Segmentation in vertebrates: clock and gradient finally joined.

The vertebral column is derived from somites formed by segmentation of presomitic mesoderm, a fundamental process of vertebrate embryogenesis. Models on the mechanism controlling this process date back some three to four decades. Access to understanding the molecular control of somitogenesis has been gained only recently by the discovery of molecular oscillators (segmentation clock) and gradients of signaling molecules, as predicted by early models. The Notch signaling pathway is linked to the oscillator and plays a decisive role in inter- and intrasomitic boundary formation. An Fgf8 signaling

Albumin, steroid hormones and the origin of vertebrates.

Albumin, the major serum protein, binds a wide variety of lipophilic compounds including steroids, other lipophilic hormones and various phytochemicals and xenobiotics that bind to receptors for steroids and other lipophilic hormones. Despite albumin's low affinity (K(d) approximately 10(-4) M to 10(-6) M) for these lipophilic compounds, the high concentration of albumin in serum makes this protein a major carrier of steroids and lipophilic hormones and a regulator of their access to receptors. Albumin also functions as a sink for xenobiotics, diminishing the binding of xenobiotics to hormone

Genetic aspects of embryonic eye development in vertebrates.

The vertebrate eye comprises tissues from different embryonic origins, e.g., iris and ciliary body are derived from the wall of the diencephalon via optic vesicle and optic cup. Lens and cornea, on the other hand, come from the overlying surface ectoderm. The timely action of transcription factors and inductive signals ensure the correct development of the different eye components. Establishing the genetic basis of eye defects has been an important tool for the detailed analysis of this complex process. One of the main control genes for eye development was discovered by the analysis of the all

Cisplatin: nephrotoxic action in vertebrates and its prevention.

1. The kidney of frog and black sculpin appeared to be much less sensitive to the toxic action of CP in comparison with rat and pigeon. 2. The impairment of renal function after CP administration resulted in increased serum urea in rat, uric acid in pigeon and magnesium in black sculpin. 3. Kidney swelling is important feature of CP nephrotoxicity in rat and pigeon but not in frog and fish. 4. Pretreatment with choline chloride, PAH, furosemide and ethacrynic acid reduced the nephrotoxic action of CP in rat but did not prevent the accumulation of platinum in renal tissue which appeared to be a

Preferred speeds in terrestrial vertebrates: are they equivalent?

Terrestrial animals have 'preferred speeds' within each gait, that are used much more frequently than others for moving along the ground. Energy costs reach minimal values at these speeds within each gait. In this study we asked whether these speeds are mechanically equivalent among different animals (i.e. speeds where the same levels of peak muscle stress occur). If so, this would help in establishing a link between the energetics and the mechanics of the active muscles at these speeds, providing a first step in understanding why energy costs are minimal. We also asked whether peak muscle str

Neurobiological bases of rhythmic motor acts in vertebrates.

The general principles governing the nervous control of innate motor acts in vertebrates are discussed. Particular consideration is given to the control of locomotion in both mammals and lower vertebrates. One in vitro model of the lamprey central nervous system has been developed. It can be maintained in vitro for several days and the motor pattern underlying locomotion can be elicited in isolated sections of the spinal cord. These findings now allow a detailed analysis of the underlying neural mechanisms. The hypothesis that different parts of the network controlling locomotion can be used i

Molecular evolution of aspartic protease gene family in vertebrates.

Aspartic proteases are a gene superfamily with diverse functions in vertebrates, including pepsinogens (Pgs), the precursors of the major gastric enzyme pepsin. Despite their physiological importance, the evolutionary history of this gene family has been studied only in a fragmented manner. Here, we conducted comprehensive phylogenetic and genomic synteny analyses using whole-genome data from 75 vertebrate species to systematically elucidate the molecular evolution of aspartic protease genes. We identified Pg genes in cartilaginous fishes (elasmobranchs) for the first time, demonstrating that

Century-old chromatin architecture revealed in formalin-fixed vertebrates.

Gene expression is regulated by changes in chromatin architecture intrinsic to cellular differentiation and as an active response to environmental stimuli. Chromatin dynamics are a major driver of phenotypic diversity, regulation of development, and manifestation of disease. Remarkably, we know little about the evolutionary dynamics of chromatin reorganisation through time, data essential to characterise the impact of environmental stress during the ongoing biodiversity extinction crisis (20th-21st century). Linking the disparate fields of chromatin biology and museum science through their com

The unconditioned fear response in vertebrates deficient in dystrophin.

Dystrophin loss due to mutations in the Duchenne muscular dystrophy (DMD) gene is associated with a wide spectrum of neurocognitive comorbidities, including an aberrant unconditioned fear response to stressful/threat stimuli. Dystrophin-deficient animal models of DMD demonstrate enhanced stress reactivity that manifests as sustained periods of immobility. When the threat is repetitive or severe in nature, dystrophinopathy phenotypes can be exacerbated and even cause sudden death. Thus, it is apparent that enhanced sensitivity to stressful/threat stimuli in dystrophin-deficient vertebrates is a

Neuronal control of the vagina in vertebrates: A review.

