Allergens in hymenoptera venoms. X. Vespid venoms versus venom sac extracts: comparison by two-dimensional polyacrylamide gel electrophoresis.

Vespid venoms were compared to venom sac extracts by two-dimensional polyacrylamide gel electrophoresis using non-equilibrium pH gradient electrophoresis in the first dimension and sodium dodecyl sulfate electrophoresis in the second. The gels were stained with silver. Fresh venoms from four species, Vespula maculifrons, Polistes fuscatus fuscatus, P. metricus and P. exclamans, were compared with commercially available venom sac extracts from the same species. In each case the venom sac extract contained all of the proteins detected in the fresh venom plus numerous additional proteins which ar

Inflammation induced by snake venoms optimizes envenomation.

Snake envenomation activates the immune system through permeability-increasing factors that generate an acute vascular inflammatory response by opening large inflammation-activated pores (IAPs) in the microvasculature. These events facilitate the exudation of plasma from blood into body tissues. However we show that IAPs also allow macromolecules, including venom toxins, to flood directly into the bloodstream, even against an outflow of plasma solutes. Such inflammation-facilitated macromolecular absorption (IFMA) acts together with lymphatic absorption and has a physiological role in removing

Animal Venoms as Potential Source of Anticonvulsants.

Epilepsy affects millions of people worldwide, and there is an urgent need to develop safe and effective therapeutic agents. Animal venoms contain diverse bioactive compounds like proteins, peptides, and small molecules, which may possess medicinal properties against epilepsy. In recent years, research has shown that venoms from various organisms such as spiders, ants, bees, wasps, and conus snails have anticonvulsant and antiepileptic effects by targeting specific receptors and ion channels. This review underscores the significance of purified proteins and toxins from these sources as potenti

Early significant ontogenetic changes in snake venoms.

Snake venom plays a critical role in food acquisition, digestion, and defense. Venoms are known to change throughout the life of some snake species, but nothing is known about the venom composition of hatchling/neonate snakes prior to and just after their first shedding cycle, despite this being a critical time in the life of the snake. Using a cohort of Crotalus horridus and two cohorts of Crotalus adamanteus, we showed for the first time that snakes undergo significant changes in venom composition after the postnatal shedding event. The number of changes among cohorts ranged widely and there

[The threat of snake and scorpion venoms].

Venoms of snakes and scorpions pose a significant threat to the health and life of humans. The speed and range of their actions causes damage of the organ responsible for the maintenance of vital signs. Venomous snake venoms cause blood clotting disorders, tissue necrosis and hemolysis, and the release of a number of proinflammatory cytokines and impair antibody synthesis. Availability of antitoxins is limited and in the most cases supportive treatment is recommended. In turn, the venom of scorpions beside intestinal symptoms cause significant impairment of neuromuscular conduction, causing se

The constituents of Microctonus sp. parasitoid venoms.

Purified RNA transcripts from venom glands dissected from the parasitoid wasp Microctonus hyperodae were copied, cloned and sequenced using traditional dideoxy sequencing methods. Using mass spectrometry analysis of the trypsinised PAGE gel protein bands we identified the RNA transcripts for the 3 most abundant proteins found in the venom and hence obtained their full protein sequence. Other abundant transcripts were also further sequenced. To reduce the effort required to obtain transcript information we dissected venom glands from a second parasitoid, Microctonus aethiopoides (Morocco biotyp

New linear polyamine derivatives in spider venoms.

Linear free polyamines were characterized in the venom of the spiders Agelenopsis aperta, Hololena curta, and Paracoelotes birulai by RP-HPLC coupled to mass spectrometry. The several linear polyamines found were tetramine, pentamine, and hexamine derivatives. Some of these natural products were identified as N-hydroxylated, guanidylated, or acetylated compounds. In addition, the biosynthetical pathway leading to the formation of acylpolyamines in spider venoms is discussed.

Comparative enzymology of venoms from stinging Hymenoptera.

Venoms from 20 species of stinging Hymenoptera, including nine species of ants and nine species of social wasps, were quantitatively analyzed for the following enzymic activities: phospholipase A, hyaluronidase, lipase, esterase, protease, acid phosphatase, alkaline phosphatase and phosphodiesterase. Phospholipase and hyaluronidase were present in all the venoms, with activity levels generally higher among the wasps than the ants (P less than 0.05). Lipase was present in high activity in several social wasp venoms and one ant venom, in low levels in two other ant venoms and absent from four te

The biology and evolution of spider venoms.

