Phospholipase A2 dependent release of fatty acids from peroxidized membranes.

Unilamellar vesicles (liposomes) consisting of liver phosphatidylcholine and phosphatidylethanolamine were used as model membranes and subjected to lipid peroxidation. Following peroxidation samples were treated with phospholipase A2 from snake venom and subsequently analyzed for products of lipid peroxidation and of phospholipase A2 action. A significantly increased susceptibility to phospholipase A2 was noted for liposomes subjected to peroxidation, as compared to controls, which correlated with the extent of lipid peroxidation measured by the formation of thiobarbituric acid reacting produc

Immunization against experimental rabbit cysticercosis using liposome-associated antigen preparations.

Rabbits were vaccinated once, by subcutaneous and intradermal injection, with sonicates of oncospheres (TpO) or conditioned media from in vitro maintained mature metacestodes (TpMcES) of Taenia pisiformis. Extracts were either incorporated into or mixed with unilamellar liposomes (reverse phase evaporative vesicles) or emulsified in Freund's Incomplete Adjuvant (FIA). Control groups received liposomes or FIA without antigen, or antigen preparation without adjuvant. Rabbits were challenged orally two weeks after vaccination with approximately 1500 eggs of T. pisiformis and necropsied eight week

Reconstitution of the purified alpha-latrotoxin receptor in liposomes and planar lipid membranes. Clues to the mechanism of toxin action.

The receptor of alpha-latrotoxin (the major toxin of the black widow spider venom), purified from bovine synaptosomal membranes, was reconstituted into small unilamellar liposomes. These (but not control) liposomes exhibited high-affinity, specific binding of [125I]alpha-latrotoxin. In the receptor-bearing liposomes alpha-latrotoxin induced depolarization and stimulated 45Ca efflux. These responses to alpha-latrotoxin, that were observed only in the presence of external divalent cations, resembled those previously demonstrated in mammalian brain synaptosomes. The alpha-latrotoxin-activated ion

pH-dependent effects of chlorpromazine on liposomes and erythrocyte membranes.

Chlorpromazine (CPZ) is an amphipathic antipsychotic drug that binds to erythrocytes reaching in this way the central nervous system. CPZ is a basic molecule with pK=8.6. This paper reports on CPZ-induced lysis of red blood cells and liposomes. Haemolysis was tested under hypotonic conditions, in the pH range 5.0-10.0. Cell sensitivity towards CPZ increased with increasing pH. Increasing pH caused also a decrease in the critical micellar concentrations of CPZ. These results are interpreted in terms of a competition between repulsive electrostatic forces and attractive hydrophobic forces, that

Liposomes in drugs, nutraceuticals, and cosmetics.

BACKGROUND: Liposomes are spherical vesicular structures formed by lipid bilayers with a hydrophilic internal nucleus. The size varies from about 20 nanometers to several micrometers. They were initially described in the sixties by the biophysicist Alec Bangham. Since then, multiple studies have been conducted to characterize them according to the number of layers: unilamellar, bilayers, multilamellar vesicles, and vesicles containing more vesicles called multivesicular vesosomes; size: between 0.025 µm and 2.5 µm, and functionality: Drug delivery system, cosmetic, nutraceutics. Moreover, ther

Changes of the lipid membrane structures caused by chain-length-dependent doxorubicin embedment in PEGylated liposomes.

Poly(ethyl-ene glycol)-grafted (PEGylated) liposomes receive increasingly more attention due to their practical applications in delivering vaccines, nutrients and drug molecules such as doxorubicin (DOX). PEGylated liposomes have been well documented for their capability in carrying DOX as rod-like crystallites enclosed inside the unilamellar vesicles. This study addresses the previously unresolved question of whether DOX intercalates into liposome bilayers by employing simultaneous small- and wide-angle X-ray scattering (SWAXS), complemented by an integrated asymmetric flow field-flow fractio

Delivery of DTPA through Liposomes as a Good Strategy for Enhancing Plutonium Decorporation Regardless of Treatment Regimen.

