Verticillium dahliae is a widespread and destructive soilborne fungus that causes vascular wilt disease, significantly reducing cotton (Gossypium hirsutum) yield and quality. Cotton's xyloglucan-specific endoglucanase inhibitor protein (GhXEGIP1) has demonstrated effectiveness against the fungal glycoside hydrolase VdEG1, a member of the glycoside hydrolase family 12. However, the mechanisms underlying GhXEGIP1's defense against V. dahliae remain unclear. This study provides evidence that GhXEGIP1 are instrumental in plant responses to V. dahliae infection. GhXEGIP1 interacts with and regulate

The soil-borne fungal pathogen Verticillium dahliae (V. dahliae) is the causal agent of Verticillium wilt (VW), a vascular disease that severely threatens global cotton production. Although cell wall lignification represents a cornerstone of plant immunity, the precise regulatory circuits that bridge this structural reinforcement with Verticillium dahliae resistance in cotton have yet to be fully elucidated. Here, we demonstrate that the NAC transcription factor GhNAC043 is a key positive regulator of this defense. GhNAC043 expression was rapidly induced upon V. dahliae infection. Silencing Gh

Verticillium dahliae is a destructive soil-borne fungus with a broad host range, and its persistence in soil complicates control. Current measures, mainly resistant cultivars and chemicals, are limited and environmentally risky, promoting biocontrol as a green alternative. Here, we investigated the biocontrol mechanisms of Bacillus velezensis L33a against V. dahliae JR2 in tomato. In vitro assays on PDA plates at 26°C for 9 d showed that L33a inhibited JR2 by 58.6%, caused hyphal malformation and disruption, and its volatile organic compounds suppressed pathogen growth. In pot experiments, tom

Verticillium dahliae is a soilborne fungal pathogen that causes Verticillium wilt in cotton and many other crops. While siderophore-associated iron acquisition is recognized as crucial, its precise connections to fungal development, stress adaptation, and host colonization remain incompletely elucidated. This study identifies the VdOMO protein as a putative SidA-family L-ornithine N5-monooxygenase through phylogenetic and pairwise sequence analyses. Analyses of independent deletion mutants and a complemented strain showed that VdOMO contributes to conidial morphology and production, microscler

Polygalacturonase-inhibiting protein (PGIP), belonging to a group of plant defence proteins, specifically inhibits endopolygalacturonases secreted by pathogens. Herein, we showed that purified GhPGIP1 is a functional inhibitor of Verticillium dahliae and Fusarium oxysporum f. sp. vasinfectum, the two fungal pathogens causing cotton wilt. Transcription of GhPGIP1 was increased in cotton upon infection, wounding, and treatment with defence hormone and H2O2. Resistance by GhPGIP1 was examined by its virus-induced gene silencing in cotton and overexpression in Arabidopsis. GhPGIP1-silenced cotton

BACKGROUND: Verticillium dahliae is the most destructive root rot pathogen of marigold. For eco-friendly management of this disease, iron oxide nanoparticles (Fe2O3 nanoparticles) were synthesized with the seed extract of Trachyspermum ammi.
RESULTS: Synthesis of Fe2O3 NPs was confirmed by ultraviolet-visible (UV) spectrum absorption peak at 275 nm. FTIR validated functional groups on the nanoparticle surface, XRD showed a crystalline structure with an average size of 42 nm, EDX proved elemental composition (Fe 71.6%, O 15.9%), and SEM showed a spherical morphology. Cynanchum komarovii Al Iljinski is a desert plant that has been used as analgesic, anthelminthic, and antidiarrheal, but also as herbal medicine to treat cholecystitis in people. In this work, an antifungal protein with sequence homology to chitinase was isolated from C. komarovii seeds and named CkChn134. The three-dimensional structure prediction of CkChn134 indicated that the protein has a loop domain formed a thin cleft, which is able to bind molecules and substrates. The protein and CkTLP synergistically inhibited the fungal growth of Verticillium dahliae, Fusarium oxysporum, Rhizoctonia

Based on GWAS, QTL mapping, BSA and RNA‒seq jointly reveal the genetic mechanisms underlying Verticillium wilt resistance in cotton and identify three candidate genes associated with this trait. Verticillium wilt, caused by the soil-borne vascular pathogen Verticillium dahliae, is a devastating disease that severely threatens global cotton production. However, the genetic basis of resistance to this disease remains poorly uncovered, hindering the advancement of genomics-assisted breeding programs for Verticillium wilt (VW) resistance. Here, we systematically evaluated VW resistance in a recomb

Fungal ergosterol biosynthesis relies on electron carriers to activate substrates, and thus, they are recognized as targets for fungicides. In this study, we demonstrated that a cytoplasm-localized cytochrome b5-like progesterone binding protein, VdPBP1, participated in vegetative growth, virulence, and metabolism of membrane components in Verticillium dahliae, especially ergosterol biosynthesis. As a stress sensor, its protein level was dose-dependently inhibited by terbinafine. VdPBP1 directly interacted with squalene epoxidase VdERG1 on the lipid droplets. Supplementing hemin and enhancing

