and assess the causal effect of FAs on GI cancer risk through Mendelian randomization (MR). METHODS: This prospective cohort study includes 230,415 cancer-free participants from the UK Biobank. We utilized Cox regression, restricted cubic splines, and accelerated failure time models to analyze the correlation

Relationship between cardiac index and brain volume in the UK Biobank.

AIMS: Although cardiac function is associated with cognitive impairment in end-stage heart disease, the links between cardiac function and brain structure remain unclear. We aimed to examine the association between cardiac index (CI) (a measure of cardiac output adjusting for body surface area) and brain volumes in mid-to later life and how these are affected by age, sex and cardiometabolic factors. MATERIALS AND METHODS: We utilized brain and cardiac magnetic resonance imaging (MRI) data from a population-based cohort of people in the UK Biobank (a United Kingdom population-based cohort of p

Risk Factors for the Diagnosis of Progressive Supranuclear Palsy in the UK Biobank.

BACKGROUND: Progressive supranuclear palsy (PSP) is a tauopathy for which there is limited understanding of epidemiological risk factors. OBJECTIVE: The objective of this study was to identify premorbid nonmedical and medical risk factors for PSP diagnosis in a large prospective population-based cohort. METHODS: We performed a matched nested case-control study using the UK Biobank (UKB) cohort consisting of >500,000 adults aged 37 to 73 years. PSP diagnoses were identified from 2007 to censor date (August 2025) and matched 1:10 by age and sex to control subjects without parkinsonism or demen

Analgesics use and risk of pancreatitis: Result from the UK Biobank.

BACKGROUND: Pancreatitis, an inflammatory pancreatic disorder, arises from various etiologies including alcohol, tobacco, and drug use. Although the initiation of pancreatitis has been strongly linked to several medications, the association between analgesic use and pancreatitis risk remains ambiguous in population-based studies. METHODS: This prospective cohort study involved 324,982 participants from the UK Biobank. Multivariable-adjusted Cox proportional hazards models were conducted to evaluate the longitudinal association between incident pancreatitis risk and analgesics use, encompassin

Plasma Proteomic Biomarkers of Degenerative Cervical Myelopathy in the UK Biobank.

BACKGROUND AND OBJECTIVES: The diagnosis of spinal cord dysfunction in degenerative cervical myelopathy (DCM) is currently based on correlating clinical signs and symptoms with neuroimaging findings. Plasma proteomics represents an opportunity to establish objective, blood-based biomarkers, which predict DCM severity or response to surgical intervention. These methods may also support novel insights into its pathophysiological relationship with other neurological diseases, such as multiple sclerosis (MS), which also can present with myelopathy, or peripheral neuropathy (PN), which has overlapp

The role of obesity in mortality from digestive diseases in UK Biobank.

Most current studies use body mass index (BMI) or waist circumference (WC) to define obesity, and BMI, which reflects subcutaneous fat independent of visceral fat, is the most commonly used indicator of obesity. WC reflects subcutaneous and visceral fat. This research aims to investigate the effect of obesity on mortality in people with digestive diseases. According to BMI and WC, we divided patients with obesity into the following four groups: general obesity, abdominal obesity, combined obesity, and non-obese. The effects of different obesity types on mortality in a population with digestive

A spectrum of recessiveness among Mendelian disease variants in UK Biobank.

Recent work has found increasing evidence of mitigated, incompletely penetrant phenotypes in heterozygous carriers of recessive Mendelian disease variants. We leveraged whole-exome imputation within the full UK Biobank cohort (n ∼ 500K) to extend such analyses to 3,475 rare variants curated from ClinVar and OMIM. Testing these variants for association with 58 quantitative traits yielded 102 significant associations involving variants previously implicated in 34 different diseases. Notable examples included a POR missense variant implicated in Antley-Bixler syndrome that associated with a 1.76

The influence of personality on the risk of myocardial infarction in UK Biobank cohort.

Personality is a strong determinant for several health-related behaviours and has been linked to the development of cardiovascular diseases. However, the reports of personality's mediating role have been inconsistent with no data available from large population-based cohorts. The study aimed to create proxies for the Big Five personality traits, extraversion, agreeableness, conscientiousness, openness and neuroticism, to examine the longitudinal relationship between personality and myocardial infarction in the UK Biobank. The study sample comprised of 484,205 participants (55% female, 45% male

Genome-wide association study of dermatophytosis in the UK Biobank cohort.

BACKGROUND: Analyses of the hereditary propensity to dermatophytosis have revealed several proven genetic relationships. They include CARD9 deficiency, HLA-DR4 and HLA-DR8 type and genes encoding interleukin-22, defensin 2 and 4, and genetic defects in dectin-1, which increased the prevalence of dermatophytosis in families and were involved in the inheritance of susceptibility in their members. METHODS: To further investigate the genetic basis of dermatophytosis, we performed a genome-wide association study (GWAS) of the UK Biobank cohort. To identify cases of dermatophytosis, we used ICD10 c

An atlas of mitochondrial DNA genotype-phenotype associations in the UK Biobank.

Mitochondrial DNA (mtDNA) variation in common diseases has been underexplored, partly due to a lack of genotype calling and quality-control procedures. Developing an at-scale workflow for mtDNA variant analyses, we show correlations between nuclear and mitochondrial genomic structures within subpopulations of Great Britain and establish a UK Biobank reference atlas of mtDNA-phenotype associations. A total of 260 mtDNA-phenotype associations were new (P -5), including rs2853822 /m.8655 C>T (MT-ATP6) with type 2 diabetes, rs878966690 /m.13117 A>G (MT-ND5) with multiple sclerosis, 6 mtDNA associa

Relationships between estimated autozygosity and complex traits in the UK Biobank.

Inbreeding increases the risk of certain Mendelian disorders in humans but may also reduce fitness through its effects on complex traits and diseases. Such inbreeding depression is thought to occur due to increased homozygosity at causal variants that are recessive with respect to fitness. Until recently it has been difficult to amass large enough sample sizes to investigate the effects of inbreeding depression on complex traits using genome-wide single nucleotide polymorphism (SNP) data in population-based samples. Further, it is difficult to infer causation in analyses that relate degree of

Childhood febrile illness and the risk of myopia in UK Biobank participants.

PURPOSE: Historical reports suggest febrile illness during childhood is a risk factor for myopia. The establishment of the UK Biobank provided a unique opportunity to investigate this relationship. PATIENTS AND METHODS: We studied a sample of UK Biobank participants of White ethnicity aged 40-69 years old who underwent autorefraction (N=91 592) and were classified as myopic (≤-0.75 Dioptres (D)), highly myopic (≤-6.00 D), or non-myopic (>-0.75 D). Self-reported age at diagnosis of past medical conditions was ascertained during an interview with a nurse at a Biobank assessment centre. Logistic