In this study, we assessed the efficacy of unilamellar 110-nm liposomes encapsulating the chelating agent diethylenetriaminepentaacetic acid (DTPA) in plutonium-exposed rats. Rats were contaminated by intravenous administration of the soluble citrate form of plutonium. The comparative effects of liposomal and free DTPA at similar doses were examined in terms of limitation of alpha activity burden in rats receiving various treatment regimens. Liposomal DTPA given at 1 h after contamination more significantly prevented the accumulation of plutonium in tissues than did free DTPA. Also, when compa

Optimization of cationic lipid/DNA complexes for systemic gene transfer to tumor lesions.

Intravenous (i.v.) administration of cationic lipid N-[( 1-(2-3-dioleyloxy)propyl)]-N-N-N-trimethylammonium chloride (DOTMA)-based transfection complexes in mice with subcutaneous squamous cell tumors yielded plasmid delivery and expression in tumor lesions. The efficiency of gene transfer in tumors was significantly lower than in the lung. This was consistent with low plasmid levels associated with the tumor, suggesting that plasmid delivery to the tumor site was a limiting factor. Lowering the lipid/DNA charge ratio from 5:1 to 0.8:1 (+/-) did not change DNA levels in tumor but significantly

Mapping lipid-dependent operational outcomes of puroindoline-derived tryptophan-rich peptides at model membrane interfaces.

HYPOTHESIS: Membrane-active peptides are often described as "membrane targeting," yet peptide adsorption, vesicle leakage, and colloidal destabilisation are not necessarily coupled. We hypothesised that lipid composition controls access to distinct ensemble-level peptide-LUV interaction outcomes, and that sequence-derived variables bias these outcomes rather than encoding a single universal lytic mechanism. EXPERIMENTS: Three tryptophan-rich cationic peptides (PuroA, P1, W7) were examined against composition-defined large unilamellar vesicles (LUVs) representing Gram-, Gram+, fungal, red bloo

Membrane-Penetrating Molecular Device Based on a Triplex Containing Acyclic L-Threoninol Nucleic Acid Functionalized with Cholesterol.

Membrane-penetrating molecular devices are valuable biological tools. Herein, we describe membrane-targeting molecular devices based on the triplexes of poly T acyclic L-threoninol nucleic acid (L-aTNA) tethered site-specifically to cholesterol and poly A DNA. The L-aTNA was linked to cholesterol in two different orientations. The membrane-binding properties differed depending on the orientation at the cholesterol linkage. One triplex adhered to giant unilamellar vesicles and the other penetrated these vesicles. When the membrane-penetrating triplex was formed with a photoresponsive DNA, photo

Magnetic micromotors crossing lipid membranes.

Nano/micromotors are self-propelled particles that show enhanced motion upon being triggered by a stimulus. Their use in nanomedicine has been widely explored, with special focus on imaging or drug delivery. However, a thorough understanding of the requirements for more efficient locomotion is still lacking. In this paper, we assembled magnetically propelled motors of different sizes (i.e., 0.5, 1 and 4 μm) and surface chemistries (positive charge or PEGylated) and assessed their motion in the presence of giant unilamellar lipid vesicles (GUVs) of varying compositions (zwitterionic, negatively

Composition dependent effects of charged lipids on freeze-thaw induced fusion in dioleoyl phospholipid membranes.

Artificial cells based on giant unilamellar vesicles (GUVs) provide a platform for reconstituting biochemical reactions within membrane-bound compartments. However, limited membrane permeability restricts substrate exchange, leading to depletion during prolonged reactions. Freeze-thaw (F/T) treatment has been used to overcome this limitation by enabling liposome fusion and content exchange. Here, we systematically investigated F/T-induced fusion in dioleoylphosphatidylcholine (DOPC) liposomes with negatively charged dioleoylphosphatidylglycerol (DOPG) and positively charged dioleoyltrimethylam

Inhalable liposomal formulation for vasoactive intestinal peptide.

