Vestibular findings and brainstem pathology in two siblings with Cockayne's syndrome.

Two siblings with Cockayne's syndrome (CS) are described. Both showed cachectic dwarfism, photosensitivity and progressive neurological dysfunction. The damped-rotation test revealed no vestibular nystagmus in one of them, and reduced per-rotatory nystagmus in the other. Neuropathological examination in the former subject disclosed neuronal loss with fibrillary gliosis in the vestibular nuclei, whereas neuronal degeneration was not apparent in the abducens nuclei, the oculomotor nuclei and the paramedian pontine reticular formation. Our findings suggest that CS might cause vestibular dysfuncti

Experimental vestibular pharmacology: a minireview with special reference to neuroactive substances and antivertigo drugs.

Neurotransmitters and neuromodulators involved in the function of vestibular nuclei were reviewed with special reference to drugs used for treatment of motion sickness and vertigo. Biochemical, histochemical and electrophysiological studies have demonstrated that acetylcholine is a transmitter candidate from the afferent vestibular nerve to the lateral vestibular nucleus (LVN), because acetylcholine satisfies most criteria for a chemical transmitter in the central nervous system. It is unlikely, however, that monoamines such as noradrenaline, dopamine and serotonin are transmitters in the vest

Pharmacology of the vestibular system.

In the past year significant advances have been made in our understanding of the neurochemistry and neuropharmacology of the peripheral and central vestibular systems. The recognition of the central importance of excitatory amino acids and their receptors at the level of the hair cells, vestibular nerve and vestibular nucleus has progressed further, and the role of nitric oxide in relation to activation of the N-methyl-D-aspartate receptor subtype is becoming increasingly clear. Increasing evidence suggests that excessive N-methyl-D-aspartate receptor activation and nitric oxide production aft

[Afferent projections to the uvula in the cat. II. Mossy fiber projections].

The mossy fiber projections to the uvula of the cerebellum were studied by means of retrograde axonal transport of horseradish peroxidase (HRP) in the cat. Following large and small injections into the uvula, distribution of the labeled cells in the brainstem nuclei was investigated. The results showed different afferent projections between the dorsal and ventral uvula. Major sources projecting to the dorsal uvula were the peduncular, paramedian, and lateral nuclei of the pontine nuclei. Labeled cells found in the pontine nuclei amounted to 81.6% of the total number of labeled cells in cat 1.

Cerebellar corticovestibular fibers of the posterior lobe in a prosimian primate, the lesser bushbaby (Galago senegalensis).

The orginization of cerebellar corticovestibular fibers was studied in a prosimian primate (Galago senegalensis) using the Fink and Heimer ('67) method. The vestibular complex of Galago is larger than in other mammals and some higher primates. Vermis lobule IX contributes the largest number of fibers to the ipsilateral vestibular complex. Lobules VI and VIII give rise to lesser, but similar, numbers of fibers which also pass into the ipsilateral vestibular nuclei. Vermis lobule VII and the paravermal and lateral cortices contribute extremely sparse numbers of fibers to the dorsal area of the i

Mossy fiber projections from the brain stem to the nodulus in the cat. An experimental study comparing the nodulus, the uvula and the flocculus.

Mossy fiber projections from the brainstem to the nodulus were studied by means of the retrograde transport of horseradish peroxidase (HRP) in the cat. The findings indicate exclusive secondary vestibular projections to the nodulus (96.5% of the total number of labeled cells in cat 1). Major sources of such projections include the caudal half of the medial and inferior vestibular nuclei, and the dorsal half of the superior vestibular nucleus. Groups-x and -z of the vestibular nuclei give rise to minor projections. The projections from groups-f and -y, and the interstitial nucleus of the eighth

Differential mossy fiber projections to the dorsal and ventral uvula in the cat.

The brainstem afferents to the uvula were studied by using retrograde axonal transport of horseradish peroxidase in the cat. Findings indicate differential afferent projections to the ventral and dorsal uvula. Major sources projecting to the ventral uvula include the caudal parts of the medial and inferior vestibular nuclei, the x- and f-groups of the vestibular nuclei, the dorsal and central parts of the superior vestibular nucleus, the rostral dorsomedial part of the paramedian nucleus of the pontine nuclei, the caudal part of the prepositus hypoglossal nucleus, and the infratrigeminal nucle

Some afferent and efferent connections of the vestibular nuclear complex in the red-eared turtle Pseudemys scripta elegans.

In the present study some afferent, commissural, and efferent connections of the vestibular nuclear complex in the turtle Pseudemys scripta elegans were demonstrated with the HRP tracing technique. Afferent projections to the vestibular nuclei were found to arise in the nucleus of the basal optic root, the interstitial nucleus of the fasciculus longitudinalis medialis, the medial and lateral cerebellar nuclei, the perihypoglossal nuclear complex, and the reticular formation. Distinct commissural projections appeared to arise in the dorsolateral, ventromedial, and descending vestibular nuclei.

Local cerebral glucose utilization in the symptomatic thiamine-deficient rat: increases in fornix and pyramidal tract.

Local cerebral glucose utilization (LCGU) was measured in asymptomatic rats on a thiamine (B1)-deficient diet for 70 days and in symptomatic rats on a B1-deficient diet for 98 days. LCGU increased in postcommissural fornix (F), pyramidal tracts (P), and inferior internal capsule (CAI) of symptomatic, compared with asymptomatic B1-deficient rats but decreased in thalamic nuclei, auditory structures, and lesioned vestibular nuclei. B1 administration to symptomatic rats improved symptoms; decreased LCGU in F. P, and CAI; increased LCGU in the lesioned vestibular nuclei; but decreased LCGU in mamm

L-Thyroxine Improves Vestibular Compensation in a Rat Model of Acute Peripheral Vestibulopathy: Cellular and Behavioral Aspects.

