Role of AMPK and PPARα in the anti-skin cancer effects of ursolic acid.

The phytonutrient ursolic acid (UA), present in apples, rosemary, and other plant sources, has anti-cancer properties in a number of systems, including skin cancers. However, few reports have examined upstream mechanisms by which UA may prevent or treat cancer. Recent reports have indicated UA induces death of cancer cell lines via AMP-activated protein kinase (AMPK), an energy-sensing kinase which possesses both pro-metabolic and anti-cancer effects. Other studies have shown UA activates peroxisome proliferator activated receptor α (PPARα) and the glucocorticoid receptor (GR). Here, we found

Ursolic acid and mechanisms of actions on adipose and muscle tissue: a systematic review.

This systematic review aimed at addressing the ursolic acid actions as an adjunctive treatment of the obesity-mediated metabolic abnormalities. To explore our aims, we used the literature search including clinical and animal studies using the Medline and Google Scholar (up to December 2015). Out of 63 screened studies, 17 presented eligibility criteria, such as the use of ursolic acid on adiposity, energy expenditure and skeletal muscle mass in mice and humans. In the literature, we found that several physiological and molecular mechanisms are implicated in the effects of ursolic acid on obesi

Spasmolytic and antinociceptive activities of ursolic acid and acacetin identified in Agastache mexicana.

Agastache mexicana is a plant in high demand that has long been used in Mexican folk medicine to treat anxiety, insomnia, and stomachache, among other afflictions. Ursolic acid and acacetin were isolated and identified as two possible active compounds of A. mexicana aerial parts. An antinociceptive response was demonstrated in a significant and dose-dependent manner with ursolic acid and acacetin (i. p. and p. o.) in comparison to the analgesic diclofenac by using the writhing test in mice. Moreover, acacetin also produced a significant concentration-dependent spasmolytic response with major e

Ursolic acid: context-dependent redox activity on oxidative stress and its toxicological implications.

Oxidative stress is a major contributor to xenobiotic-induced tissue injury, mitochondrial dysfunction, inflammation, ferroptosis, and carcinogenesis. Ursolic acid (UA), a naturally occurring pentacyclic triterpenoid, has emerged as a context-dependent regulator of redox homeostasis. This review summarizes current evidence regarding the dual effects of UA on oxidative stress and its toxicological implications. Under non-malignant conditions, UA generally exerts antioxidant and cytoprotective effects in the majority of experimental models examined. Reduced reactive oxygen species (ROS) accumula

Ursolic acid attenuates cholestasis through NRF2-mediated regulation of UGT2B7 and BSEP/MRP2.

Ursolic acid (UA), a pentacyclic triterpenoid, exhibits various pharmacological actions, such as anti-inflammation, anti-tumor, anti-diabetes, heart protection, and liver protection. However, the role of nuclear factor E2-related factor 2 (NRF2)-mediated regulation of uridine diphosphate glucuronosyltransferase (UGT2B7) and bile salt export pump (BSEP)/multidrug resistance-associated protein 2 (MRP2) in UA against cholestatic liver injury has not been cleared. The purpose of this study is to explore the effect of UA on cholestatic liver injury and its potential mechanism. The results of the li

Ursolic Acid Hydrazide Based Organometallic Complexes: Synthesis, Characterization, Antibacterial, Antioxidant, and Docking Studies.

In thecurrent research work, eleven metal complexes were synthesized from the hydrazide derivative of ursolic acid. Metal complexes of tin, antimony and iron were synthesized and characterized by FT-IR and NMR spectroscopy. The antibacterial and antioxidant activities were performed for these complexes, which revealed that the metal complexes synthesized are more potent than their parent compounds. We observed that antioxidant activity showed by triphenyltin complex was significant and least activity have been shown by antimony trichloride complex. The synthesized metal complexes were then eval

Systematic enzyme and cofactor engineering for efficient ursolic acid biosynthesis in Yarrowia lipolytica.

Ursolic acid (UA) is a pharmaceutically valuable pentacyclic triterpenoid, but its microbial production is constrained by inefficient cytochrome P450 catalysis and limited cofactor availability. Here, we engineered an efficient α-amyrin-producing Yarrowia lipolytica chassis for UA biosynthesis through integrated enzyme engineering, cofactor optimization, and metabolic flux balancing. Screening of heterologous plant cytochrome P450 monooxygenases identified CYP716A48 from Olea europaea as the most efficient α-amyrin oxidase in Y. lipolytica. Fusion of OeCYP716A48 with its redox partner AtCPR1 u

Ursolic acid inhibits leucine-stimulated mTORC1 signaling by suppressing mTOR localization to lysosome.