Inhalation of vasoactive intestinal peptide (VIP) was suggested as promising treatment option of various lung diseases like asthma and pulmonary hypertension. However, the medical use of peptides is limited by their short half-life due to rapid enzymatic degradation in the airways. For that reason, we recently developed unilamellar nano-sized VIP-loaded liposomes (VLL). Now we investigated their applicability for inhalation purposes. After nebulisation by a mouthpiece ventilation inhaler we found the particle size almost unaffected, being in a size range appropriate for bronchiolar deposition;

Liposome Based Drug Delivery for the Management of Psoriasis - A Comprehensive Review.

Psoriasis has been considered as a chronic inflammatory skin disease which leads to the dysfunction of immune systems. According to the World Psoriasis Day consortium, psoriasis affects around 125 million individuals globally or about 2% to 3% of the overall population. Most of the conventional drug delivery systems primarily attempt to relieve symptoms of psoriasis and are ineffective in providing targeted action and higher bioavailability because of the drug's short half-life and instability, as well as they lack safety and efficacy. The shortcomings of conventional drug delivery systems giv

Effect of Selected Anionic and Cationic Drugs Affecting the Central Nervous System on Electrical Properties of Phosphatidylcholine Liposomes: Experiment and Theory.

Interactions between phospholipid membranes and selected drugs affecting the central nervous system (CNS) were investigated. Small, unilamellar liposomes were used as biomimetic cell membrane models. Microelectrophoretic experiments on two-component liposomes were performed using the electrophoretic light scattering technique (ELS). The effect of both positively (perphenazine, PF) and negatively (barbituric acid, BA) charged drugs on zwitterionic L-α-phosphatidylcholine (PC) membranes were analyzed. Experimental membrane surface charge density (δ) data were determined as a function of pH. Quan

Electroformation of Giant Unilamellar Vesicles: Investigating Vesicle Fusion versus Bulge Merging.

Partially ordered stacks of phospholipid bilayers on a flat substrate can be obtained by the evaporation of a spread droplet of phospholipid-in-chloroform solution. When exposed to an aqueous buffer, numerous micrometric buds populate the bilayers, grow in size over minutes, and eventually detach, forming the so-called liposomes or vesicles. While observation of vesicle growth from a hydrated lipid film under an optical microscope suggests numerous events of vesicle fusion, there is little experimental evidence for discriminating between merging of connected buds, i.e., a shape transformation

Protocol for microfluidic-based high-precision general polarization fluorescence microscopy of lipid packing in membrane vesicles.

Lipid packing plays a critical role in membrane organization and function, but its precise quantification remains challenging in fluidic environments. Here, we present a protocol to integrate confocal fluorescence generalized polarization (GP) imaging with a polydimethylsiloxane (PDMS)-based microfluidic platform. We describe steps for lipid preparation, giant unilamellar vesicle (GUV) fabrication, microfluidic trapping, and GP analysis using polarity-sensitive dyes. This protocol enables robust measurement of membrane order and is readily adaptable to diverse lipid compositions and aqueous en

Phosphatidylethanolamine modulates α-synuclein membrane-binding behavior.

Interaction with lipid membranes is important in the physiological and pathological functioning of α-synuclein (αS) in brain neuronal cells. In this study, we investigated the effect of lipid composition on the membrane-binding behavior of αS using multiple biophysical techniques. Circular dichroism measurement revealed that, although negatively charged phospholipids are necessary for αS to bind to small unilamellar vesicles, the presence of phosphatidylethanolamine (PE) significantly enhances α-helical structure formation, specifically within the first 35 αS residues. To obtain residue-level

Protocol for building synthetic protocell membranes that sense redox using synthetic phospholipids and natural lipids.

Sensing is a critical function of artificial cells; however, this is challenging to realize using bottom-up approaches. Here, we present a protocol for building protocell membranes that sense cues important for redox biochemistry and signaling by combining synthetic phospholipids and natural lipids. We detail procedures for building giant unilamellar vesicles as protocell models that fluoresce in response to the biologically significant redox agents peroxynitrite, hydrogen peroxide, and hydrogen sulfide. For complete details on the use and execution of this protocol, please refer to (i) Gutier