Unilateral vestibular lesions induce a vestibular syndrome, which recovers over time due to vestibular compensation. The therapeutic effect of L-Thyroxine (L-T4) on vestibular compensation was investigated by behavioral testing and immunohistochemical analysis in a rat model of unilateral vestibular neurectomy (UVN). We demonstrated that a short-term L-T4 treatment reduced the vestibular syndrome and significantly promoted vestibular compensation. Thyroid hormone receptors (TRα and TRβ) and type II iodothyronine deiodinase (DIO2) were present in the vestibular nuclei (VN), supporting a local a

Nonphosphorylated neurofilament protein is expressed by scattered neurons in the vestibular and precerebellar brainstem.

Vestibular information is essential for the control of posture, balance, and eye movements. The vestibular nerve projects to the four nuclei of the vestibular nuclear complex (VNC), as well as to several additional brainstem nuclei and the cerebellum. We have found that expression of the calcium-binding proteins calretinin (CR) and calbindin (CB), and the synthetic enzyme for nitric oxide synthase (nNOS) define subdivisions of the medial vestibular nucleus (MVe) and the nucleus prepositus (PrH), in cat, monkey, and human. We have asked if the pattern of expression of nonphosphorylated neurofil

[Vestibular compensation. Review of the literature and clinical applications].

Vestibular compensation is an excellent model for the study of plasticity of the adult central nervous system. Therefore it has been the subject of several studies in humans and animals, which will be briefly summed up by the authors. Lesions of the labyrinth or vestibular neurectomy are immediately followed of postural and oculomotor disorders, as well as by dynamic deficits of the various vestibular reflexes (vestibulo-ocular and vestibulonucal reflexes). While the former problems always recede in all species, the restoration of the dynamic properties of vestibular reflexes largely depends u

Structural and functional connectivity mapping of the vestibular circuitry from human brainstem to cortex.

Structural and functional interconnections of the bilateral central vestibular network have not yet been completely delineated. This includes both ipsilateral and contralateral pathways and crossing sites on the way from the vestibular nuclei via the thalamic relay stations to multiple "vestibular cortex" areas. This study investigated "vestibular" connectivity in the living human brain in between the vestibular nuclei and the parieto-insular vestibular cortex (PIVC) by combined structural and functional connectivity mapping using diffusion tensor imaging and functional connectivity magnetic r

Spinocerebellar ataxias types 2 and 3: degeneration of the pre-cerebellar nuclei isolates the three phylogenetically defined regions of the cerebellum.

The pre-cerebellar nuclei act as a gate for the entire neocortical, brainstem and spinal cord afferent input destined for the cerebellum. Since no pathoanatomical studies of these nuclei had yet been performed in spinocerebellar ataxia type 2 (SCA2) or type 3 (SCA3), we carried out a detailed postmortem study of the pre-cerebellar nuclei in six SCA2 and seven SCA3 patients in order to further characterize the extent of brainstem degeneration in these ataxic disorders. By means of unconventionally thick serial sections through the brainstem stained for lipofuscin pigment and Nissl material, we

Advanced progress of vestibular compensation in vestibular neural networks.

Vestibular compensation is the natural process of recovery that occurs with acute peripheral vestibular lesion. Here, we summarize the current understanding of the mechanisms underlying vestibular compensation, focusing on the role of the medial vestibular nucleus (MVN), the central hub of the vestibular system, and its associated neural networks. The disruption of neural activity balance between the bilateral MVNs underlies the vestibular symptoms after unilateral vestibular damage, and this balance disruption can be partially reversed by the mutual inhibitory projections between the bilatera

Lateral medullary syndrome following injury of the vestibular pathway to the core vestibular cortex: Diffusion tensor imaging study.

OBJECTIVE: The parieto-insular vestibular cortex (PIVC) is a core region of vestibular input into regions of the cortex. The vestibular nuclei have reciprocal connections with the PIVC. However, little is known about injury of the core vestibular pathway to the PIVC in patients with dorsolateral medullary infarctions. In this study, using diffusion tensor tractography (DTT), we investigated injury of the neural connections between the vestibular nuclei and the PIVC in patients with typical central vestibular disorder. METHODS: Eight consecutive patients with lateral medullary syndrome and 10

Synaptic mechanisms of interaction between Deiters' nucleus and the nuclei of some cranial nerves.

The effects of stimulation of the vestibular nerve, spinal trigeminal nucleus, facial and hypoglossal nuclei of the cranial nerves on the neuronal activity in the lateral vestibular nucleus of Deiters were studied in cats anaesthetized with pentobarbitone. Stimulation of these nuclei was found to produce antidromic and synaptic activation of Deiters' neurons. Descending axon collaterals of the vestibular neurons to these brainstem structures were revealed. Stimulation of the VIIIth nerve, spinal trigeminal and facial nuclei evoked mono- and polysynaptic excitatory postsynaptic potentials in De

Morphometric analysis of the vestibular complex in the rat.

The characteristics of the four major vestibular nuclei and accessory cell groups in the rat have been studied in serially cut horizontal sections. Camera lucida drawings of the vestibular nuclei and their neurons were made in these sections and subjected to a computerized image analysis. The dimensions (volume and length) and the number of cells of each vestibular nucleus were obtained, as well as morphometric parameters of their neurons (cross-sectional area, maximum and minimum diameter, and shape). These parameters were statistically analyzed by comparing the cell population from different