Verticillium wilt, caused by the soilborne fungus Verticillium dahliae, and iron (Fe) deficiency are major stresses impacting valuable crops in the Mediterranean basin. The plant response to these stresses consists of overlapping pathways that, coupled with the emerging use of endophytic fungi as biological control agents (BCAs) and plant biostimulants, enables the study of their potential to mitigate co-occurring biotic stresses and nutritional deficiencies such as Fe deficiency. To this end, Arabidopsis thaliana plants of the "Columbia" ecotype grown in calcareous soil were inoculated with t

BACKGROUND: Verticillium longisporum is a soil-borne vascular pathogen infecting cruciferous hosts such as oilseed rape. Quantitative disease resistance (QDR) is the major control means, but its molecular basis is poorly understood so far. Quantitative trait locus (QTL) mapping was performed using a new (Bur×Ler) recombinant inbred line (RIL) population of Arabidopsis thaliana. Phytohormone measurements and analyses in defined mutants and near-isogenic lines (NILs) were used to identify genes and signalling pathways that underlie different resistance QTL.
RESULTS: QTL for resistance to V. lon

In the last decades, the plant innate immune responses against pathogens have been extensively studied, while biocontrol interactions between soilborne fungal pathogens and their hosts have received much less attention. Treatment of Arabidopsis thaliana with the nonpathogenic bacterium Paenibacillus alvei K165 was shown previously to protect against Verticillium dahliae by triggering induced systemic resistance (ISR). In the present study, we evaluated the involvement of the innate immune response in the K165-mediated protection of Arabidopsis against V. dahliae. Tests with Arabidopsis mutants

Eggplant verticillium wilt, caused by Verticillium spp., is a severe eggplant vascular disease. Solanum sisymbriifolium, a wild species of eggplant that is resistant to verticillium wilt, will be beneficial for genetically modifying eggplants. To better reveal the response of wild eggplant to verticillium wilt, proteomic analysis by iTRAQ technique was performed on roots of S. sisymbriifolium after exposure to Verticillium dahliae, and some selected proteins were also validated using parallel reaction monitoring (PRM). After inoculation with V. dahliae, the phenylalanine ammonia lyase (PAL) an

Verticillium wilt, caused by Verticillium dahliae Kleb., is a serious threat to cotton (Gossypium spp.) crop production. To enhance our understanding of the plant's complex defensive mechanism, we examined colonization patterns and interactions between V. dahliae and two cotton species, the resistant G. barbadense and the susceptible G. hirsutum. Microscopic examinations and grafting experiments showed that the progression of infection was restricted within G. barbadense. At all pre- and postinoculation sampling times, levels of salicylic acid (SA) were also higher in that species than in G. h

Salicylic acid (SA) and auxin often exert antagonistic effects during pathogen challenge, and SA's inhibitory effect on auxin-mediated signaling pathway plays a crucial role in plant defense mechanisms. However, whether SA-mediated immunity can be enhanced through the inhibition of auxin signaling pathway remains unclear. Cotton GhIAA43, a member from the Aux/IAA family, was a resistance-related gene against Verticillium dahliae attack. Here, we clarified the role of GhIAA43 in regulating the auxin and SA-mediated pathways to improve the plant resistance by using the RNA interference (RNAi)-Gh

Bacillus subtilis NCD-2 demonstrates exceptional biocontrol potential against cotton Verticillium wilt. While previous studies have established its direct antifungal activity (e.g., inhibiting Verticillium dahliae mycelial growth and spore germination), our work reveals a novel mechanism: NCD-2 primes systemic resistance in cotton by activating plant immune-signaling pathways. Firstly, transcriptional profiling uncovered that NCD-2 triggers a defense response in roots analogous to V. dahliae infection, allowing cotton to maintain a more balanced state when confronted with pathogen attacks. Acer truncatum wilt caused by Verticillium dahliae is a severe soilborne disease that poses a threat to the cultivation of this plant in China. The present study explored the biocontrol efficiency and underlying antagonistic mechanism of Pseudomonas aurantiaca ST-TJ4 against V. dahliae. In vitro, strain ST-TJ4 exhibited excellent inhibitory effects on V. dahliae, causing mycelial deformation. This strain significantly suppressed the production of V. dahliae conidia and microsclerotia. Moreover, the application of ST-TJ4 reduced the incidence of Verticillium wilt in A. truncatum saplings in bot

Verticillium dahliae is a prominent generator of plant vascular wilting disease and sulfur (S)-enhanced defense (SED) mechanisms contribute to its in-planta elimination. The accumulation of S-containing defense compounds (SDCs) including elemental S (S(0) ) has been described based on the comparison of two near-isogenic tomato (Solanum lycopersicum) lines differing in fungal susceptibility. To better understand the effect of S nutrition on V. dahliae resistance both lines were supplied with low, optimal or supraoptimal sulfate-S. An absolute quantification demonstrated a most effective fungal

In this study, we identified a defense-related major latex protein (MLP) from upland cotton (designated GhMLP28) and investigated its functional mechanism. GhMLP28 transcripts were ubiquitously present in cotton plants, with higher accumulation in the root. Expression of the GhMLP28 gene was induced by Verticillium dahliae inoculation and was responsive to defense signaling molecules, including ethylene, jasmonic acid, and salicylic acid. Knockdown of GhMLP28 expression by virus-induced gene silencing resulted in increased susceptibility of cotton plants to V. dahliae infection, while ectopic
