Efficient Introduction of Genetic Elements at Specific Insertion Loci.

While some genetic modifications are conducted at the native loci of the manipulated genes, "neutral" loci for insertion of (heterologous) genetic elements are equally important for a versatile model system. This, e.g., applies for insertion of genetic complementation constructs or for transplantation of heterologous metabolic pathways. Here, the most frequently used insertion loci of Ustilago maydis is presented, namely the so-called ip locus and three loci encoding secreted proteases like the upp3 locus. The strategy of counter selection provides additional benefits in strain generation as i

U. maydis Protoplast Generation and Transformation.

Ustilago maydis is a dimorphic basidiomycete that exists as a yeast-like haploid form and a filamentous dikaryotic form. The yeast-like morphology is exploited for protoplast-based, PEG (polyethylene glycol)-mediated transformation, which is a simple and reliable means for introducing DNA into protoplasts. In U. maydis, this approach has been widely used for gene deletion/insertion, complementation, and transgene expression. In short, U. maydis yeast-like cells are grown to mid-log phase, and their cell walls are enzymatically digested in the presence of an osmotic stabilizer to generate stabl

Mannosylerythritol Lipid and Ustilagic Acid Analysis Using Thin-Layer Chromatography.

Secondary metabolites produced by Ustilago maydis, like mannosylerythritol lipids and ustilagic acid, are usually secreted together, leading to a glycolipid mixture. This makes it difficult to distinguish which compounds are present in the mixture or not. Here, a simple and fast approach for analysis of mannosylerythritol lipids and ustilagic acid using thin-layer chromatography is shown, enabling the separation and visualization of those from the mixture. A step-by-step protocol from the induction of such glycolipid synthesis in U. maydis over the isolation and their separation via thin-layer

A chromosome-level genome assembly of Thecaphora frezzii, cause of peanut smut, reveals the largest genome of the true smut fungi.

Peanut smut, caused by the fungus Thecaphora frezzii, is a significant disease of peanuts in Argentina. Infected plants have seeds replaced by a mass of dark teliospores, reducing yield and seed quality. Following successful in vitro culture of the fungus in its haploid stage, we produced a chromosome-level genome assembly of the species. We compare this genome to those of 49 other species of true smut fungi, or Ustilaginomycetes, including species of medical, agricultural, and industrial importance, some of which are known as pathogens and others only as saprotrophic yeasts. At almost 39 Mb,

Cross-reactivity of cryptococcal antigen lateral flow assay with basidiomycetous yeasts.

The cryptococcal antigen lateral flow assay (CrAg LFA) is a widely available point-of-care test for the diagnosis of cryptococcal meningitis. Although there have been some reports of cross-reactivity with Trichosporon species, the extent of cross-reactivity among basidiomycetous yeasts has not been widely explored. We tested a range of basidiomycete yeast isolates using a CrAg LFA to examine potential cross-reactivity, with positive results for all isolates belonging to Trichosporon and Naganishia species, and for some isolates of Cutaneotrichosporon, Filobasidium and Ustilago species. Studying Cell Wall-Associated Effectors in U. maydis.

Fungal cell wall-binding secreted effector proteins play pivotal roles in plant-microbe interactions and cell wall remodeling in Ustilago maydis. Accurate assessment of their localization and binding mode is essential for understanding their biological function. Here, a protocol combining immunolocalization with fluorescence microscopy and immunoprecipitation (IP) under both native and denaturing (SDS-containing) conditions is described. Immunolocalization provides direct visualization of protein distribution at the cell wall, while comparative IP experiments reveal the strength and nature of

Microtubule-based transport in filamentous fungi.

Defects in microtubule-based transport are implicated in many neuropathologies. The filamentous fungi Aspergillus nidulans and Ustilago maydis are valuable models for studying transport owing to their yeast-like genetic and biochemical tractability and metazoan-like dependence on microtubule-based transport for cellular trafficking. In these organisms the role of microtubules in nuclear positioning is well studied, but recent work has expanded the range of cargos to include endosomes, messenger RNA, secretory vesicles, peroxisomes, and nuclear pore complexes, reflecting the diversity of metazo

MGL_3741 gene contributes to pathogenicity of Malassezia globosa in pityriasis versicolor.

Dihydroxyacid dehydratase (DHAD) is a key enzyme in biosynthetic pathway of isoleucine and valine. This pathway is absent in human but exists in various organisms such as fungi. Using RNA-seq analysis in this study, we identified MGL_3741gene which encodes DHAD protein in Malassezia globosa (M. globosa). Furthermore, we found that mentioned gene is homologous to the Ustilago maydis, Saccharomyces cerevisiae, Aspergillus flavus, and Aspergillus fumigatus ILV3P. For understanding the probable role of this gene in pathogenicity of M. globosa, we applied Real-time PCR to investigate the differenti

A lipophilic cation protects crops against fungal pathogens by multiple modes of action.