BACKGROUND: At present, there is an increased interest in the vaginal microbiome. It is believed that microbes play equally important roles in the vagina, including the modulation of neuronal pathways, as in the gut. However, in man as well as in animals, the vagina is the least well-studied part of the female reproductive system. The vagina, a fibromuscular tract, having two main functions, i.e., childbirth and sexual intercourse, is mainly innervated by the pudendal nerve and the pelvic splanchnic nerves (the uterovaginal nerve plexus) containing sympathetic, parasympathetic and nociceptive

Immunohistochemical detection of hepatocyte nuclear factor-4α in vertebrates.

Hepatocyte nuclear factor-4α (HNF4α) presents in multiple isoforms generated using alternative promoter (P1 and P2) and splicing. Neither conservation of tissue distribution of HNF4α isoforms, nor presence of alternative promoter usage is known. In this study, to detect the expression of HNF4α in some species of animals, we have applied monoclonal antibodies against P1 (K9218) and P2 (H6939) promoter-driven and P1/P2 promoter-driven H1415 HNF4α for immunohistochemistry and western blot analysis. Antibody K9218 was observed in the hepatocytes, proximal tubules of the kidney, and epithelial cell

"Sickling" in vertebrates: Animal studies vs. sickle cell disease.

Before the description of sickled cells in humans, erythrocytes of normal deer were found to deform, or sickle in vitro. Sickling required oxygenation and alkalization; human erythrocytes sickle because their abnormal sickle hemoglobin (HbS) polymerizes following deoxygenation in vivo and in vitro. HbS and some deer hemoglobins polymerize because of specific amino acid contacts between hemoglobin beta-globin chains, although different amino acid residues form the contact points of the polymer. Hemoglobin precipitation is found in other vertebrates. Most often it is a benign in vitro phenomenon

Low incidence of atrial septal defects in nonmammalian vertebrates.

The atrial septum enables efficient oxygen transport by separating the systemic and pulmonary venous blood returning to the heart. Only in placental mammals will the atrial septum form by the coming-together of the septum primum and the septum secundum. In up to one of four placental mammals, this complex morphogenesis is incomplete and yields patent foramen ovale. The incidence of incomplete atrial septum is unknown for groups with the septum primum only, such as birds and reptiles. We found a low incidence of incomplete atrial septum in 11 species of bird (0% of specimens) and 13 species of

Transposon vectors for gene-trap insertional mutagenesis in vertebrates.

The function of most vertebrate genes remains unknown or uncertain. Insertional mutagenesis offers one approach to identify and understand the function of these genes. Transposons have been used successfully in lower organisms and plants for insertional mutagenesis, but until activation of the Sleeping Beauty (SB) transposon system, there was no indication of active DNA-based transposons in vertebrates. Investigator-driven insertional mutagenesis in vertebrates has relied on retroviral insertions or selection of low-frequency integration of naked DNA in ES cell lines. We have combined the high

Current studies on tooth enamel development in lower vertebrates.

The tooth enamel is a unique tissue in the human body on many characteristic features, in both morphological and developmental aspects. This tissue is widely distributed in the vertebrate world. This hard tissue is divided into two groups, mainly from the developmental feature. One is the enamel in the upper classes of the adult amphibian, called true or ectodermal enamel. The other mainly distributes in fish (both of osteichytyes and chondroichtyes), called enameloid or mesodermal enamel. Enameloid seen in fish has many characteristics compared with enamel, although enameloid after eruption i

AA amyloidosis in vertebrates: epidemiology, pathology and molecular aspects.

AA amyloidosis is a prototypic example of systemic amyloidosis: it results from the prolonged overproduction of SAA protein produced in response to chronic inflammation. AA amyloidosis primarily affects the kidneys, liver, spleen, gastrointestinal tract, leading to a variety of symptoms. First, this review examines AA amyloidosis in humans, focusing on pathogenesis, clinical presentation, and diagnosis and then in animals. In fact AA amyloidosis is the only systemic amyloidosis that has been largely documented in a remarkable number of vertebrate species: mammals, birds, and fishes, especially

Generating prophylactic immunity against arboviruses in vertebrates and invertebrates.

The World Health Organization recently declared a global initiative to control arboviral diseases. These are mainly caused by pathogenic flaviviruses (such as dengue, yellow fever and Zika viruses) and alphaviruses (such as chikungunya and Venezuelan equine encephalitis viruses). Vaccines represent key interventions for these viruses, with licensed human and/or veterinary vaccines being available for several members of both genera. However, a hurdle for the licensing of new vaccines is the epidemic nature of many arboviruses, which presents logistical challenges for phase III efficacy trials.