Spiders are diverse, predatory arthropods that have inhabited Earth for around 400 million years. They are well known for their complex venom systems that are used to overpower their prey. Spider venoms contain many proteins and peptides with highly specific and potent activities suitable for biomedical or agrochemical applications, but the key role of venoms as an evolutionary innovation is often overlooked, even though this has enabled spiders to emerge as one of the most successful animal lineages. In this review, we discuss these neglected biological aspects of spider venoms. We focus on t

Immunomodulatory properties of molecules from animal venoms.

The immune system can amplify or decrease the strength of its response when it is stimulated by chemical or biological substances that act as immunostimulators, immunosuppressants, or immunoadjuvants. Immunomodulation is a progressive approach to treat a diversity of pathologies with promising results, including autoimmune disorders and cancer. Animal venoms are a mixture of chemical compounds that include proteins, peptides, amines, salts, polypeptides, enzymes, among others, which produce the toxic effect. Since the discovery of captopril in the early 1980s, other components from snakes, spi

Phosphodiesterases (PDEs) from Snake Venoms: Therapeutic Applications.

BACKGROUND: Snake venom, a highly poisonous and active venomous snake's secretion, is a complex mixture of inorganic cations, carbohydrates, lipids, proteins, peptides, toxins and hydrolytic enzymes of importance including Phosphodiesterases (PDEs). These snake venom hydrolytic enzymes interfere in different physiological processes. Snake venom PDEs have several roles to metabolize extracellular nucleotides and to regulate nucleotide based intercellular signalling mechanisms including platelet aggregation, which can lead to death and debilitation in cardiac arrest and strokes in patients havin

Animal Venoms have Potential to Treat Cancer.

The resistance to chemotherapeutics by the cancerous cells has made its treatment more complicated. Animal venoms have emerged as an alternative strategy for anti-cancer therapeutics. Animal venoms are cocktails of complex bioactive chemicals mainly disulfide-rich proteins and peptides with diverse pharmacological actions. The components of venoms are specific, stable, and potent and have the ability to modify their molecular targets thus making them good therapeutics candidates. The isolation of cancer-specific components from animal venoms is one of the exciting strategies in anti-cancer res

The Biochemical Toxin Arsenal from Ant Venoms.

Ants (Formicidae) represent a taxonomically diverse group of hymenopterans with over 13,000 extant species, the majority of which inject or spray secretions from a venom gland. The evolutionary success of ants is mostly due to their unique eusociality that has permitted them to develop complex collaborative strategies, partly involving their venom secretions, to defend their nest against predators, microbial pathogens, ant competitors, and to hunt prey. Activities of ant venom include paralytic, cytolytic, haemolytic, allergenic, pro-inflammatory, insecticidal, antimicrobial, and pain-producin

Protocols for peptidomic analysis of spider venoms.

Spider venom contains a complex mixture of components with a large range of molecular masses (0.1-60 kDa) exhibiting a diverse array of actions. Most of these components are proteinaceous molecules - biologically active proteins and peptides. Proteomics profiling of spider venoms (the components with MW >10 kDa) could be achieved through conventional 2-DE-based proteomics methods combined with MS or MS/MS detection. Peptidomic profiling (of the components with MW below ~10 kDa) is usually achieved through off-line separation by a combination of ion-exchange and reverse-phase chromatography, an

Proteome and peptidome profiling of spider venoms.

Spider venoms are an important source of novel molecules with different pharmacological properties. Recent technological developments of proteomics, especially mass spectrometry, have greatly promoted the systematic analysis of spider venom. The enormous diversity of venom components between spider species and the lack of complete genome sequence, and the limited database of protein and peptide sequences make spider venom profiling a challenging task and special considerations for technical strategies are required. This review highlights recently used methods for spider venom profiling. In gen

Identification of bradykinins in solitary wasp venoms.

Bradykinins were identified in three solitary wasp venoms. Purification and characterization of the venom extract of the scoliid wasp Megacampsomeris prismatica led to the identification of bradykinin and threonine(6)-bradykinin as the major peptide components. The survey of a number of extracts from solitary wasp venom by MALDI-TOF MS revealed that the venoms of two other scoliid wasps, Campsomeriella annulata annulata and Carinoscolia melanosoma fascinata, also contained Thr(6)-BK as one of the major components. Thus, this study showed the presence of bradykinins in some of the solitary wasp