The emerging resistance of crop pathogens to fungicides poses a challenge to food security and compels discovery of new antifungal compounds. Here, we show that mono-alkyl lipophilic cations (MALCs) inhibit oxidative phosphorylation by affecting NADH oxidation in the plant pathogens Zymoseptoria tritici, Ustilago maydis and Magnaporthe oryzae. One of these MALCs, consisting of a dimethylsulfonium moiety and a long alkyl chain (C18-SMe2+), also induces production of reactive oxygen species at the level of respiratory complex I, thus triggering fungal apoptosis. In addition, C18-SMe2+ activates

The Corn Smut ('Huitlacoche') as a New Platform for Oral Vaccines.

The development of new alternative platforms for subunit vaccine production is a priority in the biomedical field. In this study, Ustilago maydis, the causal agent of common corn smut or 'huitlacoche'has been genetically engineered to assess expression and immunogenicity of the B subunit of the cholera toxin (CTB), a relevant immunomodulatory agent in vaccinology. An oligomeric CTB recombinant protein was expressed in corn smut galls at levels of up to 1.3 mg g-1 dry weight (0.8% of the total soluble protein). Mice orally immunized with 'huitlacoche'-derived CTB showed significant humoral resp

Hook is an adapter that coordinates kinesin-3 and dynein cargo attachment on early endosomes.

Bidirectional membrane trafficking along microtubules is mediated by kinesin-1, kinesin-3, and dynein. Several organelle-bound adapters for kinesin-1 and dynein have been reported that orchestrate their opposing activity. However, the coordination of kinesin-3/dynein-mediated transport is not understood. In this paper, we report that a Hook protein, Hok1, is essential for kinesin-3- and dynein-dependent early endosome (EE) motility in the fungus Ustilago maydis. Hok1 binds to EEs via its C-terminal region, where it forms a complex with homologues of human fused toes (FTS) and its interactor FT

Characterization of tryptophan aminotransferase 1 of Malassezia furfur, the key enzyme in the production of indolic compounds by M. furfur.

Malassezia yeasts are responsible for the widely distributed skin disease Pityriasis versicolor (PV), which is characterized by a hyper- or hypopigmentation of affected skin areas. For Malassezia furfur, it has been shown that pigment production relies on tryptophan metabolism. A tryptophan aminotransferase was found to catalyse the initial catalytic step in pigment formation in the model organism Ustilago maydis. Here, we describe the sequence determination, recombinant production and biochemical characterization of tryptophan aminotransferase MfTam1 from M. furfur. The enzyme catalyses the t

An apoplastic peptide activates salicylic acid signalling in maize.

Localized control of cell death is crucial for the resistance of plants to pathogens. Papain-like cysteine proteases (PLCPs) regulate plant defence to drive cell death and protection against biotrophic pathogens. In maize (Zea mays), PLCPs are crucial in the orchestration of salicylic acid (SA)-dependent defence signalling. Despite this central role in immunity, it remains unknown how PLCPs are activated, and which downstream signals they induce to trigger plant immunity. Here, we discover an immune signalling peptide, Z. mays immune signalling peptide 1 (Zip1), which is produced after salicyl

A co-evolved peptide-GPCR system senses host entry to drive fungal infection.

A successful infection requires pathogens to recognize the specific host environment in order to reprogram their physiology accordingly. One major way in which eukaryotic cells sense their surroundings is via G-Protein Coupled Receptors (GPCRs), which share a seven-transmembrane architecture and G-protein-mediated downstream signaling. While mammalian GPCRs are well-characterized and represent important drug targets, their fungal counterparts remain poorly understood. Here we uncover a GPCR-based mechanism that allows the corn pathogen Ustilago maydis to sense whether it has entered plant tiss

Smut infection of perennial hosts: the genome and the transcriptome of the Brassicaceae smut fungus Thecaphora thlaspeos reveal functionally conserved and novel effectors.

Biotrophic fungal plant pathogens can balance their virulence and form intricate relationships with their hosts. Sometimes, this leads to systemic host colonization over long time scales without macroscopic symptoms. However, how plant-pathogenic endophytes manage to establish their sustained systemic infection remains largely unknown. Here, we present a genomic and transcriptomic analysis of Thecaphora thlaspeos. This relative of the well studied grass smut Ustilago maydis is the only smut fungus adapted to Brassicaceae hosts. Its ability to overwinter with perennial hosts and its systemic pl