Ursolic acid (UA), a pentacyclic triterpenoid widely found in medicinal herbs and fruits, has been reported to possess a wide range of beneficial properties including anti-hyperglycemia, anti-obesity, and anti-cancer. However, the molecular mechanisms underlying the action of UA remain largely unknown. Here we show that UA inhibits leucine-induced activation of the mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway in C2C12 myotubes. The UA-mediated inhibition of mTORC1 is independent of Akt, tuberous sclerosis complex 1/2 (TSC1/2), and Ras homolog enriched in brain (Rheb), s

Suppression of hepatitis B virus x protein-mediated tumorigenic effects by ursolic Acid.

This study investigated the potential effects of natural products ursolic acid (UA) and oleanolic acid (OA) against HBx-mediated tumorigenic activities in vitro and in vivo. HBx transactivated Sp-1 and Smad 3/4 in Huh7 and FL83B hepatocytes and induced cell migration of Huh7 and HepG2. HBx also induced MMP-3 secretion in Huh7 and acted against TGF-β-induced apoptosis in Hep3B. UA almost completely blocked the HBx-mediated effects, while OA had a partial inhibitive effect. Utilization of specific MAPK inhibitors and immunoblotting demonstrated that UA selectively activated MAPK signaling in cer

Macrophage-targeted Hyaluronic-acid Coated Liposomes Bearing Ursolic Acid for Visceral Leishmaniasis: In Vitro Characterisation and Pharmacokinetic Analysis.

Ursolic acid (UA), a natural triterpene, demonstrated promising activity against visceral leishmaniasis (VL); however, its clinical use is limited by poor aqueous solubility (< 5.64 µg/mL), low permeability, hindered oral bioavailability (< 3%), and extensive first-pass metabolism. Hence, hyaluronic acid-coated ursolic acid-bearing liposomes (HA-UA-Lips) were formulated using the thin-film hydration method integrated with Box-Behnken Design (BBD). HA-UA-Lips displayed a vesicle size of 200.2 ± 5.2 nm, PDI of 0.164 ± 0.041, and zeta potential of -21.39 ± 0.27 mV. The surface topography analysis

Inhibitory Effect of Ursolic Acid on the Migration and Invasion of Doxorubicin-Resistant Breast Cancer.

The cause of death in most breast cancer patients is disease metastasis and the occurrence of multidrug resistance (MDR). Ornithine decarboxylase (ODC), which is involved into multiple pathways, is closely related to carcinogenesis and development. Ursolic acid (UA), a natural triterpenoid compound, has been shown to reverse the MDR characteristics of tumor cells. However, the effect of UA on the invasion and metastasis of tumor cells with MDR is not known. Therefore, we investigated the effects of UA on invasion and metastasis, ODC-related polyamine metabolism, and MAPK-Erk-VEGF/MMP-9 signali

Ursolic acid induces the production of IL6 and chemokines in both adipocytes and adipose tissue.

Adipose tissue inflammation plays an important role in the regulation of glucose and lipids metabolism. It is unknown whether Ursolic acid (UA) could regulate adipose tissue inflammation, though it can regulate inflammation in many other tissues. In this study, 3T3-L1 adipocytes, DIO mice and lean mice were treated with UA or vehicle. Gene expression of inflammatory factors, chemokines and immune markers in adipocytes and adipose tissue, cytokines in cell culture medium and serum, and inflammation regulatory pathways in adipocytes were detected. Results showed that UA increased the expression

The Supramolecular Organogel Formed by Self-Assembly of Ursolic Acid Appended with Aromatic Rings.

Ursolic acid (UA) as a natural ursane-triterpenoid has rich pharmacological activities. We have found that it possesses aggregation properties and could self-assemble into organogels. Based on the aggregation property of ursolic acid in suitable solvents, its derivative appended with aromatic rings by amide groups was synthesized. The property of self-assembly into organogel was studied in this paper. The results revealed that this derivative could form supramolecular gel in halogenated benzene and also gelate chloroform in the presence of toluene or p-xylene. By Fourier-transform infrared spe

Effects of Betulinic Acid and Ursolic Acid on IL-17-Induced CCL20 Release in Normal Human Epidermal Keratinocytes.

Psoriasis is a chronic inflammatory skin disease characterized by erythema, infiltration, and scaling, which is mainly caused by interleukin (IL)-17. The use of molecular targeted drugs in specific therapies offers high efficacy; however, high medical costs and a significant risk of side effects highlight the need for novel therapeutic agents. We previously observed that Morus alba extract (MAE) suppressed IL-17-induced CCL20 mRNA expression in normal human epidermal keratinocytes (NHEKs). In this study, we focused on the IL-17 signaling pathway and investigated the effects of pentacyclic trit
